Antioxidant Effects of Salicornia bigelovii Seed Extracts

  • The objective of this study was to compare antioxidant effects of different extracts of Salicornia bigelovii seed. DPPH radical scavenging activity, chelating activity of ferrous, superoxide anion radical-scavenging activity, hydroxyl radical scavenging activity, and contents of total polyphenol and flavonoid were measured for different extracts. Ethanol extract (10 mg/mL) was found to have the maximum DPPH radical scavenging and superoxide anion radical scavenging activities (65.26 and 62.36%, respectively). However, hot water extract (10 mg/mL) showed the highest hydroxyl radical scavenging and chelating activities (51.50 and 53.24%, respectively). Total phenolics and flavonoid contents in the ethanol extract were 522.8 and $480.35{\mu}g/mg$, respectively. They were 401.51 and $352.60{\mu}g/mg$, respectively, in the hot water extract. These results indicate that ethanol and hot water extracts of S. bigelovii seed have good potential to be used as a new source of antioxidants for functional foods, cosmetics, and pharmaceuticals.

Effects of Antioxidative, DPPH Radical Scavenging Activity and Antithrombogenic by the Extract of Sancho (Zanthoxylum Schinilolium) (산초(Zanthoxylum Schinifolium) 뿌리, 줄기 및 잎 추출물의 항산화, DDPH Radical 소거 작용 및 항혈전 효과)

  • Effects of root, stem and leaf extract of sancho (Zanthoxylum schinifolium) on the inhibition of lipid peroxidation in the hepatic microsome of rat, DPPH radical scavenging activity and activated partial thromboplastin times (APTT) were examined in vitro. The highest inhibition of hepatic microsomal lipid peroxidation was observed by ethyl acetate fraction than that of methylene chloride fraction of the root and stem extracts. The high inhibition of lipid peroxidation was determined in the leaf, the root and the stem in order. The DPPH radical scavenging activity of ethyl acetate fraction was higher than that of n-butanol fraction and it was similar to the root and the steam extract. It was similar to the inhibition of hepatic microsomal lipid peroxidation. The DPPH radical scavenging activity was the highest in 2.500mg/mL of ethyl acetate fraction and it was 4.4 fold higher than that of $\alpha-tocopherol$, as an antioxidant standard. The DPPH radical scavenging activity was dependent on the extract concentration in the range of 0.125-5.000 mg/mL. The throm-boplastin times were higher than that of n-butanol fraction and it was similar to the root and the steam extracts. The leaf extract showed the highest antithrombogenic effect followed by the stem and then the root extract. The activated partial thromboplastin times were ependent on the extract concentration in the range of 0.100-2.000 mg/mL. Consequently, the effects of antioxidative, DPPH radical scavenging activity and antithrombogenic of Z. schinifolium was observed due to the inhibition of lipid peroxidation and the DPPH radical scavenging activity by methylene chloride, n-butanol and ethyl acetate fraction of the leaf extract. Radical Scavenging Activity of Ethanol Extracts and Solvent Partitioned Fractions from Various Red Seaweeds (홍조류 에탄올 추출물 및 다양한 용매 분획물의 라디칼 소거능)

  • The EtOH extracts of red seaweeds (Symphyocladia latiuscula, Chondrus ocellatus and Carpopeltis affinis) and solvent partitioned fractions were investigated for their 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and 1,1-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging effects and the total phenolic contents were correlated with ABTS and DPPH radical scavenging activities. The EtOH extracts and their solvent partitioned fractions exhibited strong ABTS and DPPH radical scavenging activities. Among the solvent partitioned fractions obtained from n-Hexane (HX), methylenchloride (MC), ethylacetate (EA), and buthanol (BuOH), the HX fraction from C. affinis showed higher radical scavenging activities than other fractions. Total phenolic contents showed significant correlation ($r^2$ = 0.709) with ABTS radical scavenging activity. The results of this study suggest that the strong radical scavenging activity of HX fraction from C. affinis is a promising natural antioxidant for healthcare products.

Enhanced DPPH Radical Scavenging Activity of Lactobacillus plantarum K-21 Isolated from Kimchi and its Various Antioxidant Effects (김치유래 Lactobacillus plantarum K-21의 DPPH 라디칼 제거활성 증진 및 다양한 항산화 효과)

