Enzyme Production Related to Alcohol Metabolism from Thermophilic Fungus Thermoascus aurantiacus (호열성 사상균 Thermoascus aurantiacus의 알코올분해대사 관련 효소학적 특성)

  • Thermophillic fungus Thermoascus aurantiacus showed excellent growth and produced high amount of alcohol oxidase and catalase in a pectin medium. Besides, the strain produced enzymes which related with pectin or alcohol decomposition. We detected extracellular pectin esterase (EC 3.1.1.11) activity and, both intracellular and extracellular pectinase (EC 4.2.2.10) activity, as pectinolytic enzymes produced by T. aurantiacus. The production of methanol decomposition enzymes, such as alcohol oxidase (AOD, EC 1.1.3.13), alcohol dehydrogenase (ADH, EC 1.1.1.1), formaldehyde dehydrogenase (FADH, EC 1.2.1.1) and formate dehydrogenase (FDH, EC 1.2.1.2) follows by pectin esterase reaction which is converted to methanol. We concluded that T. aurantiacus has pectinolytic and alcohol - oxidative enzymological mechanism which produced carbon dioxide as a final material, started from pectin.

Regulation of NAD+- Specific Isocitrate Dehydrogenase from Pythium ultimum

  • The $NAD^+$-specific activity of a dual coenzyme-specific isocitrate dehydrogenase (IDH; EC 1.1.1.41) from the primitive fungus Pythium ultimum was investigated to elucidate the regulatory factors that may influence the intracellular distribution of carbon and the availability of intermediates, e.g. citrate, for fatty acid synthesis. Inhibition of $NAD^+$-IDH activity by diphospho- and triphosphonucleotides (ATP, ADP, and GTP) reflected the sensitivity of this enzyme to cellular energy charge even though monophosphonucleotides (AMP and GMP) had little effect on activity. NADPH, but not NADH, substantially inhibited $NAD^+$-IDH activity, showing noncompetitive inhibition with isocitrate. Oxalacetate and ${\alpha}$-ketoglutarate showed competitive inhibition with isocitrate, while citrate and cis-aconitate showed mixed-noncompetitive inhibition with isocitrate. Inhibition by these substances ranged from 29 to 46% at 10 mM. The inhibitory effect of oxalacetate was increased synergistically by glyoxylate, which alone caused 31% uncompetitive inhibition at 10 mM, and a mixture of the two substances at 1 mM each showed 98% inhibition of $NAD^+$-IDH activity. The regulation of $NAD^+$-IDH in Pythium ultimum seems to be a complex process involving mitochondrial metabolites. The addition of glyoxylate (3 mM) and oxalacetate (3 mM) to the culture medium resulted in the production of 49% more lipid by P. ultimum.

Inhibition of glutamate dehydrogenase and insulin secretion by KHG26377 does not involve ADP-ribosylation by SIRT4 or deacetylation by SIRT3

  • We investigated the mechanisms involved in KHG26377 regulation of glutamate dehydrogenase (GDH) activity, focusing on the roles of SIRT4 and SIRT3. Intraperitoneal injection of mice with KHG26377 reduced GDH activity with concomitant repression of glucose-induced insulin secretion. Consistent with their known functions, SIRT4 ribosylated GDH and reduced its activity, and SIRT3 deacetylated GDH, increasing its activity. However, KHG26377 did not affect SIRT4-mediated ADP-ribosylation/inhibition or SIRT3-mediated deacetylation/activation of GDH. KHG26377 had no effect on SIRT4 protein levels, and did not alter total GDH, acetylated GDH, or SIRT3 protein levels in pancreatic mitochondrial lysates. These results suggest that the mechanism by which KHG26377 inhibits GDH activity and insulin secretion does not involve ADP-ribosylation of GDH by SIRT4 or deacetylation of GDH by SIRT3.

Changes of Enzyme Activity in Nitrogen Metabolism on Induced Association of N. muscorum with Cultured Tobacco Cells (N. muscorum과 담배 배양세포의 공생유도에 따른 질소대사에 관여하는 효소활성의 변화)

  • Investigations on the liability of nitrogen usuage by Nostoc muscorum that has nitrogen fixing ability, and cultured tobacco cells as they were associately cultured on nitrogen-free media and effects of polyamine on the associated culture condition were carried out. In addition, measurement on the activity of nitrate reductase, glutamine synthetase, glutamate dehydrogenase and glutamate synthase that take part in the metabolic pathway of nitrogen fixation product were performed. Among enzymes participating in the metabolic pathway of nitrogen fixation products, the activity of nitrogen reductase stimulated five times in associated culture, and that of glutamine synthetase of N. muscorum increased two times after heterocyst differentiated. Activity of glutamate dehydrogenase increased markedly when cultured tobacco cells were solely incubated on nitrogen-free media, but inhibited when cultured associately. And, glutamate synthase was showed the highest activity in 0.1 mM of spermine treated group.

Effects of Chunggansan on Detoxication of Alcohol by Activity of Enzyme in Rats (청간산(淸肝散)이 흰쥐의 알코올 대사(代謝) 효소(酵素) 활성도(活性度)에 미치는 영향(影響))

  • Chunggansan was tested for the effects on detoxication mechanism of alcohol. Chunggansan was treated firstly into samples, and then ethanol intoxicated animal models were set with them. The administration of Chunggansan to the rats increased proportionally in alcohol dehydrogenase activities in liver in relation to the level of concentration and days of treatment. Especially, the alcohol dehydrogenase was the most active when the concentration of extract was 200mg/kg and it was 7th day. The enzyme activities of alcohol dehydrogenase and aldehyde dehydrogenase in liver highly increased in Chunggansan pre-medicating group compared to that of ethanol treated group. Also, the blood ethanol concentration in rats was considerably decreased. In conclusion, Chunggansan recovers the damage of liver due to acute alcohol intoxication by the increased enzyme activities of alcohol dehydrogenase and aldehyde dehydrogenase.

