THE USE OF NEAR INFRARED REFLECTANCE SPECTROSCOPY(NIRS) TO PREDICT CHEMICAL COMPOSITION ON MAIZE SILAGE

  • Microbiological examination of silage is of little value in gauging the outcome of silage, and so chemical analysis is more reliable and meaningful indicator of quality. On the other hand chemical assessments of the principal fermentation products provide an unequivocal basis on which to judge quality. Livestock require energy, protein, minerals and vitamins from their food. While fresh forages provide these essential items, conserved forages on the other hand may be deficient in one or more of them. The aim of the conservation process is to preserve as many of the original nutrients as possible, particularly energy and protein components (Woolford, 1984). Silage fermentation is important to preservation of forage with respect of feeding value and animal performance. Chemical and bacteriological changes in the silo during the fermentation process can affect adversely nutrient yield and quality (Moe and Carr, 1984). Many of the important chemical components of silage must be assayed in fresh or by extraction of the fresh material, since drying either by heat or lyophilisation, volatilises components such as acids or nitrogenous components, or effects conversion to other compounds (Abrams et al., 1987). Maize silage dorms the basis of winter rations for the vast majority of dairy and beef cattle production in Uruguay. Since nutrient intake, particularly energy, from forages is influenced by both voluntary dry matter intake and digestibility; there is a need for a rapid technique for predicting these parameters in farm advisory systems. Near Infrared Reflectance Spectroscopy (NIRS) is increasingly used as a rapid, accurate method of evaluating chemical constituents in cereals and dried forages. For many years NIRS was applied to assess chemical composition in dry materials (Norris et al., 1976, Flinn et al., 1992; Murray, 1993, De Boever et al., 1996, De la Roza et al., 1998). The objectives of this study were (1) to determine the potential of NIRS to assess the chemical composition of dried maize samples and (2) to attempt calibrations on undried samples either for farm advisory systems or for animal nutrition research purposes in Uruguay. NIRS were used to assess the chemical composition of whole - plant maize silage samples (Zea mays, L). A representative population of samples (n = 350) covering a wide distribution in chemical characteristics were used. Samples were scanned at 2 nm intervals over the wavelength range 400-2500 nm in a NIRS 6500 (NIRSystems, Silver Spring, MD, USA) in reflectance mode. Cross validation was used to avoid overfitting of the equations. The optimum calibrations were selected on the basis of minimizing the standard error of cross validation (SECV). The calibration statistics were R$^2$ 0. 86 (SECV: 11.4), 0.90 (SECV: 5.7), 0.90 (SECV: 16.9) for dry matter (DM), crude protein (CP), acid detergent fiber (ADF) in g kg$\^$-1/ on dry matter, respectively for maize silage samples. This work demonstrates the potential of NIRS to analyse whole - maize silage in a wide range of chemical characteristics for both advisory farm and nutritive evaluation.

Evaluation of Application of Possibility of Visual Surveillance System for Cow Heat Detection

  • This study was conducted to evaluate a visual surveillance system. The advancement of recording technology and network service make it easy to record and transfer the videos. Moreover, progressed recognition technology help to make a distinction each other. Cows show distinguishing behaviors during their estrus period. The mounting is one of the behaviors. The result was different depending on the breed of the cows and the size of the farm. In the case of Korean native cattle, the estrus detection rate was 71.15%, however, dairy cows, the estrus detection rate was 39.38%. At the farms having below 6 modules, the estrus detection rate was 87.41%. On the other hand, at the farms having over 6 modules, the estrus detection rate was 77.78%. With the proper progress, the visual surveillance system can be used to detect heat detection.

