황칠나무 추출물을 함유한 폴리우레탄 나노섬유의 제조 및 특성 (Preparation and Characterization of Polyurethane Nanofibers Containing Dendropanax morbiferus Extracts)

  • In this study, we first analyzed the total polyphenol and flavonoid contents and the antioxidant activity of Dendropanax morbiferus extracts obtained from different parts using different solvents. We then investigated the possibility of producing Dendropanax morbiferus leaf extract loaded polyurethane (PU) nanofibers via electrospinning. The total polyphenol and flavonoid content and the antioxidant activity of the Dendropanax morbiferus leaf extracts obtained distilled water were found to be higher than those of the extracts obtained under other conditions, and it was therefore used to prepare the extracts/PU nanofibers by varying the concentrations of both the Dendropanax morbiferus leaf extract and PU. The most appropriate fiber morphology were when 1.5% and 1.0% of the Dendropanax morbiferus leaf extracts were added to 12% and 13% PU, respectively. Fourier-tranform infrared (FT-IR) spectroscopy and X-ray diffraction (XRD) analyse reveal the successful incorporation of the Dendropanax morbiferus leaf extracts into the PU matrix. In addition, the nanofibers containing this leaf extract were confirmed to exhibit antibacterial properties against Staphylococcus aureus, suggesting the potential usefulness of nanofacial masks containing the Dendropanax morbiferus leaf extract.

황칠나무(Dendropanax morbiferus Lev.) 잎 에탄올 추출물의 항균 및 항산화활성 (Antimicrobial and Antioxidant Activities of Ethanol Leaf Extract of Dendropanax morbiferus Lev.)

  • In the study reported here, the antimicrobial and antioxidant activities of the ethanol leaf extract of Dendropanax morbiferus Lev. from Jeju Island was investigated. Of the 14 strains of 12 species of microorganisms tested, the extract exhibited antimicrobial activity observed against seven Gram-positive bacteria of four species, but not against six Gram-negative bacteria and a yeast strain. Using the disc diffusion method, the diameter of the inhibition zone increased with application of the extract with every strain and the highest growth inhibition was exhibited with Staphylococcus aureus KCTC 1916 at 5 mg/ml. The minimal inhibitory concentration of the extract (MIC) by turbidity was 2.5 mg/ml against Bacillus cereus KACC 12672 and 15 mg/ml when with Enterococcus faecalis KCTC 3206. The minimum bacterial concentration (MBC) values defined as being $${\geq_-}99.9%$$ reduction in viable cells against the tested strains was higher than the MIC values. Time killing curves using the optimum MIC were performed on seven strains incubated for 48 hr. The growth of B. cereus KACC 12672 was detected after 12 hr and no significant growth was found in the others strains after 48 hr (p<0.05). The 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity of the ethanol leaf extract was similar to that of butylated hydroxyanisole (BHA) at the same concentration. These results indicate that leaf extract of D. morbiferus Lev. can be utilized as a natural preservative and an antioxidant.

Anti-Melanogenic Effect of Dendropanax Morbiferus and Its Active Components via Protein Kinas e A/Cyclic Adenos ine Monophosphate-Responsive Binding Protein-and p38 Mitogen-Activated Protein Kinase-Mediated Microphthalmia-Associated Transcription Factor Downregulation

  • Dendropanax morbiferus H. Lev has been reported to have some pharmacologic activities and also interested in functional cosmetics. We found that the water extract of D. morbiferus leaves significantly inhibited tyrosinase activity and melanin formation in α-melanocyte stimulating hormone (MSH)-induced B16-F10 cells. D. morbiferus reduced melanogenesis-related protein levels, such as microphthalmia? associated transcription factor (MITF), TRP-1, and TRP-2, without any cytotoxicity. Two active ingredients of D. morbiferus, (10E)-9,16-dihydroxyoctadeca-10,17-dien-12,14-diynoate (DMW-1) and (10E)-(?)-10,17-octadecadiene-12,14-diyne-1,9,16-triol (DMW-2) were identified by testing the anti-melanogenic effects and then by liquid chromatography-tandem mass spectrometry (LC/MS/MS) analysis. DMW-1 and DMW-2 significantly inhibited melanogenesis by the suppression of protein kinase A (PKA)/cyclic AMP (cAMP)-responsive binding protein (CREB) and p38 MAPK phosphorylation. DMW-1 showed a better inhibitory effect than DMW-2 in α-MSH-induced B16-F10 cells. D. morbiferus and its active component DMW-1 inhibited melanogenesis through the downregulation of cAMP, p-PKA/CREB, p-p38, MITF, TRP-1, TRP-2, and tyrosinase. These results indicate that D. morbiferus and DMW-1 may be useful ingredients for cosmetics and therapeutic agents for skin hyperpigmentation disorders.

