Incidence of Alternaria Species Associated with Watermelon Leaf Blight in Korea

  • Alternaria leaf blight is one of the most common diseases in watermelon worldwide. In Korea, however, the Alternaria species causing the watermelon leaf blight have not been investigated thoroughly. A total of 16 Alternaria isolates was recovered from diseased watermelon leaves with leaf blight symptoms, which were collected from 14 fields in Korea. Analysis of internal transcribed spacer (ITS) region, glyceraldehyde-3-phosphate dehydrogenase (GAPDH), and RNA polymerase II second largest subunit (RPB2) were not competent to differentiate the Alternaria isolates. On the contrary, analysis of amplicon size of the histone H3 (HIS3) gene successfully differentiated the isolates into three Alternaria subgroups, and further sequence analysis of them identified three Alternaria spp. Alternaria tenuissima, A. gaisen, and A. alternata. Representative Alternaria isolates from three species induced dark brown leaf spot lesions on detached watermelon leaves, indicating that A. tenuissima, A. gaisen, and A. alternata are all causal agents of Alternaria leaf blight. Our results indicate that the Alternaria species associated watermelon leaf blight in Korea is more complex than reported previously. This is the first report regarding the population structure of Alternaria species causing watermelon leaf blight in Korea.

First report of anthracnose crown rot caused by Colletotrichum siamense on strawberry in Korea

  • Anthracnose crown rot (ACR) has been observed in greenhouses during the nursery and harvest seasons in Gangwon Province, Korea. Infected plants showed black leaf spot, dark sunken pink conidial masses on petioles, wilting, and eventually death. Five isolates were obtained from the lesions of strawberry plants and were identified as a Colletotrichum gloeosporioides species complex based on their cultural and morphological characteristics. Multilocus sequence analysis of actin, calmodulin, chitin synthase, glyceraldehyde-3-phophate dehydrogenase genes, and internal transcribed spacer rDNA regions showed that the isolates formed a monophyletic group with the type strain of C. siamense. Pathogenicity tests were performed on the isolate, and Koch's postulates were performed to verify the relationship between Colletotrichum sp. and the strawberry plant variety Seolhyang. The isolate was pathogenic to strawberry plants, which exhibited typical ACR symptoms. Based on morphological characteristics, pathogenicity, and DNA sequence analyses, the fungus isolated in Korea was identified as C. siamense. This is first time C. siamense has been confirmed in ever-bearing strawberry varieties in Korea.

A Study on Real-Time Defect Detection Using Ultrasound Excited Thermography (초음파 서모그라피를 이용한 실시간 결함 검출에 대한 연구)

  • The UET(ultrasound excited thermography) for the ,eat-time diagnostics of the object employs an infrared camera to image defects of the surface and subsurface which are locally heated using high-frequency putted ultrasonic excitation. The dissipation of high-power ultrasonic energy around the feces of the defects causes an increase In temperature. The defect's image appears as a hot spot (bright IR source) within a dark background field. The UET for nondestructive diagnostic and evaluation is based on the image analysis of the hot spot as a local response to ultrasonic excited heat deposition. In this paper the applicability of VET for fast imaging of defect is described. The ultrasonic energy is injected into the sample through a transducer in the vertical and horizontal directions respectively. The voltage applied to the transducer is measured by digital oscilloscope, and the waveform are compared. Measurements were performed on four kinds of materials: SUS fatigue crack specimen(thickness 14mm), PCB plate(1.8 mm), CFRP plate(3 mm) and Inconel 600 plate (1 mm). A high power ultrasonic energy with pulse durations of 250ms Is injected into the samples in the horizontal and vertical directions respectively The obtained experimental result reveals that the dissipation loss of the ultrasonic energy In the vertical injection is less than that in the horizontal direction. In the cafe or PCB, CFRP, the size of hot spot in the vortical injection if larger than that in horizontal direction. Duration time of the hot spot in the vertical direction is three times as long as that in the horizontal direction. In the case of Inconel 600 plate and SUS sample, the hot spot in the horizontal injection was detected faster than that in the vertical direction

Analysis of the Cause of the Twig Blight on Mulberry Based on the Weather Conditions (뽕나무 재배기간 중 기상환경과 눈마름병 발생관계 분석)

  • The twig blight of mulberry caused by Fusarium lateritium occurs mainly in Buan area, which is the main area of mulberry. The initial symptom on twigs showed dark edge blight and dark orange tiny particles on the spot, and it was withered and died. In the case of mulberry twig blight, the difference in the degree of occurrence between the years is intense, and an average of 2.7% occurred in Buan in 2013, but in 2014, it was difficult to search for the generated fields. In order to find the cause of the difference in the incidence of mulberry twig blight, we investigated and analyzed the weather conditions during cultivation and as a result, the occurrence of the disease increased under the condition that the temperature of the spring was lower and the number of days passing below the winter freezing point increased. In addition, it was analyzed that the occurrence of the next year increases when the rainfall is high and the number of sunshine is low after removing the harvested branches. Therefore, in the event that weather conditions causing twig blight diseases have elapsed, it is necessary to prevent the disease by spraying the registered applied fungicide prophylactically.