Use of the Non-electrophoretic Method to Detect Testis Specific Protein Gene for Sexing in Preimplantation Bovine Embryos

  • Testis-specific protein (TSPY) is a Y-specific gene, with up to 200 copy numbers in bulls. In order to make bovine embryo sexing under farm condition more feasible, the possibility of using a non-electrophoretic method to detect the TSPY gene for sexing bovine early embryos was examined. Primers were designed to amplify a portion of the TSPY gene and a common gene as an internal control primer. PCR optimization was carried out using a DNA template from bovine whole blood. Furthermore, embryo samples were diagnosed by this method and the sexing results were contrasted with those of the Loop-Mediated Isothermal Amplification (LAMP) method. The results showed that TSPY was as reliable a sexing method as LAMP. Forty-three morula and blastocyst embryos collected from superovulated donor dairy cattle were sexed by this method, and twenty-one embryos judged to be female embryos were transferred non-surgically to recipients 6 to 8 days after natural estrus. Out of 21 recipients, 9 were pregnant (42.86%) and all delivered female calves. The results showed that the sex predicted by this protocol was 100% accurate. In conclusion, the TSPY gene was a good male specific marker and indicated that a non-electrophoretic method was feasible and accurate to detect the TSPY gene for sexing preimplantation bovine embryos.

유우농장에서 발생한 소바이러스성 백혈병의 병리학적 및 혈청학적 조사 (Pathological and serological detection of bovine viral leukosis in a dairy farm in Jeonbuk province)

  • Bovine viral leukosis is a viral disease of cattle characterized by the development of tumors in the lymphatic tissue. A female Holstein, 3-year-old, was submitted for diagnosis at the Diagnostic laboratory, Chonbuk National University. Clinical sign of the affected animal showed emaciation, enlargement of superficial lymph node and mild diarrhea. Remarkable lesions were enlargement of many internal lymph nodes. Histopathology revealed excessive neoplastic lymphoid cells characteristic of BVL infection. Subsequently, serums from all cattle were collected and serological examination was done where a 85% seropositive rate was detected using ELISA test. ELISA method showed a comparatively 75% higher detection rate than the agar gel immunodiffusion (AGID) test (85% vs 40%). Serologically positive cattle were variably detected in all ages from under 1 year to over 6 year of age. Hematological examination consistently showed leukocytosis and a differential lymphocytosis of seropositive cattle. Detailed comparative pathological and serological data diagnosed the presence of bovine viral leukosis.

  • Total 51 calves born from both 28 seropositive and 23 seronegative dams were subjected to study both prenatal and postnatal infections of bovine leukemia virus (BLV), and the duration of passive colostral antibody by means of immunodiffusion (ID) test. All calves were tested for precolostral and postcolostral periods by 16 months of age. The results were as follows: 1. Of 28 precolostral sera of the calves born from infected dams, one appeared positive, indicating in utero BLV infection from the dam. 2. BLV-antibody test for the postcolostral sera of the calves born from seropositive or seronegative dams showed that the colostral antibody of the calves disappeared from 2 to 6 months of age, and the increase of the number of seropositive calves initiated from 3 to 4 months of age indicated postnatal infection.

Salmonella dublin에 의한 소의 살모넬라증의 발생 (An Acute Outbreak of Calf Salmonellosis Caused by Salmonella dublin)

  • An epizootic of calf diarrhea occurred in an extensive dairy farm located in Gangwondo province in December of 1985. The patients showing fever, chills, complete anorexia and watery diarrhea sometimes dysentery were concentrated among the calves ranged from 8 days to 3 months old and many of them died in acute course. The five carcases were examined by pathological and bacteriological means. The predominant gross lesions were edematous swelling of the mesenteric lymph nodes, profuse catarrhal or diphtheritic enteritis and extensive purulent bronchopnenomonia or lobar fibrinous pneumonia. Microscopically the lesions of the liver and spleen were characterized by coagulative necrotic foci and granulomatous nodules. The spleen and lymph node showed also proliferative changes of reticular cells and involution of intestinal lymphatic nodules were noticed. The strains of Salmonella(S) species isolated from the liver, spleen, bile juice, peritoneal fluid and thoracic fluid were identified serologically as S. dublin. These clinical and pathological findings of the disease were those of acute enteritis form of salmonellosis and characterized by high morbidity and mortality among the calf herd. The report also signifies the first description of an epizootic of bovine salmonellosis caused by S. dublin in Korea.

