함초 추출물의 세포사멸 유도에 의한 난소암세포에서 항암 효과 (Anti-cancer effects of Salicornia herbacea extract in OVCAR-3 cells by induction of apoptosis pathway)

  • 본 연구에서는 천연유래 물질로부터 apoptosis를 유도하여 항암 활성이 있는지에 관한 실험으로 사람 난소암 세포주인 OVCAR-3 세포에 함초의 유기용매별 추출물을 처리하여 결과를 확인하였다. MTS 측정으로 세포 생존율을 확인한 결과 DCM 분획물에서 농도별로 유의적인 세포수의 감소를 보였으며, annexin V/FITC-PI 염색에 의해 apoptosis 유도로 세포가 사멸함을 확인하였다. DCM 분획물 처리는 세포주기에서 Sub-G1기의 증가로 세포증식이 억제됨을 보여준다. 세포의 내인성 경로에 관여하는 Bcl-2 family에 속하는 Bax, Bak, Bcl-2, Bcl-xL의 상호작용을 mRNA 수준에서 확인한 결과, DCM 분획물처리에서 Bax, Bak가 증가하고, Bcl-2의 감소를 동반하여 세포사멸의 신호전달 경로가 진행됨을 확인할 수 있었다. 본 연구 결과를 바탕으로 함초를 이용하여 여성의 난소암에 예방적 기능성 식품을 개발할 수 있을 것으로 판단되며, 함초의 DCM 분획물에 대해 깊이 있는 심층적 연구가 요구된다.

조릿대의 잎과 줄기 추출물 분획의 염증 및 비만 억제 효과 비교 (Comparative Study for Anti-inflammatory and Anti-obesity Effect of Fractions from Leaf and Stem of Sasa Borealis)

  • Sasa borealis has long been used in folk remedies and traditional Korean medicine for analgesic, sedative and anticarcinogenic purposes. To excavate novel natural compounds for treating obesity, we separated leaf and stem and fractionated. We screened phenolic contents, anti-oxidative and anti-inflammatory properties with the fractions. Dichloromethane (DCM) and ethyl acetate (EA) fraction from both leaf and stem exhibited high anti-oxidative properties demonstrated by DPPH, ABTS assay in accordance with their phenolic contents. In addition, DCM and EA fraction from leaf inhibited nitric oxide (NO) production as well as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in Raw 264.7 cell. Finally, DCM and EA fraction from both leaf and stem significantly inhibited 3T3-L1 preadipocyte differentiation and decreased intracellular lipid level. These results suggest that DCM and EA fraction from Sasa borealis may contain therapeutic agent for obesity by attenuating inflammation and oxidative stress.

Anti-invasive Activity against Cancer Cells of Phytochemicals in Red Jasmine Rice (Oryza sativa L.)

  • Red rice contains pharmacological substances including phenolics, oryzanol, tocotrienol and tocopherol. Recently, red rice extract has been employed as a source of antioxidants for inhibition of tumor growth. This study was carried out to evaluate the anti-invasion effects of red rice extract fractions on cancer cells. It was found that at $100{\mu}g/ml$ of crude ethanolic extract (CEE), hexane fraction (Hex) and dichloromethane fraction (DCM) could reduce HT1080 and MDA-MB-231 cancer cell invasion. Hex and DCM revealed higher potency levels than CEE, whereas an ethyl acetate fraction (EtOAc) had no effect. Gelatin zymography revealed that Hex decreased the secretion and activity of matrix metalloproteinase-2 and -9 (MMP-2 and-9). In contrast, the DCM fraction exhibited slightly effect on MMPs secretion and had no effect on MMPs activity. Collagenase activity was significantly inhibited by the Hex and DCM fractions. High amounts of ${\gamma}$-oryzanol and ${\gamma}$-tocotrienol were found in the Hex and DCM fractions and demonstrated an anti-invasion property. On the other hand, proanthocyanidin was detected only in the CEE fraction and reduced MDA-MB-231 cells invasion property. These observations suggest that proanthocyanidin, ${\gamma}$-oryzanol and ${\gamma}$-tocotrienol in the red rice fractions might be responsible for the anti invasion activity. The red rice extract may have a potential to serve as a food-derived chemotherapeutic agent for cancer patients.

3T3-L1 지방세포에 대한 와송 디클로로메탄 추출물의 지질 대사 개선에 관한 연구 (The Lipid Efflux Effects of Dichloromethane Extract from Orostachys japonicus in 3T3-L1 Adipocyte Cells)

