NMR study of the interaction of T$_4$ Endonuclease V with DNA

  • In order to obtain insight into the mechanism by which DNA containing a thymine photo-dimer is recognized by the excision repair enzyme, T$_4$ endonuclease V, we have taken NMR study of this protein and its complex with oligonucleotides. The conformations of five different DNA duplexes DNA I : d(GCGGATGGCG).d(CGCCTACCGC), DNA II d(GCGGTTGGCG) .d(CGCCAACCGC), DNA III : d(GCGGT ^ TGGCG) .d(CGCCAACCGC), DNA IV d(GCGGGCGGCG).d(CGCCCGCCGC) and DNA V d(GCGGCCGGCG) . d(CGCCGGCCGC) were studied by $^1$H NMR. The NMR spectra of these five DNA duplexes in the absence of the enzyme clearly show that the formation of a thymine dimer within the DNA induces only a minor distortion in the structure, and that the overall structure of B type DNA is retained. The photo-dimer formation is found to cause a large change in chemical shifts at the GC7 base pair, which is located at the 3'-side of the thymine dimer, accompanied by the major conformational change at the thymine dimer site. The binding of a mutant T$_4$ endonuclease V (E23Q), which is unable to digest DNA containing a thymine dimer, to the DNA duplex d(GCGGT ^ TGGCG)ㆍd(CGCCAACCGC) causes a large down-field shift in the imino proton resonance of GC7. Therefore, this position is thought to be either the crucial point of the interaction wi th T$_4$ endonuclease V, or the si to of a conformational change in the DNA caused by the binding of T$_4$ endonuclease V. Usually, it is very difficult to assign NMR peaks in DNA * protein complex because of severe peak overlaps. In order to overcome these peak overlaps, we used a method of deuterium incorporation.

  • The main purpose of this study is to identify the shallow gas origin in the KSSM zone. Based on the results of gas composition and isotope in the headsapace gas, the shallow gas is mainly composed of methane and carbon and deuterium isotopes (${\delta}^{13}CCH_4$ and ${\delta}DCH_4$) of methane has ranged from -93.4%o to -70.9%, and from -228%o to -199%o in each. These results imply that shallow gas has predominately biogenic source by $CO_2$ reduction rather than thermogenic. The carbon isotopic separation (${\varepsilon}_c$) between methane and carbon dioxide $(CO_2)$ has a range of 54.4 to 72.2, it also supports biogenic origin of shallow gas.

Old Water Contributions to a Granitic Watershed, Dorim-cheon, Seoul

  • It is reported that the intensity of rainfall will likely increase, on average, over the world on 2000. For water resources security, many studies for flow paths from rainfall or snowmelt to subsurface have been conducted. In Korea, few isotopic studies for characterizations of flow path have been undertaken. For a better understanding of how water derived from atmosphere moves to subsurface and from subsurface to stream, an analysis of precipitation and stream water using oxygen-18 and deuterium isotopes in a small watershed, Dorim-cheon, Seoul, was conducted with high resolution data. Variations of oxygen-18 in precipitation greater than 10‰ (δ18Omax = −1.21‰, δ18Omin = −11.23) were observed. Isotopic compositions of old water (groundwater) assumed as the stream water collected in advance were −8.98‰ and −61.85‰ for oxygen and hydrogen, respectively. Using a two-component mixing model, hydrograph separation of the stream water in Dorim-cheon was conducted based on weighted mean value of δ18O. As a result, except of instant dominance of rainfall, contribution of old water was dominant during the study period. On average, 71.3% of the old water and 28.7% of rainfall contributed to the stream water. The results show that even in the small watershed, which is covered with thin soil layer in granite mountain region, the stream water is considerably influenced by old water inflow rather than rainfall.

