Suppression of the Development of Delayed-Type Hypersnsitivity to Sheep Erythrocytes by Serum from BCG-Infected Mice (BCG 감염(感染)마우스 혈청(血淸)에 의(依)한 지연성(遲延性) 과민반응(過敏反應) 억제(抑制))

  • The immunosuppressive effect of serum from mice heavily infected with BCG(BCG-serum) was evaluated. The delayed-type hypersensitivity reaction to sheep erythrocytes was depressed when BCG-serum was administered both systemically and locally at the same time of challenge. This study shows that serum of animal infected with mycobacteria contains a factor responsible for immunosuppression and factor may be important in understanding the mechanisms of allergy in mycobacteria] infections.

The Effect of Honey Bee Venom Aqua-Acupuncture by its Production from Different Regions on Decreased Immune Response (산지별(産地別) 봉독액약침자극(峰毒液藥鍼刺戟)이 면역기능저하(免疫機能低下)에 미치는 영향(影響))

  • Objectives : This study was purposed to investigate the effect of honey bee venom aqua-acupuncture on decreased immune response by its production from Korea, North America and China. Methods : ICR-mouse was exposed with four minute sublethal 60Co- $\gamma$ -ray irradiation onto the body and got a series of each country's 0.1 ml honey bee venom aqua-acupuncture to Choksamni(ST36) every other day for three times. Thereafter the numbers of WBC, delayed type hypersensitivity, hemagglutinin titers, hemolysin titers, quantitation of T-cell and B-cell, lymphocyte transformation, natural killer cell activity, interleukin-2 productivity and interferon productivity were measured. Results : The numbers of WBC, delayed type hypersensitivity, hemagglutinin titers, hemolysin titers, quantitation of T-cell and B-cell and lymphocyte transformation were increased respectively with statistical significance in Korean, North American and Chinese honey bee venom aqua-acupuncture groups as compared with the control group. The natural killer cell activity was increased with statistical significance in Korean honey bee venom aqua-acupuncture group, and Interleukin-2 productivity and interferon productivity were not shown any statistical significance in all experimental groups as compared with the control group. Conclusions : According to the results, honey bee venom aqua-acupuncture has significant effect on decreased immune response, and Korean bee venom can be substituted for North American and Chinese one.

  • Background: Disialoganglioside GD2 is a tumor-associated antigen that is overexpressed on tumor cells of neuroectodermal origin, such as melanoma, small cell lung carcinoma and neuroblastoma. Immunity against GD2 has anti-tumor activities, but GD2 is poorly immunogenic. Anti-idiotypic antibodies that mimic GD2 may induce more effective immune responses than GD2 antigen itself, because they are protein antigens and are known to be able to break immune tolerance. In our previous study, we produced anti-idiotypic antibodies mimicking GD2 (3A4 and 3H9), which induced humoral immunity. However, cellular immunity is essential to eradicate tumor cells in vivo as well as humoral immunity. In the present study, we investigated whether these anti-idiotypic antibodies 3A4 and 3H9 could induce cellular immunes responses. Methods: BALB/C mice were immunized with anti-idiotypic antibody 3A4 or 3H9, or normal mouse IgG as a negative control. Lymphoproliferative responses, cytokine production responses, and delayed-type hypersensitivity reactions were measured in mice immunized with the anti-idiotypic antibodies. Results: Both the anti-idiotypic antibody 3A4 and 3H9 induced GD2-specific lymphoproliferative responses and $IFN-{\gamma}$ production of lymph node lymphocytes in BALB/C mice. Only anti-idiotypic antibody 3H9 induced significant GD2-specific delayed-type hypersensitivity in the mice. Conclusion: These results show that anti-idiotypic antibodies 3A4 and 3H9 have the potentiality of inducing GD2-specific cellular immune responses that cannot be induced by the native antigen GD2 itself.

  • The effects of intravenous administration of of C. parvum on immune cells in mice were investigated. Significant delayed type hypersensitivity parvum was given intravenously before antigen sensitization. The depressed effect of C. parvum was completely abolished by treatment of cyclophosphamide. However, delayed type hypersensitivity was not depressed when C. parvum given after antigen. This depression is considered to be attributable to the generation of suppressor cells which were either suppressor T cell or C. parvum activated macrophage, because C. parvum inhibited the proliferation of S. typhimurium in vivo and also induced splenomegaly and hepatomegaly, known as manifestation of macrophage activation. Serum antibody titers were not significantly affected by C. parvum regardless of time of C. parvum administration.

