Osteoporosis is one of the major health problem affecting postmenopausal women. Estrogen deficiency results in an increase in bone turnover, lead to bone resorption and an increase risk of fracture. The aim of this study was to evaluate the effects of Platycodon grandiflorum A. extract (PG) in bone metabolism in ovariectomized estrogen-deficient rats. Two groups were surgically ovariectomized (OVX). The third group was sham operated. Sprague-Dawley female rats were randomly assigned to the following groups : sham-operated rats (Sham), ovariectomized control rats (OVX-control), ovariectomized rats supplemented with PG at 50mg/kg body wt (OVX-PG50) The Platycodon grandiflorum extracts were orally administrated at 1mL per day. The ovariectomy caused a decreasing in the levels of collagen content in bone, cartilage and skin tissues. However PG group, supplementation with Platycodon grandiflorum extract, were increased the level of collagen content in bone, cartilage and skin tissues than OVX-control group. PG group had a higher content of pyridinoline in collagen than OVX-control group. Alkaline phosphatase activity on serum were decreased after supplemented with the PG extract. These results might be expected that Platycodon grandiflorum is believed to be possible protective effects in postmenopausal bone loss.
배경: 골다공증은 골대사의 불균형으로 인해 골 흡수가 골 형성보다 많아져 골밀도가 감소함으로서 발생한다. 골다공증의 이상적인 치료목표는 골형성을 증가시키거나 골소실을 방지하여 골량을 현 상태로 유지하는 것이다. 따라서 향후 발생되는 골소실을 에방하는 것이 골다공증의 원칙적이고 효과적인 치료방법이 될 것이다. 본 연구에서는 흑효모 중 $Aureobasidium$$pullulans$으로부터 유래한 폴리칸(베타-글루칸)이 중년여성의 골대사에 미치는 영향을 규명하고자 하였다. 연구방법: 골대사에 대한 폴리칸의 효과를 규명하기 위해 12주간의 무작위배정, 이중눈가림, 플라세보 대조 임상연구를 수행하였다. 총 60명(폴리칸 투여군30명, 플라세보 투여군 30명)의 중년 여성 피험자가 등록되어 이 중 총 58명의 피험자가 최종적으로 12주간의 임상연구를 종료하였다. 결 과: 폴리칸(150 mg/d) 투여 12주 후, 폴리칸 투여군은 요 중 Deoxypyridinoline (DPD) 농도가 유의적인 감소를 보였다($P$=0.014). 혈청 중 Osteocalcin(OSC) 농도는 두 군 모두에서 유의적으로 증가하였으며, bone-specific alkaline posphatase (bALP) 와 collagen type 1 cross-linked C-telopeptide (CTx)는 유의적 변화가 보이지 않았다. 폴리칸은 골밀도(BMD)와 혈청 부갑상선 호르몬(iPTH)에 대해 유의적인 변화를 보이지 않았으나, 24시간 요 중 Ca 배설량은 폴리칸 투여군에서 유의하게 감소되었다($P$=0.028). 또한 폴리칸 투여군에서 고밀도지단백 콜레스테롤(HDL-cholesterol) 농도의 증가 경향 및 중성지방(triglyceride)의 유의적인 감소가 보였다. 임상연구 기간 중에 발생한 이상반응은 두 군간에 유의적인 차이를 보이지 않았다. 결 론: 본 연구에서는 폴리칸이 골대사 및 지질에 대해 일부 개선효과가 있음을 보여주었다. 그러나, 골다공증 예방 측면에서 보다 장기적인 임상연구와 피험자 수를 확대하여 골대사 및 지질대사에 대한 폴리칸의 예방적 효과를 규명할 필요가 있을것으로 사료된다.

The aim of this study was to evaluate the effects of Capsosiphon fulvecense extract (SCF) on the collagen content of the connective tissues and alkaline phosphatase activity of serum in ovariectomized estrogen-deficient rats. Three groups were surgically ovariectomized (OVX). The fourth group was sham operated. Sprague-Dawley female rats were randomly assigned to the following groups : sham-operated rats (Sham), ovariectomized control rats (OVX-control), ovariectomized rats supplemented with CsF at 50mg/kg body wt (OVX-CSF50) and ovariectomized rats supplemented with CsF at 200mg/kg body wt (OVX-CSF200). The Capsosiphon fulvecense extracts were orally administrated at 1mL per day. The ovariectomy caused a decreasing in the levels of collagen content in bone, cartilage, skin and lung tissues. However CSF groups, supplementation with Capsosiphon fulvecense extract, were increased the level of collagen content in bone, cartilage, skin and lung tissues than OVX-control group. Alkaline phosphatase activity also were increased and calcium levels were decreased than OVX-control on serum. These results suggest that Capsosiphon fulvecense supplementation prevents postmenopausal bone loss, thus it may be used possibly to improve the quality of life in menopausal women.

