아토피 피부염을 유발한 마우스에서 백선피지부자복합방의 항염증 효과 (Anti-inflammatory effect of Baecksunpijibujabokhap-bang in Atopic dermatitis model mice)

  • Objectives : In order to investigate the efficacy of BJBB on atopic dermatitis, various anti-inflammatory factors were studied. Methods : In-vitro, inflammatory mediators, such as MTT and nitric oxide were detected after the addition of LPS with or without BJBB in Raw 264.7 cells. In-vivo, in order to verify the effectiveness of BJBB in atopic dermatitis animal model, its role in inflammation factors and histological changes were observed in NC/Nga mice. Results : BJBB showed cell viability of 100% or higher in all concentration in Raw 264.7 cells. BJBB inhibited LPS-induced productions of inflammatory mediators nitric oxide in RAW 264.7cells. BJBB treated group showed significant decrease in the expression of IL-1b, IL-6 and TNF-a by 40%, 80% and 44% respectively. Also the group showed decrease in the transcription of IL-1b, IL-6 and TNF-a mRNA in spleen by 41%, 93% and 39% respectively. BJBB treated group showed significant decrease in WBC, neutrophil, lympocyte and monocytes immune cell ratio in blood by 54%, 63%, 57% and 86% respectively. BJBB treated group showed decrease in the expression of IgG by 39% respectively. Also, infiltration of adipocytes into skin was suppressed and the thickness of epidermis and dermis were relatively decreased in the BJBB treated group. Conclusion : BJBB has an anti-inflammatory effects in NC/Nga mouse. Thus, these results suggested a beneficial effect of BJBB in treatment with Atopic dermatitis and inflammatory.

Protective effect of Lycium barbarum leaf extracts on atopic dermatitis: in vitro and in vivo studies

  • BACKGROUND/OBJECTIVES: Atopic dermatitis (AD) is a chronic disease with an increasing incidence globally; therefore, there is a growing demand for natural compounds effective in treating dermatitis. In this study, the protective effects of Lycium barbarum leaves with and without chlorophyll (LLE and LLE[Ch-]) on AD were investigated in animal models of AD and HaCaT cells. Further, we investigated whether LLE and LLE(Ch-) show any differences in physiological activity. MATERIALS/METHODS: AD was induced by 2,4-dinitrochlorobenzene (DNCB) for three weeks, while NC/Nga mice were fed LLE or LLE(Ch-) extracts for 7 weeks. Serum immunoglobulin E (IgE) and cytokine (tumor necrosis factor [TNF]-α, interleukin [IL]-6, and IL-4) concentrations and the degree of DNA fragmentation in lymphocytes were examined. A histopathological examination (haematoxylin & eosin staining and blue spots of toluidine) of the dorsal skin of mice was performed. To elucidate the mechanism of action, the expression of the thymus and activation-regulated chemokine (TARC) and macrophage-derived chemokine (MDC) were measured in HaCaT cells. RESULTS: Serum IgE and cytokines (TNF-α and IL-6) levels as well as DNA fragmentation of lymphocytes were significantly decreased in AD-induced mice treated with LLE or LLE(Ch-) compared to those of the control group. The epidermal thickness of the dorsal skin and mast cell infiltration in the LLE group significantly reduced compared to that in the control group. The LLE extracts showed no cytotoxicity up to 1,000 ㎍/mL in HaCaT cells. LLE or LLE(Ch-)-treated group showed a reduction of TARC and MDC in TNF-α-and IFN-γ-stimulated HaCaT cells. CONCLUSIONS: These results suggest that LLE potentially improves inflammation by reducing the expression of chemokines that inhibit T helper 2 cell migration. LLE(Ch-) showed similar effects to LLE on blood levels of IgE, TNF-α and IL-6 and protein expression in HaCat cells, but the ultimate effect of skin improvement was not statistically significant. Therefore, both LLE and LLE(Ch-) can be used as functional materials to alleviate AD, but LLE(Ch-) appears to require more research to improve inflammation.

Lactobacillus bifermentans로 발효한 율피의 항아토피 효과 (Anti-atopic Effects of Castanea crenata Inner Shell Extracts Fermented by Lactobacillus bifermentans)