A Study on Applicability of SP Creep Testing for Measurement of Creep Properties of Zr-2.5Nb Alloy (Zr-2.5Nb 합금의 크리프 물성 측정을 위한 SP 크리프 시험의 적용성에 대한 연구)

  • The pressure tubes made of cold-worked Zr-2.5Nb alloy are subjected to creep deformation during service period resulting in changes to their geometry such as longitudinal elongation, diameter increase and sagging. To evaluate integrity of them, information on the material creep property of the serviced tubes is essential. As one of the methods with which the creep property is directly measured from the serviced components, small punch(SP) creep testing has been considered as a substitute for the conventional uniaxial creep testing. In this study, applicability of the SP creep testing to Zr-2.5Nb pressure tube alloy was studied particularly by measuring the power law creep constants, A, n. The SP creep test has been successfully applied fur other high temperature materials which have isotropic behavior. Since the Zr-2.5Nb alloy has anisotropic property, applicability of the SP creep testing can be limited. Uniaxial creep tests and small punch creep tests were conducted with Zr-2.5Nb pressure tube alloy along with finite element analyses. Creep constants obtained by each test method are compared. It was argued that the SP creep test result gave results reflecting material properties of both directions. But the equations derived in the previous study for isotropic materials need to be modified. Discussions were made fur future research directions for application of the SP creep testing to Zr-2.5Nb tube alloy.

Biosynthesis of L-Azetidine-2-Carboxylic Acid In Actinoplanes ferrugineus

  • L-Azetidine-2-carboxylic acid (A-2-C), a four-membered cyclic imino acid has been identified in certain plants, and the microorganism Actinoplanes ferrugineus. The imino acid A-2-C has a physiological significance as an antgaonist of proline during peptide synthesis. The biosynthetic mechanism for the formation of A-2-C has not been studied in any detail. By using various amino acids such as methionine and S-adenosyl-L-methionine labeled with deuterium or carbon-14, the details of the biosynthetic pathway and a possible mechanism for the formation of L-A-2-C in .4. ferrugineus have been unravelled, Both in vivo and in vitro experimental results suggest the biosynthesis of L-A-2-C is mediated by a confactor containing a carbonyl group, probably pyridoxal Phosphate. S-Adenosyl-L-methionine, which seems to be the direct biosynthetic substrate, has undergone a f-displacement by an ${\alpha}$-amino group of the amino acid portion of the substrate S-adenosyl-L-methionine potentially via a vinylglycine intermediate. The overall stereochemical events at the ${\beta}$-carbon of the substrate have been shown to inversion of configuration. The overall stereochemical events at the -position of the sub- strate have also been shown to occur with inversion of configuration. The ${\beta}$, ${\gamma}$-elimination reaction of the substrate seems to follow a cisoidal-type mechanism and the addition portion of the reaction a transoidal-type mechanism . The assignment of the proton NMR of A-2-C has been deduced by apply- ing NOE difference experiments, Gd(III) line-broadening experiments and 2D-NOESY experiments of regio-and stereospecificially deuterated A-2-C's.

  • The dryer (ten per unit) are operating to remove tritium in PHWR(Pressurized Heavy Water Reactor). There are coming out heavy water adsorbent from operating the dryer (95 drums for ten year per unit) The amount of radioactivity of heavy water adsorbent almost exceed ninety times more than disposal limit-in-itself showed by The Ministry of Science and Technology. It has to be disposed whole radioactive waste products, however there are problems of increase at the expense of their permanent disposal. In this research, We have studied how to remove kinds of nuclear materials and amount of tritium with in heavy water adsorbent. As the result we could develop disposal equipment and apply it. D20 adsorbent have to contain below Gamma nuclide O.3Bq/g and tritium 100Bq/g "The Regulation for disposal of the radioactivity wastes" showed by The Ministry of Science and Technology. There fore. So as to remove amount of tritium and kinds of nuclear materials (DTO) we needed a equipment. Also we have studied how to remove effectively radioactivity with in Adsorbent. As cleaning heavy water adsorbent and drying on each condition (temperature for drying and hours for cleaning). Because there is something to return heavy water adsorbent by removing impurities within adsorbent when it is dried o high temperature. After operating, we have been applying this research to the way to dispose heavy water adsorbent. Through this we could reduce solid waste products and the expense of permanent disposal of radioactive waste products and also we could contribute nuclear power plant run safely. According to the result we could keep the best condition of radiation safety super vision and we could help people believe in safety with Radioactivity wastes control for harmony with Environment.