The purpose of this study was to investigate the relation between NEAP (net rate of endogenous noncarbonic acid production) from dietary potassium and protein intakes, and bone mineral density and urinary deoxypyridinoline (DPD) in Korean women. The subjects were 276 women aged $18\sim81$, and they were asked about general characteristics and nutrient intake using 24-hr recall method. Also, their bone mineral density of spine and femoral neck were measured using DEXA. Urinary DPD analysis was done in 106 postmenopausal women. The average energy, protein and potassium intake were 1503.0 kcal, 57.7 g and 60.8 mEq, respectively. Mean NEAP [54.5$\times${protein intake (g)/potassium intake (mEq)}-10.2] was 44.6. NEAP showed a significantly negative correlation with spine bone mineral density of the subjects after controlling for confounding factors (age, height and weight) (p<0.05). The lowest quartile of protein intake had the highest spine bone mineral density after controlling for confounding factors (p<0.05). There was no significant correlation between urinary DPD and calculated NEAP. These results provided positive relation between a ratio of lower protein to higher potassium dietary intake and bone mineral density.
The purpose of this study was to determine which differences in the source of protein (soy vs casein) and isoflavones in soy protein are responsible for the differential effects of bone marks and hormones in growing female rats. Forty-two 21-day-old Sprague-Dawley female rats were randomly assigned to one of three groups, consuming casein (control group), soy protein isolate (57 mg isoflavones/100 g diet), or soy protein concentrate (about 1.2 mg isoflavones/100 g diet). All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone formation was measured by serum osteocalcin and alkaline phosphatase (ALP) concentrations. And bone resorption rate was measured by deoxypyridinoline (DPD) crosslinks immunoassay and corrected for creatinine. Serum osteocalcin, growth hormone, estrogen and calcitonin were analyzed using radioimmunoassay kits. Diet did not affect weight gain and mean food intake. Food efficiency ratio was lower in the soy protein groups. The soy isolate group had a higher ALP and osteocalcin concentration and lower crosslinks value than the casein group. Therefore, the soy isolate groups had a higher bone formation/resorption ratio than the casein group. And, the soy group had significantly higher growth hormone than the casein group. The findings of this study suggest that soy protein and isoflavones are beneficial for bone formation in growing female rats. Therefore, exposure to these soy protein and isoflavones early in life may have benefits for osteoporosis prevention.
Taurine supplementation has been shown to have a beneficial effect on femur bone mineral content in ovariectomized rats. It therefore seemed desirable to find out whether the beneficial effect of taurine on ovariectomized rats fed calcium deficient diet could also be reproduced. Forty female Sprague-Dawley rats were divided into two groups, One group was OVX and the other group received sham operation (SHAM), and received either control diet or a taurine supplemented diet for 6 weeks. All rats were fed on calcium deficient diet (AIN-93: 50% level of calcium) and deionized water. Bone mineral density (BMD) and bone mineral content (BMC) were measured in spine and femur. The serum and urine concentrations of calcium and phosphorus were determined, Bone formation was measured by serum osteocalcin and alkaline phosphatase (ALP) concentrations, Bone resorption rate was measured by deoxypyridinoline (DPD) crosslinks immunoassay and corrected for creatinine. Urinary calcium and phosphorus excretion, osteocalcin in blood and cross link value were not significantly different among the groups. Within the OVX group, the taurine supplemented group had not higher femur bone mineral content than the control group. This study established the need for a study on the taurine effect on bone with different calcium levels.
