Development of Stable Culture Techniques for Kenaf (Hibiscus cannabinus L.) (케나프 생산성 향상을 위한 안정 재배기술 개발)

  • Kenaf (Hibiscus cannabinus L.) was recognized as a potential source of forage. But the domestic cultivation techniques are not set standards yet. So we tested the basic culture techniques during 2012~2013 for getting a high yield and good grade forage production system. The best seeding method for mechanized planting (corn planter used) was hill seeding with $20{\times}20cm$ seeding distance. When we treated hill seeding with $20{\times}20cm$, the yield what we could get was 13,641 kg/10a and it was 32% more than that of conventional practice hill seeding with $20{\times}30cm$ seeding distance. The proper seeding date for getting high yield was May 1. In May 1, the yield per 10a was reached 13,423 kg, and it was 30% more than that of seeding at May 30. More over the crude protein content which was important factor for determinating forage nutritive value was 12.7% and it was higher 1.8% (relatively 16.5% high) than that of May 30 seeding. The most effective herbicide for kenaf was Fluazipof-p-butyl. It's herbicidal rate was 97% and phyto-toxicity was less than 5%. Regional adaptability for Jeollabuk-Do including Imsil Gun, Kochang Gun and Sunchang Gun were identified that the stable cultivation were possible in these area with average yield 12,400 kg/10a and it was about 1.7 times as compared to corn harvest.

A study on the relationship of self-efficacy to stressors and stress adaptation in dental hygiene students (치위생과 학생의 자기효능감 수준에 따른 스트레스 요인의 적응방법에 관한 연구)

  • Objectives : The purpose of this study was to examine the relationship of the self-efficacy of dental hygiene students to their stressors and ways of stress-adaptation patterns. Methods : The subjects in this study were dental hygiene juniors in four selected colleges located in the metropolitan area. Results : 1. They got a mean of 3.22 in self-efficacy. They gave the highest mark(3.50) to an item 'I can attain it if I set a primary goal.' 2. They got a mean of 3.18 in stressors. Among the stressors, task assignments(3.74) were identified as the greatest stressor, followed by the curriculum(3.25), learning environments(3.16), prospects of employment (3.07) and test anxiety(2.95). 3. They got a mean of 2.02 in stress-adaptation method. They got 2.31 and 1.72 in long-term and short-term adaptation respectively, which showed that long-term stress adaptation method were more prevailing than short-term ones. 4. As a result of analyzing whether there would be any gaps in self-efficacy according to general characteristics, statistically significant gaps were found in that regard according to experience of preparing for college admission after leaving high school, academic standing, satisfaction with the department of dental hygiene and prospects of employment(p<.05). 5. As a result of checking the relationship of their self-efficacy to their stressors and ways of stress adaptation method, there were statistically significant differences in that aspect according to examinations and prospect of employment(p<0.05). As for ways of stress adaptation method, there were statistically significant differences in long-term adaptation method (p<0.05). Conclusions : As it's found that the level of the self-efficacy of the dental hygiene students was linked to the efficiency of their ways of stress adaptation method, the development and implementation of programs geared toward boosting the self-efficacy of dental hygiene students are required to teach them to successfully cope with various kinds of stress that they are likely to face after getting a job.

THE NONDESTRUCTIVE MEASUREMENT OF THE SOLUBLE SOLID AND ACID CONTENTS OF INTACT PEACH USING VIS/NIR TRANSMITTANCE SPECTRA

  • Since the SSC(soluble solid contents) and titratable acidity of fruit are highly concerned to the taste, the need for measuring them by non-destructive technology such as NIR(Visual and Near-infrared) spectroscopy is increasing. Specially, in order to grade the quality of each fruit with a sorter at sorting and packing facilities, technologies for online measurement satisfying the tolerance in terms of accuracy and speed should be developed. Many researches have been done to develop devices to measure the internal qualities of fruit such as SSC, titratable acidity, firmness, etc. with the VIS(Visual)/NIR(Near Infrared) reflectance spectra. The distributions of the SSC, titratable acidity, firmness, etc. are different with respect to the position and depth of fruit, and generally the VIS/NIR light can interact with fruit in a few millimeters of pathlength, and it is very difficult to measure the qualities of inner flesh of fruit. Therefore, to measure the average concentrations of each quality factor such as SSC and titratable acidity with the reflectance-type NIR devices, the spectra of fruit at several positions should be measured. Recently, the interest about the transmittance-type VIS/NIR devices is increasing. NIR light can penetrate through the fruit about 1/10-1/1,000,000 %. Therefore, very intensive light source and very sensitive sensor should be adopted to measure the transmitted light spectra of intact fruit. The ultimate purpose of this study was to develop a device to measure the transmitted light spectra of intact fruit such as apple, pear, peach, etc. With the transmittance-type VIS/NIR device, the feasibility of measurement of the SSC and titratable acidity in intact fruit cultivated in Korea was tested. The results are summarized as follows; A simple measurement device which can measure the transmitted light spectra of intact fruit was constructed with sample holder, two 500W-tungsten halogen lamps, a real-time spectrometer having a very sensitive CCD array sensor and optical fiber probe. With the device, it was possible to measure the transmitted light spectra of intact fruit such as apple, pear and peach. Main factors affecting the intensity of transmitted light spectra were the size of sample, the radiation intensity of light source and the integration time of the detector. Sample holder should be designed so that direct light leakage to the probe could be protected. Preprocessing method to the raw spectrum data significantly influenced the performance of the nondestructive measurement of SSC and titratable acidity of intact fruit. Representative results of PLS models in predicting the SSC of peach were SEP of 0.558 Brix% and R2 of 0.819, and those in predicting titratable acidity were SEP of 0.056% and R2 of 0.655.

