섬진강-광양만 하구 기수 재첩 (Corbicular japonica)의 분자 계통유전학적 분석 (Molecular Phylogenetic Analysis of the Brackish Water Clam (Corbicular japonica) from Seomjin River to Gwangyang Bay, South Korea)

  • 재첩은 하구생태계에 서식하는 종으로, 우리나라에는 섬진강 하구에서 가장 많이 분포하는 것으로 알려져 있다. 섬진강 하구에 서식하는 재첩은 담수와 해수생태계의 변화를 잘 반영하는 종이다. 재첩은 외부형태만으로는 종 동정이 어려운 분류군에 속한다. 본 연구에서는 섬진강 서식 재첩의 종 동정을 위해 형태적 관찰뿐만 아니라 미토콘드리아 DNA의 COI 유전자 기반 DNA 바코딩을 통해 분석하였다. 그 결과 재첩 간 다른 두 종을 확인하였으며, COI 염기서열의 다중배열 분석결과 약 98%의 높은 상동성을 보였다. 재첩과 내 다양한 종들과 계통수 분석으로 계통유 전학적 위치를 확인한 결과, 본 연구에서 사용한 재첩은 C. japonica, C. fluminea와 하나의 계통군으로 묶여졌고 진화적 거리는 0.003 이하로 나타났다. 또한 재첩속인 C. leana와 0.089, C. fluminalis와 0.096으로, 재첩과 내 3종과는 약 0.2로 C. japonica에 비해 상대적으로 진화적 거리가 먼 것으로 나타났다. 계통유전학적 분석을 통해 본 연구에서 사용한 재첩은 C. japonica이고, 그중 한 개체가 C. fluminea인 것으로 확인되었다. 이러한 연구 결과는 DNA 바코딩을 이용한 섬진강 하류 지역에 서식하는 재첩의 COI 시컨스를 통해 계통유전학적 정보를 제공하여 형태학적 분석에 어려움이 있는 종에 대한 중요한 자료로 활용될 것이다.

Real-time PCR 분석법을 이용한 옥돔과 옥두어의 종 판별법 개발 (Development and Validation of Real-time PCR to Determine Branchiostegus japonicus and B. albus Species Based on Mitochondrial DNA)

  • 미토콘드리아 게놈에 존재하는 시토크롬C 산화효소 서브유닛 I (cytochrome C oxidase subunit I, COI) 유전자의 DNA 염기서열을 기반으로 하는 종 판별은 수산물 자원의 지속적인 개발과 어류 다양성 보존을 위해 폭넓게 적용되고 있다. 본 연구에서는 한국에서 소비되는 옥돔과 가짜 옥돔으로 둔갑하는 옥두어의 종 판별을 위한 분석법을 개발하였다. 옥돔과 옥두어, 두 종의 종 판별과 검증을 위해 미토콘드리아 게놈의 DNA 염기서열 차이를 이용하여 real-time PCR법에 의해 분석하였다. 미토콘드리아 DNA 서열의 생물정복학적 분석에서 옥돔과 형태학적 옥돔 유사종인 옥두어, 두 종 사이에 COI 유전자 내에서 상당히 유사한 DNA 서열 부분과 일부 서열 변화 부분이 확인되었다. 명확하게 종 판별을 하기 위해 COI 유전자 내에서 일부 변화된 서열에서 종 특이적 프라이머를 디자인하였다. 10 개체의 옥돔과 옥두어에서 게놈 DNA을 추출하여 옥돔과 옥두어의 종 특이적 프라이머를 이용하여 real-time PCR 시스템에 의해 분석되었다. 이러한 real-time PCR 시스템을 이용한 genomic DNA 기반의 분자 기술은 동물 조직의 분류학적 분류를 위한 신뢰할 수 있는 방법을 제공한다. 옥돔판별을 위해, 옥돔 DNA에서 옥돔 종 특이적 프라이머를 이용한 Ct 평균값($21.85{\pm}3.599$)과 옥두어 DNA에서 옥돔 종 특이 프라아머를 이용한 Ct 평균값($33.49{\pm}1.183$) 차이를 나타내었다. 그리고 옥두어판별을 위해, 옥두어 DNA에서 옥두어 종 특이적 프라이머를 이용한 Ct 평균값($22.49{\pm}0.908$)과 옥돔 DNA에서 옥두어 종 특이 프라아머를 이용한 Ct 평균값($33.93{\pm}0.479$)을 통해 옥돔과 옥두어의 각 종 특이 프라이머의 효율성, 특이성 및 교차 반응성 측정은 통계적으로 유의한 차이를 보여 주었다. 제안된 방법은 10개의 상용 샘플로 검증이 되었다. 따라서, threshold cycle (Ct) value와 같은 real-time PCR 결과 분석에 의해 종 판별이 가능하였다.

