S$_H$2 Reaction on Silicon-Carbon Bond in the Photoreactions of 2, 3-Benzo-1, 1-diphenyl(or dimethyl)-1-sila-2-cyclobutene with Carbonyl Compounds

  • The photoreaction of 2, 3-benzo-1, 1-diphenyl (or dimethyl)-1-sila-2-cyclobutene (9 or 10) with an aldehyde or ketone results in 1:1 cycloadduct of [4 + 2] type. In the reactions of 2, 3-benzo-1, 1-dimethyl-1-sila-2-cyclobutene (10) with acetone and butanone, another 1:1 adducts (13) were also formed, respectively. The following facts indicate that the formation of adduct involves an attack of a triplet carbonyl compound on the silicon of the benzosilacyclobutene, an $S_H2$ process. (1) Even when the reaction of 9 with acetophenone was carried out under conditions such that more than 99% of incident light was absorbed only by acetophenone using the filter solution of aq. cupric sulfate, the same adduct was still formed. (2) When the reaction of 9 with acetone was carried out under oxygen atmosphere, only trace amount of adduct was formed.

충북지역 토마토 시설재배지의 풋마름병균(Ralstonia solanacearum) 오염도 및 분리균주의 특성 (Contamination Level of Ralstonia solanacearum in Soil of Greenhouses Cultivating tomato Plants in Chungbuk Province and Characteristics of the Isolates)

  • 충북지역 토마토 시설재배지 토양의 풋마름병 병원균(Ralstonia solanaceaum)의 오염도 및 분리균의 특성을 조사하였다. 총 133개 조사 토양 중 27개 토양에서 선택배지로부터 R. solanacearum이 분리되어, 전체 시설재배지 토양의 20.3%가 풋마름병균에 오염되어 있음을 나타냈다. 오염 토양에서 병원균의 밀도는 토양 g당 $10^{2.4}$ cfu로 조사되었다. 병든 토마토로부터 분리한 12균주를 포함한 총 71개의 분리균주는 모두 레이스 1이었고, 35개 균주가 생리형 3으로, 36개 균주가 생리형 4로 구분되었다. 전체 분리균의 88% 이상이 각각 oxolinic acid 0.5$\mu\textrm{g}$/ml, streptomycin 25 $\mu\textrm{g}$/ml, tetracycline 5 $\mu\textrm{g}$/ml and cupric sulfate 375 $\mu\textrm{g}$ml(1.5 mM)을 함유한 nutrient agar에서 생장이 억제되었고, 전체균주의 11.3%, 4.2% and 5.6%가 각각 감수성균주의 최소억제농도보다 10배 이상의 oxolinic acid, streptomycin, tetracycline가 함유된 배지에서 생장하였다. 5개 균주는 2개의 살세균제에 동시에 그리고 1개 균주는 3개의 살세균제 모두에 저항성을 보였다.

Herpetosiphon geysericola 균주의 Amylase 생성 (Production of Amylases from Herpetosiphon geysericola)

  • 퇴비 숙성초기에 고온성 cellulose 분해이용균으로 분리된 Herpetosiphon geysericola CUM 317균주는 전분분해 효소인 ${\alpha}-amylase,\;{\beta}-amylase$ 및 glucoamylase를 모두 생성한다. 이 균을 사용하여 그 배양조건을 달리하여 각 amylase의 생성관계를 서로 비교한바 50℃의 밀기울 고체배지나 $40^{\circ}C$의 액체배지상에서 ${\beta}-amylase$는 배양초기 10시간만에 최대의 생성력가를 보였는 반면, ${\alpha}-amylase$와 glucoamylase는 30 내지 40시간 정도의 배양말기에 최대를 이루었다. Polypeptone을 함유한 액체배지에 탄소원의 첨가나 무기질소원의 첨가는 전반적으로 amylase들의 생성이 크게 저하되었으나 cellulose에 의해서 glucoamylase의 경우 150% 정도 증가되었다. 액체배지에 $CuSO_4$를 첨가해 줌으로서 ${\alpha}-amylase$만의 생성증가 효과를 얻었고 $CdSO_4$에 의하여 ${\beta}-amylase$만의 생성증가가 있었으며, 그리고 $CaCl_2$에 의하여 glucoamylase만의 증가효과가 있은 반면, 상대적으로 ${\beta}-amylase$의 급격한 감소가 일어났다. 이들 amylase들의 최적 효소생성 pH는 7.5였으며, 최적온도는 ${\alpha}-amylase$와 glucoamylase의 경우 $40^{\circ}C$였고 ${\beta}-amylase$$30^{\circ}C$였다.

