Safety Evaluation of Bifidobacterium breve IDCC4401 Isolated from Infant Feces for Use as a Commercial Probiotic

  • Previously, our research group isolated Bifidobacterium breve IDCC4401 from infant feces as a potential probiotic. For this study, we evaluated the safety of B. breve IDCC4401 using genomic and phenotypic analyses. Whole genome sequencing was performed to identify genomic characteristics and investigate the potential presence of genes encoding virulence, antibiotic resistance, and mobile genetic elements. Phenotypic analyses including antibiotic susceptibility, enzyme activity, production of biogenic amines (BAs), and proportion of D-/L-lactate were evaluated using E-test, API ZYM test, high-performance liquid chromatography (HPLC), and D-/L-lactic acid assay respectively. The genome of B. breve IDCC4401 consists of 2,426,499 bp with a GC content of 58.70% and 2,016 coding regions. Confirmation of the genome as B. breve was provided by its 98.93% similarity with B. breve DSM20213. Furthermore, B. breve IDCC4401 genes encoding virulence and antibiotic resistance were not identified. Although B. breve IDCC4401 showed antibiotic resistance against vancomycin, we confirmed that this was an intrinsic feature since the antibiotic resistance gene was not present. B. breve IDCC4401 showed leucine arylamidase, cystine arylamidase, α-galactosidase, β-galactosidase, and α-glucosidase activities, whereas it did not show production of harmful enzymes such as β-glucosidase and β-glucuronidase. In addition, B. breve IDCC4401 did not produce any tyramine, histamine, putrescine, cadaverine, or 2-phenethylamine, which are frequently detected BAs during fermentation. B. breve IDCC4401 produced 95.08% of L-lactate and 4.92% of D-lactate. Therefore, our findings demonstrate the safety of B. breve IDCC 4401 as a potential probiotic for use in the food industry.

Whole Genome Sequence of Lactiplantibacillus plantarum HOM3204 and Its Antioxidant Effect on D-Galactose-Induced Aging in Mice

  • Lactiplantibacillus plantarum, previously named Lactobacillus plantarum, is a facultative, homofermentative lactic acid bacterium widely distributed in nature. Several Lpb. plantarum strains have been demonstrated to possess good probiotic properties, and Lpb. plantarum HOM3204 is a potential probiotic strain isolated from homemade pickled cabbage plants. In this study, whole-genome sequencing was performed to acquire genetic information and predict the function of HOM3204, which has a circular chromosome of 3,232,697 bp and two plasmids of 48,573 and 17,060 bp, respectively. Moreover, various oxidative stress-related genes were identified in the strain, and its antioxidant activity was evaluated in vitro and in vivo. Compared to reference strains, the intracellular cell-free extracts of Lpb. plantarum HOM3204 at a dose of 1010 colony-forming units (CFU)/ml in vitro exhibited stronger antioxidant properties, such as total antioxidant activity, 2,2-diphenyl-1-picrylhydrazyl radical scavenging rate, superoxide dismutase activity, and glutathione (GSH) content. Daily administration of 109 CFU Lpb. plantarum HOM3204 for 45 days significantly improved the antioxidant function by increasing the glutathione peroxidase activity in the whole blood and GSH concentration in the livers of D-galactose-induced aging mice. These results suggest that Lpb. plantarum HOM3204 can potentially be used as a food ingredient with good antioxidant properties.

첨가제 처리가 알팔파 사일리지의 품질에 미치는 영향 (Effects of the Additives on the Quality of Alfalfa Silage)

