The Factors on Somatic Embryogenesis of Soybean [Glycine max. (L.) Merrill]

  • To enhance in vitro plantlet regeneration efficiency of soybean through embryogenesis, the culture conditions such as material part and size of immature seed, 2,4-D, pH and solidifying agents for somatic embryogenesis were investigated. Somatic embryogenesis was induced from the immature embryo, immature cotyledon and embryonic axis explants of the immature seed on MS medium supplemented with 2.0 mg/L 2,4-D. The highest rate (up to 22.9%) of somatic embryogenesis was obtained from the immature cotyledon, following embryonic axis and the immature embryo. The rate varied with the developmental stages of seed. The maximum rate (25.4%) of embryogenesis was obtained from 3-4 mm length of the seed (after 25 days of flowering). The optimum concentration of 2,4-D for embryogenesis was 10 mg/L. The optimum pH was at 5.8 and solidifying agent for medium was better with 0.4% gelrite than with agar. For rapid multiplication of shoot tips from the germinating somatic embryos, they were cultured on MS medium containing 2 mg/L indole-3-butyyic acid (IBA) and 1 mg/L 6-benzyladenine (BA). After then somatic embryos with one and three cotyledons were transferred to the growth regulator free medium. The medium exhibited the higher rate (ca. 50%) of development than the multiplication medium.

  • For the investigation of resources in Codonopsis species, the studies were carried out to evaluate the pharmaco-constituents from the leaves of Codonopsis lanceolata (Campanulaceae) whose roots have been used to antitussive, expectorant, detoxicate, tonic, edible, etc. as a folk medicine in Korea. From the EtOAc and BuOH fractions of MeOH extract, three flavonoid compounds, $luteolin-7-O-{\beta}-_D-glucopyranoside$$(C_{21}H_{20}O_{1i},\;mp\;254{\sim}255^{\circ}$, compound 1), $luteolin-5-O-{\beta}-_D-glucopyranoside$$(C_{21}H_{20}O_{1i},\;mp\;279{\sim}281^{\circ}$, compound 2) and luteolin $(C_{15}H_{10}O_6,\;mp\;327{\sim}330^{\circ}$, compound 3) were isolated and identified on the basis of their physicochemical properties and spectroscopic evidences(UV, IR, $^1H-NMR$, $^{13}C-NMR$, MS etc.) in comparison with authentics respectively.

Effects of Applying Microbial Additive Inoculants to Spent Mushroom Substrate (Flammulina velutipes) on Rumen Fermentation and Total-tract Nutrient Digestibility in Hanwoo Steers (팽이버섯 부산물 발효에 따른 한우 거세우 반추위 성상 및 소화율에 미치는 영향)

  • We inoculated a spent mushroom substrate from Flammulina velutipes (SMSF) with a microbial additive and assessed the effects on chemical composition, ruminal fermentation parameters, and total-tract nutrient digestibility. In Exp. 1, three cannulated Hanwoo steers were used in an in situ trial to determine the degradation kinetics of dry matter (DM) and crude protein (CP). In Exp. 2, three Hanwoo steers were randomly assigned to experimental diets according to a $3{\times}3$ Latin square for a 3-week period (2 weeks for adaptation and 1 week for sample collection). Experimental diets included the control diet (3.75 kg/d formulated concentrate mixture + 1.25 kg/d rice straw), SMSF diet (3.19 kg/d formulated concentrate mixture + 1.25 kg/d rice straw + 0.56 kg/d SMSF), and inoculated SMSF (ISMSF) diet (3.19 kg/d formulated concentrate mixture + 1.25 kg/d rice straw + 0.56 kg/d ISMSF). The chemical composition of ISMSF did not differ from that of SMSF. Microbial additive inoculation decreased pH (P<0.05) and improved preservation for SMSF. The percentages of DM, neutral detergent fiber (NDF), and acid detergent fiber (ADF) in ISMSF were slightly lesser than those in SMSF. Ruminal fermentation characteristics and total-tract nutrient digestibility were not affected by diet. Overall, microbial additive inoculation improved preservation for SMSF and may allow improved digestion in the rumen; however, the total digestible nutrients (TDN) of SMSF and ISMSF diets were slightly lesser than the control diet. The ISMSF can be used as an alternative feedstuff to partially replace formulated concentrate feed.