Spermatogenic and Antioxidant Potential of Mucuna prureins (L.) in Epididymal Spermatozoa: A Dose Dependent Effect

  • The study aim is to investigate the free radicals scavenging and spermatogenic potentials, as well as to analyze any reproductive toxicity of ethanolic extract of Mucuna prureins (M. pruriens) Linn. in spermatozoa, under different dosages in normal male rat. Normal rats were randomly selected and suspension of the extract was administered orally at the dosages of 150, 200 and 250 mg/kg body weight of the different groups of male rats (n=6) once in a day for 60 days and grouped as group II, III and IV respectively. Saline treated rats served as control -group I. On the $60^{th}$ day the animals were sacrificed and the epididymal sperm were subjected to various analyses like level of ROS production, LPO, enzymatic and non enzymatic antioxidant, morphology, morphometry, chromosomal integrity and DNA damage. Results showed significant reduction in ROS production and peroxidation and significant increase in both enzymic and non-enzymic antioxidants in all concentration treated groups when compared with control. Results from all the drug treated groups showed good sperm morphology, increased sperm count and motility. There was no DNA damage and showed normal chromosomal integrity even in 250 mg/kg dose. When compared with control all the three extract treated groups showed increased ROS scavenging activity. However, group II (200 mg/kg) showed significant changes in all the parameters. From the present study it was confirmed that the M. pruriens has potential to improve the sperm qualitatively and quantitatively through scavenging the excess ROS with any adverse side effects. These observations suggest that ethanolic seed extract of M. pruriens may serve as anti-oxidant that can exploit to treat the oxidative stress mediated male factor infertility.

  • The ability to develop curriculums is a crucial factor in evaluating the expertise of a teacher who teaches culture & art education. Establishing a one-year plan for classes is an effort to create a well-designed curriculum for the year and also to foresee the big picture of classes in the corresponding year. A curriculum should not be composed of merely educational content or a series of knowledge and skills. It should be well-designed, based on principles of a coherent plan. This study examines organizational principles on which common curriculums are based on and looks at how a curriculum can be designed, especially for cartoon & animation classes, as part of Culture & Art education, and which factors should be considered in planning. In the process of forming such a curriculum, these steps should be followed: considering educational standards for cartoon animation classes; determining the learning experience, organizing the learning experience; and, lastly, evaluating the level of learning. In addition, effective teaching strategies that reflect the characteristics of a class on cartoon animation should be formulated. This study suggests actual examples of an effective annual curriculum for cartoon animation classes based on all the factors presented above.

Effects of replacing fish oil with palm oil in diets of Nile tilapia (Oreochromis niloticus) on muscle biochemical composition, enzyme activities, and mRNA expression of growth-related genes

  • Background: Due to the continuous demand for fish coupled with decline in capture fisheries, there is the need to increase aquaculture production to meet the demand. Aquaculture is faced with high cost of feeding since fish oil and fish meal are expensive. In view of this, there are calls to explore alternatives that are cheap and reliable. Objectives: This study on Oreochromis niloticus was conducted to evaluate the effects of replacing fish oil (FO) with palm oil (PO) at 0%, 25%, 50%, 75%, and 100% on muscle fatty acid and proximate composition as well as growthrelated enzyme activities and mRNA expression. Methods: Oreochromis niloticus were fed five experimental diets (33% crude protein and 10% crude lipid) for 8 weeks. Feed had variation in fish oil and palm oil contents. After the 8 weeks feeding trial, five fish were sampled from each tank (15 from each treatment) and euthanized using an excess dose of tricaine methane sulfonate (MS-222 at 200 mg/L). Fatty acid and enzyme activities were analyzed using standard protocols. Also, RT-qPCR was used to quantify the expression levels of selected growth-related genes. Results: Fish fed 25% PO recorded the least muscle protein content and was significantly lower than the group fed 100% PO. Paired box protein 7 (Pax-7) enzyme activity was significantly higher in the group fed 50% PO compared to the groups fed 25% PO and 100% PO, while caplain-3 (Capn-3) was significantly lower in the group fed 0% PO compared to all other groups. There was a significant difference among treatments with respect to mRNA expression of Pax-7 and Capn-3. Group fed 25% PO had significantly lower mRNA expression of Pax-7, while the group fed 75% PO recorded significantly higher mRNA expression of Capn-3 compared to groups fed 0% PO, 25% PO, and 100% PO. Pearson's correlation analysis revealed that Igf-I and Igf-II mRNA expression have significant correlation with n-3 polyunsaturated fatty acids content in muscle. Conclusion: The results suggest muscle protein content could be modified if FO is replaced with PO. Also, mRNA expression of Pax-7 and Capn-3 is affected by replacing FO with PO.

