EVALUATION OF GENETIC TOXICITY FROM ENVIRONMENTAL POLLUTANTS IN DAPHNIA MAGNA AND CHIRONOMUS TENTANS FOR APPLICATION IN ECOLOGICAL RISK ASSESSMENT

  • The genetic toxicity of environmental pollutants, namely, nonylphenol (NP), bisphenol A (BPA) and chloropyriphos (CP) was investigated in aquatic sentinel species, freshwater crustacean, Daphnia magna, and larva of aquatic midge, Chironomus tentans, using Comet assay. Physiological effect of such pollutants was also investigated by studying the specimens' rates of reproduction, growth and survival. Acute toxicity results showed that, as expected, Daphnia was more sensitive than Chironomus to chemical exposure. The order of acute toxicity was CP > NP > BPA in D. magna and NP > CP > BPA in C. tentans. BPA may exert a genotoxic effect on D. magna and C. tentans, given that DNA strand breaks increased in both species exposed to this compound, whereas NP- and CP-induced DNA damage occurred only in C. tentans. In vivo genotoxic data obtained in aquatic sentinel species could provide valuable information for freshwater quality monitoring. The experiments with NP-exposed D. magna showed that the pollutant has long-term effects on reproduction, whereas no short-term effect on DNA integrity was found, being an example of a false-negative result from the biomarkers perspective. This result could be interpreted that other mechanism than genetic alteration might be involved in NP-induced reproduction failure in D. magna. False-positive results from the genotoxic biomarker obtained in BPA-exposed D. magna and in NP-exposed C. tentans make it difficult to use DNA integrity as an early warning biomarker. However, as the mere presence of genotoxic compounds, which are potentially carcinogenic, is of high concern to human and ecosystem health, it could also be important to rapidly and effectively detect genotoxic compounds in the aquatic system in ways that do not necessarily accompany a higher level of alteration. Considering the potential of D. magna and C. tentans as bioindicator species, and the importance of genotoxic biomarkers in ecotoxicity monitoring, DNA damage in these species could provide useful information for environmental risk assessment.

A Study on Damage from the Distributed Imports with the Infraction of the Country of Origin Indication (원산지표시위반 수입물품 유통으로 인한 피해의 연구)

  • This study represents the current situation on the Korean Government's investigation into the imports with the infraction of the country of origin indication law, showing how much those imports affect the purchasers as well as the domestic competitors in Korea. According to the HS 4 unit, the range of imports with the country of origin indication should be 55.2% of all imports, but it turned out there were actually 87.6% of them, according to the number of the import declarations in the first half year of 2010. The government's investigation on the infraction of the country of origin indication is conducted in the stages of customs and distribution into market. As a result of recent 4 years government's investigation into the imports with the infraction of the country of origin indication, the rate of inspection was averagely 3.6% of the reported number of imports in the stage of customs, the rate of the infraction of the country of origin indication cases was 0.14% of the total reported number, and 3.85% of the total actually inspected number. The investigation in the stage of distribution was below 20% level of the stage of customs inspection, on the number of caught cases basis. A survey of 4 categorized imports such as Watches, Tools, Glasses frames, and Clothes, which are assumed to happened frequently with the infraction cases, shows that the decreased sales rate of the domestic competitors was averagely 18.8% and the extent of the damage on consumers was 34.7% of the purchased prices, with both rates resulted from the investigation in the stage of distribution into market.

Superior Cervical Sympathetic Ganglion Block may not Influence Early Brain Damage Induced by Permanent Focal Cerebral Ischemia in Rats (상경부교감신경절블록은 백서의 영구국소뇌허혈에서 초기의 뇌손상에는 영향을 미치지 못한다)

  • Background: Cerebral blood vessels are innervated by sympathetic nerves from the superior cervical ganglion (SCG). The purpose of the present study was to evaluate the neuroprotective effect of superior cervical sympathetic ganglion block in rats subjected to permanent focal cerebral ischemia. Methods: Thirty male Sprague-Dawley rats (270-320 g) were randomly assigned to one of three groups (control, lidocaine and ropivacaine). A brain injury was induced in all rats by middle cerebral artery occlusion with a nylon thread. The animals of the local anesthetic group received $30{\mu}l$ of 2% lidocaine or 0.75% ropivacaine in the SCG. Neurologic scores were assessed 24 hours after brain injury. Brain samples were then collected. The infarct and edema ratios were measured by 2.3.5-triphenyltetrazolium chloride staining. Results: There were no differences in the death rates, neurologic scores, or infarction and edema ratios between the three groups. Conclusions: These findings suggest that superior cervical sympathetic ganglion block may not influence the brain damage induced by permanent focal cerebral ischemia in rats.