건조 황칠나무 부위별 추출물을 이용한 견직물의 염색성과 기능성 (Dyeability and Functionality of Dried Dendropanax morbiferus Extracts)

  • This study aims to examine the possibility of using dried Dondropanax morbiferus extract as a functional dye. The leaves and branches of were extracted with distilled water and 30% ethanol, and the dyeability and functionality of silk fabrics were examined according to the color characteristics of the extract and dyeing conditions. As a result of analyzing the ultraviolet and visible light absorption spectrum of the extract, it was possible to confirm the peak of flavonoid belonging to polyphenol, and the peak of riboflavin expressing yellow color was confirmed. Adsorption equilibrium was observed at 4% dyeing concentration and 60 minutes of dyeing time, and as the temperature increased, dyeing amount increased without color change of Y-series. Aluminum mordanting also increased the yellow color. The color fastness of washing and UV irradiation was low, but the color fastness of rubbing was evaluated as relatively good. The silk fabric dyed with the distilled water extract of the leaves showed a 99.9% bacteriostatic reduction against Staphylococcus aureus and Klebsiella pneumoniae, showing excellent antibacterial properties.

고초균-젖산균의 순차적 복합 발효를 통한 복합 기능성 물질 함유 황칠나무 추출물의 생산 (Production of Dendropanax morbiferus extract containing multi-functional ingredients by serial fermentation using Bacillus subtilis HA and Lactobacillus plantarum KS2020)

  • 황칠나무 추출물의 순차적 복합발효를 통한 복합 기능성 물질들을 강화시킨 발효소재를 개발하였다. 황칠 추출물에 MSG와 glucose를 첨가한 후 1차 B. subtillis HA 발효를 통해 γ-PGA와 단백질 분해효소를 생성시켰다. 이후 skim milk와 glucose, yeast extract를 추가적으로 첨가하여 30℃에서 L. plantarum KS2020에 의한 2차 복합 발효를 통해서 기능성 물질인 GABA를 생산하였다. 황칠 추출물의 고초균 발효에서 생성된 점질물 및 점조도은 발효 2일 4.21±0.04 Pa·s, 4.1%±0.48%로 높은 점조도 값과 점질물을 함량을 보였다. pH 및 산도는 각각 8.9±0.01, 0.00±0.00이었으며 생균수는 7.27 log CFU/mL에서 발효 2일에 9.50 log CFU/mL로 높은 생균수를 보였다. 순차적 복합 발효가 진행되면서 고초균의 생균수는 4.30 log CFU/mL로 크게 감소하였으며 젖산균은 9.12 log CFU/mL로 증가하였다. Protease 활성 및 tyrosine 함량은 각각 0.8 unit/g, 57 mg%로 측정되었다. 이후 연속적 복합 발효 과정에서 발효 5일 차 106 mg% 정도로 증가되었으며, SDS-PAGE를 이용하여 casein 단백질이 분해된 것을 확인하였다. GABA 정성분석 결과 skim milk 5%만을 첨가한 조건을 제외하고 glucose, yeast extract가 추가로 첨가된 모든 조건에서 전구물질 MSG가 소진되면서 GABA로 전환되었다. 이중 최적 조건을 골라 정량분석을 진행하였으며 18.29 mg/mL의 GABA 함량을 나타내었다. 최종 황칠 복합 발효물은 γ-PGA, peptides, 고농도 GABA를 함유한 발효소재로써 다양한 식품의 원료 활용이 기대된다.

황칠, 닥나무, 꾸지뽕 혼합 추출물의 항당뇨 효과 (Antidiabetic Effects of Mixed Extract from Dendropanax morbiferus, Broussonetia kazinoki, and Cudrania tricuspidata)

