Identification of Anti-Cancer Targets of Eco-Friendly Waste Punica granatum Peel by Dual Reverse Virtual Screening and Binding Analysis

  • Background: Punica granatum (family: Lythraceae) is mainly found in Iran, which is considered to be its primary centre of origin. Studies on pomegranate peel have revealed antioxidant, anti-inflammatory, anti-angiogenesis activities, with prevention of premature aging and reducing inflammation. In addition to this it is also useful in treating various diseases like diabetes, maintaining blood pressure and treatment of neoplasms such as prostate and breast cancer. Objectives: In this study we identified anti-cancer targets of active compounds like corilagin (tannins), quercetin (flavonoids) and pseudopelletierine (alkaloids) present in pomegranate peel by employing dual reverse screening and binding analysis. Materials and Methods: The potent targets of the pomegranate peel were annotated by the PharmMapper and ReverseScreen 3D, then compared with targets identified from different Bioassay databases (NPACT and HIT's). Docking was then further employed using AutoDock pyrx and validated through discovery studio for studying molecular interactions. Results: A number of potent anti-cancerous targets were attained from the PharmMapper server according to their fit score and from ReverseScreen 3D server according to decreasing 3D scores. Conclusion: The identified targets now need to be further validated through in vitro and in vivo studies.

시뮬레이션에 의한 컨테이너 물류시스템의 분석에 관하여(BCTOC를 중심으로) (An Analysis of Container Logistics System by Computer Simulation)

  • Because of the sharp increase of its export and import container cargo volumes contrast to the lack of related Container Terminal facility, equipment and inefficient procedure, there is now heavy container cargo congestions in Pusan Container Terminal. As a result of such a situation, many container ships avoid their calls into Pusan port. This is a major cause that in tum kads to weakening intemational competitiveness of the Korean industry. This study, therefore, aims are to make a quantitative analysis of Container Terminal System through the computer simulation, especially focusing on its 4 sub-system of a handling system, 'it is checked whether the current operation is being performed effectively through the computer simulation. The overall findings are as folIows; Firstly, average tonnage of the ships visiting the BCTOC was 32,360 G/T in from January '96, to may '96. The average arrival interval and service time of container ships at BCTOC are 5.63 hours and 18.67 hours respectively. Ship's arrival and service pattern at BCTOC was exponential distribution with 95% confidence and Erlang-4 distribution with 99% confidence. Secondly, average waiting time and number of ships was 9.9 hours, 235 ships(38%) among 620 ships. Number of stevedoring container per ship was average 747.7 TED, standard deviation 379.1 TEU and normal distribution with 99% confidence. Thirdly, from the fact that the average storage days of containers at BCTOC are 2.75 days (3.0 days when import, 2.5 days when export). it is founds that most containers were transfered to the off-dock storage areas with the free periods(5 days when import, 4 days when export), the reason for which is considered to be the insufficient storage area at BCTOC. Fourthly, in the case of gate in-out at BCTOC, occupied containers and emptied containers are 89% and 11% respectively in the gate-in, 75% and 25% seperately in the gate-out. Finally, from the quantitative analysis results for container terminal at BCTOC, ship's average wating time of ships was found to be 20.77 hours and berth occupancy rate(${\rho}$) was 0.83. 5~6 berths were required in order that the berth occupancy rate(${\rho}$) may be maintained up to 60% degree.

위그선의 연안여객운송시장 도입을 위한 정책 연구 (A Study on the Policy for Introduction of WIG Craft into Coastal Passenger Service)

  • 본 연구는 상용화를 앞두고 있는 위그선의 국내 연안여객운송시장 도입시 문제점을 사전 검토하고 효과적인 상용화를 위한 정책을 제시함에 있다. 차세대 해상운송수단으로 개발중인 위그선은 현재 연안여객시장에서 활동중인 초고속 선박보다 매우 빠른 속도로 운항하면서도 비행기보다 저렴하고 연비가 매우 우수해서 경제적인 운송수단으로 기대받고 있다. 위그선의 상용화는 국내 연안여객 운송시장에서 시작될 것으로 예상되며, 따라서 본 연구에서는 효과적인 위그선의 연안여객운송시장 도입 방안을 도출하기위해 AHP기법을 이용했다. 그 결과 위그선 관련법 정비, 계류장 설치 및 운영, 위그선 관제 및 통제, 조종사 고용 및 훈련, 위그선 유지보수, 위그선 선급인증 순으로 우선순위가 나타났다.

