Tethered DNA shear dynamics in the flow gradient plane: application to double tethering

  • We examine the wall contact of a $3\;{\mu}m$ tethered DNA chain's free end under shear with a focus on developing schemes for double-tethering in the application of making scaffolds for molecular wires. At this scale our results are found to be highly dependent on small length scale rigidity. Chain-end-wall contact frequency, mean fractional extension deficit upon contact, and standard deviation in extension upon contact are examined for scaling with dimensionless flow strength, Wi. Predictions made using a one dimensional approximation to the Smoluchowski equation for a dumbbell and three dimensional dumbbell simulations produce extension deficit, standard deviation, and frequency scaling exponents of -1/3, -1/3, and 2/3, respectively whereas more fine-grained Kratky-Porod (KP) simulations produce scaling exponents of -0.48, -0.42, and 0.76. The contact frequency scaling of 2/3 is derived from the known results regarding cyclic dynamics Analytical scaling predictions are in agreement with those previously proposed for ${\lambda}-DNA$. [Ladoux and Doyle, 2000, Doyle et al., 2000]. Our results suggest that the differences between the dumbbell and the KP model are associated with the addition of chain discretization and the correct bending potential in the latter. These scaling results will aide future exploration in double tethering of DNA to a surface.

  • Attention deficit/hyperactivity disorder (ADHD) is a neurodevelopmental disorder characterized by hyperactivity, inattention, and impulsiveness. Current estimates suggest that 4-12% of school age children are affected by ADHD, which hampers proper social relationship and achievements in school. Even though the exact etiology of the disorder is still in the middle of active investigation, the availability of pharmacological treatments for the disorder suggest that at least the symptoms of ADHD are manageable. To develop drugs with higher efficacy and fewer side effects, it is essential to have appropriate animal models for in vivo drug screening processes. Good animal models can also provide the chances to improve our understanding of the disease processes as well as the underlying etiology of the disorder. In this review, we summarized current animal models used for ADHD research and discussed the point of concerns about using specific animal models.

  • Meteorological and soil water content data measured from semiarid watersheds of Lucky Hills and Kendall during the summer rainy and winter periods were used to study the interrelationships between the controlling variables of the evapotranspiration, and to evaluate the effects of variables on daily actual evapotranspiration (ET) estimation. Simple and multiple linear regression (MLR) analyses were employed to evaluate the order of importance of the meteorological and soil water factors involved. The information gained was used for MLR model development. Theavailable energy and vapor pressure deficit were found to be the important variables to estimate actual ET (AET) for both periods and at both watersheds. Therefore, the important variables of evapotranspiration process in these semiarid watersheds appear to be simply the components of energy term in available energy and aerodynamic term in vapor pressure deficit of Penman potential evapotranspiration (PET) equation.

  • 본 연구에서는 일반 상호보완이론(Generalized Complementary Relationship, GCR)을 활용하여 실제증발산량을 추정하고, 추정한 실제증발산량기반 가뭄지수로부터 미국 전역에 대한 가뭄을 해석하는 것이다. 월강수량, 최고 최저기온, 이슬점온도 등의 필요한 기상자료는 Parameter-elevation Relationships on Independent Slopes Model(PRISM)으로부터 수집하였으며, 1981년부터 2015년까지 총 35년의 미국 전역에 대한 실제증발산량을 추정하였다. 대상지역의 유역평균 강수량과 유출량의 차(P-Q)와 North American Land Data Assimilation System(NLDAS-2) Noah 지표모형(Land surface models)으로 산정한 실제증발산량과 비교 검증하였다. GCR로부터 증발산 부족량(ET Deficit, ETD)을 산정하고 이를 표준정규화하여 미국 전역에 대해 Standardized Evapotranspiration Deficit Index(SEDI)를 산정하였다. 본 연구로부터 GCR 기반 실제증발산량은 P-Q의 값과 상관계수가 0.94로 매우 높은 상관성을 보였으며, NLDAS-2 Noah모형의 실제증발산량보다 다소 크게 추정하는 경향을 보였다. SEDI와 Standard Precipitation Index(SPI)의 상관성은 지속시간이 클수록 더 크게 나타났다. 증발산 상호보완이론활용 실제증발산기반 SEDI이 강수자료를 사용하지 않고서도 적절한 가뭄해석에 이용될 수 있을 것으로 판단된다.

