채소추출물이 Listeria monocytogenes의 생육에 미치는 영향 (The Effects of Vegetable Extracts on the Growth of Listeria monocytogenes)

  • The inhibitory effects of vegetables, collected from Jinju district, on the growth of Listeria monocytogenes were investigated. Among 12 vegerables, garlic, leek, onion and cabbage were remarkably effective to inhibit the growth of the strain. Especially with the addition of extract equivalent 0.25g of raw garlic per ml broth, the population of the strain was decreased wignificantly, and completely inhibited with the addition of 0.625g or more. During the incubation with the addition of extract equivalent 0.25g of raw garlic per ml broth, the degree of inhibition was gradually increased with the lapse of time, and completely inhibited the growth of the strain after 48 hrs. While the pH value of treated broth remained unchanged almost, those of control were slightly reduced.

  • To investigate the effect of light intensity on the growth of shoots and roots, water and chlorophyll contents in the ginseng leaf were determined at 5%, l0%, 20%, 30% light transmittance rate (LTR) with 1,2 and 4 year-old ginseng plants in the field. Stem length, size of the leaf, water and chlorophyll contents were decreased as the increase of the light intensity in all ages of ginseng plants. The degree of decrease was severe in the one year-old ginseng compared to that of 2, or 4 year-old ginseng, while there was no difference between the 2 and 4 year-old plants. Root weight per plant was highest at 5% LTR in the one year-old plants, while it was at 20% LTR in the 2 and 4 year-old plants. Generally, demand of light for the growth of one year-old ginseng was lower than those of 2 or 4 year-old plants there was no difference of it among the ginseng plants older than 2 years.

수수로부터 allelopathy성 물질의 기내선별 (In vitro bioassay for allelopathic substances of Sorghum ( Sorghumbicolor L.))

  • These experiments were conducted to determine the effects of Sorghum allelopathic substances on the callus growh of several weeds and crops. 1. When substances extracted from allelopathic Sorghum(Sorghum bicolor L.) were treated on medium, growth of callus of several weeds and crops were in-hibited. The degree of inhibition differed depending on the genotypes, ranging from 50 to 90% com-pared with that of control. 2. The extracts of above 5% Sorghum inhibited the callus growth of Che-nopodium albun L., Commelina communis L., and .Ammaranthus retroflexus L.and showed in-hibition rate of above 70% in callus growth. These results indicate that we could investigate theallelopaihy effect by using in vitro system. 3. The suitable explant for callus induction fromallelopathic plants was immature embryos, the callus induction rate differed depending on the geno-type, growth regulators and concentrations. In general, the addition of 2, 4-D and NAA onto medium increased the rate and amount of callus.

단수수의 파종기에 따른 절간별 당도차이 (Differences of Internode Brix Degree on Different Seeding Date in Sweet Sorghum)