  • 본 연구에서는 DNCB 도포를 통해 아토피 피부염을 유발시킨 NC/Nga mice에 율피 발효물을 각각 0.1%, 1%, 5%로 도포하고 항아토피 효능을 평가하였다. 율피 발효물의 유효성분은 HPLC 분석 결과, gallic acid와 ellagic acid가 각각 10.18 mg/g, 2.14 mg/g 함량을 나타내었다. 실험군 간의 유의적 체중 변화는 관찰되지 않았으며, 육안 평가를 통해 홍반, 가려움과 피부건조, 부종과 혈종, 짓무름 그리고 태선화와 같은 일반적인 아토피 피부염 증상의 심각도를 측정한 점수 결과에서는 DNCB 도포를 통해 아토피 피부염을 유발한 atopic dermatitis(AD)군($13.17{\pm}0.40$)과 비교 시, 아토피 피부염 유발 후 5% 율피 발효물을 도포한 fermented Castanea crenata inner shell(FCS) 5군에서 $7.50{\pm}0.43$ 으로 나타나 아토피 피부염 증상이 빠르게 개선되는 것을 확인하였다(P<0.01). 피부 측정 기기인 MPA5 580을 이용하여 피부멜라닌지수, 피부홍반지수, 피부수분지수를 측정한 결과, AD군은 각각 $296.61{\pm}8.11$, $264.33{\pm}10.15$, $3.15{\pm}0.54$를 나타내었으며 FCS 5군은 각각 $105.86{\pm}2.92$, $199.56{\pm}4.22$, $17.62{\pm}1.75$로 개선효과가 유의적으로 나타났다(P<0.05). 각 실험군 mice의 면역반응을 확인하기 위해 비장 무게(mg)를 체중(g)으로 나누어 산출한 비장 수치의 경우, 아토피 피부염 유발로 인해 AD군에서 증가된 비장 수치($5.55{\pm}0.31$)가 FCS 0.1군, FCS 1군, FCS 5군에서는 각각 $4.53{\pm}0.24$, $3.80{\pm}0.40$, $3.32{\pm}0.24$로 나타나 율피 발효물의 도포 농도에 의존적으로 감소되는 경향을 나타냈으며 특히 FCS 5군에서는 유의적으로 낮게 나타났다(P<0.05). 이는 율피 발효물이 아토피 피부염 상태에서 증가된 비장 내 T-림프구를 효과적으로 억제할 수 있음을 나타낸다. Quantitative real-time PCR을 통해 피부조직에서 IL-$1{\beta}$, TNF-${\alpha}$와 같은 염증 유전자의 발현 변화를 분석한 결과, 두 유전자 모두 정상군의 relative quantification값이 1일 때 AD군은 각각 1.30, 1.26으로 증가되었으며 FCS 5군에서는 1.11, 0.93으로 나타나 율피 발효물 도포에 의한 유의적인 감소(P<0.05)를 확인할 수 있었다. 아토피 피부염의 면역학적 지표인 혈청 내 IgE 함량을 분석한 결과에서는 각 실험군 간의 유의적 차이는 보이지 않았으나 AD군($1,628.71{\pm}202.59ng/mL$)과 비교 시, 율피 발효물을 도포한 FCS 0.1군, FCS 1군, FCS 5군에서 각각 $1,530.15{\pm}198.70ng/mL$, $1,462.15{\pm}83.79ng/mL$, $1,187.47{\pm}140.09ng/mL$로 나타나 율피 발효물 도포에 의해 감소되는 경향을 확인할 수 있었다. 따라서 율피 발효물은 아토피 피부염 증상 개선을 위한 기능성 천연물의 소재로 활용될 수 있을 것이라 생각된다.

천연 탄산 온천수의 아토피 유발 쥐 모델에 대한 혈소판 응집 억제 효능 평가 (The anti-coagulation effect of natural carbonated hot spring water on DNCB-induced NC/Nga mice.)

  • Objectives:Spring water is widely known to relax muscles by promoting blood circulation. This study was conducted to analyze theantithrombotic effect of naturally carbonated hot spring water (NCHW) to assess its influence on blood circulation. Methods:Atopic dermatitis was induced in the skin of the mice used in this experiment. NCHW was applied, and the antithrombotic effect was assessed and compared with that of other interventions. The positive control group was treated orally with aspirin. Results:After 3 weeks of exposure to NCHW, the experimental groupshowed a significant antithrombotic effect. NCHW also produced inhibitory responses to both collagen- and ADP-induced platelet aggregation, whereas the group given aspirin reacted only to collagen-induced platelet aggregation. Conclusions:The experiment demonstrated the intrinsic antithrombotic effects of NCHW compared with those of artificially carbonated water, tap water, and aspirin. This result suggests the possibility that NCHW can be used as a supportive and alternative treatment for vascular diseases.