  • 본 연구는 와송 유기용매 추출물중 DCM층을 이용하여 지방세포로 유도된 3T3-L1세포에 대한 지질대사 개선효과를 확인하는 연구이다. 세포독성을 확인하기 위하여 MTS-assay를 이용하여 6가지 분획의 와송 유기용매 추출물(EtOH, Hexane, DCM, EToAc, BuOH, H2O)에 대한 독성검사를 실시하여 세포에 대한 안정성을 확인하였다. 그 결과 DCM추출물이 전 농도에 걸쳐 안정성이 있음을 확인하고, 지질대사 개선효과를 확인하기 위하여 DCM층을 사용하였다. 우선 지질대사 개선효과를 확인하기 위하여 지질 배출능을 측정하였다. 지방세포로 유도된 3T3-L1세포에 대하여 DCM추출물을 처리한 후 oil-red O염색을 실시하여 지질 축적량을 평가한 결과 지질 배출능이 개선된 것을 확인할 수 있었다. 또한 지질 배출능 분석을 위하여 지질수송 단백질인 ABCA1, ABCG1의 mRNA발현을 real-time PCR로 확인한 결과 DCM추출물 처리 시 유의적인 발현 증가가 나타났음을 확인 하였다. 따라서 본 연구를 통하여 와송 DCM추출물이 3T3-L1 지방세포에 대하여 지질대사 개선효과가 있음을 확인하였다.

Anti-inflammatory effects of ethanol extract from Orostachys japonicus on modulation of signal pathways in LPS-stimulated RAW 264.7 cells

  • In this study, powder of Orostachys japonicus A. Berger (O. japonicus) was extracted with 95% ethyl alcohol and fractionated using a series of organic solvents, including n-hexane (hexane), dichloromethane (DCM), ethylacetate (EtOAc), n-butanol (BuOH), and water ($H_2O$). We investigated the anti-inflammatory effects of these O. japonicus extracts on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Their effects on the expression of inflammatory mediators and transcription factors were analyzed by Western blotting. DCM fraction significantly inhibited formation of reactive oxygen species (ROS) as well as nitric oxide (NO) in LPS-stimulated RAW 264.7 cells. Phosphorylation of the pro-inflammatory transcription factor complex nuclear factor-kappa B (NF-${\kappa}$B) p65 and expression of inducible nitric oxide synthase (iNOS), one of its downstream proteins, were also suppressed by DCM fraction. These effects were regulated by upsteam proteins in the mitogen-activated protein kinase (MAPK) and phosphoinositide 3-kinase/Akt (PI3K/Akt) signaling pathways. Taken together, our data suggest that O. japonicus could be used as a potential source for anti-inflammatory agents.

Anticancer Effects of Leek Kimchi on Human Cancer Cells

  • The anticancer effects of leek (buchu in Korean) kimchi were evaluated in the human cancer cells: AGS gastric adenocarcinoma cells, HT-29 human colon adenocarcinoma cells and HL-60 leukemia cells. The leek kimchi (fermented for 6 days at 15$^{\circ}C$) was fractionated into 7 groups: methanol extract, hexane extract, methanol soluble extract MSE), dichloromethane (DCM) fraction (fr.), ethyl acetate fr., butanol fr. and aqueous fr. Most of the leek kimchi tractions inhibited the growth of AGS and HT-29 cancer cells in a dose dependent manner. In particular, the DCM fr. showed the highest inhibitory effect among the tractions. Treatment with the DCM fr. (0.1 mg/mL) reduced the survival rates of AGS and HT-29 cancer cells to 19% and 37% of the controls, respectively. Moreover the DCM fr. of the leek kimchi arrested G2/M phase in the cell cycle and induced apoptosis in HL-60 human promyelocytic leukemia cells. These results indicate that the leek kimchi exerted an anticancer effect on those human cancer cells, and that the DCM fr. arrested G2/M phase in the cell cycle and induced apoptosis in the leukemia cells.

Comparative Antitumor Activity of Different Solvent Fractions from an Auricularia auricula-judae Ethanol Extract in P388D1 and Sarcoma 180 Cells

  • The objective of this study was to evaluate and compare the antitumor activity of different solvent fractions (ethanol, dichloromethane, ethyl acetate, butanol and water) of the Auricularia auricula-judae 70% ethanol extract on the P388D1 macrophage and sarcoma 180 cells. A dose-dependent antitumor activity of each solvent fraction (from 0.01 mg/ml to 0.3 mg/ml) was shown against both cell types. These cytotoxic effects of all the tested fractions were confirmed on the MTT and SRB assays, without statistical differences each other. $IC_{50}$ value of dichloromethane fraction was 94.2 ${\mu}g/ml$ against sarcoma 180 cells lower than any other solvent fractions. The potent antitumor effect of the dichloromethane (DCM) fraction was also found against solid tumor in BALB/c mice. The splenomegaly and higher splenic index were found in tumor-bearing mice, with the DCM fraction returning to the negative control values. Thus, the results indicated the dichloromethane fraction may have potential ingredients as antitumor candidates.