Sequence Divergence and Phylogenetic Investigation of the Nymphalidae (Lepidoptera: Papilionoidea) Occurring in South Korea

  • As a first step toward understanding the divergence and relationships of the Nymphalidae (Lepidoptera: Papilionoidea) occurring in South Korea, cytochrome oxidase subunit I (COI), 16S ribosomal RNA (16S rRNA), and elongation factor-$1{\alpha}$ (EF-$1{\alpha}$) that comprise 3,501-3,716 bp were either sequenced (55 species) or the sequences were obtained from GenBank (23 species). The concatenated sequence divergence of six nymphalid subfamilies ranked in the following order: Danainae (10.3%), Satyrinae (9.5%), Limenitidinae (8.0%), Apaturinae (7.0%), Nymphalinae (6.7%), and Heliconiinae (6.2%). As has been reported in previous large scale international studies, the subfamilial relationships of (((((Limenitidinae + Heliconiinae) + (Nymphalinae + Apaturinae)) + Satyrinae) + Libytheinae) + Danainae) were also confirmed, except for the switched positions between Danainae and Libytheinae, and supported all subfamilies and tribe monophylies. Unlikely consistent phylogenetic relationships among genera within the majority of tribes in Nymphalidae, a conflicting relationship within the subfamily Apaturinae was obvious, presenting Apatura as sister to either Mimathyma or (Mimathyma + (Sephisa + (Hestina + Sasakia))), and both of these relationships are unconventional. Within the subfamily Limenitidinae, the genus Neptis was consistently revealed as a paraphyletic with respect to the genus Aldania, requiring further taxonomic investigation of the genus. Although limited, current sequence information and phylogenetic relationships are expected to be helpful for further studies.

A New Species of Eudactylopus (Copepoda: Harpacticoida) from the South Coast of Korea Based on Morphological and Molecular Evidence

  • A new species of Eudactylopus Scott A., 1909 is described from the southern coast of Korea. The specimens were collected using a light trap set overnight at the entrance near a pier. Eudactylopus yokjidoensis n. sp. is similar to E. andrewi Sewell, 1940 and E. spectabilis (Brian, 1923) in two key respects: similar length of proximal and distal inner setae on female P2 enp-2, and modification of two subapical setae on male P2 endopod. However, E. yokjidoensis can be differentiated from the two species by following morphological characteristics: in females, the length ratio of cephalothorax/2nd-4th thoracic somites combined is smaller in E. yokjidoensis than other two species (1 : 0.8 vs. 1 : 1); antennule has nine segments (vs. 7-segmented in E. andrewi); P2 to P4 each bears a process in medial distal margin of basis, while it is just smooth in E. spectabilis; in males; the length ratio of cephalothorax to 2nd-4th thoracic somites combined is smaller in E. yokjidoensis than other two species (1 : 0.6 vs. 1 : 1 in E. andrewi and 1 : 0.8 in E. spectabilis); and P5 exopod has a comb-like innermost seta, while it is bipinnate seta in E. spectabilis. To prove the Korean species of Eudactylopus to be new, full descriptions of both sexes are given here, and the claim is supported by distinct genetic differences between E. yokjidoensis and E. spectabilis (22.3-22.7%) in the mitochondrial gene cytochrome oxidase subunit I(mtCOI) sequence.

