개용환(改容丸)의 항염 및 항산화 효과에 대한 실험적 연구 (The Experimental Study on Anti-inflammation and Anti-oxidation of GaeYongHwan)

  • Objectives : This study was performed to investigate the anti-inflammatory and anti-oxidantic effects of GaeYongHwan(GYH) extract which has been used for patients with acnes. Methods : Anti-inflammatory and anti-oxidantic effects of GYH extract were tested in terms of inhibitory ability of Nitric oxide(NO) production, 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging activity and anti-bacterial effects against Propionibacterium acnes(P. acnes). Results : 1. All GYH treated groups did not show cytotoxicity. 2. Treatment with $100{\mu}g/m{\ell}$ of GYH extract lowered production levels of NO significantly compared to non-treated control or normal. 3. All of GYH treated groups did not show DPPH free radical scavenging activities. 4. All of GYH treated groups did not show anti-baterial action against P. acnes. Conclusions : These results imply that GYH extract has anti-inflammatory effect to treat acnes.

흰목이 버섯 배양액의 항산화 활성 규명 (In Vitro Evaluation for Antioxidant Activities of Culture Broth of Tremella fuciformis)

  • To investigate antioxidant activity of the culture broth from submerged culture of Tremella fuciformis, we preferentially analyzed the chemical composition of culture broth, which was mainly composed of carbohydrate (296.39 mg/g) and protein (9.24 mg/g), respectively. Also, contents of polyphenols, flavonoids and flavonols were 16.63 mg/g, 9.19 mg/g and 83.74 ${\mu}/g$, respectively. Next, we examined the scavenging abilities on DPPH (1,1-diphenyl-2-picrylhydrazyl) radical, ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid) radical, the reducing power, and chelating ability on ferrous ions. All antioxidant activities of the culture broth were increased in proportion to its concentrations. The $IC_{50}$ values were in order as follows ABTS radical scavenging activity < OPPH radical scavenging activity < chelating power. Accordingly, these results suggest that pharmacological function of T. fuciformis might be due to, at least partially, their protective effects against oxidation and the culture broth of T. fuciformis was free radical inhibitors or scavengers at low concentration, involving possibly in termination of free radical reaction as primary antioxidants.

수순(水醇)추출법으로 조제된 오공 약침액의 생리활성 효과 (Biological Activities of Scolopendrid Pharmacopuncture)

  • Reactive Oxygen Species(ROS) are continuously produced at a high rate as a by-product of aerobic metabolism. Since tissue damage by free radical increases with age, the reactive oxygen species(ROS) such as hydrogen peroxide($H_2O_2$), nitric oxide(NO). Several lines of evidence provided that ROS appears to cause to develop aging-related various diseases such as cancer, arthritis, cardiovascular disease. Our reserch objective was to examine the in vitro biological activity of Scolopendrid Pharmacopuncture, including the total poly-phenol content, DPPH radical scavenging, ABTS radical scavenging, Superoxide dismutase(SOD)-like activity, Nitrite scavenging ability. The total poly-phenol contents of Scolopendrid Pharmacopuncture was 35.859mg/L. Elctron donation ability on DPPH was 36.82%. The 2,2'-azinobis-3-ehtlbezothiazoline-6-sulfonic acid radical decolorization (ABTS) was 84.7%. The superoxide dismutase (SOD)-like activities of Scolopendrid Pharmacopuncture was 44.33%. The nitrite scavenging effects were pH dependent, and were highest at pH 1.5(45.2%) and lowest at pH 6.0(11.3%). We conclude that Scolopendrid Pharmacopuncture may be useful as potential sources of antioxidant.