  • 본 시험은 첨가제 처리가 알팔파 사일리지의 품질에 미치는 영향을 구명하기 위하여 1996년 6월부터 12월까지 축산기술연구소 초지사료과에서 개화중기에 수확된 알팔파(Vernal)를 사일리지로 조제시 무처리, 개미산, 당밀 및 젖산균 제제 (inoculant A 및 inoculant B)등 5처리 3반복의 난괴법 배치로 수행하였으며 그 결과는 다음과 같다. 첨가제 처리로 알팔파 사일리지의 조단백질 및 NFE 함량은 대조구에 비해 높게 나타났고, 조회분 및 조섬유 함량은 대조구에서 11.5 및 39.6%로 높았다. ADF 및 NDF 함량은 대조구에서 36.2 및 48.6%로 첨가제 처리구보다 높았으며 사일리지 조제전에 비해 약 4% 증가하였다. 사일리지의 pH는 대조구에서 5.45로 가장 높았고 개미산 및 inoculant A 처리구에서 4.35 및 4.32로 유의적으로 낮게 나타났다. 또한 건물 함량은 대조구에서 유의적으로 낮았으며 건물손실률은 대조구에서 높았으며 첨가제 처리구간에는 유의적인 차이가 없었다. 암모니아태 질소 함량은 개미산 처리구에서 가장 낮았고 젖산균수는 무처리 및 개미산 처리구에서 낮은 경향을 보였다. 첨가제 처리에 따른 유기산 조성에 있어서는 inoculant A 및 inoculant B 처리구에서 총산 및 젖산 함량이 높게 나타났으며 품질등급에 있어서도 젖산균제 처리구가 가장 높았다. 이상의 결과를 종합하여 볼 때 고품질의 알팔파 사일리지를 조제할때는 첨가제 처리가 효과적이며 안전성, 기계부식 문제를 고려할 때 살포가 용이한 젖산균제의 이용이 바람직한 것으로 생각된다.

파프리카 발효즙의 화장품 소재개발 연구 (Development of Cosmetic Ingredient by Fermented Paprika Juice)

  • 본 연구에서는 천연물 화장품 원료의 유기용매 추출법과 미생물을 이용한 발효법의 장단점을 보완하기 위해, 생(生) 천연물에 미생물을 이용한 발효를 통해 과즙을 만드는 새로운 제조법으로 화장품 소재를 개발하였다. 천연물은 다양한 색상과 비타민이 풍부한 것으로 알려진 파프리카 중 두 가지 색상(빨간색, 녹색)을 선정하였고, 발효에 사용된 미생물로서 당분해 효소능이 있는 유산균(Lactobacillus plantarum)을 이용해 파프리카에 접종하여 발효하였다. 먼저 고압으로 착즙한 파프리카 생즙 2종과 유산균으로 발효한 발효즙 2종의 생리활성변화를 확인해 보았다. 총 페놀함량과 총 플라보노이드 함량 측정 실험 결과, 파프리카 생즙보다 발효즙에서 모두 함량이 높았으며 특히 빨간색 파프리카 발효즙에서 가장 높은 함량을 나타내었다. 프리라디칼 소거효과와 지질과산화 억제효과 역시 파프리카 생즙에 비해 발효즙에서 우수한 항산화 효과가 나타났으며, 그중 빨간색 파프리카 발효즙의 효과가 가장 높았다. 항산화 효과가 높은 빨간색 파프리카 발효즙을 이용해 MMP-1 발현실험을 한 결과 농도 의존적으로 MMP-1 mRNA와 MMP-1 단백질의 발현을 억제하였다. 노화에 따른 glycation(당화현상) 실험의 경우, 파프리카 발효즙에서 상대적으로 anti-glycation 효과로 최종 당화산물(advanced glycation end-products, AGEs) 생성 억제 활성이 높아 항산화효과와 밀접한 연관성을 나타내었다. 또한, 세포노화 지표물질인 senescence-associated ${\beta}$-galactosidase (SA-${\beta}$-gal) 활성을 사람 섬유아세포(HDF)를 이용하여 실험한 결과, 파프리카 발효즙을 처리하였을 때 노화에 의해 염색된 세포의 수가 감소하여 세포의 senescence를 억제하는 것을 확인할 수 있었다. 결론적으로, 파프리카는 생즙보다 유산균을 이용한 발효즙이 항산화 효과와 항노화 효능이 우수했으며, 그중 빨간색 파프리카 발효즙이 가장 우수한 항산화와 항노화 효능을 가져 이를 이용한 항산화 및 항노화의 천연 신소재로 적용될 수 있을 것으로 사료된다.