  • Dengropanax morfiferus (D), Broussonitia kazinoki (B), and Cudriania tricuspidata (E), a widely cultivated species in South Korea, has been used as traditional medicine to treat numerous diseases. In this study, we evaluated the antidiabetic effects in a various signaling mechanisms using mixed extract and major component contents were analyzed by HPLC in the combined extracts from Dengropanax morfiferus, Broussonitia kazinoki, and Cudriania tricuspidata (DBCE). DBCE inhibited ${\alpha}$-glucosidase and ${\alpha}$-amylase activation and showed potent antioxidant effects, which are evaluated using DPPH, ABTS, and SOD assay. Cytokines, which are released by inflammatory cells in pancreatic islets, are involved in the pathogenesis of type 1 diabetes mellitus. DBCE showed the protective effects in RINm5F cells against cytokines-induced damage by suppressing inducible nitric oxide (NO) synthase and COX-2 expression and NO production. Insulin resistance is the primary characteristic of type 2 diabetes. Therefore, the regulatory effect of DBCE on glucose uptake and production are investigated in insulin-responsive human HepG2 cells. DBCE stimulated glucose uptake, prevented Glut2 and phosphor-IRS1 downregulation induced by high glucose (HG, 30 mM). Moreover, DBCE pretreatment diminished glucose levels, PEPCK and G6Pase overexpression provoked by HG. These findings suggest that DBCE might be used for diabetes treatment through alpha-glucosidase or alpha-amylase activity regulation, pancreatic beta cell protection, hepatic glucose sensitivity improvement. Cytokines, which are released by inflammatory cells' infiltrations around the pancreatic islets, are involved in the pathogenesis of type 1 diabetes mellitus.

Comparison of Biological Activities of Dendropanax morbiferus by Different Cultivation Areas in Korea

  • Dendropanax morbifera H.Lev is an evergreen tree that lives in subtropical climates. About 75 species of D. morbifera live in East Asia, but only one species live in Korea(Korean dendropanax). D. morbifera belonging to Aralicacae family 98% of D. morbifera are distributed in Jeollanam-do in the Korea and the grow wild in Gyeongsangnam-do and Jeju Island. The harvest time and usage of D. morbifera were recorded in traditional medicinal books. The roots and stems of D. morbifera had been used for traditional medicine to treat migraine, menstrual irregularity and skin disease. And D. morbifera leaves are contain flavonoids and polyacetylene compounds. In this study, we were investigated the physiological activity of D. morbifera by different areas collected at the same time, and compared to characteristics of plants. D. morbifera collected from Jeollanam-do (Goheung-gun), Gyeongsangnam-do (Namhae-gun) and Jeju Island, and dried at $50^{\circ}C$ for three days. We used dried D. morbifera powder for antioxidant tests. Each sample was extracted with hot water under the same conditions. The contents of total polyphenols and total flavonoids from D. morbifera were identified. Also, we performed to DPPH radical scavenging activity, ABTS cation radical scavenging activity and Superoxide anion scavenging activity efficacy for antioxidant activity determination. The contents of total polyphenols and total flavonoids in hot water extract of D. morbifera harvested from Gyeongsangnam-do and Jeollanam-do were higher than Jeju. However, D. morbifera harvested from Gyeongsangnam-do and Jeollanam-do showed no significant difference those content of total polyphenols and total flavonoids. And the antioxidant capacity was showed the similar patterns in antioxidant activity.

황칠나무 추출물의 고초균 발효물로 제조된 가쓰오부시 단백가수분해물의 Lactobacillus plantarum 발효를 통한 고농도 GABA 생산 (Production of highly enriched GABA through Lactobacillus plantarum fermentation of katsuobushi protein hydrolyzate made from Dendropanax morbiferus extract fermented by Bacillus subtilis )

  • 참치액 소스를 가공할 때 가쓰오부시(가다랑어 훈연 숙성) 열수 추출물을 주원료로 사용하며, 이때 부산물로 생산되는 가쓰오부시 단백질을 이용하여 가수 분해산물인 peptide 강화소재를 제조하며 이를 젖산발효를 통한 기능성 물질 GABA가 증진된 기능성 발효소재를 개발하였다. 황칠나무 추출물 25%에 glucose 2%, skim milk 1%를 첨가한 제한배지에서 1차 고초균 발효 1일 동안 B. subtilis HA의 생균수는 8.83 log CFU/mL로 크게 증가하였고, pH 7.53과 산도 0.06%를 나타내었다. 고초균 발효물의 protease 활성은 발효 전 1.55 unit/mL에서 발효 후 102.22 unit/mL로 크게 증가하였고, 이에 가쓰오부시 부산물을 10% 첨가하여 60℃에서 3시간 동안 단백질 분해한 결과 tyrosine 함량이 분해 전 72.94 mg%에서 분해 3시간 후 156.85 mg%로 2배 이상 증가하였다. 고초균 발효물을 이용하여 가수분해시킨 가쓰오부시 단백질 분해물에 추가적으로 MSG 10%, glucose 3%, yeast extract 0.5%를 첨가하여 정치배양으로 젖산균 발효를 수행하였다. L. plantarum KS2020의 생균수는 0일 차 7.65 log CFU/mL에서 발효 7일 차 9.33 log CFU/mL까지 크게 증가하였고, B. subtilis HA의 생균수는 0일 차 5.98 log CFU/mL에서 1일 차부터 생균수가 검출되지 않았다. 젖산균 발효물의 pH 및 산도의 변화는 0일 차 pH 7.14, 산도 0.19%에서 1일 차에 pH는 5.77로 감소하고 산도는 1.92%로 증가한 후 발효 3일 차에 산도는 0% 및 pH 8.00 이상을 나타내었다. 젖산발효 1일 차부터 GABA의 전환이 보이면서 발효 3일 차에 고농도 GABA 생성을 나타내었다. GABA 및 glutamic acid의 함량을 HPLC로 분석한 결과, GABA의 함량은 3,139.58 mg/100 g으로 매우 높은 값을 나타냈으며, glutamic acid의 함량은 83.44 mg/100 g이었다. 결과적으로 황칠나무 추출물을 이용한 고초균 발효물을 효소원으로 이용하여 가쓰오부시 부산물의 단백질을 효과적으로 고온에서 단기간에 분해하여 peptide를 생성시켰으며, 연속적으로 젖산발효를 통해 약 3.1%의 GABA가 생성되었다. 최종 가쓰오부시 단백질의 가수분해물을 이용한 젖산균 발효물은 산도가 0%이며 풍미, probiotics, peptides, 고농도 GABA를 함유한 복합 기능성 발효소재로써 소스 등 다양한 식품의 건강소재로 활용이 기대된다.

