Enzymatic Characteristics in the Bioconversion of D,L-ATC to L-Cysteine (D,L-ATC로 부터 L-Cysteine으로의 Bioconversion에 관여하는 효소의 특성)

  • The bioconversion of D, L-20aminothiazoline-4-carboxylic acid (D, L-ATC) to L-cysteine was investigated. After the intracelluar enzyme of a Pseudomonas species was inducibly formed by addition of D, L-ATC in the middle of culture, the cells were isolated and treated with sonication to prepare the crude enzyme solution. The results indicated that the cysteine was produced only in the form of L-isomer from D,L-ATC and its production could be enhanced several tens times by addition of managanese ions which were required as a cofactor in this enzymatic reaction. Bedies, this reaction suffered from the feedback inhibition of L-cysteine. On the other hand, since L-cysteine-decomposing enzyme coexisted in the crude enzyme solution, most of the L-cyseine formed disappeared in the absence of its inhibitor. However, hydroxylamine was found to be a potent inhibitor which could successfully prevent the decomposition of L-cyseine.

  • The reaction steps involved in the bioconversion of a chemically synthesized precursor, $D,L-2-amino-{\Delta}^2-thiazoline-4-carboxylic$ acid (D,L-ATC), to L-cysteine and the properties of the involved enzymes were investigated. It was found that the conversion consisted of two steps, i. e., D,L-ATC to S-carbamyl-L-cysteine (S-C-L-cysteine) and S-C-L-cysteine to L-cysteine, and the S-C-L-cysteine was an intermediate between them. While the enzymes involved in the reactions were induced by the addition of D,L-ATC as an inducer, S-C-L-cysteine induced only the enzyme involved in the latter step. The conversion of S-C-L-cysteine to L-cysteine could be also carried out in the presence of hydroxylamine and its rate was much faster than that by the corresponding enzyme. On the other hand, L-cysteine (or L-cystine) was decomposed to evolve $H_2S$ by the enzyme considered to be a kind of desulfhydrase. However, hydroxylamine was a perfect inhibitor for this enzyme.

Bioconversion of D,L-ATC to L-cysteine Using Whole Cells (D,L-ATC의 L-cysteine으로의 생물학적 전환반응에서의 균체이용 기술)

  • In the conversion of D.L-2-amino-$\Delta^2$-thiazoline-4-carboxylic acid(D,L-ATC) to L-cysteine using Pseudomonas sp. CU6. the effects of surfactants on whole cells and the stabilities of cellfree enzyme solution and continuous reactor packed with immobilized whole cells were investigated. The enzymatic reaction was little accomplished by whole cells without adding surfactants, whereas it was well carried out with SDS or Triton X-loo comparable to the case using cell-free enzyme solution. Enzyme activity of the cell-free solution was lost by 50% after 7 hours of storage at $30^{\circ}C$, but not at all under an anaerobic condition by sparging nitrogen gas. On the other hand. effect of nitrogen gas did not appear in a continuous reactor using immobilized whole cells, and hydroxylamine, an inhibitor of L-cysteine desulfhydrase, lowered the enzyme stability.

A Study on the relative Efficiency of ATC Towers in Domestic Airports (국내공항 관제탑의 상대적 효율성에 관한 연구)

  • Air Traffic Control Tower is one of the most important units in Airport operation. It provides services related to safe and efficient traffic flows that control aircraft on the ground maneuvering area and terminal airspace. Also It is responsible for managing of ground operators. The major objective of this study is to evaluates relative efficiency of ATC towers in Domestic airports using data envelopment analysis so that it helps the ATC authority to improve the tower efficiency, to decide the level of benchmarking target and to establish the best alternative. The results of this study are the following; First, as a result of analysis for the potential improvement, it has analysed that the common problems of each ATC tower are to increase its number of flight and to reduce its number of runway followed by airside area, the number of air traffic controller and the number of stand. Second, it has shown that the each tower in RKPC(8), RKPT(5), RKPK(l) and RKSS(l) are used as the reference set. Especially, the tower in RKPC analyzed as a relatively efficient unit is the most main target for the towers in RKTU, RKTH, RKPS and RKTY to do bench marking and to set up the strategy for improving relative efficiency of the tower. Third, tower is actually not able to control the input and output data in this study except the number of controller, so that ATC authority is recommended to improve inefficiency of the towers through handling the number of controller.

The mitochondrial genome of Tremoctopus violaceus (Octopoda, Tremoctopodidae) and its phylogenetic consideration

  • The complete mitochondrial genome of Tremoctopus violaceus was sequenced to analyze its organization and phylogenetic status within the order Octopoda. The mitochondrial genome of T. violaceus had a structure and organization similar to that of other Octopoda. The content of the nucleotides A, C, G, and T was 31.68 %, 7.71 %, 20.02 %, and 40.58 %, respectively. All protein-coding genes (PCG) began with the ATG codon, excluding ND4 and ATP6, which began with ATC and ATT, respectively, and terminated with TAG, TAA, TA, or T. Codons for isoleucine were the most used codons, whereas those for arginine were used the least. Two extra tRNAs, trnN and trnL, were found in the control region. These tRNAs have a D-armless structure. The control region had excess A + T content (83.16 %) and a stem-loop structure with two elements, which is reported for the first time in Octopoda by our study. Bayesian inference using 13 PCG revealed that Octopus and Octopodidae were polyphyletic, and that Tremoctopodidae diverged relatively earlier within Octopoda. The mitochondrial genome of T. violaceus and its characteristics may help to understand the evolutionary history of Octopoda and establish a marine biodiversity conservation strategy.

  • An elecmc arc cracking reaclor is developed for the productiol~o f ieusuble fuel gas by the thennal destruction of waste oil. The churaclensucs of product gas and ~esiduesf rom arc crachng of wnste lubr~cat~nogil are sludird. Thc product gas is mainly coruposcd of hydrogen 135-4076), acetylene (13-4076), ethylene 13-476) and olher hgdrocnrbons. The contenr of carbon monomde, one or the main product in a conventional low-temperature Lhennal cracking umt, 1s very slnvll in lhis atc cracking expcnmcnt. Total calocctic wlue of product gas shows 11,000-13.000 kcizlkg, which is hiph cnough to use as a ~ L I I I Cga~ s . and the concentralions oC loxic gases arc well below the rcguliltury emission critena The GCIMS analysis of llquld-phase residues shows that the high rnalccular welgllt hydrocilrbons in the waste oil arc cracked into the low malecular weight hydrocarbons snd hydroem,. The dehydrogcnntion is found lo be Lhe main cracking rcacuon due lo the high temperalure ~ ~ ~ d ubcyc edle ctric arc. The average parucle size of soot as the solid-phase residue is 10 3 wm, and the conlents of cabon a ~ hdea vy metals are abovc 60% and under 0.01 ppm, respecttrely.