  • 단수수의 만숙과 조숙종인 각품종을 4월 5일, 15, 25일과 5월 5, 15일에 파종하여 품종간의 생육과 간 절위별의 Brix도를 경시적으로 조사하였던 결과를 요약하면 다음과 같다. 1. 파종기가 늦을수록 출현일수와 출수기간은 단축되었으나 5월 15일 파종에서는 출수기가 오히려 지연되었고 초장은 파종기가 늦어질수록 직선적으로 길어졌으며 조기파종일수록 분벽수는 많은 편이었으나 유효경 비율은 오히려 낮아지는 경향이었다. 2. 절간발육은 지상 제5절까지는 절간장이 급격히 길어지고 그 위로 갈수록 점차 짧아지며 절간직경과 중량은 기부위에서 선단으로 갈수록 가늘고 가벼웠다. 3. 간중의 Brix도는 같은 조사일자에서는 파종기가 늦을수록 낮은 경향이었고 출수전까지는 완만한 증가를 보이다가 출수 직후에는 급격한 증가를 보였으며 후기파종의 경우에는 그 증가속도가 더욱 빠른 편이었다. 4. 간 부위별 Brix도는 출수전까지는 기부위에서 중부, 상부위로 갈수록 높은 편이었으나 출수후 이삭의 고습기까지는 중부위에서 가장 높고 상부위, 기부위 순으로 낮았으며 한 줄기의 절위간에 있어서는 지상으로부터 제6또는 제7절 위의 Brix도가 가장 높았다. 5. Brix도가 15%이상이 되는 시기는 4월 5일부터 5월 5일까지의 파종때는 출수후 40∼45일쯤이었으나 5월 15일 파종때에는 출수후 35일쯤에 이미 17%이상의 Brix도를 나타내어 남부지방의 단수수 파종적기는 5월 15일경이라고 생각되었다. 6. 4월 5일부터 4월 25일까지의 파종에서는 파종후 140일경 즉 출수후 50일경까지도 당원료 이용가치선인 Brix도 15%미만이었으나 5월 5일과 15일 파종에서는 파종후 140일경에 18%내외의 Brix도를 보였다. 7. 당분축적이 왕성할 파종후 130∼140일경에도 강우에 의한 토양수분의 과다는 간중 Brix도를 현저히 낮게 하는 것으로 생각되었으며 만숙종에 비하여 조숙종에서 더욱 예민하게 작용하는 것 같았다. 8. 절간장과 절간직경 및 Brix기간에는 부의 상관관계이며 기타 형질간에는 정의 상관관계가 있었으나 그중 절간직경과 절간중간에는 고도의 정상관관계가 인정되었다.

High Density Cell Culture of Mucor sp. KCTC 8405P for Production of ${\gamma}-Linolenic$ Acid in Fed-Batch Culture

  • In order to produce ${\gamma}-linolenic$ acid by Mucor sp. KCTC 8405P. the fungus was cultivated in fed-batch culture with two phases. i.e., growth in yeast-like form and induction to hyphal growth by pH shift of the culture medium during cultivation. The synchronous growth of the fungus into the appropriate sizes was important for the high density cell culture of this dimorphic fungus. Dissolved oxygen concentration in the medium did not affect degree of unsaturation of fatty acids and ${\gamma}-linolenic$ acid content. Under the culture conditions applied in this experiment. the fungus was found to produce 100 g/l dry mycelia containing 40% of the lipids, where ${\gamma}-linolenic$ acid comprised about 9% of the total extractable fatty acids.

Effect of prohexadium-calcium on growth, lodging and yield of proso millet (Panicum miliaceum L.)

  • The proso millet is vulnerable to lodging due to high plant height and shallow root. A lodging results in a hard mechanical harvesting and yield loss. One of solutions on this problem is inhibition of internode elongation. The objective of this study was to set up use time and dose of prohexadium-calcium, is growth inhibitor. The experimental variety was Ibaekchal. The experiment design was a split-plot design with three replications. The treatments were as follow: Main-plots were 25 and 35 day after sowing(DAS) as use time and sub-plots were 0%, 50%, 100%(diluted solution of 1000 times, $1000{\ell}\;ha^{-1}$), 150% as dose. The amount of nitrogen, phosphate and potassium fertilization were 90, 70, $80kg\;ha^{-1}$, respectively. The size of high ridge and plant spacing were $90{\times}30cm$ and $60{\times}15cm$, respectively. Proso millet was sown on June 9, 2016 by hands and was adjusted at 2 plant per hill. The growth survey of vegetative growth stage was conducted at 1 day before treatment and with one week interval after treatment. Data were collected: (1) grain yield: weight of grain in $kg\;ha^{-1}$, (2) 1000 grain weight: average weight of 1000 grain, (3) plant height: distance from soil to top of panicle or leaf in cm, (4) ear length: distance from top of stem to top of ear in cm, (5) stem diameter: diameter of second internode (6) degree of lodging: percentage of lodging area, etc. Analyses of variance were performed using R version 3.3.1(https://www. r- project. org). The Duncan's multiple range test(DMR) was used to separate treatment means at P < 0.05. There was a significant difference in plant height and number of stem among the use time and dose of prohexadium-calcium during vegetative growth stage. At 25 DAS, the difference with no treatment increased until 25 day after treatment and decreased since then. The difference in number of stem increased until 18 day and decreased since 25 day. At 35 DAS, the difference with no treatment in plant height and number of stem increased until 22 day after treatment and decreased since then. We assumed that the effect of prohexadium-calcium was inhibition of internode elongation and promotion of tillering, continued untel 25day after treatment. At 25 DAS, the degree of lodging deceased to 100%, 30%, 10% and 0% as dose increased. At 35 DAS, the degree of lodging decreased to 100%, 20%, 0% and 0% as dose increased. At 25 DAS, the yield was 2910, 2710, 3190, $2310kg\;ha^{-1}$ among dose. At 35 DAS, the yield was 2750, 2630, 2220, $2050kg\;ha^{-1}$. We recommend that the optimum use time and dose of prohexadium-calcium for proso millet is 1000 times diluted solution of $1000{\ell}$ per ha at 25 day after sowing.

