AITC induces MRP1 expression by protecting against CS/CSE-mediated DJ-1 protein degradation via activation of the DJ-1/Nrf2 axis

  • The present study aimed to examine the effect of allyl isothiocyanate (AITC) on chronic obstructive pulmonary disease and to investigate whether upregulation of multidrug resistance-associated protein 1 (MRP1) associated with the activation of the PARK7 (DJ-1)/nuclear factor erythroid 2-related factor 2 (Nrf2) axis. Lung function indexes and histopathological changes in mice were assessed by lung function detection and H&E staining. The expression levels of Nrf2, MRP1, heme oxygenase-1 (HO-1), and DJ-1 were determined by immunohistochemistry, Western blotting and reverse transcription-quantitative polymerase chain reaction. Next, the expression of DJ-1 in human bronchial epithelial (16HBE) cells was silenced by siRNA, and the effect of DJ-1 expression level on cigarette smoke extract (CSE)-stimulated protein degradation and AITC-induced protein expression was examined. The expression of DJ-1, Nrf2, HO-1, and MRP1 was significantly decreased in the wild type model group, while the expression of each protein was significantly increased after administration of AITC. Silencing the expression of DJ-1 in 16HBE cells accelerated CSE-induced protein degradation, and significantly attenuated the AITC-induced mRNA and protein expression of Nrf2 and MRP1. The present study describes a novel mechanism by which AITC induces MRP1 expression by protecting against CS/CSE-mediated DJ-1 protein degradation via activation of the DJ-1/Nrf2 axis.

Characterization of the Transglycosylation Reaction of 4-α-Glucanotransferase (MalQ) and Its Role in Glycogen Breakdown in Escherichia coli

  • We first confirmed the involvement of MalQ (4-${\alpha}$-glucanotransferase) in Escherichia coli glycogen breakdown by both in vitro and in vivo assays. In vivo tests of the knock-out mutant, ${\Delta}malQ$, showed that glycogen slowly decreased after the stationary phase compared to the wild-type strain, indicating the involvement of MalQ in glycogen degradation. In vitro assays incubated glycogen-mimic substrate, branched cyclodextrin (maltotetraosyl-${\beta}$-CD: G4-${\beta}$-CD) and glycogen phosphorylase (GlgP)-limit dextrin with a set of variable combinations of E. coli enzymes, including GlgX (debranching enzyme), MalP (maltodextrin phosphorylase), GlgP and MalQ. In the absence of GlgP, the reaction of MalP, GlgX and MalQ on substrates produced glucose-1-P (glc-1-P) 3-fold faster than without MalQ. The results revealed that MalQ led to disproportionate G4 released from GlgP-limit dextrin to another acceptor, G4, which is phosphorylated by MalP. In contrast, in the absence of MalP, the reaction of GlgX, GlgP and MalQ resulted in a 1.6-fold increased production of glc-1-P than without MalQ. The result indicated that the G4-branch chains of GlgP-limit dextrin are released by GlgX hydrolysis, and then MalQ transfers the resultant G4 either to another branch chain or another G4 that can immediately be phosphorylated into glc-1-P by GlgP. Thus, we propose a model of two possible MalQ-involved pathways in glycogen degradation. The operon structure of MalP-defecting enterobacteria strongly supports the involvement of MalQ and GlgP as alternative pathways in glycogen degradation.

결빙된 전기체 헬리콥터의 비행성 향상을 위한 강인 제어 설계 (Robust Control Design for Handling Quality Improvement of Iced Full-scale Helicopter)

  • 악천후나 기계적 결함에 의한 비행성(Handling Qualities) 저하는 그 상황에 익숙하지 못한 조종사에게 치명적인 위험을 줄 수 있다. 특히 결빙의 경우에는 실제로 사고가 빈번하게 일어나는 원인인 만큼 중요하게 고려해야 할 사안이다. 대다수의 기존 연구들은 결빙에 따른 공기역학적 성능 변화와 그에 따른 모델링 방법을 제시하거나 결빙을 방지하는 방법을 주로 다루었으나, 본 연구에서는 전기체(full-scale) 헬리콥터에 발생하는 결빙으로 인한 비행 성능 저하를 능동적으로 보상하는 비행제어기를 설계하고자 한다. 본 연구에서는 먼저 UH-60 헬리콥터에 발생하는 결빙으로 인한 비행성 저하를 CONDUIT이라는 프로그램을 통해 보이고, 이러한 비행성 저하를 보상하기 위해 RS-LQR(Robust Servomechanism Linear Quadratic Regulation) 기법을 사용한 강인 제어기를 설계하였다. 시뮬레이션을 통해 제안한 강인 제어기가 헬리콥터가 결빙된 상황에서도 Level 1 비행성을 유지하는 것을 보였다.

Residual capacity assessment of in-service concrete box-girder bridges considering traffic growth and structural deterioration

  • The existing concrete bridges are time-varying working systems, where the maintenance strategy should be planned according to the time-varying performance of the bridge. This work proposes a time-dependent residual capacity assessment procedure, which considers the non-stationary bridge load effects under growing traffic and non-stationary structural deterioration owing to material degradations. Lifetime bridge load effects under traffic growth are predicated by the non-stationary peaks-over-threshold (POT) method using time-dependent generalized Pareto distribution (GPD) models. The non-stationary structural resistance owing to material degradation is modeled by incorporating the Gamma deterioration process and field inspection data. A three-span continuous box-girder bridge is illustrated as an example to demonstrate the application of the proposed procedure, and the time-varying reliability indexes of the bridge girder are calculated. The accuracy of the proposed non-stationary POT method is verified through numerical examples, where the shape parameter of the time-varying GPD model is constant but the threshold and scale parameters are polynomial functions increasing with time. The case study illustrates that the residual flexural capacities show a degradation trend from a slow decrease to an accelerated decrease under traffic growth and material degradation. The reliability index for the mid-span cross-section reduces from 4.91 to 4.55 after being in service for 100 years, and the value is from 4.96 to 4.75 for the mid-support cross-section. The studied bridge shows no safety risk under traffic growth and structural deterioration owing to its high design safety reserve. However, applying the proposed numerical approach to analyze the degradation of residual bearing capacity for bridge structures with low safety reserves is of great significance for management and maintenance.