Identification and Pharmacological Analysis of High Efficacy Small Molecule Inhibitors of EGF-EGFR Interactions in Clinical Treatment of Non-Small Cell Lung Carcinoma: a Computational Approach

  • Inhibition of EGFR-EGF interactions forms an important therapeutic rationale in treatment of non-small cell lung carcinoma. Established inhibitors have been successful in reducing proliferative processes observed in NSCLC, however patients suffer serious side effects. Considering the narrow therapeutic window of present EGFR inhibitors, the present study centred on identifying high efficacy EGFR inhibitors through structure based virtual screening strategies. Established inhibitors - Afatinib, Dacomitinib, Erlotinib, Lapatinib, Rociletinib formed parent compounds to retrieve similar compounds by linear fingerprint based tanimoto search with a threshold of 90%. The compounds (parents and respective similars) were docked at the EGF binding cleft of EGFR. Patch dock supervised protein-protein interactions were established between EGF and ligand (query and similar) bound and free states of EGFR. Compounds ADS103317, AKOS024836912, AGN-PC-0MXVWT, GNF-Pf-3539, SCHEMBL15205939 were retrieved respectively similar to Afatinib, Dacomitinib, Erlotinib, Lapatinib, Rociletinib. Compound-AGN-PC-0MXVWT akin to Erlotinib showed highest affinity against EGFR amongst all the compounds (parent and similar) assessed in the study. Further, AGN-PC-0MXVWT brought about significant blocking of EGFR-EGF interactions in addition showed appreciable ADMET properties and pharmacophoric features. In the study, we report AGN-PC-0MXVWT to be an efficient and high efficacy inhibitor of EGFR-EGF interactions identified through computational approaches.

옥포만내 퇴적물, 해수 및 참굴 중의 유기주석 화합물의 분포 (Distribution of Organotin Compounds in Sediments, Seawater and Oysters (Crassostrea gigas) in Okpo Bay)

  • 대형조선소가 위치한 옥포만내 퇴적물, 해수, 참굴(Crassostrea gigas) 중의tributyltin(TBT)과 triphenyltin(TPhT), 그리고 분해산물의 농도를 분석하였다. 퇴적물과 참굴 내 TBT 농도는 각각 5~2,650 ng/g, 387~l,190ng/g dry wt.(as Sn)으로 검출되었다. 해수 중의 TBT 화합물은 19~84 ng/ $\ell$ 농도범위를 나타내었다. 조선소 내 dry-dock에 인접한 정점에서 최고값을 보이며, 조선소로부터 거리가 멀어질수록 감소하는 농토분포를 나타내었다. 이는 조선소가 옥포만내 주 오염원임을 의미한다. 퇴적물 중의 TBT 화합물의 구성비는 약 57%로 dibu-tyltin(DBT)와 monobutyltin(MBT)보다 높게 나타났다 만을 가로지르는 방파제를 경계로 하여 부틸주석 화합물(butyltin)의 농도가 급격하게 감소하여, 방파제가 옥포만 내 유기주석 화합물의 분포에 영향을 미치고 있음을 나타내었다. 페닐주석화합물은 참굴 중 nd-52 ng/g으로 검출되었으며, 퇴적물과 해수의 전 정점에서 검출한계 이하의 값을 보였다. 참굴 중의 TBT 농도는 대조구인 해금강의 시료보다 높은 값을 보였고, 생물농축계수(biological concentration factor, BCF)는 1.1$\times$$10^4$의 값을 보였다.

Grillage Method Applied to the Planning of Ship Docking

  • When blocks are supported on a dock, huge reaction forces concentrated at the supports cause structural damage owing to local stress concentrations. Thus, the supports should be arranged to avoid local failure from the reaction forces by redistributing those forces. Docking analyses to determine the proper blocks and their support arrangements are introduced so that the local stresses are minimized to warrant the safety of the docking supports. Local stresses enforced by the support arrangement should be evaluated by finite element analysis (FEA). However, it is difficult to consider an accurate 3D geometry of the blocks in the finite element model because the structural design information is too complicated to determine within several days using the FEA model. This paper presents a simplified FE model to evaluate the safety of the arrangement of supports using a simplified grillage element. The grillage element can be efficiently used to obrain the reaction forces in docking analysis becasuse the reaction forces at the supports are enough to assess the safety of block. Since a simplified grillage model of the entire ship cannot accurately calculate the local stresses, an optimized modeling method based on the grillage element was introduced. The local reaction forces obtained by the proposed approach and three-dimensional FEA were discussed for typical types of ships. It is shown that the reaction forces obtained by the present grillage model are in reasonably good agreement with the FEA model.