Positive association of breastfeeding on respiratory syncytial virus infection in hospitalized infants: a multicenter retrospective study

  • Background: Breastfeeding reportedly reduces the overall frequency of infections. Respiratory syncytial virus (RSV), the most common respiratory pathogen in infants, involves recurrent wheezing and has a pathogenic mechanism related to airway structural damage. Purpose: This study aimed to investigate whether breastfeeding has a beneficial effect against RSV-induced respiratory infection compared to formula feeding among infants in Korea. Methods: We retrospectively reviewed the medical records of infants under 1 year of age who were admitted with RSV infection between January 2016 and February 2018 at the department of pediatrics of 4 hospitals. We investigated the differences in clinical parameters such as cyanosis, chest retraction, combined infection, fever duration, oxygen use, oxygen therapy duration, intensive care unit (ICU) admission, and corticosteroid treatment of exclusive breast milk feeding (BMF), artificial milk formula fed (AMF), and mixed feeding (MF) groups. Results: Among the 411 infants included in our study, 94, 161, and 156 were included in the BMF, MF, and AMF groups, respectively. The rates of oxygen therapy were significantly different among the BMF (4.3%), MF (8.1%), and AMF (13.5 %) groups (P=0.042). The odds ratios (ORs) for oxygen therapy was significantly higher in the AMF group than in the BMF group (adjusted OR, 3.807; 95% confidence interval, 1.22-11.90; P=0.021). The ICU admission rate of the BMF group (1.1%) was lower than that of the MF (3.5%) and AMF (4.5%) groups; however, the dissimilarity was not statistically significant (P=0.338). Conclusion: The severity of RSV infection requiring oxygen therapy was lower in the BMF than the AMF group. This protective role of human milk on RSV infection might decrease the need for oxygen therapy suggesting less airway damage.

Degradation Damage Evaluation for Turbine Structural Components by Electrochemical Reactivation Polarization Test (전기화학적 재활성화 분극시험에 의한 터빈부재의 열화손상 평가)

  • The extent of materials deterioration can be evaluated accurately by mechanical test such as impact test or creep test. But it is almost impossible to extract a large test specimen from in-service components. Thus material degradation evaluation by non-destructive method is earnestly required. In this paper, the material degradation for virgin and several aged materials of a Cr-Mo-V steel, which is an candidated as structural material of the turbine casing components for electric power plant, is nondestructively evaluated by reactivation polarization testing method. And, the results obtained from the test are compared with those in small punch(SP) tests recommended as a semi-nondestructive testing method using miniaturized specimen. In contrast to the aged materials up to 1,000hrs which exhibit the degradation behaviors with increased ${\Delta}[DBTT]_{SP}$, the improvement of mechanical property can be observed on the 2,000hrs and 3,000hrs aged materials. This is because of the softening of material due to the carbide precipitation, the increase of ferritic structures and the recovery of dislocation microstructure by long-time heat treatment. The reactivation rates($I_R/I_{Crit},\;Q_R/Q_{Crit}$) calculated by reactivation current densityt ($I_R$) and charge($Q_R$) in the polarization curves exhibit a good correlation with ${\Delta}[DBTT]_{SP}$ behaviors.

Effects of Talmyung-san on the Cultured Rat Myocardiac Cell and Vascular Smooth Muscle Cell (탈명산(奪命散)이 배양심근세포(培養心筋細胞) 및 혈관평골근세포(血管平滑筋細胞)에 미치는 영향(影響))

  • Objectives : Talmyung-san(TMS) has been used for treatment of brain diseases in Chinese traditional medicine. However, little is known about the mechanism by which TMS rescues brain cells from ischemic damages. To elucidate the protective mechanisms of TMS, we execute experiments. Methods : The effects of TMS on ischemia/reperfusion-induced cytotoxicity and generation of nitric oxide(NO) are investigated in primary neonatal myocardial cells and A7rS, aortic smooth muscle cell line. Results : Ischemia/reperfusion itself induces severe myocardial cell death in vitro. However, treatment of the cells with TMS significantly reduces both ischemia/reperfusion-induced myocardial cell death and LDH release. In addition, pretreatment of TMS before reperfusion recovers the lose of beating rates alter ischemia/reperfusion. For a while, the water extract of TMS stimulates myocardial cells to produce NO in a dose dependent manner and it protects the damage of ischemia/reperfusion-induced myocardial cells. Furthermore, the protective effects of the water extract of TMS is mimicked by treatment of sodium nitroprusside, an exogenous NO donor. NG-monomethyl-L-arginine (NGMMA), a specific inhibitor of nitric oxide synthase(NOS), significantly blocks the protective effects of TMS on the cells after ischemia/reperfusion. In addition, on ischemia the water extract of TMS induce NO in A7r5 cell. Conclusions : Taken together, we suggest that the protective effects of TMS against ischemia/reperfusion-induced myocardial damages may be mediated by NO production of myocardial and vascular smooth muscle cell during ischemic condition.