Osteoporosis is a disease that often occurs in postmenopausal women. The purpose of this study was to examine the positive effects of Cheongukjang powder containing menaquinone-7 (MK-7) on bone health circulation in postmenopausal women. Fifty-six postmenopausal women were recruited into a randomised double-blind placebo-controlled trial. The participants were randomly assigned into two groups. For 12 weeks, one group received $800{\mu}g$ MK-7 in the form of Cheongukjang packs (CMK-7), and the other group received the same amount of identical-looking placebo packs containing barley meal for 12 weeks. Femoral bone mineral density (BMD), bone-specific alkaline phosphatase (ALP), deoxypyridinoline (DPD), osteocalcin (OSC), serum Ca, and serum P were measured at baseline (0 weeks) and 12 weeks. After 12 weeks in the CMK-7 group, it was found that BMD, serum Ca, and serum P had increased above the baseline (p<0.0001, p=0.0028, p<0.0001), whereas bone-specific ALP, DPD, and OSC had decreased below the baseline (p=0.0003, p<0.0001, p<0.0001). Therefore, MK-7 taken as Cheongukjang is expected to prevent osteoporosis in postmenopausal women.
The effect of calcium and boron supplementation on bone strength was determined in growing and ovariectomized (OVX) Sparague-Dawley rats. Rats were divided into 9 groups and fed diet with different intake levels of calcium and boron for 4 weeks. About fifty percentages of rats in each group were OVX and the others were sham-operated. The rats were fed same diets after operation for 8 weeks. The feed intake, body weight gain, and FER were significantly higher in OVX rats than those in sham-operated ones. Serum osteocalcin, bone formation biomarker, was significantly increased with increment in calcium and boron intakes. Serum estradiol was lower in OVX rats than in sham-operated ones. Bone mineral density of femur was significantly lower in OVX rats than in other group. The breaking forces of bones were not significantly different among the groups. The urinary excretion of deoxypyridinoline, osteolytic marker was significantly increased with increment in calcium intake and ovariectomy. The urinary calcium excretion was significantly increased with increment in calcium intake, but decreased with increment in boron intake. According to theses results, the boron supplementation resulted in higher serum osteocalcin and lower urinary calcium excretion. Therefore, it could be suggested that the boron supplementation may be complementary and useful to calcium nutrition for bone health.
Purpose: This study was done to identify the effects of weight-bearing exercise(WBE) on bone metabolism. Method: WBE was performed for 12 weeks by healthy college women. Bone-related parameters were measured four times during this period by evaluating the immunoradiometric assay and enzyme immunoassay. Bone mineral densities(BMDs) were measured by dual energy x-ray absorptiometry before and after the WBE program. Data was analyzed using t-test, paired t-test, $x^2$-test, and repeated measures ANOVA. Result: Osteocalcin, a bone formation marker, increased more in the experimental group than in the control group based on the interaction between time and group(F=3.29 p=.024). Little difference between the two groups was found for the other parameters: urinary deoxypyridinoline, insulin-like growth factorI, parathormone, serum calcium, and serum phosphorus without showing any time interaction between the groups. The femoral trochanter BMD rose in the experimental group while that of the control group fell, showing a significant difference for BMD(t=3.06 p=.005). However, there was no significant difference between the two groups for changes in BMD of the forearm, lumbar spine, femoral neck, and femoral ward's triangle. Conclusion: These findings supported the WBE is beneficial for increasing bone formation in college women and long-term application is needed to substantiate the effects of WBE as a intervention in promotion of bone-health.
An important related question is whether arginine has influence bone metabolism. The effect of arginine supplements on bone markers and related hormones were studied in young female Sprague-Dawley rats fed either an arginine supplemented diet or control diet. Twenty four rats (body weight 83${\pm}$5 g) were randomly assigned to one of two groups, consuming casein or casein with supplemented arginine diet. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone formation was measured by serum osteocalcin and alkaline phosphatase (ALP) concentrations. And bone resorption rate was measured by deoxypyridinoline (DPD) crosslinks immunoassay and corrected for creatinine. Serum osteocalcin, growth hormone, estrogen, insulin-like growth factor-1 (IGF-1), parathyroid hormone (PTH) and calcitonin were analyzed using radioimmunoassay kits. The weight gain and mean food intake were not affected regardless of diets. The rats fed arginine-supplemented diet had not significantly different in ALP, osteocalcin, crosslinks value, PTH, estradiol, and IGF-1 compared to those fed casein diet group. The arginine-supplemented group had significantly higher growth hormone and calcitonin than casein group. This study suggests that arginine is beneficial for bone formation in growing female rats. Therefore exposure to diet which rich in arginine early in life may have benefits for bone formation and osteoporosis prevention.