  • Vitamin D dependent rickets type 1A (VDDR1A) is an autosomal recessive disorder caused by mutations in CYP27B1. Clinical findings are growth retardation, hypotonia, muscle weakness, hypocalcemic seizures, and radiological features of rickets. We aimed to present the VDDR1A case with a genetic study of CYP27B1. The 14-month-old boy was admitted to the hospital due to a seizure. Serum calcium, phosphorus, alkaline phosphatase, parathyroid hormone (PTH), 25(OH) vitamin D, and 1,25(OH)2 vitamin D values were 5.1 mg/dL, 3.7 mg/dL, 705 IU/L, 429 pg/mL, 24.9 ng/mL, and 8.8 pg/mL, respectively. Radiological study showed cupping and fraying of the distal ulna and radius. The molecular genetic study revealed that the patient had a compound heterozygous mutation, $Phe443Profs^*24$ and c.589+1G>A, in CYP27B1. Genetic analysis of the family members presented that the mother was heterozygous for the mutation c.589+1G>A, and that the father was heterozygous for $Phe443Profs^*24$. The patient was treated with calcium lactate and calcitriol. Until now, six Korean patients with VDDR1A have been studied. Including this case, Korean patients with VDDR1A were found to have only three different mutations in 14 alleles, indicating that the mutation in the CYP27B1 gene is homogeneous in the Korean population.

One Alpha-hydroxycholecalciferol Improves Growth Performance, Tibia Quality, and Meat Color of Broilers Fed Calcium- and Phosphorus-Deficient Diets

  • An experiment was conducted to evaluate the effects of one alpha-hydroxycholecalciferol ($1{\alpha}$-OH $D_3$) on growth performance, tibia quality, and skin and meat color in broilers fed calcium (Ca)- and phosphorus (P)-deficient corn-soybean meal diets. A total of 288 male Ross broilers, at 21 days of age, were randomly assigned to three treatments with eight cages per treatment. Three levels of $1{\alpha}$-OH $D_3$ (0, 5, and 10 ${\mu}g$/kg) were added to a basal diet (0.50% Ca, 0.13% non-phytate phosphorus (NPP), and 0.35% total phosphorus (tP)) without vitamin $D_3$. As a result of this study, the addition of $1{\alpha}$-OH $D_3$ increased body weight gain (p<0.001), feed intake (p = 0.007), feed efficiency (p<0.001), tibia weight (p = 0.002), length (p<0.001), breaking-strength (p = 0.012), ash (p<0.001), Ca (p<0.001), and P content (p = 0.004). Dietary $1{\alpha}$-OH $D_3$ enhanced breast meat yellowness (p = 0.015) and the length and weight of the small intestine of the broilers. Moreover, $1{\alpha}$-OH $D_3$ decreased serum Ca concentration (p = 0.074) and breast meat redness (p = 0.010). These results indicate that the $1{\alpha}$-OH $D_3$ improves growth, tibia quality, and meat color in broilers fed Ca- and P-deficient corn-soybean meal diets.

Effects of Water Temperature and Photoperiod on the Oxygen Consumption Rate of Juvenile Dark-banded Rockfish, Sebastes inermis (수온과 광주기에 따른 볼락, Sebastes inermis 치어의 산소 소비율)

