Inhibitory activity of Lactobiocin on the skin inflammation and acnes (Lactobiocin의 피부 염증 및 여드름 저해효과에 관한 연구)

  • The purpose of this study was to evaluate bacteriocin activity against human flora. Lactobiocin, a bacteriocin produced by Lactococcus sp. HY 449, inhibited the growth of Starphylococcus epidermidis, Starphylococcus aureus, Streptoccoccus pyogenes and Propionibacterium acnes. When crude bacteriocin was added to indicator cells during logarithmic growth, the optical density(O.D 650nm) of cells without bacteriocin increase after 5h of incubation. Whereas in the presence of bacteriocin, the O.D of cell suspensions decreased. The similar patterns were observed for absorbance readings at 280 nm and 260 nm. The release of cellular components when cell were treated with Lactobiocin suggests some degree of membrane damage or cell lysis. Scanning electron microscopy of cells following treatments with Lactobiocin in PBS buffer revealed disruptures of cell morphology. These results indicate that bacteriocin appears to cause cell lysis of tested strains. In cytotoxicity on human fibroblast, LD$\_$50/ of Lactobiocin was ca. 50 mg/ml and no change was observed cell proliferation at the same concentration. Any irritation and allergic reaction did not observed when evaluated by human patch test for Lactobiocin.

  • Tropical tasar silkworm, Antheraea mylitta (Lepidoptera: Saturniidae) is a polyphagous silk producing forest silkworm of commercial importance in India. Forest dependent people rear its larvae on different forestry host plants twice or thrice in a year for small household income. Larvae of A. mylitta feeds on many forest tree species, but always show a great degree of selectivity as a function of its behavioural responses to physical structure and chemical features of the host plants. Cocoon crop of A. mylitta is influenced by heterogeneity of tasar food plants and climatic conditions of the habitat. The role of host plants, temperature, humidity, rainfall, photoperiod and climatic variables on the growth and development of insects have clearly been demonstrated. This article entails an in-depth analysis on ecological and nutritional aspects of A. mylitta, which may provide selective information to researcher and forest managers, who are particularly associated with livelihood improvement of the poor people in forested area through location specific forest insect industry.

The effect of Indole acetic acid on the accumulation of Cd2+ and growth of Cd2+ - treated Commelina communis L. (닭의장풀에 Cd2+ 처리시 Cd2+ 흡수와 생장에 미치는 indole acetic acid의 영향)

  • 3-weeks old Commelina was transferred to and grown in Hoagland solution (Control, $100\muM$ $Cd^{2+}$, $100\muM$ $Cd^{2+}$+ $100\muM$ IAA, $100\muM$ $Cd^{2+}$+ $100\muM$ IAA + 2 mM sucrose) for 3 weeks and then the effects of indole acetic acid (IAA) on the accumulation of $Cd^{2+}$ and growth of $Cd^{2+}$-treated Commelina were investigated. In the treatment of $Cd^{2+}$, $Cd^{2+}$ was uptaked to 1.74, and 51.36 ${\mu}g/g$ frwt. at the first week, but for three weeks, 0.51 and 34,53 ${\mu}g/g$ frwt, in leaf and stem respectively. When IAA was treated along with $Cd^{2+}$, $Cd^{2+}$ was uptaked to 0.18 and 8.63 ${\mu}g/g$ fiwt, at the first week, and for the incubation of 3 weeks, 0,51 and 45.0 ${\mu}g/g$ fiwt. in leaf and stem. In case of $Cd^{2+}$+IAA+sucrose, $Cd^{2+}$ was uptaked to 1.45 and 18.33 ${\mu}g/g$ frwt. at the first week, but for 3 weeks, 0,51 and 25.45 ${\mu}g/g$ fiwt. in leaf and stem. Likewise $Cd^{2+}$ uptake, the growth was also affected by $Cd^{2+}$ and IAA. During the incubation of 3 weeks, $Cd^{2+}$ reduced the stem growth about 8% in all weeks, but the treatment of IAA recovered the inhibition of stem growth caused by $Cd^{2+}$ to the degree of the control Therefore, it could be concluded that IAA altered the pattern of $Cd^{2+}$ uptake and the growth which were supposed to change $Cd^{2+}$ toxicity.

  • A polymerized organic-inorganic complexation route is introduced for the synthesis of oxide ceramic powders. Polyvinyl alcohol was used as the organic carrier for precursor ceramic gel. Porous and soft powders, which have a high specific surface area, were obtained after calcinating the aerated precursors. The PVA content and its degree of polymerization had a significant influence on the homogeneity of the final powder. In particular, attrition milling process with the porous powder resulted in ultra-fine particles. In the case of the preparation of cordierite powder, nano-size powder, which has a high specific surface area of 181 ㎡/g, was obtained by the milling process. The complexation route was also applied to the synthesis of unstable phase in room temperature like beta-cristobalite, high temperature form of silica.

  • Utilization of yeast extract and formation of byproduct metabolite were investigated for hyperthermophilic archaeon Sulfolobus solfataricus (DSH 1617). In both batch and fed-batch cultivations of S. solfataricus, maximal cell density, {{{{ { NH}`_{4 } ^{ +} }}} ion production and pH change were highly dependent on the ratio of yeast extract to glucose in the medium. Variation of {{{{ { NH}`_{4 } ^{ +} }}}} ion level was identified as a major cause of pH change during cultivation, and acidification of culture broth was attributed to consumption of {{{{ { NH}`_{4 } ^{ +} }}}} ions rather than formation of acid byproducts. It was also observed that increase of {{{{ { NH}`_{4 } ^{ +} }}}} ion concentrations in the medium resulted in greater degree of growth inhibition.