Safety Assessment on Long-term Radiological Impact of the Improved KAERI Reference Disposal System (the KRS+)

  • The Korea Atomic Energy Research Institute (KAERI) has developed geological repository systems for the disposal of high-level wastes and spent nuclear fuels (SNFs) in South Korea. The purpose of the most recently developed system, the improved KAERI Reference Disposal System Plus (KRS+), is to dispose of all SNFs in Korea with improved disposal area efficiency. In this paper, a system-level safety assessment model for the KRS+ is presented with long-term assessment results. A system-level model is used to evaluate the overall performance of the disposal system rather than simulating a single component. Because a repository site in Korea has yet to be selected, a conceptual model is used to describe the proposed disposal system. Some uncertain parameters are incorporated into the model for the future site selection process. These parameters include options for a fractured pathway in a geosphere, parameters for radionuclide migration, and repository design dimensions. Two types of SNF, PULS7 from a pressurized water reactor and Canada Deuterium Uranium from a heavy water reactor, were selected as a reference inventory considering the future cumulative stock of SNFs in Korea. The highest peak radiological dose to a representative public was estimated to be 8.19×10-4 mSv·yr-1, primarily from 129I. The proposed KRS+ design is expected to have a high safety margin that is on the order of two times lower than the dose limit criterion of 0.1 mSv·yr-1.

Comparative Evaluation of Mn(II) Framework Substitution in MnAPSO-34 and Mn-impregnated SAPO-34 Molecular Sieves Studied by Electron Spin Resonance and Electron Spin Echo Modulation Spectroscopy

  • MnAPSO-34 and Mn-impregnated SAPO-34(Mn-SAPO-34) sample were prepared with various manganese contents and studied by electron spin resonance(ESR) and electron spin echo modulation(ESEM). Electron spin echo modulation analysis of 0.07mo1 % Mn(relative to p) in MnAPSO-34 with adsorbed D$_{2}$O shows two deuteriums at 0.26 nm and two at 0.36 nm from Mn. This suggests that two waters hydrate an MnO$_{4}$ configuration with a D-O bond orientation for the waters as expect for a negatively charged site at low manganese content (0.1 mol%), the ESR spectra of MnAPSO-34 and MnH-SAPO-34 exhibit the same parameters(g 2.01 and A 89 G), but the spectra obtained from MnAPSO-34 samples are better resolved. The decomposition temperature of as-synthesized MnAPSO-34 were in the range of 200-600 $^{\circ}C$ of the morpholine which is 12 $^{\circ}C$ higher than that in as-synthesized MnH-SAPO-34. Infrared spectra showed that the position of a band at 3450 $cm^{-1}$ / shifted about 15 $cm^{-1}$ / toward higher energy in MnAPSO-34 versus MnH-SAPO-34. The modulation depth of the two-pulse ESE of MnAPSO-34 with adsorbed D$_{2}$O is deeper than that of MnH-SAPO-34 with adsorbed D$_{2}$O. Three-pulse ESEM of MnAPSO-34 and MnH-SAPO-34 with adsorbed deuterium oxide shows that the local environments of manganese in the hydrated samples are different, suggesting that Mn(II) is framework substituted in MnAPSO-34 since it obviously occupies an extraframework position in MnH-SAPO-34.

  • In this study, a mathematical model for optimal operation of the fusion fuel cycle is developed based on scheduling approach. The fusion fuel cycle consists of a system for storing and supplying deuterium and tritium, and receiving and separating process after the fusion reaction. Except that tritium is a radioactive material, most of these processes consist of catalytic reactions and separation process. For these reasons, it is possible to apply scheduling approach which is also widely utilized to chemical plants to derive the optimal operating scenarios. The developed model determined the optimal regeneration cycle to minimize the amount of tritium inside the vacuum pumps. Based on the characteristics of various device in the fusion reactor, the optimal tritium plant operation scenario is evaluated. The formulated model was applied to the actual tokamak scenario and utilized to analyze the fuel flow and balance of ITER fuel cycle.

  • The ITER fuel cycle is designed for DT operation in equimolar ratio. It involves not only a group of fuelling system and torus cryo-pumping system of the exhaust gases through the divertor from the torus in tokamak plant, but also from the exhaust gas processing of the fusion effluent gas mixture connected to the hydrogen isotope separation in cryogenic distillation to the final safe storage & delivery of the hydrogen isotopes in tritium plant. Tritium plant system supplies deuterium and tritium from external sources and treats all tritiated fluids in ITER operation. Every operation and affairs is focused on the tritium inventory accountancy and the confinement. This paper describes the major fuel cycle processes and interfaces in the tritium plant in aspects of upcoming technologies for future hydrogen and/or hydrogen isotope utilization.