  • Lactic Acid Bacteria (LAB) are among the representative probiotics that have been used for a long time in fermented food. Although there are many studies on detecting the radical scavenging activity of LAB, few studies have been conducted on the environmental factors that improve scavenging activity. This study investigated the environmental factors affecting the DPPH radical scavenging and various antioxidant activities of Kimchi-derived Lactobacillus plantarum K-21 with antihypertensive and radical scavenging activities. The optimal conditions for scavenging DPPH radicals were glucose 2%, bactopeptone 0.5%, Tween 80 0.05%, L-cysteine 0.05%, and an initial pH 6.5 at 35℃. Under optimal conditions, the DPPH radical scavenging activity was 94.8±2.2%, which was 1.5 times higher than that of the basic medium. In addition, L. plantarum K-21 had other antioxidant activities; ABTS radical scavenging (93.6±1.5%), hydroxyl radical scavenging (8.5±0.9%), metal chelating (65.9±0.5%), NO scavenging (53.1±19%), SOD-like (25.1±1.5%), and reducing power (11.7±1.4%) activities were detected. Therefore, L. plantarum K-21 may act not only as a starter for lactic acid-fermented foods with improved functionality but also as a drug for various diseases caused by oxidative stress.

1,1-Diphenyl-2-picrylhydrazyl Radical Scavenging Compounds of Fraxini Cortex

  • The radical scavenging effect of the MeOH extract of Fraxini Cortex on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical was examined. The $CH_2Cl_2$-and n-BuOH-soluble fractions of MeOH extract showed the promising DPPH radical scavenging effects, and further purified by silica gel, Sephadex LH-20 column chromatography, and reversed-phase C-18 MPLC to yield five coumarins, esculetin (1), fraxidin (2), fraxetin (3), fraxidin $8-O-{\beta}-D-glucopyranoside$ (fraxin methyl ether) (5), esculin (6), and a secoiridoid oleuropein (4), and a coumarin-secoiridoid escuside (7). Compounds 1, 3, and 4 showed potent DPPH radical scavenging effects, exhibiting $IC_{50}$ values of 14.68, 9.64, and $22.03\;{\mu}M$, respectively. Compounds 6 and 7 also showed moderate effects with $IC_{50}$ values of 147.79 and $72.73\;{\mu}M$, respectively. L-Ascorbic acid was used as a positive control and exhibited the $IC_{50}$ value of $50.31\;{\mu}M$.

Antioxidant Effects and Inhibitory Effect on NO Synthesis by Extracts of Canavalia lineata (해녀콩 (Canavalia lineata (THUNB.) DC.) 추출물의 항산화 효과 및 NO 생성 억제 효과)

  • Scavenging effects of DPPH radical and hydroxyl radical, inhibition of linoleic acid oxidation, inhibition of NO synthesis and iNOS expression were tested with extracts and chromatographic subfractions of Canavalia lineata obtained at Jeju island. Chloroform extract and its subfractions gave moderate effects on scavenging DPPH radical and hydroxyl radical, They also inhibited linoleic acid oxidation, and NO synthesis. Inhibition of NO synthesis resulted from the repression of iNOS gene expression. Ethyl acetate extract and its subfractions showed excellent effects on scavenging DPPH radical and hydroxyl radical, while they were cytotoxic.

Experimental Study on Antioxidative Effect of Luffae Fructus Retinervus and Their Effects on Cytokines to 3T3-L1 Cell Lines (사과락(絲瓜絡)의 항산화효과 및 3T3-L1분화 시 Cytokine류에 미치는 영향)

  • In recent year, We are concerned in anti-aging, disease-protection, long-life, many method are used in solving this problem. Recently, We heard that Luffae Fructus Retinervus(LFR) has effect of anti-aging, disease-protection, long-life. So I let made a experiment for this result. The purpose of this study is to; 1) the anti-oxidant effect of Luffae Fructus Retinervus(LFR) used for 3 methods, those are DPPH radical scavenging activity, Nitric oxide(NO) radical scavenging activity, Superoxide anion radical scavenging activity, 2) cultivation 3T3-L 1 Preadipocytes and Protein chip used for ProteoPlexTM 16-Well Murine Cytokine Array Kit. We measured level of DPPH radical scavenging activity. And we experienced that the ability of DPPH radical's elimination was increased by rising concentration of LFR. When the concentration of LFR was 5 mg/ml, the ability of DPPH radical's elimination was Maximum. We measured level of Nitric oxide(NO) radical scavenging activity. And we founded that the ability of NO radical's elimination was significant when concentration of LFR was from 1.25 mg/ml to 2.5 mg/ml. We measured level of Superoxide anion radical scavenging activity. And we founded that the ability of Superoxide anion radical's elimination was maximum when concentration of LFR was 0.3125 mg. When we inspected Antioxidative Effects with BSA, we experienced that ability of defense was increased by rising concentration of LFR. We known the immunity of LFR about 3T3-L1 Preadipocytes and gained the increase of Cytokines(IL-2, IL-4, GM-CSF) without IL-12p70, $INF-{\gamma}$, $TNF-{\alpha}$ So I guess that Luffae Fructus Retinervus(LFR) has effects of anti-aging, disease-protection, long-life, etc.