번행초 추출물의 당뇨관련 효소에 관한 저해 활성 (Inhibitory Activity on the Diabetes Related Enzymes of Tetragonia tetragonioides)

  • 본 연구에서 당뇨병의 예방, 치료제 및 건강기능 식품으로서 산업적 활용가능성을 평가하기 위한 기초연구로서 번행초 추출물을 사용하여 당뇨관련 효소인 $\alpha$-amylase, $\alpha$-glucosidase, $\beta$-glucosidase 그리고 aldose reductase에 대한 저해활성을 조사하였다. 번행초의 HX 분획물은 인간 타액, 돼지 췌장 $\alpha$-amylase에 대해 5 mg/mL 농도에서 각각 62.23%, 88.74%로 높은 저해효과를 나타냈으며, $\alpha$-glucosidase에 대해서도 HX 분획물이 1 mg/mL 농도에서 60.27%로 높은 저해효과를 나타냄으로써 현재 혈당강하제로 사용되고 있는 acarbose와 1-deoxynorjirimycin보다 높은 억제 효과를 보였다. 그리고 만성 합병증에 의한 시력장애, 신경성장애, 신장기능장애, 심장기능 장애 등의 심각한 당뇨병성 합병증 유발과 관련이 있는 aldose reductase에 대한 억제활성을 조사한 결과, 1 mg/mL 농도에서 번행초 DCM 분획과 HX 분획물이 각각 51.95%, 47.22%로 높은 저해활성을 나타냄으로써 당뇨병과 그로 인한 합병증의 예방에도 효과가 있는 것으로 판단된다. 전 세계적으로 당뇨병 환자가 증가추세에 있어 당뇨병 치료제의 시장 규모는 더욱 확대될 가능성이 높은 가운데 지금까지 보고되지 않은 번행초의 항당뇨 효능의 우수성을 검증함으로써 바이오산업육성뿐만 아니라 신소득 작물로 순화재배를 통한 농가소득 향상에도 기여할 것으로 기대된다.

HPLC와 LC/MS에 의한 식육내 잔류 설파제의 동시 분석법 (Simultaneous analysis of sulfonamides in beef and pork by high performance liquid chromatography and electrospray ionization mass spectrometry)

  • A multiresidual analysis was performed to determine 12 sulfonamides(sulfacetamide, sulfadiazine, sulfisomidine, sulfathiazole, sulfapyridine, sulfamerazine, sulfamethazine, sulfamonomethoxine, sulfisoxazole, sulfamethoxazole, sulfaquinoxaline, and sulfadimethoxine) in beef and pork simultaneously. The multiresidual analysis for the sulfonamides currently used was able to analyze 5 kinds of sulfonamides at the same time. The method of this 12 sulfonamides multiresidual analysis in this study was matrix solid-phase dispersion(MSPD) by high performance liquid chromatography (HPLC) and liquid chromatography mass spectrometry (LC/MS). The recovery rate of the materials was measured by MSPD method with 3 different extraction solvents; Dichloromethane, DCM: Ethylacetate(3:1), DCM:EA(9:1). Also, samples (84 beef and 205 pork samples) which were positive by EEC-4 plate test from 2001 to 2003 were tested to investigate the kinds of sulfonamides using HPLC. The results from the study were as follows; 1. The recovery rate of the materials was measured by MSPD method with 3 different extraction solvents; Dichloromethane, DCM:Ethylacetate(3:1), DCM:EA(9:1). The method of extraction solvent with DCM:ethyl acetate(9:1) was the most excellent(87.7∼99.3%) in separation and reappearance. 2. In the LC/MS analysis. of sulfonamides, signal to noise ratio was showed relatively high in the positive mode and special ion in the quality analysis was determined via [M+H]$\^$+/ and m/z 156. A spectrum of sulfonamides was showed from all 12 sulfonamides. 3. The samples positive by the EEC-4 plate, a screening test method, were categorized by sulfonamides through Charm II and confirmed the kinds of sulfonamides through HPLC. 1) Among 84 beef samples positive by EEC-4 plate, 20 samples were positive by Charm II and identified as 7 sulfamethazine, 9 sulfadimethoxine, 1 sulfamonomethoxine and 3 unknown status. 2) Among 205 pork samples positive by EEC-4 plate, 42 samples were positive by Charm II and identified as 19 sulfamethazine, 1 sulfadimethoxine, 4 sulfamonomethoxine and 5 unknown status.

저근백피성분의 생리활성에 관한 연구(III) - 디클로드메탄분획의 항암작용 - (Studies on the Biological Activities of the Constituents of Ailanthi Cortex Radicis III - Antitumor activities of dichloromethane fration -)

  • The cytotoxic activities of methanolic extract and its fractions of Ailanthi Cortex Radicis and column chromatographic eluates of its dichloromethane fraction (DCM fr.) were investigated. DCM fr. Showed the strongest cytotoxicity against hepatoma cells. Furthermore, the active equates 1-3, 8 and 9 were obtained. Effects on free radical generation and the growth of vascular endothelial cells were tested to elucidate the action mechanism of anticancer activity. Eluates 1-3 stimulated free radical generation, while eluates 8 and 9 showed no changes. Especially, eluates 8 and 9 efffectively inhibited the proliferation of vascular endothelial cells in a dose- dependant manner. It is speculated that the anticancer effects of eluates 1-3, 8 and 9 might be due to free radical generation and inhibition of endothelial cell growth, respectively.