IMMUNIES, a unique polyherbal extract, exhibits antiproliferative activity and improves tumor-bearing canine patients: a pilot study

  • Dog owners seek treatment when their pets develop cancer. IMMUNIES is traditional herbal medicine-based figment made of 10 natural herbs, designed to maintain host immune function. The major component of IMMUNIES is Dendropanax morbiferus. This clinical pilot study monitored the toxicity and efficacy of IMMUNIES. Four senile dogs with spontaneously occurring mammary and liver cancers were enrolled in this study and treated orally daily for 3 months, and their blood/urine biochemical profiles were examined each month. IMMUNIES was well tolerated during the treatment period. Blood urea nitrogen, creatinine, alanine aminotransferase, alkaline phosphatase, and C-reactive protein levels decreased in all four dogs, whereas red blood cells and hematocrit were within the normal range. IMMUNIES also changed the expression of several molecular targets in the anticancer pathway, such as pro-NAG-1, p53, and cyclin D1. Although the tumors did not completely respond to IMMUNIES, the biochemical profiles and clinical examination showed a stabilized cancer status for 3 months. Thus, IMMUNIES was found to be safe and well-tolerated in the dosage range tested and exhibited cancer antiproliferative activity in canine cancer. Future studies should address other potential benefits of IMMUNIES, including correlative assessments of immune function, quality of life, and owner satisfaction.

유방암 세포에서 황칠나무 잎 추출물의 MAPK 경로를 통한 apoptosis 유도 (Apoptosis Induction by Dendropanax morbiferus Leaves Extract in Human Breast Cancer Cells SK-BR-3 through MAPK Pathway)

  • 황칠나무 잎 추출물(DPL)은 염증, 당뇨병, 암 등 다양한 질병에 예로부터 약으로 사용되어 왔다. 특히 여러 종류의 암세포에 세포사멸 효과가 있는 것으로 밝혀졌지만 유방암 세포 SK-BR-3에 대한 세포사멸 효과는 아직 밝혀지지 않았다. 따라서 본 연구에서는 in vitro 실험을 통해 DPL이 유방암 세포 SK-BR-3에서의 증식 억제와 apoptosis를 통한 세포사멸 효과를 확인하였다. DPL이 세포 생존율에 미치는 영향을 조사하기 위해 MTT 분석을 수행했고 세포 생존율이 농도의존적으로 감소하는 것을 확인했다. DAPI 염색은 DPL에 의해 발생한 세포 형태학적 변화와 apoptotic body의 증가에 미치는 영향을 조사하기 위해 수행되었다. 이를 보완하기 위해 annexin V/PI 염색 후 유세포 분석을 통해 세포 apoptosis 비율의 증가를 확인했다. Apoptosis 관련 단백질들을 확인하기 위해 Western blot을 수행하였으며, cleaved-PARP과 pro-apoptosis 단백질인 Bax의 증가와 anti-apoptosis 단백질인 Bcl-2의 감소를 확인하였다. MAPK 경로 단백질 p-ERK1/2, p-JNK 및 p-p38의 발현 수준의 변화도 확인하였으며 p-p38의 유의미한 증가가 관찰되었다. 본 연구 결과에 따르면 DPL은 유방암 세포 SK-BR-3에서 p-p38 MAPK 경로 조절을 통해 세포사멸 효과를 유도하는 것으로 사료된다.