The Effect of Total Patellectomy in the Prosthetic Replacement of Proximal Tibia (경골 근위부 종양에서 인공 삽입물 사용시 슬개골 전적출술이 관절기능 회복에 미치는 영향)

  • The purpose of this study is a comparative evaluation of range motion, especially extension deficit between the group of total patellectomy and that of intact patella, after reconstruction of the patellar tendon in the prosthetic replacement of a proximal tibia. Between 1990 and 1994, 15 patients who had a primary malignancy on proximal tibia were operated on. All patients were evaluated clinically and radiographically. Two patients were excluded because one had a deep infection treated with arthrodesis of the knee and the other was a composite allograft. The mean follow-up of the 13 patients was 27 months(15-47), including 10 osteosarcomas, 1 chondrosarcoma, 1 malignant fibrous histiocytoma and 1 malignant giant cell tumor. Eleven patients had a resection of the proximal tibia and 2 had an extracapsular total knee resection with distal femur. Reconstruction of the defect was done in 8 cases with a custom-made Link Endo-Model Total Rotation Knee Joint Prosthesis, and in 5 with How Medica Modular Resection System (HMRS). We used two methods to reconstruct the ligamentum patellae. Fixation of the patellar tendon to the prosthesis only with suturing and/or stapling(group SS) was done in 7. Transposition of gastrocnemius muscle to enhance fixation and to cover the prosthesis(group TG) was done in 6. Regardless of fixation methods, total patellectomy was done in 5 either to lengthen the patellar tendon or to make primary skin closure easier or for both. In 8 cases, patella was left intact or resurfaced with polyethylene prosthesis. Active extension was measured while the patient was in a sitting position. There is no statistically meaningful difference in terms of extension deficit (Wilcoxon rank test, p=0.8800) between patellectomy group and intact patella group, and between group of fixation only with suturing and that of gastrocnemius transposition. Two cases of extension deficit over 30 degree were seen in group SS and in the group of intact patella. Conclusively, total patellectomy could be an option without increasing the risk of extension deficit when primary skin closure is difficult or patellar tendon is a little bit short to be fixed. There is no rating in the Enneking system of functional evaluation that this finding into consideration.

  • Objective : The purpose of this study was to provide for the intervention and outcome measurement tools of children with Attention Deficit Hyperactivity Disorder (ADHD) through ICF model. Methods : The systematic review methods were used. Papers published in the journal between January, 2000 and July, 2014 were searched through MEDLINE/PubMed, Sciencedirect, Ovid. The main terms searched were 'ADHD, Children, intervention, outcome measure', and 8 papers were analyzed. Results : 1. The subjects of ADHD were pure ADHD (75.8%), ADHD with dyslexia (9.1%), ADHD with conduct disorder (5.8%), ADHD with tic disorder (3.8%), ADHD with DCD (3.0%), ADHD with emotional disorder (2.5%). 2. The nonpharmacologic intervention of ADHD were functioning and disability (80%) and contextual factors (20%). Most frequently used intervention were body function and structure (60%). 3. The outcome measurement tools of ADHD were functioning and disability (80.5%) and contextual factors (19.5%). Most frequently used outcome measurement tools were body function and structure (70.8%). Conclusion : This study can provide information on the intervention and outcome measurement tools of ADHD.