가다랑어 (Katsuwonus pelamis) 자어의 국내 출현 (Occurrence of skipjack tuna (Katsuwonus pelamis) larvae in Korean waters)

  • Three specimens of larvae (5.2~7.8 mm in standard length (SL)), of the family Scombridae, were collected from the southeastern waters off Jeju Island in August, 2014. These specimens were identified to Katsuwonus pelamis have melanophores distributed on the 1st dorsal-fin spines and post ventral margin on caudal peduncle. An analysis of the cytochrome oxidase submit I (COI) sequences (652 base pairs) of mitochondrial DNA showed that our specimens must be K. pelamis, because their sequences were concordant with those of the adult K. pelamis (genetic distance= 0.000~0.002), followed by Auxis rochei (genetic distance= 0.061~0.063) and Euthynnus affinis (genetic distance= 0.077~0.079). During the larval stage, K. pelamis differed from Thunnus spp. species in melanophores distribution period of 1st dorsal-fin spines, lower jaws and ventral margin on caudal peduncle. In conclusion, occurrence of K. pelamis larvae means Korean waters are potential spawning ground of K. pelamis.

Revealing hidden diversity in the Sheathia arcuata morphospecies (Batrachospermales, Rhodophyta) including four new species

  • The freshwater red algal genus Sheathia contains species with heterocortication (both bulbous and cylindrical cells covering the main axis) and homocortication (only cylindrical cells). When the genus was proposed, the species with heterocortication were revised, but all specimens with homocortication were assigned to Sheathia arcuata with the caveat that it may represent a species complex. Recent studies have described new species with homocortication and S. arcuata has been rendered paraphyletic. In the current study, new sequences of the rbcL and 5′ region of the cytochrome c oxidase subunit I markers were combined with previously published data to construct a robust phylogeny and circumscribe new species. Four new species, S. abscondita, S. californica, S. plantuloides, and S. transpacifica are proposed. Examination of morphological characters among homocorticate species show no diagnostic characters to distinguish among species, whereas S. plantuloides is only known from sporophytes (chantransia) so it lacks the typical morphological characters derived from the gametophytes for comparison. Although DNA sequence data would be needed to make a positive species identification, geography could be employed to narrow the identification to one or two species. The genus is geographically widespread having been recorded from oceanic islands and five continents, whereas the individual species typically occur on a single continent. With this study, the number of species recognized in Sheathia is raised to 17; seven heterocorticate and 10 homocorticate, making this genus one of the most species rich in the Batrachospermales. As well, the resulting phylogeny provides insights into the evolution of heterocortication in Sheathia.

A Phylogenetic Study in Some Long-Horned Beetles (Coleoptera: Cerambycidae) Using Mitochondrial COI Gene and 16S rRNA Sequences

  • Two regions of mtDNA genome, cytochrome oxidase subunit I (COI) and 165 ribosomal RNA (165 rRNA) genes, were sequenced for 15 species of the long-horned beetle belonging to four subfamilies and geographic samples of mulberry longicorn beetle, Apriona germari, from two localities in Korea. Ten samples of A. germari collected from Suwon and Busan revealed three COI haplotypes ranging in nucleotide divergence of 0.3% to 0.5%, and the two populations shared one common COI haplotype (80%). The sequence divergence among 15 species of the long-horned beetle was much higher in COI gene (12.3%∼39.4%) than 16S rRNA gene (7.2% to 23.1), and the maximum value in the COI gene is exceptional compared with other relevant studies, including that of Coleoptera. The greatly increased divergence in the COI gene, in facto was stemmed from a peculiar sequence of Prionus insularis belonging to Prioninne, divergence of which ranges from 31.2% to 39.3% from other species. We discussed possible reason of the divergence in this species. Due to the abnormality of COI gene divergence, decrease in phylogenetic signal was severe in COI nucleotide and, subsequently, the converted amino acid sequences, rendering us to put more confidence on the 16S5 rRNA gene data. Although the molecular phylogeny confidently supports the monophyletic origin of Lepturinae, the presence of discrepancy between molecular data and traditional taxonomic views also is a testable hyothesis. One such discrepancy includes taxonomic position of Sophronica obrioides and Theophilea cylindricollis belonging to Lamiinae.

