Stability of Danggwisu-san (Dangguixu-san) Water Extract, a Herbal Medicine, Under Various Storage Conditions (보관조건에 따른 당귀수산의 성분 변화)

  • Objectives Danggwisu-san (Dangguixu-san) is a herbal prescription frequently used to treat pain or swelling caused by contusion. To determine the expiration period through scientific methodology, stability of Danggwisu-san (Dangguixu-san) water extract, a herbal medicine, was examined under various storage conditions and periods. Methods Danggwisu-san (Dangguixu-san) was stored either at room temperature ($23{\pm}2^{\circ}C$), under a refrigerating condition ($4^{\circ}C$) or under a freezing condition ($-18{\pm}2^{\circ}C$) for 0, 1, 2, 3 or 4 weeks and then freeze-dried. Total phenol and total flavonoid amounts were investigated; contents of amygdalin (Prunus persica), paeoniflorin (Paeonia lactiflora), and glycyrrhizin (Glycyrrhiza uralensis) - the marker compounds of Danggwisu-san (Dangguixu-san) - were also analyzed through high performance liquid chromatography (HPLC). Results No significant change in total phenol and total flavonoid amounts was observed under the indicated storage conditions. Moreover, the contents of marker compounds, i.e. amygdalin, paeoniflorin, and glycyrrhizin, did not alter significantly under the indicated conditions, as well. Conclusions Danggwisu-san (Dangguixu-san) was found to be stable up until 4 weeks under the indicated conditions. Further studies on efficacy and long-term stability are warranted to establish the expiration period of Danggwisu-san (Dangguixu-san).

A Case Report of Chronic Insomnia Diagnosed Blood Stasis (어혈로 변증된 불면증 환자의 치험례)

  • Objectives: Insomnia is a common disease in the general population. The purpose of this paper was to suggest that insomnia subtype of blood stasis can be effectively treated with Danggwisu-san and acupuncture. Methods: This is a clinical case report of a patient with chronic insomnia that was diagnosed with blood stasis subtype. I treated him with Danggwisu-san (Dangguixu-san) two to three times daily and with acupuncture and moxibustion two to three times weekly. Results: After short-term treatment, the patient was gradually recovering from deteriorating chronic insomnia and multiple accompanying symptoms such as severe constipation, spontaneous bleeding and edema, darkened face, depressive moods, etc. Conclusions: Results revealed a possibility that chronic insomnia subtype of blood stasis may be successfully treated with Danggwisu-san and acupuncture and moxibustion.

Healing Effect of Danggwisu-san (Dangguixu-san) on Femur Fractured Mice (당귀수산(當歸鬚散)이 대퇴골절 유발 생쥐에 미치는 영향)

  • Objectives This study was designed to evaluate the effects of Danggwisu-san (Dangguixu-san, DG) on bone repair from femur fracture in mice. Methods Mice were randomly divided into 4 groups (normal, control, positive control and DG 300 mg/kg-treated group). In order to investigate the effects of DG on gene expressions in experimental animals with fracture, we measured the levels of bone morphogenetic protein-2 (BMP2), cyclooxygenase-2 (COX2), Sox9, collagen type II alpha 1 chain (Col2a1), runt-related transcription factor 2 (Runx2), osterix genes. After the cytotoxicity test, we analyzed the levels of expression of osteocalcin and Runx2, and tumor necrosis factor-α (TNF-α), a pro-inflammatory cytokine. The process of fusion in the fracture was also investigated by gross examination. Results Through in vivo BMP2, COX2 gene expression significantly decreased. Sox9 significantly increased. Col2a1, Runx2, osterix gene expression also increased as well, but there was no statistical significance. The degree of unilateral fracture fusion investigated by gross examination was significantly faster than those of the other groups. Through in vitro the level of TNF-α in macrophages was increased by DG in a dose-dependent mannerand and 250 and 500 ㎍/mL showed statistical significance. Osteocalcin and Runx2 genes expressions increased when DG was treated in osteoblasts. Conclusions DG promotes the healing of the fracture through the expression of bone repair-related genes and TNF-α production. This study may set the foundation for the clinical application of DG to the patients with bone fractures.