홀스타인 독우(犢牛)에 발생(發生)한 질산염중독(窒酸鹽中毒) (Nitrate Poisoning Occurred in Holstein Calves)

  • Nitrate poisoning was observed in Holstein calves raising for beef purpose in a dairy-farm of Gyeongbug province. The results observed were summarized as follows: 1. These calves had been fed mainly cornstalk, and a qualitative reaction by "dephenylamine blue test" for nitrate was positive in the serum of the affected calves and in the cornstalk provided. 2. In the clinical signs, the affected calves were suddenly recumbent without premonition and then dyspnea, followed by death. Death also occurred after inappetence, depression, dyspnea, cyanosis of mucous membrane, and terminal anoxic convulsions were observed. 3. In the macroscopical findings, the blood was dark brown (chocolate in color) and more or less poor coagulative. Petechiae of epicardium, severe cloudy swelling of liver, cloudy swelling and hemorrhage of kidney, congestion and hemorrhage of spleen, congestion and edema of lung, and congestion af abomasum and small intestine were observed in these cases. 4. Microscopical changes observed in these cases were peripheral congestion of lobules, centrilobular necrosis and sinusoidal dilatation in liver, hydropic degeneration of hepatic cells, hemorrhage and edema of kidney, necrosis of convoluted-tubular epithelium, and dilatation of Bowman's space and convoluted tubule. There were also congestion and hemorrhage of spleen, hemorrhage and edema of lung, cloudy swelling of myocardium, and congestion and hemorrhage of various organs.

소유역의 토지이용에 따른 비점원오염 부하량 (Nonpoint Source Pollution Loadings from Land Uses on Small Watersheds)

  • Nonpoint source (NPS) pollution from small watersheds has recently been brought into attention as a potential pollutant to streams and tnbutaries, as majority of them are experiencing water quality degradation. This necessiates the quantification of NPS loadings from agricultural and forested lands. And this study attempts to quantify daily loadings from forested and farm lands using hydrologic and water quality monitoring. The hydrologic monitoring program consists of five water level gauging stations along creeks and stream at the Banweol reservoir watershed having 1220 hectare in size. Water sam pies were taken and analyzeel periodically at the streamf low gaging sites and tributaries. Soil samples were also taken and the chemical constitutes analyzed. The primary results indicate that the major sources of pollution were small villages and dairy farms on the watersheds, constituting two-third of total nutrient loadings to the reservoir. However, the loadings from paddies and upland areas may cause a problem to the water quality of the reservoir and stream as the measured levels of total nitrogen and phosporus are not low enough to ignore. Further studies are needed to quantify the effects of landuses and treatments at a watershed scale.

  • 충남대학교(忠南大學校) 농과대학(慶科大學) 부속동물사육장(附屬動物飼育場)에서 사육중인 한국재래산양(韓國在來山羊) 및 면양(緬羊) 각(各) 3 두(頭)에 대하여 방목중(放牧中)에 있어서 forage material의 섭식상(攝食狀)을 연구할 목적(目的)으로 고안(考案)한 방법에 의하여 식도루관형성술(食道瘻管形成術)을 실시하여 다음과 같이 보고(報告)한다. 1. 고안(考案)된 식도루관형성술의 시술(施術)이 용이(容易)하였으며 섭식상(攝食狀)의 연구에 적합(適合)하였다. 2. 본(本) 법(法)에 의한 식도루관은 장착(裝着)이 확실(確實)하여 장기간(長期間)(1년이상(年以上))에 걸쳐 공시(供試)할 수 있었다.

  • A tractor attached TMR(model 430) mixer has been developed in a previous study. However, the mixer was found to be improved through field applications in its capacity, manufacturing cost, ergonomic design and power-train requirement. The TMR mixer was modified into a model TMR500, approved by Institute of Agricultural Mechanization, as follows : 1. Roughage cutting system was seperated from the mixer, resulting in the 33% reduction of manufacturing cost. 2. Enlarged hopper capacity enabled to feed 60 heads at a batch. 3. Hydraulically controlled gate system saved ergonomic man power. 4. Power transmission system was changed from a chain-sprocket system(27:1) to the gear-train reduction system(38.6:1) to satisfy the recommended use of 540rpm PTO input.