Effect of High Dietary Copper on the Morphology of Gastro-Intestinal Tract in Broiler Chickens

  • An experiment was conducted to study the effects of high dietary copper supplementation on the gastrointestinal tract morphology of broiler chickens. Eighty 3-week-old broiler chicks were divided randomly into eight groups of four dietary treatments and over three week were fed isoenergetic and isonitrogenous diets that contained 0, 100, 250, or 500 mg/kg of supplemental copper from cupric sulfate. The copper supplementation in the broiler diet up to 250 mg/kg did not significantly influence broilers' performance. A high dietary copper supplementation of 500 mg/kg did significantly depress growth and feed conversion in the broilers (p<0.05). Copper supplementation more than 250 mg/kg in the broiler diet significantly influenced the morphology of the GI tract, as shown by severe oral lesions and gizzard erosion. It also significantly depressed the villi height and significantly thickened the muscular layer in the duodenum (p<0.05). The severely damaged villi were observed by scanning electronic microscope from the duodenum samples of broilers fed a 500 mg/kg copper supplemented diet. The 500 mg/kg copper supplemented diet also significantly influenced the plasma constituents. Plasma glucose concentration was significantly depressed (p<0.05).

유기물 첨가제와 펄스-역펄스 전착법을 이용한 구리 Via Filling에 관한 연구 (Copper Via Filling Using Organic Additives and Wave Current Electroplating)

  • 반도체 소자의 집적도가 높아짐에 따라 3D SiP에 대한 관심이 높아지고 전기도금법을 이용한 구리 via filling이 활발히 연구되어왔다. Via filling시 via 입구와 바닥에 전류밀도 차이로 인해 via 내부에 결함이 발생하기 쉽다. 여러 가지 유기물 첨가제와 전류인가 방식의 변화를 통한 via filling을 하였다. 첨가된 유기물은 PEG, SPS, JGB, PEI를 사용하였다. 유기물이 첨가된 용액을 이용하여 펄스와 역펄스 방법을 이용하여 via filling을 하였다. 유기물의 첨가에 따른 도금된 구리 입자의 크기 및 형상에 관하여 고찰하였으며 도금 후 via 시편의 단면을 FESEM으로 관찰하였다. JGB에 비하여 PEI를 사용한 경우 치밀한 도금층을 얻을 수 있었다. 2 step via filling을 사용한 경우 via filling 시간을 단축시킬 수 있었다.

머위잎 추출액의 염색성 (A study on the chemical and dyeing properties of Petasites japonicus leaf extract)

  • This study was carried out to investigate the chemical and dyeing properties of Petasites japonicus leaf extract under the various extracting and dyeing conditions such as temperature, time, the ratio of water and pH, repetition of dyeing in silk and cotton fabrics with Petasites japonicus leaf. The results were as follows: 1. It was found that λ$_{\max}$ of color solution extracted by Petasites japonicus leaf has two peaks at 290nm and 323nm. 2. The optimum extracting conditions were studied at 10$0^{\circ}C$, 40min., pH 7 and 1 : 20(the ratio of water and Petasites japonicus leaf), the optimum dyeing temperature, dyeing time, dyeing pH and repetition of dyeing were 10$0^{\circ}C$, 60min., pH 7, repetitions of three times, respectively. 3. Silk and cotton fabrics dyed with Petasites japonicus leaf extract were colored yellowish orange. The colory Petasites japonicus leaf extract in silk and cotton fabrics were deeped by same-mordanting with aluminum potassium sufate and cupric sulfate. 4. Washing fastness of silk fabrics was 4∼5 grade, but cotton fabrics was 3∼4 grade, so washing fastness of silk fabrics washed with neutral detergent was excellent.

Effects of Dietary Garlic Powder and Copper on Cholesterol Content and Quality Characteristics of Chicken Eggs

  • This experiment was conducted to investigate the dietary effect of garlic powder (GP) and copper (Cu) on laying performances and the cholesterol content and quality characteristics of eggs during storage in laying hens. A total of one hundred and eighty, 50-wk-old, Hy-Line Brown layers were divided into 6 groups with 3 replicates per group (10 layers per replicate) and fed one of six diets containing GP 0%, GP 1%, GP 3%, GP 5%, Cu 200 ppm, or GP 3%-Cu 200 ppm for 5 wks. There were no differences in the laying performances and feed intakes bertween treatments. Eggshell strength, eggshell thickness and yolk color were also not affected by feeding of GP and Cu. With increasing dietary GP, Haugh unit was linearly increased after 2 wk of storage (p<0.05). The levels of serum total cholesterol in hens fed diets containing GP or Cu were lower than that of the control (p<0.05), but high density lipoprotein-cholesterol was not influenced by dietary GP or Cu. The content of egg yolk cholesterol from hens fed diets containing GP or Cu was significantly decreased from that of the control, except for the GP 1% group. Based on the results of this experiment, the decrease of Haugh unit during storage was alleviated by feeding of GP. The feeding of GP or Cu alone and in combination altered the cholesterol fractions in serum and reduced the content of egg yolk cholesterol in laying hens.

