Melatonin Attenuates Mitochondrial Damage in Aristolochic Acid-Induced Acute Kidney Injury

  • Aristolochic acid (AA), extracted from Aristolochiaceae plants, plays an essential role in traditional herbal medicines and is used for different diseases. However, AA has been found to be nephrotoxic and is known to cause aristolochic acid nephropathy (AAN). AA-induced acute kidney injury (AKI) is a syndrome in AAN with a high morbidity that manifests mitochondrial damage as a key part of its pathological progression. Melatonin primarily serves as a mitochondria-targeted antioxidant. However, its mitochondrial protective role in AA-induced AKI is barely reported. In this study, mice were administrated 2.5 mg/kg AA to induce AKI. Melatonin reduced the increase in Upro and Scr and attenuated the necrosis and atrophy of renal proximal tubules in mice exposed to AA. Melatonin suppressed ROS generation, MDA levels and iNOS expression and increased SOD activities in vivo and in vitro. Intriguingly, the in vivo study revealed that melatonin decreased mitochondrial fragmentation in renal proximal tubular cells and increased ATP levels in kidney tissues in response to AA. In vitro, melatonin restored the mitochondrial membrane potential (MMP) in NRK-52E and HK-2 cells and led to an elevation in ATP levels. Confocal immunofluorescence data showed that puncta containing Mito-tracker and GFP-LC3A/B were reduced, thereby impeding the mitophagy of tubular epithelial cells. Furthermore, melatonin decreased LC3A/B-II expression and increased p62 expression. The apoptosis of tubular epithelial cells induced by AA was decreased. Therefore, our findings revealed that melatonin could prevent AA-induced AKI by attenuating mitochondrial damage, which may provide a potential therapeutic method for renal AA toxicity.

Classifier Combination Based Source Identification for Cell Phone Images

  • Rapid popularization of smart cell phone equipped with camera has led to a number of new legal and criminal problems related to multimedia such as digital image, which makes cell phone source identification an important branch of digital image forensics. This paper proposes a classifier combination based source identification strategy for cell phone images. To identify the outlier cell phone models of the training sets in multi-class classifier, a one-class classifier is orderly used in the framework. Feature vectors including color filter array (CFA) interpolation coefficients estimation and multi-feature fusion is employed to verify the effectiveness of the classifier combination strategy. Experimental results demonstrate that for different feature sets, our method presents high accuracy of source identification both for the cell phone in the training sets and the outliers.

  • In this article, we have done some analysis about the collision-avoidance specialty of “pressing complexion” and “pressing danger”in the meet process of two boats, and offered a mathematic model of differential coefficient countermeasure decision for collision-avoidance, which adapt to the right complexion. The basal idea is, in the right condition whatever do the coming boat do, and our boat will always adept dynamic, continuous and the best countermeasure. When both the controlling capabilities of two boats have advantage and inferior position, we can working-out with the qualitative differential coefficient countermeasure.

  • The foam concrete was fabricated by adding the foaming agent which composite ordinary Portland cement with plant and animal protein into cement paste, and the electromagnetic wave absorption properties were studied for the first time as well. The studies showed that the electromagnetic waves can be absorbed by multiple reflections and scattering within the porous material. Thickness and filling ratio have a great influence on the electromagnetic wave absorbing properties in 2-18 GHz of the foam concrete, the greater the thickness, the better the performance of absorption; filling ratio was about 52 vol.%, the absorbing properties achieved the best.

Overexpression of Long Non-Coding RNA HOTAIR Promotes Tumor Growth and Metastasis in Human Osteosarcoma

  • Human osteosarcoma usually presented a high tendency to metastatic spread and caused poor outcomes, however, the underlying mechanism was still largely unknown. In the present study, using a series of in vitro experiments and an animal model, we investigated the roles of HOX antisense intergenic RNA (HOTAIR) during the proliferation and invasion of osteosarcoma. According with our results, HOTAIR was commonly overexpressed in osteosarcoma, which significantly correlated with advanced tumor stage, highly histological grade and poor prognosis. In vitro and in vivo experiments demonstrated that knockdown of HOTAIR could notably suppress cellular proliferation, inhibit invasion and decrease the secretion of MMP2 and MMP9 in osteosarcoma. Collectively, our results suggested that HOTAIR might be a potent therapeutic target for osteosarcoma.