소리쟁이 (Rumex crispus)의 부정근과 모상근 배양에 의한 Anthraquinone 생산성 (Anthraquinone Productivity by the Cultures of Adventitious Roots and Hairy Roots from Curled Dock (Rumex crispus))

  • 형질전환된 소리쟁이 모상근의 기내 배양시 그 생장 특성과 anthraquinone생산성을 조사하기 위하여 소리쟁이의 모상근을 Agrobacterium rhirogenes strain $A_4$ 접종에 의하여 잎 절편으로부터 유기하여 부정근과 비교하였다. 잎 절편에서의 부정근 형성은 5 $\mu$M NAA를 첨가한 MS배지에서 가장 양호하였다. Paper electrophoresis를 통한 opine분석 결과 모상근에서만 mannopine이 검출되고 부정근에서는 검출되지 않았다. 부정근과 모상근 모두에서 2차근인 측근 근단은 호르몬을 첨가하지 않은 배지에서 신장만 할 뿐 분지하지 않았으나, 1차근인 주근의 근단은 많은 측지를 내며 빠르게 생장하였다. Anthraquinone 함량 증대에 따른 부정근과 모상근의 생장에는 MS기본 배지가 가장 양호하였다. 부정근과 모상근 배양시 생장량과 anthraquinone함량은 5% sucrose를 포함한 0.05 $\mu$M NAA와 0.1 $\mu$M kinetin으로 조성된 MS 액체 배지에서 가장 높은 것으로 나타났으며, 모상근의 anthraquinone함량은 배양 일수에 따라 증가하다가 배양 25일 이후에는 감소하였다.

Efficient Data Management for Hull Condition Assessment

  • Performing inspections for Hull Condition Monitoring and Assessment as stipulated in IACS unified requirements and IMO's Condition Assessment Scheme (CAS) IMO Resolution MEPC.94(46), 2001, Condition Assessment Scheme, IMO Resolution MEPC.111(50), 2003, Amendments to regulation 13G, addition of new regulation 13H involves a huge amount of measurement data to be collected, processed, analysed and maintained. Information to be recorded consists of thickness measurements and visual assessment of coating and cracks. The amount of data and increasing requirements with respect to condition assessment demand efficient computer support. Currently, due to the lack of standardization for this kind of data, the thickness measurements are recorded manually on ship drawings or tables. In this form, handling of the measurements is tedious and error-prone and assessment is difficult. Data reporting and analysis takes a long time, leading to some repairs being performed only at the next docking of the ship or making an additional docking necessary. The recently started ED funded project CAS addresses this topic and develops-as a first step-a data model for Hull Condition Monitoring and Assessment (HCMA) based on XML-technology. The model includes simple geometry representation to facilitate a graphically supported data collection as well as an easy visualisation of the measurement results. In order to ensure compatibility with the current way of working, the content of the data model is strictly confined to the requirements of the measurement process. Appropriate data interfaces to classification software will enable rapid assessment by the classification societies, thus improving the process in terms of time and cost savings. In particular, decision-making can be done while the ship is still in the dock for maintenance.

ESTIMATION OF DUCTILE FRACTURE BEHAVIOR INCORPORATING MATERIAL ANISOTROPY

  • Since standardized fracture test specimens cannot be easily extracted from in-service components, several alternative fracture toughness test methods have been proposed to characterize the deformation and fracture resistance of materials. One of the more promising alternatives is the local approach employing the SP(Small Punch) testing technique. However, this process has several limitations such as a lack of anisotropic yield potential and tediousness in the damage parameter calibration process. The present paper investigates estimation of ductile fracture resistance(J-R) curve by FE(Finite Element) analyses using an anisotropic damage model and enhanced calibration procedure. In this context, specific tensile tests to quantify plastic strain ratios were carried out and SP test data were obtained from the previous research. Also, damage parameters constituting the Gurson-Tvergaard-Needleman model in conjunction with Hill's 48 yield criterion were calibrated for a typical nuclear reactor material through a genetic algorithm. Finally, the J-R curve of a standard compact tension specimen was predicted by further detailed FE analyses employing the calibrated damage parameters. It showed a lower fracture resistance of the specimen material than that based on the isotropic yield criterion. Therefore, a more realistic J-R curve of a reactor material can be obtained effectively from the proposed methodology by taking into account a reduced load-carrying capacity due to anisotropy.

목재 바이오매스를 활용한 이중유동층 가스화기의 SNG 생산 (SNG Production from Wood Biomass with Dual Fluidized-Bed Gasifier)

  • 목재 바이오매스를 이용한 가스화 공정은 고열량의 합성가스를 통해 알콜류, SNG 등 다양한 에너지 자원으로 변환시킴으로써 자원의 재순환에 기여할 수 있으며, $CO_2$ 등의 온실가스를 감소시킴으로써 지구온난화 방지에 기여할 수 있다. 본 연구에서는 이중유동층 가스화기에 목재 바이오매스를 투입하여 가스화기의 최적운전 조건을 도출하고, SNG 생산효율을 검증함으로써 이중유동층 가스화기에 대한 국내 상용화 기반을 마련하고자 하였다. 목재 바이오매스에 대한 가스화기의 최적 운전조건 도출 결과, 운전온도 $826^{\circ}C$에서 Steam 투입량 1,334g/hr, Air 투입량 5.56L/min일 때 탄소전환율이 81%로 확인되었으며, SNG 생산을 위한 $CH_4$가스 농도를 확인한 결과, 92%로 나타났다.