Effects of Soy Isoflavone on Performance, Meat Quality and Antioxidative Property of Male Broilers Fed Oxidized Fish Oil

  • This study was conducted to determine the effect of dietary soy isoflavone (ISF, 0 and 20 mg/kg) on performance, meat quality and antioxidative property in male broilers. Six replicates of 45 birds (42 d old) were used for each treatment. The birds were fed soybean meal-free diets containing 3% oxidized fish oil (acid value, 6.76 mg potassium hydroxide/g; and peroxide value, 6.18 meq/kg) for 3 wk. The results showed that average daily gain, feed intake, feed conversion and carcass traits were not affected by soy ISF. Compared with the control group, breast muscle color redness value and water holding capacity were increased (p<0.05) by ISF supplementation. The activity of plasma catalase was increased by supplementing with 20 mg ISF/kg diet. In breast muscle, Broilers fed 20 mg ISF/kg had decreased production of malondialdehyde and lactic acid. The ISF supplementation elevated total antioxidative capacity and activities of total superoxide dismutase and glutathione peroxidase. The results indicated that dietary ISF could increase redness and water holding capacity of meat, and antioxidative property of meat in male broilers fed oxidized fish oil diet.

Quality Improvement of Pork Loin by Dry Aging

  • This study aimed to investigate the effects of dry aging on the quality of pork loin. Longissimus lumborum muscles were dissected from the right half of five pork carcasses and were used as the control samples. The left halves of the carcasses were aged at 2±1℃ and a relative humidity of 80% for 40 d. The total aerobic bacteria count was similar between the control and dry-aged pork loin (p>0.05). Lactic-acid bacteria was absent in both the control and dry-aged pork loins. Dry-aged pork loin contained low moisture and high protein and ash compared to the controls (p<0.05). The pH was higher and cooking loss was lower in dry-aged pork loin compared to that in the control (p<0.05). Flavor related compounds, such as total free amino acid, hypoxanthine, and inosine of pork loin were higher in dry-aged pork loin; whereas, inosine 5'-monophosphate and guanosine 5'-monophosphate were low in dry-aged pork loin than control (p<0.05). There was no difference in carnosine and anserine content between dry-aged pork loin and the control (p>0.05). Dry-aged pork loin had lower hardness and shear force and received higher core in sensory evaluation than the control (p<0.05). According to the results, dry aging improved textural and sensorial quality of pork loin.

Formulation and Characterization of Antigen-loaded PLGA Nanoparticles for Efficient Cross-priming of the Antigen

  • Background: Nanoparticles (NPs) prepared from biodegradable polymers, such as poly (D,L-lactic acid-co-glycolic acid) (PLGA), have been studied as vehicles for the delivery of antigens to phagocytes. This paper describes the preparation of antigen-loaded PLGA-NPs for efficient cross-priming. Methods: NPs containing a similar amount of ovalbumin (OVA) but different sizes were produced using a micromixer-based W/O/W solvent evaporation procedure, and the efficiency of the NPs to induce the cross-presentation of OVA peptides were examined in dendritic cells (DCs). Cellular uptake and biodistribution studies were performed using fluorescein isothiocyanate (FITC)-loaded NPs in mice. Results: The NPs in the range of $1.1{\sim}1.4{\mu}m$ in size were the most and almost equally efficient in inducing the cross-presentation of OVA peptides via $H-2K^b$ molecules. Cellular uptake and biodistribution studies showed that opsonization of the NPs with mouse IgG greatly increased the percentage of FITC-positive cells in the spleen and lymph nodes. The major cell type of FITC-positive cells in the spleen was macrophages, whereas that of lymph nodes was DCs. Conclusion: These results show that IgG-opsonized PLGA-NPs with a mean size of $1.1{\mu}m$ would be the choice of biodegradable carriers for the targeted-delivery of protein antigens for cross-priming in vivo.