AC Modeling of the ggNMOS ESD Protection Device

  • From AC analysis results utilizing a 2-dimensional device simulator, we extracted an AC-equivalent circuit of a grounded-gate NMOS (ggNMOS) electrostatic discharge (ESD) protection device. The extracted equivalent circuit is utilized to analyze the effects of the parasitics in a ggNMOS protection device on the characteristics of a low noise amplifier (LNA). We have shown that the effects of the parasitics can appear exaggerated for an impedance matching aspect and that the noise contribution of the parasitic resistances cannot be counted if the ggNMOS protection device is modeled by a single capacitor, as in prior publications. We have confirmed that the major changes in the characteristics of an LNA when connecting an NMOS protection device at the input are reduction of the power gain and degradation of the noise performance. We have also shown that the performance degradation worsens as the substrate resistance is reduced, which could not be detected if a single capacitor model is used.

항공기 재료 성형시의 손상진전에 관한 연구 (A Study on the Damage Propagation of an Aircraft Material During Forming)

  • In this paper damage propagation of a material during forming is investigated with the concept of continuum damage mechanics. An isotropic damage model based on the theory of materials of type N is adopted to describe the damage process of a ductile material with large elasto-viscoplastic deformation. The stiffness degradation of the loaded material is chosen as a damage measure. The highly nonlinear equilibrium equations are reduced to the incremental weak form and approximated by the total Lagrangian finite element method. To simulate contact condition, extended interior penalty method with modified coulomb friction law is adopted. The displacement control method along with the modified Riks' continuation technique is used to solve the incremental iterative equations. As numerical examples, upsetting problem and backward extrusion problem are simulated and the results of damage propagation and $J_2$ stress contours with and without friction are presented.

  • This paper proposes the development of delay compensator to minimize performance degradation caused by time delays in network-based real-time control systems. The delay compensator uses the time-stamp method as a direct delay measuring method to measure time delays generated between network nodes. The delay compensator predicts the network time delays of next period in the views point of time delays and minimizes performance degradation from network through considering predicted time delays. Control output considering network time delays is generated by the defuzzification of probable time delays of next period. The time delays considered in the delay compensator are modeled by using a timed Petri net model. The proposed delay prediction mechanism for the delay compensator is evaluated through some simulation tests by measuring deviation of the predicted delays from simulated delays.

  • A discrete joint finite element with joint surface degradation was developed to investigate the shear behavior of rough rock joint. Isoparametric formulation was used for facilitating the implementation of the element in existing Finite Element Codes. The elasto-plastic joint deformation model with the discontinuity constitutive law proposed by Plesha was applied to the element. The reliability of the developed finite element code was successfully testified through numerical direct shear tests conducted under both constant normal stress and constant normal displacement conditions. The result of the numerical direct shear test showed that the code can capture characteristic deformation features envisaged in the direct shear test of rough rock joint.

Formation Kinetic Study of Thermal Products of Tocopherols

  • The kinetic analyses for thermal products of alpha-, gamma- and delta-tocopherols during heating as functions of temperature and time were studied. Alpha-, gamma- and delta-tocopherols dissolved in glycerol were heated at $100{\sim}200^{\circ}C$ for $5{\sim}60$ min. The thermal products were separated by hexane extraction and analyzed by HPLC using a reversed phase ${\mu}-Bondapak$ $C_{18}-column$ with two kinds of elution solvents in a gradient mode. The formation kinetics of thermal products of tocopherols followed a first-order kinetic model. The formation rate of thermal products of tocopherols was dependent on heating temperatures and heating times. The activation energy and enthalpy for the thermal products of ${\gamma}-and$ ${\delta}-tocopherols$ were higher than those for ${\alpha}-tocopherol$ as in the case of the oxidative degradation kinetics of tocopherol. The magnitude order of the activation energy was ${\gamma}->{\delta}->{\alpha}-tocopherol$.

시멘트/슬래그/Fe(II) 시스템에 의한 NAPL TCE의 분해 특성

  • Batch slurry experiments were conducted to develop cement/slag/Fe(II) system that could treat hazardous liquid wastes containing halogenated organic solvents. Portland cement in combination with Fe(II) was reported to reductively dechlorinate chlorinated organics in a modified solidification/stabilization process. TCE (trichloroethylene) was used a model halogenated organic solvent. The objectives of this study were to assess the feasibility of using cement and steel converter slag amended with Fe(II) as a low cost abiotic reductive dechlorination and to investigate the kinetics of TCE dechlorination over a wide range of TCE concentration. From the result of screening experiments, cement/slag/Fe(II) system was identified as a potentially effective system to treat halogenated organic solvent. Kinetic studies were carried out to further investigate degradation reaction of TCE NAPL (Non Aqueous Phase Liquids) in cement/slag/Fe(II) systems by using batch slurry reactors. Degradation rate of TCE solution in this system can be explained by pseudo-first-order rate law because the prediction with the rate law is in good agreement with the observed data.