다공성 그물구조 음극을 이용한 구리 전착에 관한 연구 (II) -유기첨가제 PEG, MPS의 영향 - (Electrodeposition of Copper on Porous Reticular Cathode (II) - Effect of PEG and MPS on throwing Power-)

  • 황산과 황산구리 수용액을 전해질로 사용하여 다공성 그물구조 금속을 전기화학적으로 제조하였으며, 이때 균질 전착에 영향을 미치는 첨가제 PEG, MPS의 영향에 대해 살펴보았다. 분극실험과 기공의 평균 직경이 $250{\mu}m$의 그물구조 전극을 여러장 적층하여 throwing power를 정량화 한 결과, PEG의 경우 분극 초기에 전극에서 탈착됨으로써 오히려 throwing power를 감소시키고 MPS는 500ppm까지 용액 중 첨가량의 증가와 함께 throwing power를 향상시키는 결과를 보였다 또한 MPS와 PEG가 동시에 혼합된 용액에서는 MPS의 효과가 우선적으로 나타나며 PEG의 효과는 무시할 정도였다 그러나 MPS의 첨가는 전착층의 균열을 유도하여 기계적 강도에 좋지 못한 영향을 미치며 균열을 피하려면 50ppm 이하가 적정함을 SEM 관찰을 통해 확인할 수 있었다.

비둘기 및 수생조류(水生鳥類) 유래(由來) Salmonella typhimurium의 생물화학적(生物化學的) 특성(特性)과 plasmid profile에 관(關)한 연구(硏究) (Studies on the biochemical characteristics and plasmid profiles of Salmonella typhimurium isolated from pigeons and aquatic birds)

  • A total of 166 strains of Salmonella (S) typhimurium var copenhagen isolated from pigeons (164 strains) and aquatic birds (2 strains) were examined for the biochemical characteristics and plasmid profiles. All the strains were sensitive to ampicillin, chloramphenicol, gentamicin, kanamycin and sulfadimethoxine. But 13 strains(7.8%) were resistant to streptomycin (Sm), 2 (1.2%) to tetracycline, 2 (1.2%) to rifampicin, and 1 (0.6%) to nalidixic acid. Among drug resistant strains, only one strain resistant to Sm contained conjugative R plasmid which was fertility inhibition and incompatibility group $I_{\alpha}$. All the strains were sensitive to cobalt chloride, cupric sulfate, lead nitrate, mercuric chloride and silver nitrate. Of 166 isolates, 6 (3.6%) were resistant to sodium arsenate and 1 (0.6%) to potassium tellurite. Among 166 isolates, 1 (0.6%) was colicinogenic, 12 (7.2%) sucrose fermenters, and 166 (100%) maltose fermenters. Plasmid profiles were confirmed as being 4 or 5 plasmids, and their molecular weight ranged 3.2 to 60 megadalton (MD). All the strains harbored 60 Md plasmid. There are three patterns by the plasmid profile, 150 isolates (90.4%) were pattern I (3.2, 3.5, 33, 60Md), 14 (8.4%) pattern II (3.2, 3.5, 29, 60Md), and 2 (1.2%) pattern III (4.2, 7.8, 8.5, 15, 60Md). S typhimurium var copenhagen strains containing 60Md plasmid were resistant to killing by 90% normal guinea pig serum.

Streptomyces 속 중의 Acetanilide p-Hydroxylase에 관한 연구 (Studies on Acetanilide p-Hydroxylase in Streptomyces spp.)

  • For microbial production of acetaminophen, a popular analgesic-antipyretic from aniline, we screened various fungi and bacteria. And we succeeded to some extents in acetaminophen production by successful protoplast fusion between S. lividans and S. globisporus and also between S. rimosus and S. aureofaciens. However, more fertile results might be brought via performing the cloning of acetanilide p-hydroxylation genes of Streptomyces in yeast. This study was initiated to determine whether the acetanilide p-hydroxylase of Streptomyces is cytochrome P-450 species or non-heme iron protein species. The p-hydroxylationactivity on acetanilide in S. aureofaciens ATCC 10762 was found to be unstable on exposing to the air. However, 100,000xg supernatant of the cell free extracts which were prepared in $N_2$ atmosphere showed the p-hydroxylation activity. Characteristic absorption peak of cytochrome P-450 after reduction with dithionite and addition of CO was not observed in the region of 450nm. Moreover, metyrapone and 2, 6-dichloroindophenol did not affect this enzyme activity, but sodium azide, sodium cyanide, cupric sulfate, cadmium chloride, ${\alpha}$, ${\alpha}'-dipyridyl$, and o-phenanthroline reduced p-hydroxylase activity considerably. S. fradiae NRRL 2702 was shown to have strong p-hydroxylation activity in intact cells. This activity disappeared in its cell free extracts. In its 100,000xg supernatant, however, characteristic absorption peak of cytochrome P-450 after reduction with dithionite and addition of CO was observed at 446nm. Thus, the results herein presented suggest that acetanilide p-hydroxylase of Streptomyces aureofaciens is not related to cytochrome P-450 and may include non-heme iron protein for its activity. However, it is not clear whether acetanilide p-hydroxylase in S. fradiae belongs to the same category of S. aureofaciens.