Stalk Rot Caused by Fusarium thapsinum on Sorghum at Organic Paddy-upland Rotation System in Korea (유기농 답전윤환지 Fusarium thapsinum에 의한 수수 줄기썩음병)

  • In late June 2016, stalk rot symptoms were observed on five vatieties of sorghum (Sorghum bicolar) at organic paddy-upland rotation system in Anseong city, Korea. The initial symptom on stalk surfaces was red color with a dark red spot lesion. A fungus was isolated from the initial lesion, and cultured on potato dextrose agar. Size of microconidia mostly extend to $5-19{\times}2-{\mu}m$ in culture, with 0-1 septa and macroconidia extend to $29-52{\times}3-4{\mu}m$ with 4-6 septa. Pathogenicity was investigated using conidial suspension spray to seedling of sorghum. After 3 days of inoculation, the dark red lesion was produced on stalks. On the basis of mycological characteristics, pathogenicity, and internal transcribed spacer (ITS) rDNA sequence analysis, this fungus was identified as Fusarium thapsinum. This is the first report of stalk rot on sorghum caused by F. thapsinum in Korea.

Morphological and Genetic Characteristics of Colletotrichum gloeosporioides Isolated from Newly Emerging Static-Symptom Anthracnose in Apple

  • Filamentous fungi of the genus Colletotrichum (teleomorph, Glomerella) are considered major plant pathogens worldwide. Cereals, legumes, vegetables, and fruit trees may be seriously affected by this pathogen (1). Colletotrichum species cause typical disease symptoms known as anthracnoses, characterized by sunken necrotic tissue, where orange conidial masses are produced. Anthracnose appears in both developing and mature plant tissues (2). We investigated disease occurrence in apple orchards from 2013 to 2014 in northern Gyeongbuk province, Korea. Typical anthracnose with advanced symptoms was observed in all apple orchards studied. Of late, static fruit spot symptoms are being observed in apple orchards. A small lesion, which does not expand further and remains static until the harvesting season, is observed at the beginning of fruit growth period. In our study, static symptoms, together with the typical symptoms, were observed on apples. The isolated fungus was tested for pathogenicity on cv. 'Fuji apple' (fully ripe fruits, unripe fruits, and cross-section of fruits) by inoculating the fruits with a conidial suspension ($10^5$ conidia/ml). In apple inoculated with typical anthracnose fungus, the anthracnose symptoms progressed, and dark lesions with salmon-colored masses of conidia were observed on fruit, which were also soft and sunken. However, in apple inoculated with fungi causing static symptoms, the size of the spots did not increase. Interestingly, the shape and size of the conidia and the shape of the appressoria of both types of fungi were found to be similar. The conidia of the two types of fungi were straight and cylindrical, with an obtuse apex. The culture and morphological characteristics of the conidia were similar to those of C. gloeosporioides (5). The conidia of C. gloeosporioides germinate and form appressoria in response to chemical signals such as host surface wax and the fruitripening hormone ethylene (3). In this study, the spores started to germinate 4 h after incubation with an ethephon suspension. Then, the germ tubes began to swell, and subsequently, differentiation into appressoria with dark thick walls was completed by 8 h. In advanced symptoms, fungal spores of virtually all the appressoria formed primary hyphae within 16 h. However, in the static-symptom fungus spores, no primary hyphae formed by 16 h. The two types of isolates exhibited different growth rates on medium containing apple pectin, Na polypectate, or glucose as the sole carbon. Static-symptom fungi had a >10% reduction in growth (apple pectin, 14.9%; Na polypectate, 27.7%; glucose, 10.4%). The fungal isolates were also genetically characterized by sequencing. ITS regions of rDNA, chitin synthase 1 (CHS1), actin (ACT), and ${\beta}$-tubulin (${\beta}t$) were amplified from isolates using primer pairs ITS 1 and ITS 4 (4), CHS-79F and CHS-354R, ACT-512F and ACT-783R, and T1 and ${\beta}t2$ (5), respectively. The resulting sequences showed 100% identity with sequences of C. gloeosporioides at KC493156, and the sequence of the ${\beta}$t gene showed 100% identity with C. gloeosporioides at JX009557.1. Therefore, sequence data from the four loci studied proves that the isolated pathogen is C. gloeosporioides. We also performed random amplified polymorphic DNA-PCR, which showed clearly differentiated subgroups of C. gloeosporioides genotypes. The clustering of these groups was highly related to the symptom types of the individual strains.

