Bacterial chemotaxis is essential to the study of structure and function of bacteria. Although many studies have accumulated the knowledge about chemotaxis in the past, the motion of a single bacterium has not been studied much yet. In this study, we have developed a device microfabricated by soft lithography and consisting of microfluidic channels. The microfluidic assay generates a concentration gradient of chemoattractant linearly in the main channel by only diffusion of the chemicals. Bacteria are injected into the main channel in a single row by hydrodynamic focusing technique. We measured the velocity of bacteria in response to a given concentration gradient of chemoattractant using the microfludic assay, optical systems with CCD camera and simple PTV (Particle Tracking Velocimetry) algorithm. The advantage of this assay and experiment is to measure the velocity of a single bacterium and to quantify the degree of chemotaxis by statistically analyzing the velocity at the same time. Specifically, we measured and analyzed the motility of Escherichia coli strain RP437 in response to various concentration gradients of L-aspartate statistically and quantitatively by using this microfluidic assay. We obtained the probability density of the velocity while RP437 cells are swimming and tumbling in the presence of the linear concentration gradient of L-aspartate, and quantified the degree of chemotaxis by analyzing the probability density.

In this paper, the corona discharge is analyzed by Finite Element Method(FEM) combined with Flux-corrected Transport(FCT) algorithm. In the previous papers, Finite Difference Method(FDM) combined with FCT was used. Usually in the FDM, the regionof interest is discretized with structured grids. But to refine local regions with same resolution, much more grids are required for the structured grids than for unstructured grids than for unstructured grids. Therefore, we propose the FEM-FCT method to simulate the corona discharge. The proposed method has good flexibility in model shape and can reduce the computational cost by the local refinement where the physical quantities have steep gradients. Using the proposed method, we study the streamer growth of parallel plate electrodes which is initiated by the low and high perturbation density. We find that the varying the initial density of perturbation has very little effect on the streamer propagation. And the corona discharge of the rod-to-plane electrode is simulated. On the surface of the rod electrode, the high concentration of the electric field gives rise to many number of streamer seeds. The strong axial streamer propagate to the plane electrode. The weaker non-axial streamer repel each other and stop growing more. The results are very similar to those of the papers which used the FDM-FCT method on structured grids. Thus we can conclude that the proposed FEM-FCT method is more efficient than the conventional FDM-FCT method by virtue of the reduction in computational grids number.
Objective : The purpose of this study is to investigate the mechanism and effect of moxibustion with monkshood cake, slice & black pepper cake. objectively, to be used as the quantitative data through the measurement of temperatqre, and to grasp the thermodynamic characteristics of moxibustion with monkshood cake, slice & black pepper cake. Methods : We have selected of the moxibustion with monkshood cake, slice & black pepper cake. indirect moxibustion. We make a comparative study of the thermodynamic characteristics of moxibustion with monkshood cake, slice & black pepper cake. We examined combustion times, temperatures, temperature gradients in each period during a combustion of moxa. Results & Conclusions : 1. We can design the moxibustion with monkshood cake that it has thermodynamic characteristics of 173sec effective combustion time, $44^{\circ}C$ maximum temperature, $0.22^{\circ}C/sec$ ascending maximum temperature, if we use 3mm thickness or 3mm and below of monkshood cake and the moxa cone is formed the conical shape that the base diameter was 8mm, the height was 10mm, the density was $600mg/cm^3$. 2. We can design the moxibustion with monkshood cake that it has thermodynamic characteristics of 205~271sec effective combustion period time, $44.6{\sim}46.1^{\circ}C$ maximum temperature, $0.18{\sim}0.24^{\circ}C/sec$ ascending maximum temperature, if we use 3mm thickness of monkshood cake and the moxa cone is formed the conical shape that the base diameter was 8mm, the height was 10mm, the density was $480{\sim}720mg/cm^3$.