Functional Expression and Characterization of Acetyl Xylan Esterases CE Family 7 from Lactobacillus antri and Bacillus halodurans

  • Acetyl xylan esterase (AXE; E.C. 3.1.1.72) is one of the accessory enzymes for xylan degradation, which can remove the terminal acetate residues from xylan polymers. In this study, two genes encoding putative AXEs (LaAXE and BhAXE) were cloned from Lactobacillus antri DSM 16041 and Bacillus halodurans C-125, and constitutively expressed in Escherichia coli. They possess considerable activities towards various substrates such as p-nitrophenyl acetate, 4-methylumbelliferyl acetate, glucose pentaacetate, and 7-amino cephalosporanic acid. LaAXE and BhAXE showed the highest activities at pH 7.0 and 8.0 at 50℃, respectively. These enzymes are AXE members of carbohydrate esterase (CE) family 7 with the cephalosporine-C deacetylase activity for the production of antibiotics precursors. The simultaneous treatment of LaAXE with Thermotoga neapolitana β-xylanase showed 1.44-fold higher synergistic degradation of beechwood xylan than the single treatment of xylanase, whereas BhAXE showed no significant synergism. It was suggested that LaAXE can deacetylate beechwood xylan and enhance the successive accessibility of xylanase towards the resulting substrates. The novel LaAXE originated from a lactic acid bacterium will be utilized for the enzymatic production of D-xylose and xylooligosaccharides.

동맥경화도와 생리학적 변수들 간의 상관관계 (Correlation between Arterial Stiffness and Physiological Parameters)

  • Arterial stiffness(AS) is an important pathologic state of vascular injury. This study was carried out to elucidate the effect of physiological variables on brachial-ankle pulse wave velocity(BAPWV), index of AS. Four hundred adults(volunteers) participated in this study. Body indices, biochemical, cardiac and inflammatory markers, and right(Rt)- and left(Lt)-BAPWV were measured. Body mass index(BMI), Rt- and Lt-BAPWV, glucose, triglyceride, alkaline phosphatase(ALP), gamma-glutamyl transferase(GGT), creatinine, uric acid, troponin-I(TNI), NT-proBNP and high sensitivity C-reactive protein(hs-CRP) levels were higher than the reference value of each variable. Rt- and Lt-BAPWV were directly correlated with age, body weight, BMI, glucose, ketone, aspartate aminotransferase, alanine aminotransferase, ALP, GGT, total cholesterol, low density lipoprotein, lipoprotein(a), apolipoprotein-B, blood urea nitrogen, heart rate, TNI, creatine kinase, CK-MB, lactic dehydrogenase, myoglobin, hs-CRP, lipase, reumatoid factor, fibrinogen and D-dimer (P<0.05, P<0.01, P<0.001 or P<0.000, respectively), but inversely associated with total bilirubin, uric acid, apolipoprotein-A1 and GFR (P<0.05). These observations suggest that a variety of physiological variables may influence BAPWV, resulting in increased risk or prevention of cardiovascular and/or cerebrovascular attacks. Therefore, physiological variables affecting BAPWV should be regularly controlled.

Improvement of Fermentation and Nutritive Quality of Straw-grass Silage by Inclusion of Wet Hulless-barley Distillers' Grains in Tibet

  • In order to develop methods that would enlarge the feed resources in Tibet, mixtures of hulless-barley straw and tall fescue were ensiled with four levels (0, 10%, 20%, and 30% of fresh weight) of wet hulless-barley distillers' grains (WHDG). The silos were opened after 7, 14 or 30 d of ensiling, and the fermentation characteristics and nutritive quality of the silages were analyzed. WHDG addition significantly improved fermentation quality, as indicated by the faster decline of pH, rapid accumulation of lactic acid (LA) (p<0.05), and lower butyric acid content and ammonia-N/total N (p<0.05) as compared with the control. These results indicated that WHDG additions not only effectively inhibited the activity of aerobic bacteria, but also resulted in faster and greatly enhanced LA production and pH value decline, which restricted activity of undesirable bacteria, resulting in more residual water soluble carbohydrates (WSC) in the silages. The protein content of WHDG-containing silages were significantly higher (p<0.05) higher than that of the control. In conclusion, the addition of WHDG increased the fermentation and nutritive quality of straw-grass silage, and this effect was more marked when the inclusion rate of WHDG was greater than 20%.