한약재 단일 추출물 및 복합 추출물을 이용한 아토피성 피부염 억제 효과 (Medicinal Herb Extracts Attenuate 1-Chloro-2,4dinitrobenzene-induced Development of Atopic Dermatitis-like Skin Lesions)

  • 본 연구에서는 1-chloro-2,4-dinitrobenzene (DNCB)에 의해 유도된 아토피성 피부염 마우스 모델에서 고삼, 감초 및 백선피 추출물과 그들 복합 추출물(GGB)의 항아토피 효능을 평가하였다. 아토피성 피부염의 병리학적 형태인 홍반, 귀부종, 부종, 각질과 상처는 복합 추출물 중 고삼, 감초 및 백선피 추출물이 3:1:1로 혼합된 GGB-1 처리군에서 다른 시료 처리군들에 비하여 유의적인 억제 효능을 나타내었다. 또한 면역 기관인 비장의 무게도 GGB-1 처리군에서 감소하였고, 간의 무게는 정상군과 시료 처리군의 값이 유사한 결과를 나타내었다. 아울러 GGB-1 처리군에서 비만 세포의 침윤이 감소되었으며, 혈청 중 IgE의 농도 역시 GGB-1 처리군에서 가장 낮은 값을 보였다. 비록 단일 한약재와 그들의 혼합물 처리군에서는 면역 사이토카인(IFN-${\gamma}$, IL- 1, 4, 5, 6, 13, $1{\beta}$, and TNF-${\alpha}$) 의 값이 유의적으로 감소되었으나, 여러 가지 결과들을 종합하여 볼 때 고삼, 감초 및 백선피 추출물이 3:1:1로 혼합된 복합 추출물인 GGB-1이 아토피성 피부염의 개선에 효과적인 약물로 판단된다.

가미소풍산(加味消風散)이 염증 및 아토피피부염 동물병태에 미치는 영향 (Effect of Gami-sopungsan on Inflammation and DNCB-induced Dermatitis in NC/Nga in Mice)

  • Gami-Sopungsan (GS) is one of the traditional korean remedy. We investigated the anti-inflammation and anti-atopic dermatitis (AD) effect of GS. No cytotoxicity of GS was observed in the range of $1{\sim}100{\mu}g/m{\ell}$ on Raw 264.7 cells. The Inflammatory response of Raw 264.7 cells were induced by lipopolysaccharide (LPS), followed by GS treatment at indicated concentrations (0, 1, 10 and $100{\mu}g/m{\ell}$). At $100{\mu}g/m{\ell}$ concentration, GS showed inhibitory effect on LPS-induced nitric oxide production by 20%. Production of IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ was decreased by approximately 56%, 36% and 79%, respectively upon GS treatment at $100{\mu}g/m{\ell}$. 200 mg/kg of GS was orally administered to NC/Nga mice, where AD was induced by 1-chloro 2,4-dinitrobenzene. There were no significant difference between GS treated group and the control group on body weight and food intake changes during growth. The back skin of GS group showed decrease in erythema, pruritus, dry skin, edema, excoriation, erosion and lichenification level through naked eye observations. In addition, leukocyte infiltration and the thickness of epidermis were significantly decreased in the skin tissues (back and ear). The serum IgE levels were decreased by 28.8% in the GS treated group. The GS treated group showed remarkable inhibition of IL-4 (83%), IL-5 (95%), IL-6 (62%) and TNF-${\alpha}$ (84%) in serum, indicating that GS has similar or higher efficacy than those of the dexamethasone treated group. From the results above, we conclude that GS has significant anti-inflammation and anti-AD effects on Raw 264.7 cells and NC/Nga mice. The results should provide fundamental and valuable data for the research on natural products being developed against atopic dermatitis.

Essential Oil of Thujopsis dolobrata Suppresses Atopic Dermatitis-Like Skin Lesions in NC/Nga Mice

  • We examined the effects of essential oil from Thujopsis dolobrata Sieb. et Zucc. var. hondai Makino (EOTD) (Cupressaceae) on atopic dermatitis (AD)-like skin lesions in NC/Nga mice. Treatment with EOTD twice daily for two weeks ameliorate AD-like skin lesions induced by DNCB (2,4 dinitrochlorobenzene), and clinical scores were reduced to 7.29, 7.07, and 4.5 points in the groups treated with 1.5%, 3.0%, and 6.0% extract (p<0.01) respectively, from the 15.0 score obtained using vehicle. EOTD inhibited the infiltration of mast cells into the AD-like skin lesion in NC/Nga mice (p<0.01) and also reduced serum histamine and IgE levels (p<0.05). Furthermore, it dose-dependently inhibited the release of beta-hexosaminidase from rat basophilic leukemia RBL 2H3 cells. These results indicate that EOTD reduces AD-like skin lesions by inhibiting the production of IgE and histamine, and, thus, IgE-mediated degranulation.