  • An experiment was conducted to investigate the effects of four water temperatures (10, 15, 20, and $25^{\circ}C$) in combination with three photoperiods (24L:0D, 12L: 12D, and OL:24D) on the oxygen consumption rate of juvenile dark-banded rockfish, Sebastes inermis (mean body weight $20.5{\pm}0.7g$). The oxygen consumption rates of S. inermis were measured in triplicate for 24 hours using a continuous flow-through respirometer. Different combinations of water temperatures and photoperiods resulted in significant differences in the mean oxygen consumption rate of S. inermis (P<0.001). The oxygen consumption increased with increasing water temperatures for all photoperiod treatments (P<0.01). Mean oxygen consumption rates at 10, 15,20 and $25^{\circ}C$ ranged $178.3\sim283.5,\;386.7\sim530.7,\;529.2\sim754.3$ and $590.0\sim785.5mg\;O_2kg^{-1}h^{-1}$, respectively. $Q_{10}$ values ranged $3.17\sim5.51$ between 10 and $15^{\circ}C,\;1.87\sim2.10$ between 15 and $20^{\circ}C$ and $1.08\siml.24$ between 20 and $25^{\circ}C$, respectively. Fish held in continuous darkness (OL:24D) used consistently less okygen than fish exposed to continuous light (P<0.05). The mean oxygen consumption offish in a 12L:12D photoperiod was higher than that offish in 24L:0D and 0L:24D photoperiods under all temperature treatments except $10^{\circ}C$. The oxygen consumption of fish exposed to the 12L:12D photoperiod was significantly higher during the light phase than during the dark phase under all temperature treatments except $10^{\circ}C\;(P<0.05)$. This study provides empirical data for estimating oxygen consumption of S. inermis under given condition. This result has application for culture management and bioenergetic model for growth of this species.

Endovascular Treatment of Dural Arteriovenous Fistulas : Single Center Experience

  • Objective : Treatment of intracranial dural arteriovenous fistulas (dAVFs) remains a challenge. However, after introduction of Onyx, transarterial approach is the preferred treatment option in many centers. We report our experience of dAVFs embolization with special emphasis on transarterial approach. Methods : Seventeen embolization procedures were performed in 13 patients with dAVFs between Jan 2009 and Oct 2014. Clinical symptoms, location and type of fistulas, embolization methods, complications, radiological and clinical outcomes were evaluated using charts and PACS images. Results : All 13 patients had symptomatic lesions. The locations of fistulas were transverse-sigmoid sinus in 6, middle fossa dura in 4, cavernous sinus in 2, and superior sagittal sinus in 1 patient. Cognard types were as follows : I in 4, IIa in 2, IIa+IIb in 5, and IV in 2. Embolization procedures were performed ${\geq}2$ times in 3 patients. Nine patients were treated with transarterial Onyx embolization alone. One of these required direct surgical puncture of middle meningeal artery. Complete obliteration of fistulas was achieved in 11/13 (85%) patients. There were no complications except for 1 case of Onyx migration in cavernous dAVF. Modified Rankin scale score at post-operative 3 months were 0 in 11, and 3 in 2 patients. Conclusion : Transarterial Onyx embolization can be a first line therapeutic option in patients with dAVFs. However, transvenous approach should be tried first in cavernous sinus dAVF because of the risk of intracranial migration of liquid embolic materials. Furthermore, combined surgical endovascular approach can be considered as a useful option in inaccessible route.

Effects of The pilose antler of Cervus Korean TEMMINCK var. mantchuricus Swinhoe(DAS), herbal acupuncture solution on suppression of collagenolysis and bone resorption in mouse calvarial osteoblasts (녹용약침액이 mouse의 두개골 골아세포에서 collagen용해와 골재흡수에 미치는 효과)

  • 본 실험에서는 녹용 약침액의 항 골재흡수 속성을 조사하였다. PTH, $1,25(OH)_2D_3$와 IL-1을 각각 골재흡수 인자로 사용하여 생쥐의 두개골에서 osteoblast 세포를 격리, 배양, 그리고 자극시켰을 때 collagenolysis의 증가를 보였다. 두 가지를 동시에 사용한 결과, IL-1은 골재흡수성을 촉진시키고 재 흡수력을 생산하였다. In vitro에서의 세포독성 결과는 $1-200{\mu}g/ml$의 녹용 약침액 농도 분포에서 무세포독성을 보였다. 또한 녹용 약침액은 생쥐의 두개골 골아세포 내에서 PTH (2 unit/ml), IL-$1{\alpha}$ (1 ng/ml), $1,25(OH)_2D_3$ (10 ng/ml), IL-$1{\alpha}$ 및 IL-$1{\beta}$로 인해 유발된 collagenolysis에 대해서 대항하는 보호활동성을 나타내었다. 녹용약침액은 IL-$1{\alpha}$ 와 IL-$1{\beta}$로 인해 유발된 collagenolysis에 대항하는 보호활동성을 지녔다. DAS는 IL-$1{\alpha}$와 IL-$1{\beta}$로 인해 촉진된 골재 흡수력을 억제하는 효과를 보였다. 이와 같은 결과는 녹용약침액이 골다공증과 연관된 질환에 대해서 매우 안정적인 임상적 사용이 가능한 것을 관찰할 수 있으므로 추후 이와 관련한 지속적인 연구가 필요할 것으로 사료되었다.

