Reaction Conditions for Laccase Catalyzed Degradation of Bisphenol A

  • The oxidative degradation of BPA with laccase from Trametes versiclor was conducted in a closed, temperature controlled system containing acetate buffer for pH control. The effects of medium pH, buffer concentration, temperature and mediator on degradation of BPA were investigated. The inactivation of the enzyme by temperature and reaction product was also studied. The optimal pH for BPA degradation showed about 5. Buffer concentration did not affect BPA degradation. On the other hand, the enzyme stability was higher at low concentration buffer(25 mM). Temperature rise increased the degradation rate of BPA up to 45$^{\circ}C$. The valuable mediator of laccase for BPA was ABTS. Elevated temperature and reaction product irreversibly inactivated the enzyme.

  • Continuous degradation of formaldehyde by using a rotating disc contactor was investigated in this study. Peudomonas putida H-5K was selected as a mutant using N-methyl N'-nitro N-nitrosoguandine (250$\mu\textrm{g}$/$m\ell$), which showed 1.5 times higher ability of formaldehyde degradation than that of the parent strain. Enzyme activity for formaldehyde degradation released form Peudomonas putida H-5K showed the highest level of 6.2mo1/min/mg protein in the 2% glucose mineral medium containing 0.02% formaldehyde. Degradation of formaldehyde from the first stage in rotating disc contactor was 95% and 5% from the 4th stage when the reactor was fed with 0.02% or 0.04% formaldehyde solution at a rate of 20$m\ell$ per hour. Continuous degradation of formaldehyde using rotating disc contactor was above 95%o in the medium containing 0.04% formalchyde, at the medium feed ratc of 20$m\ell$ per hour.

  • The primary function of ZnO lightning arrester is to protect transmission and distribution equipment from overvoltages and to absorb electrical energy resulting from lightning or switching surges and form temporary overvoltage. However, ZnO lightning arrester are known to exhibit an increases in resistive current with time, the rate of increase being exacerbated with increasing applied voltage and ambient temperature. So, it is important to the leakage current measurement of ZnO lightning arrester. In addition, since the resistive leakage current caused by deterioration of ZnO lightning arrester mainly caused an increase of the third harmonic component, thereby it is possible the arrester degradation diagnosis by measuring the third harmonic component in the total leakage current. The leakage current and third harmonic component are measured and used to investigate the degradation diagnosis of ZnO element of arrester. Also the SEM photography is used to investigate the change of crystal structure of ZnO element with degradation.

Study on the Improvement of Speech Recognizer by Using Time Scale Modification (시간축 변환을 이용한 음성 인식기의 성능 향상에 관한 연구)

  • In this paper a method for compensating for thp performance degradation or automatic speech recognition (ASR) is proposed. which is mainly caused by speaking rate variation. Before the new method is proposed. quantitative analysis of the performance of an HMM-based ASR system according to speaking rate is first performed. From this analysis, significant performance degradation was often observed in the rapidly speaking speech signals. A quantitative measure is then introduced, which is able to represent speaking rate. Time scale modification (TSM) is employed to compensate the speaking rate difference between input speech signals and training speech signals. Finally, a method for compensating the performance degradation caused by speaking rate variation is proposed, in which TSM is selectively employed according to speaking rate. By the results from the ASR experiments devised for the 10-digits mobile phone number, it is confirmed that the error rate was reduced by 15.5% when the proposed method is applied to the high speaking rate speech signals.

Photocatalytic and Sonophotocatalytic degradation of alachlor using different photocatalyst

  • The degradation of alachlor has been investigated using sonolysis (US), photocatalysis (UV) and sonophotocatalysis (US/UV) using three photocatalyst viz. $TiO_2$ (mixture of anatase and rutile), $TiO_2$ (anatase) and ZnO. The effect of photocatalyst loading on the extent of degradation of alachlor has been investigated by varying $TiO_2$ (both types) loading over the range of 0.01 g/L to 0.1 g/L and ZnO loading over the range of 0.05 g/L to 0.3 g/L. The optimum loading of the catalyst was found to be dependent on the type of operation i.e., photocatalysis alone or the combined operation of sonolysis and photocatalysis. All the combined processes gave complete degradation of alachlor with maximum rate of degradation being obtained in the case of sonophotocatalytic process also showing synergistic effect at optimized loading of photocatalyst. About 50% to 60% reduction in TOC has been obtained using the combined process of sonophotocatalysis depending on the operating conditions. The alachlor degradation fitted first order kinetics for all the processes under investigation. It has been observed that the $TiO_2$ (mixtrure of anatase and rutile) is the most active photocatalyst among the three photocatalysts studied in the current work. The effect of addition of radical enhancers and scavengers on sonophotocatalytic degradation of alachlor has been investigated in order to decipher the controlling mechanism. The alachlor degradation products have been identified using LC-MS method.

Characterization of MTBE (Methyl Tertiary Butyl Ether) Utilizing Bacteria from the Gasoline Contaminated Soils (유류오염토양에서 분리된 MTBE(Methyl Tertiary Butyl Ether) 이용 균주의 MTBE 분해특성)

  • In this study, we have isolated MTBE utilizing bacteria at the gasoline contaminated soil and also MTBE degradation patterns were characterized. The 18 bacterial mono-cultures isolated from enrichment cultures were screened for MTBE degradation. Of the 18 strains, the 3 strains (Flavobacterium, Pseudomonas, and Achromobacter) have shown effective MTBE degradation. Experimental parameters affecting the growth conditions (such as temperature, pH, initial cell mass) were optimized. Experimental parameters such as temperature $30^{\circ}C$, pH 7, and initial cell mass 0.6 g/mL in optimal growth conditions for MTBE degradation. The optimal growth conditions of the isolated stains were temperature $30^{\circ}C$, pH 7, and initial cell mass 0.6 g/mL in our experiment, respectively. The first order degradation coefficients of Achromobacter, Mixed culture, Pseudomonas, and Flavobacterium were 0.072, 0.066, 0.047, and $0.032hr^{-1}$, respectively. and also, it could be expressed as a degradation rate considering cell mass (1.302, 1.019, 0.523, and 0.352 mg/TSS g/hr for each microorganism). Although Achromobacter has shown highest MTBE degradation rate, degradation rate for BTEX was relatively lower than other strains. and Mixed culture and Flavobacterium have shown similar degradation pattern for MTBE and BTEX biodegradation.