  • Experimental studies were done to know effect of insensible water loss of Hwangkitang(HKT) in mice. After administration of extract of HKT to the mice, changes of body temperature, body weight, serum Sodium and chloride ion was measured. And sametime, serum antibody to SRBC, delayed hypersensitivity and phagocytic activity of polymorphonuclear leukocytes(PMN) was measured to know immunologic effect. The results obtained as follows; 1. After exposure to $40^{\circ}C$ environment, increment of body temperature in treated group was slower than control group during first 40 minute, and no difference after 40 minute. 2. After exposured to $40^{\circ}C$ environment, decrement of body weight in treated group except administration group of 1.0g/kg extract of HKT was slower than control group during first 40 minute, and no difference after 40 minute. 3. After exposured to $40^{\circ}C$ environment, decrement of body weight in administration group of 4.0g/kg extract of HKT to mice was slower than control group during first 20 minute(p<0.005). 4. After exposure to $40^{\circ}C$ environment there was no stastical difference between treated group and control group in changes of serum sodium and chloride ion. 5. There was no statistical differences between treated group and control group in changes of serum antibody, delayed hypersensitivity and phagocytic activity of PMN.

  • Even though appropriate immune response is necessary for the survival of the individual, excessive or insufficient immune response might cause autoimmune or allergic disease respectively. So the immune response must be controlled to the degree that is beneficial for the well being of the individual. This study was undertaken to know the effects of Junsibaekchulsan(JB) on the immune system od the mouse. For the evalulation of the cell-mediated immunity(CMI), delayed-type hypersensitivity against dinitrofluorobenzene(DNFB) were measured, and humoral immunity, hemagglutinin and hemolysin titers against SRBCs(sheep red blood cells) were measured, and rosette formation of spleen cells with SRBCs were measured. For the evaluation of innate immunity, phagocytic activity of macrophages, natural killer cell activity, and reactive nitrogen and oxygen intermediates were measured. The results are as follows: 1. The administration of JB depressed the antibody formation (hemagglutinin and hemolysin) against SRBCs. 2. The administration of JB did not affect the delayed-type hypersensitivity against DNFB. 3. The administration of JB did not affect the cytotoxic activity of natural killer cells. 4. The administration of JB increased the phagocytic activity of macrophages. 5. The administration of JB increased the rosette formating cells of the spleen cells. 6. The exposure of JB induced the secretion of reactive nitrogen intermediates but administration of JB deperssed the production of reactive oxygen intermediates. Administration of JB selectively depressed the humoral immune response without affecting CMI and innate immunity. These results of JB on the immune system might be useful for the treatment of such.

Effects of Type and Amount of Dietary Fat on the Immune Status of BALB/c Mouse (식이 지방의 종류 및 함량이 마우스의 면역기능에 미치는 영향)

  • This study was performed to investigate effects of dietary fat content and degree of saturation on the function of the immune system. Sixty male BALB/c mice average-weighing 17g were divided into three dietary groups: 5% safflower oil group, 20% safflower oil group, 19.3% beef tallow & 0.7% safflower oil group. Food intake and body weight were measured every day. At 4th, 7th, 10th week after dietary treatment, organ weight measurements, delayed-type hypersensitivity test, plaque forming cell test, agglutination test, differential white cell count and histological examination of spleen were performed. Results are follows; 1) Body weight, food intake and calorie intake were not different in the three dietary groups during the experimental period($\alpha$=0.05). 2) Liver weight was significantly higher in 5% safflower oil group($\alpha$=0.05). Spleen index was slightly higher in mice fed 5% safflower oil and 19.3% beef tallow & 0.7% safflower oil. Thymus index in all mice was decreased by aging. 3) Delayed-type hypersensitivity of the mice fed 5% safflower oil and 19.3% beef tallow & 0.7% safflower oil was significantly higher than that of the mice fed 20% safflower oil. 4) The number of plaque forming cell was significantly reduced at 10th week compared to 7th week in all group($\alpha$=0.05). Although there was no difference in plaque forming cell among three groups at 10th week, 5% safflower oil group showed slightly higher plaque forming cell than 20% safflower oil group at 7th week. 5) At 4th week, agglutination test seems to be higher in 5% safflower oil group and 19.3% beef tallow & 0.7% safflower oil group compared to 20% safflower oil group. 6) Percentage of neutrophil and eosinophil was slightly reduced in 20% safflower oil group. 7) Spleen tissue was not affected by and dietary treatments. According to our results, the higher the fat content & unsaturation of the diet the lower the cell-mediated immunity of the mice. Humoral-immunity did not appear to be affected by the dietary manipulation. However humoral-immunity was decreased significantly by aging in all dietary groups.

Effects of Hyunggyeyungyotang and Kamihyunggyeyungyotang administration on the anti-inflammation, analgesia and anti-allergic reaction in mice (형개연교탕(荊芥連翹湯)과 가미형개연교탕(加味荊芥連翹湯)이 소염(消炎).진통(鎭通) 및 항(抗)알레르기에 미치는 영향(影響))