  • The polysaccharide C prepared from gum tragacanth powder (U. S. P. grade) by the precipitation method with 85% ethanol was a neutral polysaccharide, $[{\alpha}]^{30}_D-72.2$. The polysaccharide C consisted of L-rhamnose, D-xylose, L-arabinose and D-galactose in the molar ratio 2:1:17:9 (Table 1, 2, 3, ). The polysaccharide C was methylated with dimethylsulphate and 40% NaOH, and Purdies regent. The hydrolyzate of fully methlated product ($[{\alpha}]^{22}_D-102$ in chloroform, the methoxy content 40.6%) was composed of 2, 3, 5-tri-O-methyl-L-arabofuranose (I), 3,4-di-O-methyl-L-rhamnopyranose (II), 2,3-di-O-methyl-D-xylose (III), 2,3,4-tri-O-methyl-D-galactopyranose (IV), 2,4-di-O-methyl-L-arabopyranose (?), 2,4-di-O-methyl-D-galactose(VI), 2-O-methyl-D-arabinose (VII), and L-arabopyranose(VIII) (Table 4, 5, and Fig. 4). The first partial hydrolysis (A) of the polysaccharide C with 0.05N-HCl for 4.5 hours at $80-85^{\circ}C$ released only L-arabinose: the second hydrolysis (B) with 0.1N-HCl for 5 hours at $80-85^{\circ}C$, L-arabinose and D-galactose; and the third hydrolysis (C) with 0.3N-HCl at $90-95^{\circ}C$ in sealed tube, L-rhamnose, D-xylose, L-arabinose and D-galactose. From the unhydrolyzate A' were found L-rhamnose, D-xylose, L-arabinose, and D-galactose; from B' L-rhamnose, d-xylose, L-arabinose and D-galactose; and from C' D-xylose and D-galactose respectively (Table 6). The periodate consumption and formic acid production of the polysaccharide C were measured at various time intervals. After 120 hours periodat was consumed by 1.23 mole per $C_5H_8O_4$ and formic acid was produced 0.78 mole per $C_5H_8O_4$ (Table 7). Although a definite chemical structure for this polysaccharide C may not be formulated, experimental data, especially, from methylation, partial hydrolysie and determination of its molar ratio, and periodate analysis showed that the polysaccharide C is a highly branched polysaccharide and would be constructed of galactoaraban as a main chain residue and L-arabofuranose, D-galactopyranosyl $(1{\rightarrow}1)$-L-arabofuranose, D-xylopyranosyl $(1{\rightarrow}2)$-L-rhamnopyranosyl $(1{\rightarrow}1)$-L-arabofuranose, and L-rhamnopyranosyl $(1{\rightarrow}1)$-arabofuranose, and D-galactopyranosyl-$(1{\rightarrow}2)$-L-arabopyranosyl-$(1{\rightarrow}1)$-I-arabofuranose as a branch chain or end group (page 21).

  • Benzoylpyridinium and lutidinium salts containing$ [MoO(NCS)_5]^{2-}$ have been synthesized. The complexes have been characterized by spectroscopic, magnetic susceptibility and conductivity studies. Benzoylpyridinium and lutidinium oxopentaisothiocyanatomolybdates(Ⅴ) arise from d-d and charge transfer transitions. The magnetic moments of the complexes are close to the spin-only values. The complex anions are mononuclear containing N-bonded $NCS^-$ ligand.
