Report of the Genus Rhodobates (Lepidoptera, Tineidae) New to Korea

  • A tineid genus, Rhodobates Ragonot, 1895 in Myrmecozelinae is reported for the first time from Korea, with a congener, R. cupulatus Li and Xiao, 2006. The voucher specimens of the species comprise four males and four females collected from Seoul City, Sejong City, Gangwon-do, Chungcheongbuk-do, and Chungcheongnam-do. Among the previously-known species of Tineidae in Korea, Rhodobates cupulatus is similar to Psychoides gosari Kim and Bae in having the uniform coloration over the body and wings, but differs from the latter in the much larger body size. External appearance and genital features of R. cupulatus are redescribed and illustrated. A COI sequence of R. cupulatus is provided for the first time and compared in the world database of DNA barcodes. The Korean records of the species represent the first evidence of its occurrence out of the type locality. Circumstances of its collecting in Korea suggest that it is possibly feeding on dead woods in damp environments.

A New Record of Epizoic Hydroid, Ectopleura radiata (Hydrozoa: Anthoathecata: Tubulariidae), from Korea

  • In 2022, during a survey of intertidal fauna around Jindo located in the Dadohaehaesang National Park in the South Sea, epizoic hydroids attaching onto seagrass (Zostera sp.) growing on soft sediments were collected. Through taxonomic examination, an unrecorded species, Ectopleura radiata, is newly added to the hydrozoan fauna of Korea. In addition, DNA barcoding for species from Ectopleura and Tubularia clarified the distinction among morphologically indistinguishable species without gonophores. Up to now, only one species, E. crocea, has been reported in Korea. Through this study, a total of two species belonging to genus Ectopleura have been reported in Korean waters so far.

First Record of Teloganopsis chinoi (Ephemeroptera: Ephemerellidae) Based on Larval Morphology and mtDNA in Korean Peninsula, with a Checklist of Korean Ephemerellidae

  • The genus Teloganopsis Ulmer belonging to the family Ephemerellidae is only known from the species of T. punctisetae (Matsumura, 1931) in the Korean Peninsula. The members of the genus Teloganopsis are characterized by the following characteristics: head and abdominal tergites without any tubercles and complex setae; forefemur with a row of long and stout setae perpendicularly; maxilla covered with dense setae. A total of 17 species had been previously recorded in this family. Here, Teloganopsis chinoi (Gose, 1980), is newly recorded in Korea which was found under a large stone in Kyeongho river. Larval habitus, habitat, line-drawings of key characters of the species, a checklist with habitus photos of Korean Ephemerellidae, and a key to the larvae of Korean Teloganopsis are provided.

DNA Barcoding of Aegista chejuensis and Plectotropis quelpartensis (Gastropoda: Stylommatophora: Camaenidae)

  • Two land snails, Aegista chejuensis (Pilsbry and Hirase, 1908) and Plectotropis quelpartensis (Pilsbry and Hirase, 1908), are endemic to Korea and were collected from Hataedo and Jodo Islands in the Yellow Sea of South Korea, respectively. Many terrestrial snail habitats have been confirmed in Korea; however, their genetic sequences have rarely been reported. This study describes the mitochondrial cytochrome c oxidase subunit I gene (COI) sequences of two species, followed by an analysis of the genetic distance between these two species and their congeners. As a result, there was no intra-species variation in both species A. chejuensis or P. quelpartensis. However, the inter-species variation was clear (10.3-31.5%). We provide photographs and a brief diagnosis for morphological verification.

A New Record of Phyllidia varicosa (Nudibranchia: Phyllidiidae) from Korea

  • In this article, a phyllidiid nudibranch which is distributed widely in the Indo-Pacific region, Phyllidia varicose Lamarck, 1801, is reported based on a specimen collected from Seopseom Islet, Jeju Island, Korea. This species is characterized by bluish-gray dorsal tubercles with a yellow cap and three distinct ridges consisting of dorsal tubercles going from the anterior to posterior region, the presence of bluish-black pigment between the dorsal ridges, and a characteristic black longitudinal stripe along the midline of the sole. In this study, we provide a key to species belonging to the genus Phyllidia discovered in Korea, the morphological descriptions, photographs, and a sequence of partial mitochondrial cytochrome c oxidase subunit I of P. varicosa. Currently, four species of the genus Phyllidia have been reported to be present in Korea, including P. varicosa.

Three Seed Beetles (Coleoptera: Chrysomelidae: Bruchinae) New to South Korea, with DNA Barcoding Data

  • Three species of seed beetles, Acanthoscelides pallidipennis (Motschulsky, 1874), Bruchidius terrenus (Sharp, 1886), and Kytorhinus senilis Solsky, 1869, from South Korea are reported for the first time. These three species can be morphologically distinguished from other Korean bruchid species by the distinctive color pattern of the elytra, presence of subapical spines on hind femur, and exposed last three tergites of abdomen, respectively. In this study, partial sequences of mitochondrial cytochrome c oxidase subunit I from Korean specimens of these species were generated. In addition, host plants, distribution maps, and photographs of the dorsal habitus and live specimens of each species are also included.