울산공단 지역에서의 대기오염이 삼림에 미치는 영향 (I) -대기오염에 의한 곰솔피해- (The Effect on the Forest by the Air Pollution around Ulsan Industrial Complex(I) -Injury on Black Pine-)

  • To investigate the damage on the black pine(Pinus thunbergii) by the air pollution around Ulsan Industrial Complex, forty-six plots were set up and the effects of air pollution were evaluated with the injured index of black pine. The injured index of black pine showing the degree of air pollution from 1987 to 1988 indicated that the central area of Yeochun-dong and Youngjam-dong were heavily damaged. The effective indicaters of black pine damage from air pollution in the order of importance were the color change of one-year-old branch, the ratio of fallen needle of two-year-old branch, the degree of reducing needle, and the degree of growing shoot growth. There was a reliable relationship between the content of $SO_3$ and F in the air and the injured index of black pine. There was no significant correlation between the injured index of the black pine and the content of chemical properties in soil.

갈대 및 가는갯능쟁이의 발아에 미치는 NaCl의 효과 (Effect of NaCl on Germination of Artiplex gmelini and Pharagmites communis)

  • The results of the conducted experiment obtained basic data on seed germination for Phragmites communis and Atriplex gmelini were; Seed germination was not influenced till 0.5% NaCl, but at over 1.0% NaCl it dropped remarkably. The germination limit fro degree of NaCl was 2.0% in Phragmites communis. Atriplex gemelini was 2.5% and in accordance with the increase of the degree of NaCl. Germination speed showed a negative correlation being highly significant and the germination period lengthened. Compared with growing top plants, growth of roots was largely influenced by a high degree of NaCl. In accordance with the rise of temperature, the germination rate, and speed of both plants remarkably increased and the germination period was shortened. Ubride of Atriplex gmelini was germinated at the early days of picking but was not as the passing of the period. The seeds also did not germinated likewise Ubride. By a seedcoat breaking germination became 81%. During 20 min soaking treatment in conc H2SO4 seed germination possibility of 63% was known to be hard. Adequate soaking time in conc H2SO4 was 17.5 min.

  • Effects of hydrogenation in amorphous silicon rile growths on Solid Phase Crystallization (SPC) was investigated using x-ray diffractometry, energy dispersive Spectroscopy, and Raman spectrum. Interdiffusion of barium(Ba) and aluminum(Al) compounds of corning substrate was observed in both of rf sputtering and LFCVD films under the low temperature(580$^{\circ}C$) annealing. Low degree of crystallinity resulted from the interdiffusion was obtained. Highly applicable degree of crystallinity was obtained through the mechanical damage induced surface activation on amorphous silicon films. X-ray diffraction intensity of (111) orientation was used to characterize the degree of crystallinity of SPC. Nucleation and growth rate in SPC could be controllable through the employed surface treatment. IIydrogenated LPCVD films showed the superior crystallinity to non-hydrogenated sputtering films. Insignificant effects of activation treatment in sputtered film was of activation treatment in sputtered film was observed on SPC.