This study was carried out to improve of effective culture system on development of IVM/IVF/IVC bovine embryos. The cumulus-oocyte-complexes (COCs) collected from Korean cattle ovaries harvested at a local abattoir were matured in 50 ${mu}ell$ of TCM199 supplemented with 10% fetal bovine serum (FBS) and hormones (35 $\mu\textrm{g}$/$m\ell$ FSH, 10 $\mu\textrm{g}$/$m\ell$ LH, 1$\mu\textrm{g}$/$m\ell$ estradiol 17 $\beta$ under paraffin oil at 39$^{\circ}C$ in a humidified atmosphere of 5% $CO_2$in air. At 24 hrs after culture, matured oocytes were fertilized in vitro for 22~24 hrs with motile semen in which obtained by centrifugation of a frozen thawed semen on Percoll-density gradients (45% vs. 90%) at 500 g for 20 min. The presumptive zygotes were divided into three experimental groups. Single egg (Group 1), 25 (Group 2) or 50 eggs (Group 3) were cultured on cumulus cell in 50 ${mu}ell$ TCM199 supplement with 10% FBS for 6~9 days after fertilization. In vitro developmental rates into the blastocysts in the groups 2 and 3 were significantly (P<0.05) higher than those of group 1 (37,27 vs. 6%, respectively). Cell number of blastocysts obtained in groups 2 and 3 at day 8 were significantly (P${mu}ell$) resulted in higher developmental competence and cell number of bovine blastocysts produced in vitro than those the culture of single embryos with cumulus cells.

We analyzed image factors to determine the characteristic factors that need for intelligent replenishment system of the auto film processor. We processed the serial 300 sheets of radiographic films of chest phantom without replenishment of developing and fixation replenisher. We took the digital data by using film digitizer which scaned the films and automatically summed up the pixel values of the films. We analyzed characteristic curves, average gradients and relative speeds of individual film using densitometer and step densitometry. We also evaluated the pH of developer, fixer, and washer fluid with digital pH meter. Fixer residual rate and washing effect were measured by densitometer using the reagent methods. There was no significant reduction of the digital density numbers of the serial films without replenishment of developer and fixer. The average gradients were gradually decreased by 0.02 and relative speeds were also gradually decreased by 6.96% relative to initial standard step-densitometric measurement. The pHs of developer and fixer were reflected the inactivation of each fluid. The fixer residual rates and washing effects after processing each 25 sheets of films were in the normal range. We suggest that the digital data are not reliable due to limitation of the hardware and software of the film digitizer. We conclude that average gradient and relative speed which mean the film's contrast and sensitivity respectively are reliable factors for determining the need for the replenishment of the auto film processor. We need more study of simpler equations and programming for more intelligent replenishment system of the auto film processor.
The Atlantic meridional overturning circulation (AMOC), driven by high density water sinking around Greenland serves as a global climate regulator, because it transports heat and materials in the climate system. We analyzed the mechanism of AMOC on a decadal time scale simulated with the HadGEM2-AO model. The lead-lag regression analysis with AMOC index shows that the decadal variability of the thermohaline circulation in the Atlantic Ocean can be considered as a self-sustained variability. This means that the long-term change of AMOC is related to the instability which is originated from the phase difference between the meridional temperature gradient and the ocean circulation. When the overturning circulation becomes stronger, the heat moves northward and decreases the horizontal temperature-dominated density gradients. Subsequently, this leads to weakening of the circulation, which in turn generates the anomalous cooling at high latitudes and, thereby strengthening the AMOC. In this mechanism, the density anomalies at high latitudes are controlled by the thermal advection from low latitudes, meaning that the variation of the AMOC is thermally driven and not salinity driven.

Macrozoobenthos were collected from 24 sites using small grab ($0.05m^2$) in order to see the spatial distribution of them at Sueocheon stream at the northern part of Gwangyang Bay during summer season when the maximum ecological processes are occurring. A total of 44 species of macrozoobenthos occurred, and their mean density was $789individuals/m^2$. Mollusks was the dominant faunal group accounted for 43.1% and 86.8% of the whole community density and biomass, respectively. Top five dominant species were a molluscan, Corbicula japonica (40.7%), two polychaete worms, Neanthes japonica (19.3%) and Heteromastus filiformis( 8.1%), and two crustaceans, Paranthura japonica (6.9%) and Jaeropsis sp. (6.9%). Species diversity index was very low with mean value of 1.12 and maximum value was recorded at the river mouth. At the upper stream, a brackish water species such as Corbicula japonica showed their maximum population density. The overall distribution of stream macrobenthos showed a rather simple pattern along with salinity and grain size gradients with few brackish water species occurring at coarse sediments in the stream but more diverse faunas inhabiting fine sediments in the river mouth.