Platynosomum fastosum (Trematoda: Dicrocoeliidae) from Cats in Vietnam: Morphological Redescription and Molecular Phylogenetics

  • The present study was performed to reveal the morphological characteristics and molecular phylogenetic position of Platynosomum fastosum Kossack, 1910. A total 167 specimens of P. fastosum were collected in 8 (4.9%) out of 163 sets of gall-bladders and bile ducts of cats. The number of worms was 1-105 per infected cat. This species was characterized by having a long and slender body, slightly larger ventral sucker than the oral sucker, indistinct prepharynx, small pharynx, short esophagus, bifurcation midway between 2 suckers, and ceca extending to the posterior end of the body. The length of the partial sequences of ITS1 and 5.8S rDNA of P. fastosum were 990 bp, GC-rich. AT/GC ratio was 0.9, there were 9 polymorphic sites, and intraspecific variations ranged from 0.1% to 0.9%. Phylogenetic analyses by neighbor-joining phylogram inferred from ITS1 rDNA sequences revealed that the genetic distance between P. fastosum specimens ranged from 0.3 to 1.5% while the smallest interspecific distance among dicrocoeliid species was 20.9 %. The redescription and genetic characters of P. fastosum are taxonomically important to recognize future different species of the genus Platynosomum showing high intraspecific and morphological variability.

미토콘드리아 Cytochrome b 유전자의 염기서열 분석을 이용한 한국산 총알고둥(복족강, 총앙고둥과)의 지리적 변이 및 오염.비오염지역간의 유전적 다양성 (Geographic Variation and Genetic Diversity between Polluted and Unpolluted Sites of Korean Littorina brevicula(Gastropoda, Littorinidae) Based on the Mitochondrial Cytochrome b Gene Sequence)

  • 한국산 총알고둥(Littorina brevicula)의 지리적 변이를 조사하기 위하여 동해안, 남해안, 서해안에서 총 11개 집단 106개체를 대상으로 미토콘드리아 DNA cytochrome b 유전자의 염기서열을 분석하였으며, 분석 결과 총 500 bp의 염기서열을 검출하였다 검출된 염기서열을 대상으로 염기치환 유무 및 치환 장소를 비교한 결과 13종류의 haplotype으로 구분되었으며, 그 중 LbA가 주 haplotype으로 나타났다. LbA의 평균 출현빈도는 0.877이었으며, 동해안은 0.82, 남해안 0.70, 서해안 1.00으로 각각 나타나 동해안 집단이 타 집단에 비해 haplotype의 다양성이 더 높았다. 특히 오염지역과 비오염지 역간의 비교에서는 8종류의 haplotype이 구분되었으며, 역시 LbA가 주 haplotype으로 나타났다.

rDNA-ITS 분석에 의한 망태버섯속균(Dictyophora spp.)의 종간 구분 가능성 (Interspecific Distinguishability of Veiled Lady Mushrooms (Dictyophora spp.) Based on rDNA-ITS Analysis)

  • 본 연구는 망태버섯속(Dictyophora species) 균의 종 구분을 위하여 국내수집 5균주와 국외도입 6균주에 대한 rDNA ITS 영역의 PCR-RFLP 및 염기서열을 분석하였다. 그 결과, ASI 32001, 32003, 32005, 32006, 32011이 D. indusiata, ASI 32002, 32007, 32008, 32010이 D. echinovolvata, 그리고 ASI 32004와 Phallus rugulosus ASI 25007은 같은 군으로 그룹화 되었다. 이 결과는 기존에 조사된 배양온도, 배지pH, 선택배지 등 배양적 특성과 재배적, 형태적 특성에 의한 구분과 일치하는 경향이었다. 따라서 rDNA ITS 영역의 PCR-RFLP 및 염기서열분석은 Dictyophora속 보존균주의 종 구분에 있어서 선행적, 보충적 수단으로 활용하기에 충분하다고 판단된다.

한국산 꿀풀과 15 분류군에 대한 유전체양 조사 (Genome size of 15 Lamiaceae taxa in Korea)

  • 한 생물체의 전체 유전체 크기는 계통학, 육종학, 집단유전학, 진화학과 같은 많은 분야에 활용될 수 있는 기본적인 정보이다. 최근에는 전체 유전체 결정 연구에서 특히 강조되고 있는데, 이는 최소 유전체 크기를 갖는 분류군의 선택은 유전체 결정사업의 효율성과 직접적으로 연관되어 있기 때문이다. 그러므로 유전체 연구의 선행 단계로서 연구 대상 종 및 연관된 분류군들의 유전체 양의 파악은 필수적이다. 본 연구에서는 쉽고 빠르면서도 신뢰성 있는 방법으로 알려져 있는 flow cytometry를 이용하여 한반도에 자생하는 꿀풀과의 9속 15 분류군에 대한 유전체 크기를 측정하였다. 본 연구에서 유전체 양이 측정된 15 분류군들은 모두 최초로 그 유전체 양이 조사된 분류군들로서 Plant DNA C-value Database (http://data.kew.org/cvalues/)에 수록된 바 없는데, 특히 Agastache, Clinopodium, Elsholtzia, Isodon에 속하는 분류군들은 속 수준에서의 최초의 보고이다. 골무꽃(Scutellaria indica L.)은 0.37 pg (1C)의 유전체 크기를 갖는 것으로 측정되었는데, 이는 현재까지 보고된 꿀풀과 98 분류군의 유전체 양들 중 네 번째로 유전체의 크기가 작은 분류군이다. 이에 골무꽃은 향후 유전체 연구를 위해 꿀풀과를 대표할 한국 자생종으로서 우선적으로 선택하여 분석할 수 있는 종일 것이다. 조사된 분류군들 중 가장 유전체 크기가 큰 분류군은 속단(Phlomis umbrosa Turcz.; 1C=2.6 pg)으로서 이는 다배체 형성에 의한 본 종의 기원 가능성을 제시하고 있다.
