Biochemical Characteristics of a Bacteria (Bacillus pseudomycoides) Alanine Racemase Expressed in Escherichia coli (Bacillus pseudomycoides로 부터 분리된 alanine racemase 유전자의 발현 및 생화학 특성)

  • A gene encoding a putative alanine racemase in B. pseudomycoides was cloned and expressed in Escherichia coli BL21(DE3) using a pET-21 vector harbouring 6xHistidine tag. Affinity purification of the recombinant alanine racemase with a nickel resin resulted in one band by SDS-PAGE analysis. The purified enzyme showed a molecular weight of 46 kDa. The enzyme was the most active toward L-alanine and secondly D-alanine, implying that the enzyme is an alanine racemase. D-cysteine significantly inhibited the enzyme activity and also L-cysteine to a lesser extent. The enzyme was considerably activated by addition of pyridoxal-5'-phosphate (PLP), showing that 73% increase in activity was observed at 0.3 mM, compared to control. The enzyme was the most active at pH 9.0 and more stable at alkaline pHs than acidic pHs.

Effects of oscillation parameters on aerodynamic behavior of a rectangular 5:1 cylinder near resonance frequency

  • Large Eddy Simulation (LES) is used to explore the influence of vibration frequency and amplitude on the aerodynamic performance of a rectangular cylinder with an aspect ratio of B/D=5 (B: breadth; D: depth of cylinder) at a Reynolds number of 22,000 near resonance frequency. In smooth flow conditions, the research employs a sequence of three-dimensional simulations under forced vibration with diverse frequency ratios fe / fo = 0.8-1.2 (fe : oscillation frequency; fo : Strouhal frequency when the rectangular cylinder is stationary ) and oscillation amplitudes Ah/D = 0.05 - 0.3. The individual influences of fe / fo and Ah/D on the characteristics of integrated and distributed aerodynamic forces are the focal points of discussion. For the integrated aerodynamic force, particular emphasis is placed on the analysis of the dependence of velocity-proportional component C1 and displacement-proportional component C2 of unsteady aerodynamic force on amplitude and frequency ratio. Near the resonance frequency, the dependencies of C1 and C2 on amplitude are stronger than that of frequency ratio. For the distributed aerodynamic force, the increase in frequency and amplitude promotes the position of the main vortex core and reattachment to the leading edge in the streamwise direction. In the spanwise direction, vibration enhances the spanwise correlation of aerodynamic force to weaken the three-dimensional effect of the flow field, and a lower frequency ratio and larger amplitude amplify this effect.

Estimation of Weaning Age Effects on Growth Performance in Berkshire Pigs

  • Analysis for back fat thickness (BFAT) and daily body weight gains from birth to the end of a performance test were conducted to find an optimal method for estimation of weaning age effects and to ascertain impacts of weaning age on the growth performance of purebred Berkshire pigs from a closed population in Korea. Individual body weights were measured at birth (B), at weaning (W: mean, 22.9 d), at the beginning of the performance test (P: mean, 72.7 d), and at the end of the performance test (T: mean, 152.4 d). Further, the average daily gains in body weight (ADG) of 3,713 pigs were analyzed for the following periods: B to W (DGBW), W to P (DGWP), P to T (DGPT), B to P (DGBP), B to T (DGBT), and W to T (DGWT). Weaning ages ranged from 17 to 34 d, and were treated as fixed (WF), random with (WC) and random without (WU) consideration of an empirical relationship between weaning ages in the models. WF and WC produced the lowest AIC (Akaike Information Criterion) and least fractions of error variance components in multi-traits analysis, respectively. The fractions of variances due to diverse weaning age and the weaning age correlations among ADGs of different stages (when no overlapping allowed) by WC ranged from 0.09 to 0.35 and from -0.03 to 0.44, respectively. The maximum weaning age effects and optimal back fat thicknesses were attained at weaning ages of 27 to 32 d. With the exception of DGBW, the effects of weaning age on the ADGs increased (ranging from 1.50 g/d to 7.14 g/d) with increased weaning age. In addition, BFAT was reduced by 0.106 mm per increased day in weaning age. In conclusion, WC produced reasonable weaning age correlations, and improved the fitness of the model. Weaning age was one of crucial factors (comparable with heritability) influencing growth performance in Berkshire pigs. Further, these studies suggest that increasing weaning age up to 32 d can be an effective management strategy to improve growth performance. However, additional investigations of the costs and losses related to extension of the suckling period and on the extended range of weaning age are necessary to determine the productivity and safety of this practice in a commercial herd and production system.