전통 식재료의 항산화 효과 (Antioxidant Effect of Traditional food Ingredient)

  • 본 연구는 전통 식재료로 많이 사용되는 10종의 약용식물 및 채소류 추출물과 10종 혼합추출물의 항산화 효과를 살펴보기 위해서 디자인되었다. 세포 생존률은 MTS assay로, 항산화효과는 DPPH free radical 소거능, Riboflavin 유래 Superoxide 억제활성(SQA)에 의한 항산화활성을 측정하였다. 그 결과 세포생존률은 10종 약용식물 및 채소류 추출물과 10종 혼합추출물 모두 0.5mg/mL에서 대조군과 유사한 생존율을 보였다. DPPH free radical 소거능은 Cinnamomum cassia Blume, Eugenia caryophyllata Thunb. Arctium lappa, Total extract가 우수하였고, Riboflavin 유래 Superoxide 억제활성(SQA)은 Cinnamomum cassia Blume, Arctium lappa, Prunus mume Sieb. et Zucc., 우수하였으나, Total extract가 가장 우수한 항산화 효과를 보였다. 전통식재료로 사용되는 약용식물 및 채소 추출물과 혼합추출물의 항산화 효과를 비교한 결과, 단독으로 사용하는 것보다 혼합물로 사용시 항산화능이 증가되었다. 이는 항산화 기능성 소재로 활용이 가능한 것으로 사료되며, 향후 항산화 소재 개발 시 가치가 있을 것으로 사료된다.

Optimization of ultrasonic-assisted enzymatic hydrolysis conditions for the production of antioxidant hydrolysates from porcine liver by using response surface methodology

  • Objective: The objective of this study was to optimize ultrasonic-assisted enzymatic hydrolysis conditions, including enzyme-to-substrate (E/S) ratio, pH, and temperature, for producing porcine liver hydrolysates (PLHs) with the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity by using response surface methodology (RSM). Methods: The study used RSM to determine the combination of hydrolysis parameters that maximized the antioxidant activity of our PLHs. Temperature ($40^{\circ}C$, $54^{\circ}C$, and $68^{\circ}C$), pH (8.5, 9.5, and 10.5), and E/S ratio (0.1%, 2.1%, and 4.1%) were selected as the independent variables and analyzed according to the preliminary experiment results, whereas DPPH free radical scavenging activity was selected as the dependent variable. Results: Analysis of variance showed that E/S ratio, pH, and temperature significantly affected the hydrolysis process (p<0.01). The optimal conditions for producing PLHs with the highest scavenging activity were as follows: E/S ratio, 1.4% (v/w); temperature, $55.5^{\circ}C$; and initial pH, 10.15. Under these conditions, the degree of hydrolysis, DPPH free radical scavenging activity, ferrous ion chelating ability, and reducing power of PLHs were 24.12%, 79%, 98.18%, and 0.601 absorbance unit, respectively. The molecular weight of most PLHs produced under these optimal conditions was less than 5,400 Da and contained 45.7% hydrophobic amino acids. Conclusion: Ultrasonic-assisted enzymatic hydrolysis can be applied to obtain favorable antioxidant hydrolysates from porcine liver with potential applications in food products for preventing lipid oxidation.

포도즙을 첨가한 양갱의 품질특성 및 유리기 소거능 (Free Radical Scavenging Ability and Quality Characteristics of Yanggaeng Combined with Grape Juice)

  • 본 연구에서는 포도즙의 양(0 g, 50 g, 100 g, 150 g, 200 g)을 다르게 첨가하여 포도 양갱을 제조하고, pH, 당도, 색도, 기계적 물성, 총 페놀 함량, DPPH, 관능평가를 실시하였다. 포도양갱의 pH는 포도즙의 첨가량이 증가할수록 pH 값이 낮아지면서 유의적인 차이를 보였으며, 당도는 대조군이 1.47 Brix로 가장 낮게 나타났고, 포도즙 첨가량이 증가할수록 당도가 점점 높아져 유의적인 차이를 보였다. 색도는 명도값(L값)과 황색도(b값)이 포도즙 첨가량이 증가할수록 감소하였으나, 적색도(a값)는 그와 반대로 증가하였다. 물성 측정 결과, 포도즙 첨가가 증가할수록 경도는 증가하는 경향을 보였지만, 탄력성, 응집성, 점착성, 씹힘성은 모두 감소하는 경향을 보였다. 총 페놀 함량은 포도즙 첨가량이 증가할수록 높은 값을 보였으며, DPPH radical 소거능도 총 페놀 측정 결과와 마찬가지로 대조군에서 가장 낮은 활성을 나타내었으며, 포도즙 첨가가 증가할수록 높은 항산화 활성을 보였다. 관능평가 결과, 색, 향, 맛 등의 전체적인 기호도에서 포도즙 150 g 첨가군이 가장 높은 점수를 받았다. 이상의 결과를 종합하여 볼 때 항산화 실험에서 포도즙 200 g 첨가군의 결과가 가장 좋았으나, 관능검사 결과, 포도즙 150 g 첨가군을 가장 선호하는 결과를 함께 고려해 볼 때는 포도즙 150 g이 첨가된 양갱시료의 실용화가 바람직 할 것으로 사료된다.