  • Palmitoyl CoA was found to inhibit corn embryo axis glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase, which were also inhibites by polyamines. However, reversal of inhibition of both enzymes by palmitoyl CoA was made by spermine. Activity of corn embryo axis protein kinase was found to increase steadily after germination. Activation and inhibition of protein kinase were made by MgCl$_2$and all polymines, respectively. Suc results suggest that fatty acid biosynthesis and lypolysis could be regulated to some extent by polyamines in corn embryo axis.

  • A mannitol dehydrogenase (StMDH) gene was cloned from Salmonella typhimurium LT2 (KCTC 2421) and overexpressed in Escherichia coli. It has a 1,467 bp open reading frame encoding 488 amino acids with deduced molecular mass of 54 kDa, which shares approximately 36% of amino acid identity with known long-chain dehydrogenase/reductatse (LDR) family enzymes. The recombinant StMDH showed the highest activity at $30^{\circ}C$, and pH 5.0 and 10.0 for D-fructose reduction and D-mannitol oxidation, respectively. On the contrary, it has no activity on glucose, galactose, xylose, and arabinose. StMDH can catalyze the oxidative/reductive reactions between D-fructose and D-mannitol only in the presence of $NAD^+$/NADH as coenzymes. These results indicate that StMDH is a typical $NAD^+$/NADH-dependent mannitol dehydrogenase (E.C. 1.1.1.67).

Co-expression of Gamma-Aminobutyrate Aminotransferase and Succinic Semialdehyde Dehydrogenase Genes for the Enzymatic Analysis of Gamma-Aminobutyric Acid in Escherichia Coli

  • Gamma-aminobutyric acid (GABA) aminotransferase (gabT) and succinic semialdehyde dehydrogenase (gabD) genes from Pseudomonas fluorescens KCCM 12537 were cloned into a single pETDuet-1 vector and co-expressed in Escherichia coli BL21(DE3) simultaneously. The mixture of both enzymes, called GABase, is the key enzyme for the enzymatic analysis of GABA. The molecular mass of the GABA aminotransferase and succinic semialdehyde dehydrogenase were determined to be 52.8 and 46.7 kDa following computations performed with the pI/Mw program, respectively. The GABase activity between pH 6.0 and 9.0 for 24 h at $4^{\circ}C$ remained over 75%, but under pH 6.0 decreased rapidly. The GABase activity between 25 and $35^{\circ}C$ by the treatment at pH 8.6 for 30 min remained over 80%, but over $35^{\circ}C$ decreased rapidly. When the activity against GABA was defined as 100%, the purified GABase activity against 5-aminovaleric acid having a similar structure to GABA showed 47.7% and GABase activity against ${\beta}$-alanine, ${\varepsilon}$-amino-n-caproic acid, $_L$-ornithine, $_L$-lysine, and $_L$-aspartic acid showed between 0.3 to 2.3%. The GABA content was analyzed with this co-expressed GABase, compared with the other GABase which was available commercially. As a result, the content of GABA extracted from brown rice, dark brown rice, and black rice were $26.4{\pm}3.5$, $40.5{\pm}4.7$ and $94.7{\pm}9.3{\mu}g/g$, which were similar data of other GABase in the error ranges.

  • The physiological activities of cultivated Korean blue mussel (Mytilus edulis) and New Zealand green-lipped mussel (Perna canaliculus) were analyzed and compared. Both hot water extracts of blue mussel flesh (BMF) and green-lipped mussel flesh (GMF) showed increased activities of alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). BMF showed increased ADH and slightly decreased ALDH activities compared to GMF. 1,1-Diphenyl-2-picrylhydrazyl radical scavenging activity of BMF was higher than that of GMF at the same concentration. BMF and GMF showed similar inhibitory activity against angiotensin converting enzyme at a concentration of 30 mg/mL. These results suggest that cultivated Korean blue mussel has similar physiological activity with New Zealand green-lipped mussel.

Effect of Consecutive Application of Organic Matter on Soil Chemical Properties and Enzyme Activity in Potato Cultivation Soil (유기물 연용이 감자재배 비화산회토양의 화학성과 효소활성에 미치는 영향)

  • This study was carried out to evaluate effect of consecutive application of organic matter on soil chemical properties and dehydrogenase, acid phosphatase activity in non-volcanic ash soil during three cropping season. Organic matter mixture and organic fertilizer (MOF, $2,000kg\;10a^{-1}$), food waste compost (FWC, $2,000kg\;10a^{-1}$), and pig manure compost (PMC, 2,000, 4,000, and $6,000kg\;10a^{-1}$) were applied for each cropping season. Soil pH values were increased after three cropping season in all treatment. In the soils of the increased application of PMC, soil pH, total-nitrogen, available phosphate, exchangeable cations (K, Ca, and Mg), and heavy metal (Zn and Cu) contents were increased. In addition, Soil dehydrogenase activity was significantly increased in proportions to PMC application rate and cropping season during potato cultivation period. The activity was two times higher in PMC ($4,000kg\;10a^{-1}$) than control after the third cropping season. Soil dehydrogenase activity was in order of PMC>FWC>NPK+PMC>MOF. Acid phosphatase activity was higher in PMC ($6,000kg\;10a^{-1}$) than other treatment. Soil Zn content and dehydrogenase activity showed linearly correlation, which were MOF ($R^2$=0.427), FWC ($R^2$=0.427) and PMC ($R^2$=0.411, p<0.01), respectively. This study demonstrated that soil chemical properties and enzyme activity could be affected greatly by consecutive application of different organic matter in the potato cultivation field.
