Decision Supporting Methodology and System Based on Theory of Constraints for Optimal Product Portfolio Strategy in Shipbuilding Industry (제약이론을 기반으로 한 최적제품조합 의사결정 지원 방법론 및 시스템)

  • Shipbuilding is a typical 'build to order' industry. It has a business model that generates revenues from building various ships and offshore products in accordance with owner's requirements at each production stage. Under uncertainty in shipping market, it is very essential for the shipbuilder to prepare the fast and competitive decision for product portfolio strategy in order to maximize contribution margin by exploiting production facilities and constraints. In this study, we introduce the unique decision supporting methodology for the optimal product portfolio sets based on Theory of Constraints(TOC). This methodology is established by adopting the concept of Drum Buffer Rope(DBR) in constraints planning and Throughput Accounting (TA) in management accounting of TOC. In addition, Decision Supporting System(DSS) is implemented. This DSS system provides a throughput estimator with reflecting the cost structure of shipbuilding industry and a resource simulator built on heuristic algorithms to operate major constraint-resources in shipyard such as dock, quay and pre-erection area etc. Several examples are presented to show that the proposed methodology and system can effectively support the strategic decision-making process of a global shipbuilding company.

  • In a shipyard, there exist various critical decision making components pertaining to various production hindrances. The most prominent one is best-fit spatial arrangement for the minimal spatial occupancy with better pick-ability for the erection of the ship in the dock. During the present research, a concept have been conceived to evade the gap between the identification oj inter-relationships among a set of blocks to be included on a pre-erection area, and a detailed graphical layout of their positions, is called an Optimal Block Relationship Diagram A research has been performed on generation of optimal (or near Optimal) that is, with minimal scrap area. An effort has been made in the generation of optimal (or near-optimal) Optimal Block Relationship Diagram with the Goldberg's Genetic Algorithms with a representation and a set of operators are 'trained' specifically for this application. The expected result to date predicts very good solutions to test problems involving innumerable different blocks to place. The suggested algorithm could accept input from an erection sequence generator program which assists the user in defining the nature and strength of the relationships among blocks, and could produce input suitable for use in a detailed layout stage.

Understanding Rifampicin Resistance in Tuberculosis through a Computational Approach

  • The disease tuberculosis, caused by Mycobacterium tuberculosis (MTB), remains a major cause of morbidity and mortality in developing countries. The evolution of drug-resistant tuberculosis causes a foremost threat to global health. Most drug-resistant MTB clinical strains are showing resistance to isoniazid and rifampicin (RIF), the frontline anti-tuberculosis drugs. Mutation in rpoB, the beta subunit of DNA-directed RNA polymerase of MTB, is reported to be a major cause of RIF resistance. Amongst mutations in the well-defined 81-base-pair central region of the rpoB gene, mutation at codon 450 (S450L) and 445 (H445Y) is mainly associated with RIF resistance. In this study, we modeled two resistant mutants of rpoB (S450L and H445Y) using Modeller9v10 and performed a docking analysis with RIF using AutoDock4.2 and compared the docking results of these mutants with the wild-type rpoB. The docking results revealed that RIF more effectively inhibited the wild-type rpoB with low binding energy than rpoB mutants. The rpoB mutants interacted with RIF with positive binding energy, revealing the incapableness of RIF inhibition and thus showing resistance. Subsequently, this was verified by molecular dynamics simulations. This in silico evidence may help us understand RIF resistance in rpoB mutant strains.

  • The ships have a lot of electric machinery needed to supply electricity from the moment of launching coming out of dry dock to docking at jetty. Thus, the ships always have to use alternator and electric machinery that has low efficiency under the low load. Many government service ships like the MMU training ship have been spending lots of time at jetty rather than sailing at sea. These ships are operated under the condition of low load due to the operation of basic machinery at jetty and electric machineries are driven with the status of low efficiency. This paper would suggest the energy saving method for these ships. The investigation describes that shore connection is a great asset to these ships and that the flow rate control by adjusting revolution with the adoption of inverter is better than flow rate control by using throttle valve to save energy. The result is based on the investigation of cooling pump at MMU training ship.

Virtual Screening of Penicillin-derived Inhibitors for the Metallo-β-lactamase from Bacillus cereus

  • The metallo-$\beta$-lactamases ($M{\beta}Ls$) are clinically significant enzymes which readily hydrolyze most $\beta$-lactam antibiotics. Discovering potential inhibitors for the $M{\beta}Ls$ is an expensive, time consuming endeavor. Virtual screening can sieve out inhibitor candidates with incompatible features prior to synthesis, decreasing these costs. Using Autodock 4.0, the binding locations and energies of four previously-studied potential inhibitors and four additional compounds obtained from the National Cancer Institute (NCI) database were computationally calculated. Based on the docking models of these eight compounds, we then designed several hypothetical inhibitor structures, compounds A through F, and performed their respective docking experiments. The docking results for compound F showed that it binds to the zinc containing active sites with a lowest predicted binding energy of -6.70 kcal/mol, suggesting F is the most likely potential $M{\beta}L$ inhibitor.