Optimal temperature conditions of Korean freshwater Cladoceran for development of standard toxicity test methods (표준생태독성시험법 개발을 위한 한국산 물벼룩의 최적사육온도 구명)

  • Temperature is an important ambient factor affecting the physiology and metabolism of aquatic invertebrates. In this study, we studied about the survival, reproduction and growth effects of 4 different temperatures(16, 20, 24, $28^{\circ}C$) in four Korean cladoceran, Daphnia sp., Daphnia obtusa, Moina macrocopa, Simocephalus vetulus and Daphnia magna as an international standard species. All 5 water flea tested showed that molting time, fecundity and intrinsic rate of natural increase(r) in high temperature condition were higher than those in lower temperature. On the other hand, lower survivals and longer time to start of offspring were showed in high temperature. Our results suggest that the optimal temperatures of Daphnia sp.,, Moina macrocopa and Simocephalus vetulus seem to be about $20^{\circ}C$ except for Daphnia obtusa which as showed good survivals and reproductions in $16^{\circ}C$.

Studies for the Processing Factors of Pesticides during the Milling of Wheat Grain (밀의 제분에 따른 밀가루 중 농약 가공계수 산출 연구)

  • Processing factors of pesticides in milling process of wheat grain, which are consumed and imported at large quantity were examined to establish reasonable MRL of the processed food. Azinphos-methyl, chlorpyrifos, chlorpyrifos-methyl, fenitrothion, malathion, and trichlorfon were selected for the study according to annual usage and the previous detection record in wheat grain. Dipping process for pesticide application was performed in laboratory, while milling process was conducted under pilot plant system. Processing factors were calculated by analyzing residual pesticides on wheat grain and processing products as wheat flour, bran and red dog. Processing factors were 0.05 for azinphos-methyl, 0.06 for chlorpyrifos, 0.05 for chlorpyrifos-methyl, 0.07 for fenitrothion, 0.07 for malathion, 0.06 for trichlorfon, respectively. Recovery test was also performed to establish extraction efficiency of analytical procedure. The recovery value ranged from 93.2% to 98.6% with standard deviation of 0.1-0.9%.

A Study on Role and Function of the Medical Representatives (의약정보담당자(MR)의 기능 및 역할에 관한 연구)

  • Background : Aim of this study is focused on the analysis of the needed abilities of medical representatives resulting in building up the market and increasing sales. It is to propose methods to increase this ability ensuring continuous growth in market share and profit. Methods : A survey was conducted between January 6 and May 31, 2003. Using SPSS(Version 10.0), the collected data was analyzed by Hotelling T2, factor analysis. Some hypotheses were selected to include the conclusion. Some questionnaires for physicians working in hospitals or clinics and the medical representatives working in a pharmaceutical company were created and asked to them to either prove or reject those hypotheses. The results were analyzed to find the primary factors that effect the interactions between physician and the medical representatives. These factors were also studied along with the theoretical research based on published references. Results : The results were as follows. The main reasons for the physician to meet with a medical representatives were collection of product informations needed for patient treatment and collection of informations on current medical issue and as well as personal interests. The main parameters by which physicians evaluate the medical representatives are human relationship including sincerity and manners and supply of accurate and unbiased information on products. Overall, the medical representatives' perception on the importance of medical knowledge and ability to deliver it are lower than that expected by physicians. Conclusion : Medical and pharmaceutical companies' environment are changed rapidly. And those changes forced medical representatives to set new roles and competency. In order to drive away from the past 'rule of thumb' and 'adaptation to circumstance', optimal method and systemic development to train and support the medical representatives should be quipped. They will help medical representatives to be specialists in medical knowledge and to understand the exact need of health care professions. Product competitiveness will be increased and eventually successful business can be achieved through it.

Status of Researches of Excavation Damaged Zone in Foreign Underground Research Laboratories Constructed for Developing High-level Radioactive Waste Disposal Techniques (고준위방사성폐기물 처분 기술개발을 위해 건설된 해외 지하연구시설에서의 암반손상대 연구 현황)

  • In the countries operating nuclear reactors, the development of high-level radioactive waste(HLW) disposal technique is considered as an urgent and important issue for sustainable utilization of nuclear energy. In Korea, in which a low and intermediate radioactive waste repository is already operating, the construction of an underground research laboratory for in situ validation studies became a matter of interest with increasing concerns on the management of HLW. In order to construct and to operate an underground HLW repository safely in deep underground, the stability of rock mass should be guaranteed. As an important factor on rock stability, excavation damaged zone (EDZ) has been studied in many underground research laboratories in foreign countries. For accurate evaluation of the characteristics and effects of EDZ under disposal condition, it is required to use reliable investigation method based on the analysis of previous studies in similar conditions. In this study, status of foreign underground research laboratories in other countries, approaches for investigation the characteristics, size, and effect of EDZ, and major findings from the researches were surveyed and reported. This will help the accomplishment of domestic researches for developing HLW management techniques in underground research laboratory.