Variation of Growth and Yield of Silage Corn According to Maturity (숙기별 사일리지용 옥수수의 생육 및 수량변화)

  • This study was carried out at Crop Experiment Station in 1993 and 1994. The objective of this study was finding out variation of growth and yield of corn according to maturity. Hybrids of corn used in this study were early maturing corn Comet80, Elite90, Royaldent IOOT, Royaldent T$\times$llO, P3525, P3394, medium maturing corn Royaldent 120T. and late maturing corn Jungbuok, P3144W. G4743. Stalk height, leaf number, ear weight, stover weight, and TDN yield of early maturing corn were linearly increased with prolonged maturity in 1993 growing season. Leaf number, and stover weight of early and late maturing corn were linearly increased with prolonged maturity in 1994 growing season. But ear weight of late manuring corn was not increased as much as that of early manuring corn with prolonged maturity. Increase of total DM and TDN yield of late maturing corn was due to stover weight increase compared with ear weight increase of early maturing corn. Leaf number and stover weight were highly correlated with silk (Growing Degree Days) GDD.

  • This study was conducted to determine effect of fruits removal on the CO2 exchange rates (CER) and growth of ginseng plant. Fruit of 2, 4 age plant removed at 7, May. The results of these investigations are as follows. 1. The net photosynthetic rates of the ginseng bearing fruits increased to a considerably greater degree than that of the ginseng without fruit in each ages. 2. The total dry matter per plant in bearing fruit (40.24g) had produced more dry matter than that of non-fruiting plant (38.13g) , but the root 4.y matter in fruiting plant (26.2g) had produced less dry matter than that of non-fruiting plant (27.1g) in 4 age. 3. The ginseng plant in bearing fruit did not influence the dry matter of stem and leaf. 4. The maximum RGR of root (17, June) was slower than that of fruit (4, June) .

A Study on the Texture and Taste of Kimchi in Various Saltings (절임정도에 따른 배추김치의 질감과 맛에 관한 연구)

  • The effects of salting degree of cabbage on the chemical, microbiological, textural changes and taste of Korean whole cabbage Kimchi fermented at $21^{\circ}C$ was studied. Low salted Kimchi (1.8% NaCl, soaking for 2 hr. in 20% brine) was showed higher total acdity than high salted Kimchi II(3.1% NaCl, Soaking For 8 hr in 20% brine) and III(2.9% NaCl, no soaking in 20% brine). The changes in growth of lactic acid bacteria was relevant to the salt content of whole cabbage Kimchi. Kimchi I was showed higher growth rate of Lactic acid bacteria and slower death rate constant of Lactic acid bacteria than that of Kimchi II and III. In palatability test, salty taste of Kimchi II, III was more salty than that of Kimchi I ($p{\leqq}0.01$) and total evaluation of Kimchi I, II was more palatable than Kimchi III ($p{\leqq}0.05$) at 3rd's day (optimum taste). Cutting force by Rheometer was well correlated with the salt content of Kimchi.

Substrate Utilization Patterns During BTEX Biodegradation by an o-Xylene-Degrading Bacterium Ralstonia sp. PHS1

  • The biodegradation of BTEX components (benzene, toluene, ethylbenzene, o-xylene, m-xylene, and p-xylene) individually and in mixtures was investigated using the o-xylene-degrading thermo-tolerant bacterium Ralsronia sp. strain PHS1 , which utilizes benzene, toluene, ethylbenzene, or o-xylene as its sole carbon source. The results showed that as a single substrate for growth, benzene was superior to both toluene and ethylbenzene. While growth inhibition was severe at higher o-xylene concentrations, no inhibition was observed (up to 100 mg $l^-1$) with ethylbenzene. In mixtures of BTEX compounds, the PHS1 culture was shown to degrade all six BTEX components and the degradation rates were in the order of benzene, toluene, o-xylene, ethylbenzene, and m- and p-xylene. m-Xylene and p-xylene were found to be co-metabolized by this microorganism in the presence of the growth-supporting BTEX compounds. In binary mixtures containing the growth substrates (benzene, toluene, ethylbenzene. and o-xylene), PHS1 degraded each BTEX compound faster when it was alone than when it was a component of a BTEX mixture, although the degree of inhibition varied according to the substrates in the mixtures. p-Xylene was shown to be the most potent inhibitor of BTEX biodegradation in binary mixtures. On the other hand, the degradation rates of the non-growth substrates (m-xylene and p-xylene) were significantly enhanced by the addition of growth substrates. The substrate utilization patterns between PHS1 and other microorganisms were also examined.

Studies of Purification and Characterization of Epidermal Growth Factor from Human Colostrum (인유 중의 Epidermal Growth Factor 분리 및 특성에 관한 연구)

  • The purpose of this study was to purify epidermal growth factor(EGF) as growth factor from human colostrum. The effects of purified EGF fraction were directly related to the growth of cells. Results were as follows; After eliminated fat from colostrum, skim milk was obtained. We obtained the EGF fraction by performing ultrafiltration and gel filtration, and then were convicted by SDS-PAGE. The result of analysis of purified EGF fraction by high performance liquid chromatography(HPLC) was shown a peak at 31.185 min period. And it was similar with standard EGF that was shown a peak at 31.545 min. 10 ng of EGF was contained in 10 mg/mL through immunoassay to measure EGF content from isolated fraction. After SDS-PAGE, isolation degree of purified fraction was convicted through western blotting. BALB/3T3 cell was the most effectively stimulated and proliferated at 1 mg/mL concentration of the purified EGF fraction and percentage of cell proliferation was about 95%. In the case of IEC-6 cell, that was about 71%.