Optimization and production of protein hydrolysate containing antioxidant activity from tuna cooking juice concentrate by response surface methodology

  • To optimize the hydrolysis conditions in the production of antioxidant hydrolysates from tuna cooking juice concentrate (TC) to maximize the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, TC containing 48.91% protein was hydrolyzed with Alcalase 2.4 L, and response surface methodology (RSM) was applied. The optimum hydrolysis conditions included a 2.2% (w/v) Alcalase concentration and 281 min hydrolysis time, resulting in the highest DPPH radical scavenging activity of 66.49% (0.98 µmol Trolox/mg protein). The analysis of variance for RSM showed that hydrolysis time was an important factor that significantly affected the process (p < 0.05). The effects of different drying methods (freeze drying, hot air drying, and vacuum drying) on the DPPH radical scavenging activity and amino acid (AA) profiles of TC hydrolysate (TCH) were evaluated. Vacuum-dried TCH (VD) exhibited an increase in DPPH radical scavenging activity of 81.28% (1.20 µmol Trolox/mg protein). The VD samples were further fractionated by ultrafiltration. The AA profiles and antioxidant activities in terms of the DPPH radical scavenging activity, 2,2'-azino-bis(3-ethylbenzthiazoline)-6-sulfonic acid (ABTS) radical scavenging activity, ferric reducing antioxidant power, and ferrous ion chelating activity were investigated. Glutamic acid, glycine, arginine, and cysteine were the major AAs found in the TCH fractions. The highest DPPH radical scavenging activity was found in the VD-1 fraction (< 5 kDa). The VD-3 fraction (> 10 kDa) exhibited the highest ABTS radical scavenging activity and ferric reducing antioxidant power. The ferrous ion chelating activity was the highest in VD-1 and VD-2 (5 to 10 kDa). In conclusion, this study provided the optimal conditions to obtain high antioxidant activities through TCH production, and these conditions could provide a basis for the future application of TCH as a functional food ingredient.

Antioxidant Activities of Chrysanthemum frutescens, Coreopsis lanceolata, Matricaria recutica and Hieracium pilosella Flower Extracts by Extraction Solvent (추출용매에 따른 마가렛트, 큰금계국, 저먼캐모마일, 알프스민들레 꽃 추출물의 항산화 효과)

  • Present studies were conducted to investigate the effects of water and 80% ethanol solvent on antioxidant activities of flower extracts of Chrysanthemum frutescens, Coreopsis lanceolata, Matricaria recutica, and Hieracium pilosella. Extraction yields of water extracts ranged 27.15~40.25% and 80% ethanol extracts 24.92~42.84%, respectively. In all species polyphenol and flavonoid contents were higher with 80% ethanol extraction, especially in C. lanceolata. Same results were obtained with scavenging effects on DPPH and ABTS radicals and ferrous ion chelating effects. Scavenging activity of flower extracts on DPPH radicals was highest in H pilosella - higher than that of synthetic antioxidant DHT. Higher scavenging activity on ABTS radicals was observed with M. recutica. Generally Compositae flower extracts exhibited higher scavenging activity on ABTS radicals than DPPH. Scavenging activity on ABTS radicals of M. recutica and P pilosella were superior to those of ascorbic acid and BHT. Ferrous ion chelating effects were much higher with H. pilosella flower extracts by 80% ethanol. Chelating effects of 4 species were much lower than those of EDTA. In conclusion, to develop natural antioxidant from above 4 Compositae species, 80% ethanol is recommended for efficient solvent to obtain maximum antioxidant isolation and activity.

Antioxidant activity of Opuntia robusta and its total polyphenol and flavonoid contents

  • The aim of this study was to investigate the antioxidant effects of O. robusta stem extract (ORE) and to determine the total polyphenol and flavonoid contents. Free radical scavenging properties were evaluated using DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid)), and ·OH (hydroxyl radical) scavenging assays. Total polyphenol and flavonoid contents were measured using Folin-Ciocalteu reagent and aluminum chloride colorimetric methods, respectively. Active compounds of ORE were determined using high-performance liquid chromatography (HPLC). The results of the study showed that ORE exhibited DPPH, ABTS, and ·OH radical scavenging activities in a dose-dependent manner. Especially, 1,000 ㎍/mL of ORE showed the strongest radical scavenging properties against DPPH, ABTS, and ·OH. ORE contained total polyphenol content of 57.4 mg GAE/g and total flavonoid content of 5.4 mg QE/g, which may contribute to their antioxidant effects. As a result of HPLC, the contents of active compounds in ORE, dihydrokaempferol (0.65 ㎍/mL), and 3-O-methylquercetin (1.10 ㎍/mL) were confirmed. In conclusion, ORE may be useful as a functional material with antioxidant properties.