Identification of Novel SNPs with Effect on Economic Traits in Uncoupling Protein Gene of Korean Native Chicken

  • The avian uncoupling protein (avUCP) is a member of the mitochondrial transporter superfamily that uncouples proton entry in the mitochondrial matrix from ATP synthesis. The sequencing analysis method was used to identify nucleotide polymorphisms within the avUCP gene in Korean native chicken (KNC). This study identified ten single nucleotide polymorphisms (SNPs) in the avUCP gene. We analyzed the SNPs of the avUCP gene to investigate whether polymorphism in the gene might be responsible for quantitative variations in economic traits in KNC. Three significant polymorphic sites for economic traits were avUCP C+282T (mean body weight, p<0.05), avUCP C+433T (daily percent lay, p<0.05), and avUCP T+1316C (daily percent lay, p<0.05). The frequency of each SNP was 0.125 (C+282T in avUCP gene exon 1 region), 0.150 (C+433T in avUCP gene intron 1 region), and 0.15 (T+1316C in avUCP gene exon 3 region), respectively. Among the identified SNPs, one pair of SNPs (genotype CC, C+282T and TT, avUCP C+433T) showed the highest daily percent lay (p<0.05) and mean body weight (p<0.05) and the frequency was 0.067. This study of the avUCP gene could be useful for genetic studies of this gene and selection on economic traits for KNC.

Linkage Disequilibrium and Effective Population Size in Hanwoo Korean Cattle

  • This study presents a linkage disequilibrium (LD) analysis and effective population size ($N_e$) for the entire Hanwoo Korean cattle genome, which is the first LD map and effective population size estimate ever calculated for this breed. A panel of 4,525 markers was used in the final LD analysis. The pairwise $r^2$ statistic of SNPs up to 50 Mb apart across the genome was estimated. A mean value of $r^2$ = 0.23 was observed in pairwise distances of <25 kb and dropped to 0.1 at 40 to 60 kb, which is similar to the average intermarker distance used in this study. The proportion of SNPs in useful LD ($r^2{\geq}0.25$) was 20% for the distance of 10 and 20 kb between SNPs. Analyses of past effective population size estimates based on direct estimates of recombination rates from SNP data demonstrated that a decline in effective population size to $N_e$ = 98.1 occurred up to three generations ago.

  • The performance of flat-tube radiators with louvered fins was numerically investigated for different tube insertion lengths. The results of numerical analysis using CFX-11 were compared with experimental results. In this study, three types of flat-tube radiators with louvered fins were considered. An experiment was conducted to validate the numerical results. Flow rate ratio (FR) and Stotal were introduced to understand the uniformity of flow distribution easily. The results of numerical analysis revealed that the heat transfer rate and pressure drop increased as the mass flow rate increased. Further, the results showed that the heat transfer rate of sample 3 with h/D = 0.5 was higher than that of the other samples. The pressure drop increased as the insertion length toward the header part increased, and the pressure drop in the case of sample 3 appeared to be the highest. The factor Stotal showed that the uniformity of the flow distribution in the case of sample 1 with h/D = 0 was higher than that in the case of the other samples.

  • Until now, the clamp yoke of automobile has been largely manufactured by hot forging or burring process. Through the study, the precision cold forging process for clamp yoke has been analysed by using rigid-plastic finite element analysis code, DEFORM-3D. It has shown various results of the FEM simulation. An engineer should select the proper process considering the amount of product.

  • As a result of the reduction of natural aggregate, most of developed country have been studied the utilization of Blast-furnace Slag(BFS) as aggregate of concrete. bur, in korea there are only basic study about these even though other country are using Blast-furnace Slag production of Ready Mixed Concrete. According, in this study, we carried out fundamental experiments in order to know the material properties of BFS and possibilities of the BFS as fine aggregate of concrete. It is included that analysis concerning material properties of BFS as like specific gravity, absorption. unit weigth, grading including investigation of the surface shape by SEM, also, analysis concerning properties of concrete with BFS as like air-content surface slump. compressive strength .etc.

  • In this paper, the temperature rise within busbar part of high voltage GIS is calculated under the condition that the continuous current is carrying. Heat transfer by conduction and convection is considered between the current carryig conductor and SF6 gas. FLUX 2D which is a commercial electromagnetic and thermal analysis program is used. The results show reasonable temperature distribution.

  • This study uses a design of experiments to explain correlations between the objective function and design variables. Analysis of means, analysis of variance and table of orthogonal array were carried out. The change of shape design variable for TFLM based on the table of orthogonal array is made. Therefore this study carried to decide design variables for maximizing thrust force of TFLM. we showed improved design variables.