Intake Valve Temperature Effect on the Mixture Preparation in a SI Engine During Warm-up

  • A heat transfer model of the intake valve in a spark ignition engine is presented, which is calibrated with a number of the valve temperature profiles measured during engine warm-up for the gaseous fuel(propane). The valve is divided into four identical elements for which the assumption of lumped thermal mass is applied. The calibration is made so that the difference between the measued and simulated valve temperatures becomes minimal. Then the model is applied to the cases of the liquid fuel(indolene) to estimate the amount of the liquid fuel vaporized from the intake valve by assuming that fuel evaporation accounts for the deficit of the heat balance budget. The results of the model show quantitative contribution of each heat transfer source to the heat balance. The behavior of the calculated mass fraction of the fuel vaporized from the intake valve explains how the liquid fuel evaporate during engine warm-up. The mass fraction at warmed-up condition is closely related with the fraction directly targeted on the valve back by the fuel spray geometry.

  • Objectives : This experiment was designed to investigate the effect of the ODTCMT hot water extract & ultra-fine Powder on Alzheimer's Disease Model Induced by 13A. Methods : The effects of the ODTCMT hot water extract on expression of IL-l${\beta}$, IL-6, TNF-${\alpha}$ in BV2 microglial cell line treated by lipopolysacchaide(LPS) were investigated. The effects of the ODTCMT hot water extract & ultra-fine powder on the behavior of the memory deficit mice induced by scopolamine were investigated. Results : 1. The ODTCMT hot water extract significantly suppressed the production of TNF-${\alpha}$, IL-6 and IL-l${\beta}$ in BV2 microglial cell line treated with LPS. 2. The ODTCMT hot water extract & ultra-fine powder groups showed significantly inhibitory effect on the scopolamine-induced impairment of memory in the experiment of Morris water maze. Conclusions : These results suggest that the ODTCMT hot water extract & ultra-fine powder may be effective for the prevention and treatment of Alzheimer's disease. Investigation into the clinical use of the ODTCMT hot water extract & ultra-fine powder for Alzheimer's disease is suggested for future research.

A Simple Behavioral Paradigm to Measure Impulsive Behavior in an Animal Model of Attention Deficit Hyperactivity Disorder (ADHD) of the Spontaneously Hypertensive Rats

  • Impulsiveness is an important component of many psychiatric disorders including Attention-deficit/hyperactivity disorder (ADHD). Although the neurobiological basis of ADHD is unresolved, behavioral tests in animal models have become indispensable tools for improving our understanding of this disorder. In the punishment/extinction paradigm, impulsivity is shown by subjects that persevere with responding despite punishment or unrewarded responses. Exploiting this principle, we developed a new behavioral test that would evaluate impulsivity in the most validated animal model of ADHD of the Spontaneously Hypertensive rat (SHR) as compared with the normotensive "control" strain, the Wistar Kyoto rat (WKY). In this paradigm we call the Electro-Foot Shock aversive water Drinking test (EFSDT), water-deprived rats should pass over an electrified quadrant of the EFSDT apparatus to drink water. We reasoned that impulsive animals show increased frequency to drink water even with the presentation of an aversive consequence (electro-shock). Through this assay, we showed that the SHR was more impulsive than the WKY as it demonstrated more "drinking attempts" and drinking frequency. Methylphenidate, the most widely used ADHD medication, significantly reduced drinking frequency of both SHR and WKY in the EFSDT. Thus, the present assay may be considered as another behavioral tool to measure impulsivity in animal disease models, especially in the context of ADHD.

  • The primary purpose of this paper is to derive a benchmark dose lower limit (BMDL) of lead for the attention deficit/hyperactive disorder (ADHD) based on a longitudinal cohort data set which is referred to as CHEER data set. The CHEER data were recently recruited from the Ministry of Environment of S. Korea to investigate the effect of environment on children's health We first confirm the correlation of ADHD with the blood lead level using a linear mixed effect model. We report from the longitudinal characteristic of CHEER data that ADHD scores tend to have "regression to the mean". A dose-response curve of blood lead level with ADHD being the end point is derived and from this dose-response curve a few BMDLs are derived based on corresponding assumptions on the benchmark region.