Prediction of Spring Rate and Initial Failure Load due to Material Properties of Composite Leaf Spring (복합재 판스프링의 재료특성에 따른 스프링 강성변화와 초기 파단하중 예측)

  • This paper presented analysis methods for adapting E-glass fiber/epoxy composite (GFRP) materials to an automotive leaf spring. It focused on the static behaviors of the leaf spring due to the material composition and its fiber orientation. The material properties of the GFRP composite were directly measured based on the ASTM standard test. A reverse implementation was performed to obtain the complete set of in-situ fiber and matrix properties from the ply test results. Next, the spring rates of the composite leaf spring were examined according to the variation of material parameters such as the fiber angles and resin contents of the composite material. Finally, progressive failure analysis was conducted to identify the initial failure load by means of an elastic stress analysis and specific damage criteria. As a result, it was found that damage first occurred along the edge of the leaf spring owing to the shear stresses.

Effects of Bacterial Leaf Blight Occurrence on Rice Yield and Grain Quality in Different Rice Growth Stage (벼 생육시기별 흰잎마름병 발병이 쌀 수량감소 및 미질에 미치는 영향)

  • We investigated the first symptom emerging date of rice bacterial leaf blight disease during four years from 2002 to 2005. The disease occurrence date was earlier 20$\sim$30 days in 2005 than that of 2002. The damage in different rice growth stage by the bacterial disease on rice yield and grain quality in southern part of Korea was examined. The disease decreased rice yield following by increased infection rates. Slight loss in rice yield and brown head rice rate were observed at below 10% level of infected leaf area, while over 25% infected leaf area caused the significant decrease in rice yield and brown head rice rate. More than 50% of the infected leaf area rate caused 29% yield reduction in case of infection at panicle formation stage and 18% of brown head rice rate. Cooked rice quality was also affected in the diseased rice by damage in palatability score and viscosity.

Dispersal Pattern of the Black Pine Bast Scale, Matsucoccus thunbergianae (Homoptera : Margarodidae), in Korea (솔껍질깍지벌레(Matsucoccus thunbergianae)의 확산(擴散) 유형(類型))

  • The black pine bast scale (BPBS), Matsucoccus thunbergianae, causes serious damage to Japanese black pine forests in the southern part of Korean peninsula. After first detection of its damage in Kohung, Chonnam Province in 1963, the distribution has been expanding gradually year by year. Since 1983, extensive surveys for BPBS distributions have been carried out by Korea Forest Research Institute. We used cumulative data for BPBS distribution and analyzed the dispersal pattern. We divided expansions into three directions such as north along the west coast, northeast towards the inland, and east along the south coast. In the direction of northeast the dispersal rate was approximately 4.3km per year in dense forests of Japanese black pine, but it slowed down since the early 1990s. The annual dispersal rates were approximately 5.9 and 3.3km in north and east directions, respectively.

Studies on Damage, Emergence, and Overwintering of the Chestnut Curculio, Curculio sikkimensis in Chonnam Province (전남지방에 있어서 밤바구미의 피해상황, 우화 및 월동에 관한 연구)

  • This study was conducted to investigate the bionomics of Curculio sikkimensis in Chonnam province from 1981 to 1983. Damage rates of the chestunt were $26.4\%$ in Damyang, $21.1\%$ in Gwangyang, and $12.9\%$ in Seungju. Adults occurred from the mid-July to the mid-September and showed peak from the mid-August to the early-September. Larvae escaped the chestnut from the early-October to the early-November. Overwintering larvae emerged $3.3\%$ in the first year, $22.78\%$ in the second year and continously overwintered $1.67\%$ in the third year. Larvae showed the highest distribution at $1.67\%$ of soil depth during the overwintering. Mortality of overwintering larvae and course of time showed the linearly correlation. Period of each stage was 5.9 days for egg and 17.17 days for pupa. Adult longevity was 9.10 days for female and 7.03 days for male.