  • Hyunggyeyungyotang has been used for treatment of sinusitis and otitis media in oriental medicine since ancient times. It is reported that Hyunggyeyungyotang has good effects on inflammatory and allergic diseases of otorhinolaryngology in clinical medicine. Kamihyunggyeyungyotang was made by adding several herbs to Hyunggyeyungyotang which has such good effects. To investigate the effects of Hyunggyeyungyotang and Kamihyunggyeyungyotang on inflammatory, algesic and allergic diseases, the author examined the analgesic effect by acetic acid reaction, studied the anti-inflammatory effect through the experiments of the protein thermo-denaturation and circumscribed edema. Besides researched the anti-allergic effect through the vascular permeability response to Chemical Mediator and the delayed type hypersensitivity response to Picryl Chloride. The obtained results were as follows ; 1. In the analgesic effect of Hyunggyeyungyotang and Kamihyunggyeyungyotang extract by acetic acid method, both of the sample groups showed the analgesia, but didn't show useful effect. 2. In the anti-inflammatory effect on the protein thermo-denaturation, the sample groups revealed the inhibitory effect in proportion to concentration as compared with the control group. 3. In the inhibitory action on circumscribed edema induced by Caraggeenin, both of Hyunggyeyungyotang and Kamihyunggyeyungyotang administration showed the significant effect after 4 hours in comparison to the control group. 4. In the delayed type hypersensitivity response to Picryl Chloride, both of the sample groups revealed the significant effects. 5. Both of the sample groups decreased the vascular permeability induced by Histamine in comparison with the control group, but the significancy was admitted in only Hyunggyeyungyotang administration. According to above results, Hyunggyeyungyotang and Kamihyunggyeyungyotang are considered to be used for treament of the inflammatory diseases including sinusitis.

Contact Sensitivity to Dinitrochlorobenzene as a Marker Trait in the Indirect Selection for Body Mange and Coccidiosis Resistance in Broiler Rabbits

  • To determine the effects of genetic and environmental influences on cell mediated immune (CMI) responses in broiler rabbits, contact sensitivity to 2,4-Dinitrochlorobenzene (DNCB) was assessed in three temperate broiler breeds of rabbits, namely Soviet Chinchilla, New Zealand White and Grey Giant. The feasibility of using the contact sensitivity to DNCB as a marker trait in selection for disease resistance was examined. There were highly significant differences between breeds (p<0.01) in initial skin thickness and contact sensitivities to DNCB at 24, 48 and 72 hours. Initial skin thickness was greatest in the Soviet Chinchilla breed (mean 2.2484 mm), and was significantly greater (p<0.01) in males (2.4963 mm) than in females (1.7846 mm) (p<0.01). Highest contact sensitivity to DNCB was in the New Zealand White breed with mean increase in skin thickness of 1.1884, 0.9072 and 0.5879 mm at 24, 48 and 72 hours post challenge respectively. Delayed type hypersensitivity (DTH) reaction to DNCB at 24 hours post challenge had a highly significant association (p<0.01) with the incidence of body mange in rabbits. The results indicated a lowered contact sensitivity to DNCB at 24 hours post challenge was associated significantly (p<0.01) with an increase in incidence and severity of body mange, suggesting its potential value as a marker. The correlation s among contact sensitivities at 24, 48 and 72 hours were positive and highly significant (p<0.01); correlations between initial skin thickness and contact sensitivities were negative and highly significant (p<0.01). Another notable significant correlation was between body weight and delayed type hypersensitivity at 24 hours indicating that an enhanced CMI might be associated with better growth rate and general wellbeing.

An Experimental study on the effects of Insambakhab-tang on the Anti-allergic effect and Pulmonary Function of $O_3$ intoxicated Mice (인삼백합탕이 알레르기와 폐손상에 미치는 영향)

  • Experimental studies were done to research the Clinical effects of Insambakhab-tang on the Anti-allergic effect and pulmonary function of O₃ intoxicated Mice. Anti-allergic effect experiment consisted of vascular permeability responses to intradermal histamine and serotonin, 48hrs homologous passive cutaneous anaphylaxis provoked by the IgE-like antibody against egg white albumin, and delayed type hypersensitivity responses to Picryl Chloride and SRBC. Pulmonary function of O₃ intoxicated Mice experimental consisted of pulmonary thromboembolism (Sodium Arachidonate-induced and ADP-induced), lung TBA value, and serum Na/sup +/, K/sup +/, Cl/sup +/ level. The results obtained as follows; 1. In the effects of Insambakhab-tang on the pulmonary thromboembolism by Sodium Arachidonic acid and ADP, Insambakhab-tang group revealed significant effect. 2. In the effects of Insambakhab-tang on the vascular permeability responses to intradermal histamine, Insambakhab-tang group revealed significant effect. 3. In the effects of Insambakhab-tang on the vascular permeability responses to intradermal serotonine, Insambakhab-tang group revealed significant effect. 4. In the 48hrs homologous passive cutaneous anaphylaxis provoked by the IgE-like antibody against egg white albumin, Insambakhab-tang group revealed significant effect. 5. In the delayed type hypersensitivity responses to Picryl Chloride, Insambakhab-tang group revealed significant effect. 6. Insambakhab-tang group revealed significant effect on decrease of the lung TBA value of lung. 7. In the effects of Insambakhab-tang on Serum Na/sup +/, K/sup +/ Level in O₃-intoxicated Mice. Insambakhab-tang group revealed none significant effect, but In the effects of Insambakhab-tang on Serum Cl/sup +/ Level in O₃-intoxicated Mice, Insambakhab-tang group revealed significant effect.