A Case of Diphyllobothrium nihonkaiense Infection as Confirmed by Mitochondrial COX1 Gene Sequence Analysis

  • Diphyllobothrium nihonkaiense has been reported in Korea as Diphyllobothrium latum because of their close morphologic resemblance. We have identified a human case of D. nihonkaiense infection using the mitochondrial cytochrome c oxidase subunit I (cox1) gene sequence analysis. On 18 February 2012, a patient who had consumed raw fish a month earlier visited our outpatient clinic with a long tapeworm parasite excreted in the feces. The body of the segmented worm was 2 m long and divided into the scolex (head) and proglottids. It was morphologically close to D. nihonkaiense and D. latum. The cox1 gene analysis showed 99.4% (340/342 bp) homology with D. nihonkaiense but only 91.8% (314/342 bp) homology with D. latum. The present study suggested that the Diphyllobothrium spp. infection in Korea should be analyzed with specific DNA sequence for an accurate species identification.

CCl4전처치한 흰쥐에 Cyclohexane 투여가 간손상에 미치는 영향 (Effect of Cyclohexane Treatment on the Liver Damage in CCl4-Pretreated Rats)

  • TO evaluate an effect of cyclohexane treatment on the degree of liver damage, rats were induced liver damage with 10 or 17 times $CCl_4$ injection (0.1 m1/100 g body wt., 50% $CCl_4$ dis-solved in olive oil) at intervals of every other day. Cyclohexane (1.56 g/kg body wt., i.p.) was administrated to the animals at 48 hours after the last pretreatment of $CCl_4$ . Rats were sacrificed at 4 hours after injection of cyclohexane. On the basis of histopathological findings, liver weight/body weight (LW/ BW, %), activities of serum alanine aminotransferase (ALT), xanthine oxidase (XO) and akaline phosphatase (ALP), and contents of liver protein and manlondialdehyde (MDA), $CCl_4$ -pretreatment induced liver damage. And $CCl_4$ 17 times treated group showed more severe liver damage than $CCl_4$ 10 times treated group. Administration of one dose of cyclohexane to $CCl_4$ 10 times treated animals resulted in the enhanced liver damage; liver necrosis with proliferation of fibroblast and bile duct abnormality, and increase in hepatic MDA content and the activities of serum ALP and ALT, But the enhanced liver damage was not found in $CCl_4$ 17 times treated animals. Serum cyclohexanone concentrations at 4 or 8 hours after injection of cyclohexane were higher in all liver damaged groups than normal group and were somewhat higher In $CCl_4$ 17 times treated animals than $CCl_4$ 10 times treated ones. Among the oxygen free radical metabolizing enzymes, hepatic cytochrome P45O dependent aniline hydroxylase (CYPdAH) activity in cyclohexane metabolizing enzyme system was meaningfully increased by the injection of cyclohexane to the liver damaged rats, with increased Vmax and high affinity to aniline. LW/BW (%) and activities of serum XO and ALT were more significantly increased in liver damaged groups than normal group by administration of cyclohexanone. In conclusion, it is assumed that an enhancement of liver damage by injection of one dose of cyclohexane to liver damaged animals might be caused by oxygen free radicals and cyclohexanone.

Prevalence of Dirofilaria immitis Infection in Stray Cats by Nested PCR in Korea

  • The purpose of this study was to conduct a survey of Dirofilaria immitis infection among stray cats in Korea using nested PCR. We included 235 stray cats (121 females and 114 males) and evaluated each for the presence of feline heartworm infection. Blood samples were collected from 135 cats in Daejeon, 50 cats in Seoul, and 50 cats from Gyeonggi-do (Province). Of the 235 DNA samples, 14 (6.0%) were positive for D. immitis. The prevalence of infection in male cats (8/114, 7.0%) tended to be higher than that in female cats (6/121, 5.0%), but the difference was not statistically significant. In each location, 8, 2, and 4 cats were positive for infection, respectively, based on DNA testing. No significant differences in the prevalence were observed among the geographic regions, although the rate of infection was higher in Gyeonggi-do (8.0%) than Daejeon (5.9%) and Seoul (4.0%). We submitted 7 of the 14 D. immitis DNA-positive samples for sequencing analysis. All samples corresponded to partial D. immitis cytochrome c oxidase subunit I gene sequences with 99% homology to the D. immitis sequence deposited in GenBank (accession no. FN391553). To the best of our knowledge, this is the first survey using nested PCR to analyze the prevalence of D. immitis in stray cats in Korea.