Comparison of Some 3-(Substituted-Benzylidene)-1, 3-Dihydro-Indolin Derivatives as Ligands of Tyrosine Kinase Based on Binding Mode Studies and Biological Assay

  • A series of 3-(substituted-benylidene)-1, 3-dihydro- indolin-2-one, 3-(substituted-benylidene)-1, 3-dihydro- indolin-2-thione and 2, 2'-dithiobis 3-(substituted-benylidene)-1, 3-dihydro-indole derivatives was investigated as inhibitor of $p60^{c-Src}$tyrosine kinase by performing receptor docking studies and inhibitory activity toward tyrosine phosphorylation. Some compounds were shown to be docked at the site, where the selective inhibitor PP1 [1-tert-Butyl-3-p-tolyl-1H-pyrazolo[3,4-d]pyrimidine-4-yl-amine] was embedded at the enzyme active site. Evaluation of all compounds for the interactions with the parameters of lowest binding energy levels, capability of hydrogen bond formations and superimposibility on enzyme active site by docking studies, it can be assumed that 3-(substituted-benzylidene)-1, 3-dihydro-indolin-2-one and thione derivatives have better interaction with enzyme active site then 2, 2'-dithiobis 3-(substituted-benzylidene)-1, 3-dihydro indole derivatives. The test results for the inhibitory activity against tyrosine kinase by Elisa method revealed that 3-(substituted-benylidene)-1, 3-dihydro- indolin-2-thione derivatives have more activity then 3-(substituted-benylidene)-1, 3-dihydro- indolin-2-one derivatives.

The Stimulatory Effects on the Osteoblast Cells of the Root Constituents from Rumex crispus (소리쟁이 뿌리 성분이 조골세포 분화에 미치는 영향)

  • Rumex crispus (Curled Dock, Polygonaceae) is a perennial wild plant used in traditional medicine as a laxative, astringent, and to treat blood and skin disease. The ethanol extract of R. crispus was obtained and its carbohydrate contents were analyzed using high-performance anion-exchange chromatography with pulsed amperometric detection. The anabolic effects of R. crispus in human osteoblastic MG-63 cells were investigated using the WST-8 assay, alkalinephosphatase (ALP) assay, and mineralization assay. The ethanol extract increased the proliferation of MG-63 cells and stimulated ALP activity in a dose-dependent manner over a 72-hrs period. Additionally, the ethanol extract dose-dependently stimulated the formation of bone nodules in MG-63 cells treated for 12 days. The ethyl acetate fraction from the ethanol extract did not affect osteoblast viability but induced an increase in ALP activity. In conclusion, the ethanol extract of R. crispus increases the proliferation and bone-forming activity of osteoblasts, and hence it could be used in the development of bone-forming stimulatory nutraceuticals and osteoporosis-related medicines.

  • Ship salvage crane is to salvage the equipment safely, this type of crane in the shipyard's large ships or port is being used a lot of container etc. Such a salvage crane was installed on the land and it is built to use the harbour facilities. In this paper, the response amplitude operator and the wave exiting force of floating type salvage crane is calculated and it is performed to structural response estimation.

  • 대부분의 단백질-리간드 도킹 프로그램에서 리간드의 구조 유연성은 리간드의 회전 가능한 torsion angle들을 샘플링 하는데 국한된다. 따라서 도킹에 사용되는 초기 리간드 구조의 결합길이, 결합각, ring 구조 등에 따라 단백질-리간드 도킹의 성공여부가 달라질 수 있다. 실제 단백질-리간드 도킹 프로그램을 이용하여 단백질-리간드 상호작용을 연구하는 경우, 리간드의 구조를 임의의 구조 데이터베이스로부터 얻거나 다양한 리간드 3차원 구조 생성 프로그램 등을 이용하여 생성하게 된다. 따라서 리간드의 초기 구조가 단백질-리간드 도킹 프로그램의 성능에 어떤 영향을 주는지 살펴보는 것은 실제 단백질-리간드 도킹을 활용하는 경우에 단백질-리간드 도킹 프로그램의 성능이 어떨 것인지 알아볼 수 있다는 점에서 매우 중요하다. 본 연구에서는 리간드 분자의 초기 구조가 단백질-리간드 도킹에 미치는 영향을 알아보기 위해 30개의 단백질-리간드 복합체 표적에 대해 리간드의 초기 구조를 다양하게 생성하여 GalaxyDock으로 단백질-리간드 복합체 구조를 예측하였다. 도킹을 위한 리간드 분자를 만들 때 Ring library에서 여러 가능한 ring의 conformation을 가져오는 방법으로 리간드의 구조를 다양하게 만들었으며, 도킹 결과 한 가지 모델만 사용했을 때에 비해 도킹의 성공률이 70%에서 80%로 10% 증가한 것을 확인하였다. 또한 특히 구조적으로 유연한 ring이 리간드에 있는 경우 초기 구조가 도킹의 성공률에 큰 영향을 미치는 것을 확인할 수 있었다.

Effect of Applying Pressure of High Pressure Diecasting Process Using Salt core (용탕단조 시 저온염코어 적용 가압력의 영향)

  • A new concept of salt core, a melting temperature of which is lower than the solidus temperature of cast alloy, was introduced to produced an integrated casting part having a complicated inner shape or requiring under-cut in high pressure die casting or squeeze casting process. The main goal of this study is to develop a new integrated net-shape forming technology using fusible core of lower melting temperature than that of a casting alloy. This integrated net-shape forming technology would be very successful and cost-effective for producing the integrated products having a complicated inner shape or requiring under-cut. The technology for measuring and evaluating a various property of fusible core such as a thermal conductivity and thermal expansion coefficient, melting temperature was established. Also, the work space can be cleaned without a pollution inducing products.