Kinetics of Metolachlor Degradation by Zerovalent Iron (Zerovalent Iron에 의한 Metolachlor의 분해 Kinetics)

  • Metolachlor may pose a threat to surface and ground water qualities due to its high solubility in water, Zerovalent iron (ZVI) releases $e^-$ which can degrade the organochlorinated compounds. The objective of this research was to evaluate the kinetics of metolachlor degradation as affected by ZVI sources [Peerless unannealed (PU) and Peerless annealed (PA)] and ZVI levels (1 and 5%) under batch conditions at different metolachlor concentrations (200 and 1000 mg/l) and temperatures (15, 25, and $35^{\circ}C$). The effectiveness of ZVI on metolachlor degradation was assessed by characterizing the dechlorinated metolachlor byproduct molecules. Metolachlor degradation by ZVI followed the first-ordered kinetics with a higher rate constant at higher level of ZVI treatment. At 5% (w/v) of PU and PA treatment, the half-lives of metolachlor degradation were 9.93 and 6.51 h and all of the initial metolachlor were degraded in 72 and 48 h, respectively. Rate constants (k) of metolachlor degradation were higher at the lower initial metolachlor concentration. The metolachlor degradation by ZVI was temperature dependent showing that the rate constant (k) at 15, 25, and $35^{\circ}C$ were 0.0805, 0.1017, and 0.3116 /h, respectively. The ZVI-mediated metolachlor degradation yielded two byproduct molecules identified as dechlorinated metolachlor $(C_{13}H_{18}NO)$ and dechlorinated-dealkylated metolachlor $(C_{12}H_{17}NO)$. The PA ZVI was more effective than PU ZVI in metolachlor degradation.

  • Rumen liquor characteristics and disappearance rate of dry matter were studied in Kedah-Kelantan cattle and swamp buffaloes fed grass of rice straw-based diet. Cobalt-EDTA and chromium mordented fibres prepared from the faecal material were used to determine the liquid and solid particles movement in both animal species fed with rice straw. Swamp buffaloes showed a more intense rumen fermentation activity than Kedah-Kelantan cattle when both species were fed straw-based diet. The buffaloes also demonstrated faster rates of grass and straw degradation in situ. The fluid outflow rate from the rumen of buffalo ($1.06{\pm}0.19l/h$) was observed to be slower than that of cattle ($1.55{\pm}0.01l/h$). No significant differences between cattle and buffaloes were observed in rumen fluid volume and passage rate of small particles from the rumen.

Characteristic of Oxidants Production and Dye Degradation with Operation Parameters of Electrochemical Process (전기화학적 공정의 운전인자에 따른 산화제 생성과 염료 분해 특성)

  • The purpose of this study is to investigate electro-generation of free Cl, $ClO_2$, $H_2O_2$ and $O_3$ and degradation of Rhodamine B in solution using Ru-Sn-Sb electrode. Electrolysis was performed in one-compartment reactor using a dimensionally stable anode(DSA) of Ru-Sn-Sb/Ti as the working electrode. The effect of applied current (0.5-3 A), electrolyte type (NaCl, KCl, HCl, $Na_2SO_4$ and $H_2SO_4$) and concentration (0.5-2.5 g/L), air flow rate (0-3 L/min) and solution pH (3-11) was evaluated. Experimental results showed that concentration of 4 oxidants was increased with increase of applied current, however optimum current for RhB degradation was 2 A. The generated oxidant concentration and RhB degradation of the of Cl type-electrolyte was higher than that of the sulfate type. The oxidant concentration was increased with increase of NaCl concentration and optimum NaCl dosage for RhB degradation was 1.75 g/L. Optimum air flow rate for the oxidants generation and RhB degradation was 2 L/min. $ClO_2$ and $H_2O_2$ generation was decreased with the increase of pH, whereas free Cl and $O_3$ was not affected by pH. RhB degradation was increase with the pH decrease.

Effect of Rapid Chilling on Beef Quality and Cytoskeletal Protein Degradation in M. longissimus of Chinese Yellow Crossbred Bulls

  • The objective of this study was to investigate the effect of rapid chilling (RC) on beef quality and the degradation of cytoskeletal proteins. Twenty Chinese Yellow crossbred bulls were selected and randomly divided into two groups. RC and conventional chilling (CC) were applied to left and right sides of the carcasses respectively after slaughtering. To determine whether electrical stimulation (ES) treatment can alleviate the potential hazard of RC on meat quality, ES was applied to one group. The effects of RC and ES were determined by meat color, shear force and cytoskeletal protein degradation postmortem (PM). The results showed that RC decreased beef tenderness at 1 d and 3 d postmortem, but had no detrimental effect on meat color. Western blotting showed that RC decreased the degradation rate of desmin and troponin-T, but the effects weakened gradually as postmortem aging extended. Degradation rates of both desmin and troponin-T were accelerated by ES. The combination of RC and ES could improve beef color, accelerate degradation rate of cytoskeletal protein and improve beef tenderness.