Free Radical Scavenging Activity of the Seed of Phaseolus calcaratus Roxburgh

  • The seed of Phaseolus calcaratus Roxburgh (PHCR) is traditionally used for anti-pyretic and antiinflammatory effects. Although these effects are believed to be related to its antioxidant potential, little information is available for the mechanisms by which PHCR seed might scavenge free radicals or otherwise act as an antioxidant. In the present study, we purified some fractions from the ethanol extract of PHCR seed and evaluated each fraction's ability to scavenge free radicals generated by cell-free systems. We also identified active compound that is putatively responsible for free radical scavenging by analyzing NMR spectra. PHCR samples exhibited a concentration-dependent radical scavenging activity against hydroxyl radicals, superoxide anions, and DPPH radicals. Of the samples tested, a methanol-eluted sub-fraction from the PHCR extract, named $FF_4$, scavenged these radicals more effectively than the other fractions. We identified catechin-7-O-$\beta$-Dglucopyranoside as the active compound responsible for free radical scavenging potential of $FF_4$.

Synthesis of Dihydroxylated Chalcone Derivatives with Diverse Substitution Patterns and Their Radical Scavenging Ability toward DPPH Free Radicals

  • A series of dihydroxylated chalcone derivatives with diverse substitution patterns on a phenyl ring B and the para-substituents on a phenyl ring A were prepared, and their radical scavenging activities were evaluated by simple DPPH test to determine quantitative structure-activity relationship in these series of compounds. The chalcone compounds with the ortho- (i.e. 2',3'- and 3',4'-) and para- (i.e. 2,5'-) substitution patterns show an excellent antioxidant activities (80-90% of control at the concentration of 50 $\mu$M) which are comparable to those of ascorbic acid and $\alpha$ -tocopherol as positive reference materials. On the contrary, the compounds with meta- (i.e. 2',4'-, 3',5'-) substitution pattern demonstrate very dramatic decrease in activities which are around 25% of the control even at the concentration of 200 $\mu$ M (IC50 > 200 $\mu$ M). These dramatic differences could be interpreted in terms of the ease formation of fairly stable semiquinone radicals from the ortho- and parasubstituted chalcone molecules through facilitating electron delocalization. Our results indicate that the substitution patterns of two hydroxyl groups on ring B are very important structural factors for their radical scavenging activity enhancement. Meanwhile, the substituents at para-position of the phenyl ring A of chalcones have no influence on the activity.

백련(白蓮) 잎의 영양 성분 및 항산화 활성 (Nutritional Composition and Antioxidant Activity of the White Lotus (Nelumbo nucifera Gaertn) Leaf)

  • The nutritional composition of the white lotus(Nelumbo nucifera Gaertn) leaf are moisture($1.87{\pm}0.06%$), crude ash($9.57{\pm}0.07%$), crude protein($15.53{\pm}0.4%$) and crude lipid($2.42{\pm}0.1%$) per 100g. The highest mineral content was K and Ca. The principal free sugars in the white lotus leaf was sucrose, fructose and glucose. The total phenolics compounds evidenced maximal levels of $8,842.73{\pm}20.57\;g/100g$. DPPH and hydroxyl radical of scavenging ability of each sample tended to increase with increasing degrees of the sample concentration. The $IC_{50}$ values of the ethanol extract, n-hexane, chloroform, ethylacetate, butanol and water fraction from the white lotus leaf to DPPH radicals were 5.65, 8.5, 8.27, 2.03, 2.3 and $13.7\;{\mu}g/mL$ respectively. The $IC_{50}$ values of the hydroxyl radical scavenging ability of the ethanol extract, ethylacetate, butanol and water fraction were 390, 574, 327 and $378\;{\mu}g/mL$ respectively.