BALB/c 및 NC/Nga 마우스의 아토피성 피부질환모델에서 생약조성물 SPZZC의 치료효능 (Effect of SPZZC, a Composition of Herb Extracts, on Atopic Dermatitis in BALB/c and NC/Nga Mouse)

  • The aim of this study was to evaluate if a composition of herb extracts, PLX-PLS was effective to treat atopic dermatitis (AD) in mice. SPZZC is a composition of herb extracts containing the roots of Scopolia parviflora and Paeonia lactiflora, the herb of Zizania caudiflora, the fruit of Ziziphus jujuba and the leaf of Chinese arborvitae. AD in BALB/c mouse was induced by patching ovoalbumin on the backside, while it in NC/Nga mouse was induced by repeated application of 1-chloro 2,4-dinitrobenzene (DNCB). Mice were topically treated with SPZZC or Domohorn ointment on the backside for 2 weeks (BALB/c) or 1 week (NC/Nga). Scratching behavior, clinical skin severity and the levels of WBC, neutrophil, eosinophil and total serum IgE were measured. After AD induction, scores of scratching behavior and clinical skin severity and the levels of WBC, neutrophil, eosinophil and total serum IgE were increased. Treatment with SPZZC significantly decreased scores of scratching behavior and clinical skin severity in a dose dependent manner in NC/Nga and BALB/c mice. Treatment with SPZZC 2% significantly decreased also the levels of WBC, neutrophil, eosinophil and total serum IgE. Especially, treatment of SPZZC 2% reduced more rapidly score of clinical skin severity than clobetasol cream. These results suggest that the SPZZC may be an alternative substance for the management of AD.

가미미후등식장탕이 DNCB 유도 아토피 피부염 동물모델에 미치는 효과 (Effects of KMS on the DNCB induced animal Model of Atopic Dermartitis)

  • Objectives: The purpose of this study is to examine the effect of water and fermentation extracts of KMS (Kami-Mihudeongsikjang-tang) on AD (atopic dermatitis). Additionally, by applying the fermentation extracts of KMS at the first sensitization and latency period, I investgated whether it could prevent AD. Methods: In this study, to test the effect and preventive efficacy of KMSs on AD. DNCB-induced BALB/c mice of AD model was used. Through histological observation, epidermis and dermis thickness, the infiltration of eoshiphils and mast cells in epidermis and dermis were examined. We measured the serum level of IgE and IL-6, TNF-alpha, and the expressions of $NF-{\kappa}B$ and MAPK protein. In order to examine the effect of KMSs on keratinocyte, HaCaT cells were treated TNF-alpha and IFN-gamma to induce an inflammatory response. The KMSs were applied at various concentration in the experimental group. We investigated TARC expression. Results: The treatment groups were reduced epidermis and dermis thickness, inhibited the infiltration of eosinophils and mast cells, reduced the serum level of IL-6. Moreover, sfKMS group reduced serum level of TNF-alpha, inhibited the protein expressions of $NF-{\kappa}B$ and the phosphoryllation of ERK, JNK and p38. Especially sfKMS-pre group were reduced the serum level of IgE, show significant inhibition on the protein expression of $NF-{\kappa}B$ and the phosphoryllation of ERK, JNK and p38. In the experiment on HaCaT cells, sfKMS group were reduced expression of TARC. Conclusions: These result suggest that wKMS and sfKMS was effective in the treament on AD, and sfKMS would prevent AD.

천연 소재 복합물이 항아토피 피부염 및 피부재생에 미치는 영향 (Effects of Natural Herb Mixture on Anti-atopic Dermatitis and Skin Regeneration)

  • Objectives : This study aims to evaluate the effects of natural herb mixutre (NHM) on atopic dermatitis and skin regeneration using in vivo test. Methods : NHM was prepared with DW. 25% of NHM was applied to skin lesion, where atopic dermatitis was induced by DNCB in NC/Nga mice. The levels of cytokines (IL-4, IL-5, IL-6, IL-13, TNF-a, and $IFN-{\gamma}$), and IgE in serum were measured by Luminex. Immune cells (WBC, eosinophil, lymphocyte, and monocyte) in blood were counted by coulter counter. The gross investigation of atopic dermatitis index score test were performed during the NHM treatment period. Also, the histopathological change of dorsal skin was observed by H&E and M&T staining. Results : NHM showed the levels of IL-4, IL-5, IL-6, IL-13, IgE, WBC, eosinophil, lymphocyte, and monocyte in serum or blood were significantly decreased. On the contrary, the productions of FGF, and VEGF were increased in the serum. Also, atopic dermatitis index score in NHM-treated mice were observed in the similar levels to those of normal group. Histological examination demonstrated that NHM suppressed immune cell infiltration and thickening of epidermis, meanwhile the extraction induced collagen production in the dorsal skin. Conclusion : This study demonstrated that NHM is appeared to be effective on atopic dermatitis and skin regeneration efficacy based on the observations with hematologic, gross, and histologic examinations. Therefore, we suggest that NHM could be effectively used as an external therapeutics against atopic dermatitis and a consequence skin damage.