The Characteristics of Progenies derived from Lilium lancifolium and Asiatic hybrid "Dreamland" (참나리와 Asiatic hybrid "Dreamland"의 교잡 후대 특성)

  • Lilium lancifolium, which is one of the Korean native lilies, possesses several useful genetic characteristics such as growth vigor, bulbil formation and resistance to Fusarium. Hybridization was performed to insert useful traits of Asiatic hybrid "Dreamland" as the male parent with upward direction and dark brown spotless on the petal into L. lancifolium as the female parent. The rate of pollen germination of L. lancifolium and "Dreamland" were 30% and 60%, respectively. The pollen germination ratio of $F_1$ progenies at flowering date was 25-57%, which is between the parents'pollen germination ratio. The individual flowering time was between 113 days and 131 days after planting. Of 99% of all plants, the leaf shape index was more than five, which means that the leaf shape of $F_1$ plants is more influenced by female than male parent. The flower color was separated into four types. 51.5% of the progenies showed orange and 44.2% showed light orange, respectively. The colors seemed to be highly influenced by female parents. Mixed colors of parents, orange yellow and light yellow orange, were 3.1%, 1.2%, respectively. Flowering directions were segregated by three different ways, upside, side, and downside. Side direction, which is medium type between female and male parent, was 73%. All progenies showed lots of spot on petals except one offspring. The percentage of bulbil formation on leaf axil was 49.4%. Among those, the maximum number of bulbil formation was 25. The mean number of bulbil formation per plant was five.

Taxonomic Review of the Family Trachipteridae (Lampridiformes) from Korea (한국산 투라치과(Lampridiformes: Trachipteridae) 어류의 분류학적 재검토)

  • The taxonomic review of the family Trachipteridae was carried out based on the Trachipterus ishikawae (7 specimens), Zu cristatus (4 specimens), Desmodema polystictum (2 specimens) collected from the East Sea, South Sea and adjacent waters of Jeju Island of Korea from June 2006 to August 2008. Among them, D. polystictum was revealed as an unrecorded species from Korea, being characterized by the following characters: the ventral edge smooth; body color silvery white with a lot of large dark rounded spot. We proposed the new Korean name of the genus Desmodema as "Jeom-tu-ra-chi-sog", D. polystictum as "Jeom-tu-ra-chi". Z. cristatus has a elongated first six dorsal fin and pelvic fin rays. T. ishikawae has a row of sharp tubercles along the ventral edge, and straightly elongated body form. In meristic characters, the number of dorsal fin rays were 178~195 in T. ishikawae, 137~148 in Z. cristatus and 125~130 in D. polystictum, the number of vertebrae were well distinguished among three species; T. ishikawae (80~83), D. polystictum (72~74) and Z. cristatus (65~68).

Gray Mold of Kalanchoe (Kalanchoe blossfeldiana) Caused by Botrytis cinerea in Korea (Botrytis cinerea에 의한 칼란코에 잿빛곰팡이병)

  • In April of 2002, the gray mold disease occurred severely on kalanchoe (Kalanch oe blossfeldiana) grown in a nursery in Daesan-myon, Changwon City, Korea. The lesions were started with water-soaked spot and the leaves discolored from the tip and became gray or dark and gradully expanded to whole leaf. The conidia and mycelia of pathogen appearedd on flower, flower stalk and stem. Conidia were gray, 1-celled, mostly ellipsoid or ovoid in shape and were 8~17 ${\times}$5~12${\mu}m$ in size. Conidiophores were 13~34${\mu}m$ in size. The sclerotia were formed abundantly on potato-dextrose agar. The optimum temperature for sclerotial formation was $20^{\circ}C$. Pathogenicity of the causal organism was proved on Kalanchoe blossfeldiana according to Koch,s postulate. The causal organism was identified as Botrytis cinerea based on mycological characteristics. This is the first report on gray mold of Kalanchoe(Kalanchoe blossfeldiana) caused by Botrytis cinerea in Korea.

The Visualization of Unstable Combustion in Hybrid Rocket (하이브리드 로켓의 불안정 연소 특성 가시화)

  • The irregular fuel surface was observed by the visualization of hybrid rocket combustion. Even though the test condition maintained oxidizer rich environment, the irregular dark fuel surface was formed as the result of incomplete combustion. In order to investigate the correlation of the characteristics of oxidizer flow and the irregular fuel surface, various flow conditions were imposed such as swirl flow, induced swirl flow by helical fuel configuration and straight flow. Test results revealed no correlation was found between oxidizer flow condition and irregular fuel surface. And this can be a commonly observed phenomena in the tests with different fuel/oxidizer combination. Thus, the irregular fuel surface can be a result of the interaction of blowing flow of vaporized fuel and the boundary layer of oxidizer flow. A further study will be required to confirm the assumption for the formation of irregular fuel surface.