In this study, we analyze the structure of the Korea-Japan Joint Development Zone (JDZ) using gravity, magnetic, and seismic data. Gravity and magnetic data analysis confirmed that the Jeju Basin exhibits low anomalies compared to adjacent areas. We applied the total gradient to the Bouguer anomaly to identify basin boundaries, and computed the analytic signal from the total magnetic anomaly data to enhance the edges of the magnetic anomalies. The Taiwan-Sinzi Belt, exhibits high magnetic anomalies and crosses the center of the JDZ in the northeast-southwest direction; we presume that intrusive rocks are sporadic in the JDZ. The 3D inversion results of the gravity and magnetic data show a strong correlation between magnetic susceptibility and density (i.e. a low-density zone in the Jeju Basin and the Ho Basin, and a high magnetic susceptibility distribution in the Taiwan-Sinzi Belt). Comparison of the density and seismic profiles of the Jeju Basin shows that high densities are associated with sill, horst, and basement highs, whereas low densities are associated with basement low and grabens. These results suggest that interpretations based on seismic, gravity and magnetic data can effectively reveal the subsurface structure of the JDZ.
Delta-endotoxin in Bacillus thuringiensis var. finitimus, HD-1, HD-9 and HD-73 strains were isolated by NaBr, CsCl and Renografin density gradients. The purity of the toxin was about 98%. The purified o-endotoxin was analyzed by SDS-PAGE, electron microscopic observation and bioassay. According to SDS-PAGE, the molecular weight of subunits of the o-endotoxin were about 66,000 and 130,000 daltons. The shapes of the crystal toxin observed by TEM except finitimus strain were bipyramidal. When the purified endotoxin was bioassayed against tobacco horn worm, the entomocidal activities ($1{\mu}g/ml$) of HD-1 and HD-73 strains were, respectively, 60% and 100% at nine days after treated. The molecular weights of the plasmids isolated from B. thuringiensis were various from 0.5 to 120 Kb. The numbers of plasmids in HD-1, HD-9 and HD-73 strains were 12, 3 and 11, respectively, but B. thuringiensis var. finitimus strain had no plasmid.

During sintering of very porous green bodies, as obtained by compaction of hard powders - such as tungsten carbide or ceramics - or by injection moulding, important shrinkage occurs. Due to heterogeneous green density field, gravity effects, friction on the support, thermal gradients, etc., this shrinkage is often non-uniform, which' may induce significant shape changes. As the ratio of compact dimension to powder size is very high, the mechanics of continuum is relevant to model such phenomena. Thus numerical techniques, such as the finite element method can be used to simulate the sintering process and predict the final shape of the sintered part. Such type of simulation has much been developed in the last decade firstly for hot isostatic pressing and next for die compaction. Finite element modelling has been recently applied to free sintering. The simulation of sintering should be based on constitutive equations describing the thermo-mechanical behaviour of the material under any state of stress and any temperature which may arise within the sintering body. These equations can be drawn either from experimental data or from micromechanical models. The experiments usually consist in free sintering and sinter-forging tests. Indeed applying more complex loading conditions at high temperature under controlled atmosphere is delicate. Micromechanical models describe the constitutive behaviour of aggregates of spheres from the deformation of two-sphere contact either by viscous flow or grain boundary diffusion. Such models are not able to describe complex microstructure and mechanisms as observed in real materials but they can give some basic information on the formulation of constitutive equations. Practically both experimental and theoretical approaches can be coupled to identify the constitutive equations. Such procedure has been performed for modelling the sintering of compacts obtained by die pressing of a mixture of tungsten carbide and cobalt powders. The constitutive behaviour of this material during sintering has been described by a linear viscous constitutive model, whose functions have been fitted from results of free sintering and sinter-forging experiments. This model has next been introduced in ABAQUS finite element code to simulate the sintering of heterogeneous green compacts of various geometries at constant temperature. Examples of simulations are shown and compared with experiments.
