네리네 대량증식을 위한 자구 저장 및 순화재배 (Condition of Storage and Growth Characteristics on Bulb Propagation of Nerine by Tissue Culture)

  • Nerine 대량증식을 위한 자구 저장 및 순화재배 조건 등에 관한 연구를 수행하였다. 저장온도와 기간별 자구 무게 참모율은 저온보다 고온에서 증가하였다. 따라서 조직배양 자구의 적정 저장온도와 기간은 86.4%의 출현율을 보이는 $15^{\circ}C$에서 12주가 적절하였다. 또한 기내 생산 자구의 적정토양순화 시기는 휴면타파온도인 $22^{\circ}C$이상 유지가 가능한 6월 상순이 적절하였다. 순화 재배시 효율성 향상을 위한 적정 광원은 적색과 청색의 혼합파장에서 네리네 생육과 광합성 능력이 우수하였다.

재조합 Saccharomyces cerevisiae에서 Inulinase와 Invertase의 발현과 분비에 미치는 배양조건의 영향

  • The effects of medium pH and culture temperature on the expression and secretion of inulinase and invertase were investigated with recombinant Saccharomyces cerevisiae cells. These cells were obtained by transformation of 2$\mu$-based plasmids pYI10 and pYS10 which contain Kluyveromyces marxianus inulinase gene (INU1A) and S. cerevisiae invertase gene (SUC2), respectively, in the downstream of GAL1 promoter. The expression level and localization of inulinase and invertase were not affected significantly by the initial medium pH: secretion efficiencies of inulinase and invertase into the medium were about 90% and 60%, respectively, in the pH ranges of 4.0 to 6.5. However, the expression and secretion of both enzymes were strongly dependent on the culture temperature. The highest expression (7.7 units/mL) and secretion (6.7 units/mL) of inulinase were observed at 28$\circ$C and 30$\circ$C. As a consequence of decreased localization of inulinase in the periplasmic space, the secretion efficiency increased from 68% at 20$\circ$C, to 95% at 35$\circ$C,. The total expression level and secretion efficiency of invertase increased from 19 units/mL and 55% at 20$\circ$C to 25 units/mL and 68% at 35$\circ$C, respectively. Irrespective of the culture temperature, the invertase activity in the cellular fraction (periplasmic space and cytoplasmic fractions) was kept constant at around 33-45%.

Preparation of Novel Quaternary Plastomers with Extremely Low Glass Transition Temperature

  • In this study, novel quaternary plastomers consisting of ethylene, 1-hexene, high ${\alpha}$-olefin, and divinylbenzene were prepared using Zr metallocene catalyst, borate type cocatalyst, and tri-isobutylaluminium. The molar ratio changes of 1-hexene and high ${\alpha}$-olefin (1-octene, 1-decene, and 1-dodecene) had an effect on the properties of the quaternary plastomers. The structure of the quaternary plastomers was characterized using $^1H$ NMR. Molecular weight properties, crystallinity, and thermal properties of the plastomers were determined by GPC, WAXS, and DMA, respectively. Compared with the terpolymers prepared in our previous study, molecular weight and molecular weight distribution of the quaternary polymers were very similar, whereas glass transition temperature ($T_g$) was very low. Also, hardness and tensile properties of the quaternary plastomers were measured.

Electric Stimulation(음이온 pad)이 생쥐난자의 성숙 및 수정난의 난할에 미치는 영향 (The Effect of Electric Stimulation(anion pad) on the Maturation of Follicular Oocytes and the Cleavage of Fertilized Embryos of the Mouse)

  • In the present study, mouse follicular oocytes and 2-cell embryos(late -zygote stage embryos included) were cultured on the electric pad for electric stimulation in the culture incubator. In addition, follicular oocytes and embryos were tested for maturation and development under higher temperature condition($39^{\circ}C$).Mouse follicular oocyte maturation were not affected by anion electric stimulation and there is no significant difference in GBVD and MI between the control and experiment group after 4hr culture. In the embryo culture, it was found that more morula and blastocyst were found in the electric stimulation group rather than the control(96hr). This may seem to be caused with cytoplasmic $Ca^{2+}$ transient rise by electric stimulation(anion pad). On the other hand higher temperature incubation ($39^{\circ}C$) on the anion pad caused all the embryos degenerated within $12h{\sim}24hr$ culture. This was quite different from large animal embryos(bovine, pig, sheep), in which beneficial effect of high temperature incubation for oocyte maturation and embryo development were found.

  • Helicobacter pylori is a major cause of gastric-associated diseases. In our previous study, the Adhesin/CTXA2B was expressed as insoluble recombinant chimeric protein derived from the H. pylori adhesin genetically coupled to CTXA2B subunit in Escherichia coli. Since it is very important to optimize IPTG concentration, culture temperature and composition of medium to maximize cell growth and productivity, these conditional growth factors were determined for increasing the productivity of the expressed Adhesin/CTXA2B chimeric protein in Escherichia coli JM101 carrying pTEDhpa/ctxa2b. Our data demonstrate that optimal medium for increased production of chimeric protein was a YCP/Glu medium composed of 2% yeast extract, 1% casamino acid, phosphate solution [0.3% $KH_2P0_4$, 0.4% $Na_2HP0_4$, 0.25% ($NH_4)_2HPO_4$], and 0.5% glucose. In addition, optimal concentration of IPTG was 1 mM and culture temperature, $37^{\circ}C$.

왕우럭조개의 모패관리와 성숙촉진을 위한 가온 효과 구명 (Study on the Management of Broodstock and Effect of Raise the Temperature for Promotion of Maturity in Sulf Clam, Tresus keenae)

  • 패류 인공종자생산에서 모패관리의 중요성이 커져가고 있으며 최적의 모패 관리 조건과 원하는 시기에 종자생산을 할 수 있는 성숙유도 조건을 구명하는 하는 것이 매우 중요하기 때문에 본 연구에서는 왕우럭 조개의 모패관리 최적조건과 성숙 촉진을 위한 가온 효과를 조사하였다. 모패관리는 실내사육에 비하여 실외사육이 비만도와 생존율 측면에서 더 효과적인 것으로 나타났으며 관리 측면에서도 인위적인 먹이생물 공급이 없으며 자연환경 조건에서 사육하기 때문에 관리가 수월하다는 장점이 있는 것으로 나타났다. 그리고 동계가온을 통한 성숙촉진 유도는 비만도와 생식소 관찰 등을 통하여 수온 18℃ 이상에서 2개월 정도 사육하였을 경우 성성숙이 이루어지는 것으로 나타나 성숙관리가 다른 패류에 비해 용이하다는 것을 알 수 있었다.

손의 한랭자극이 인체생리반응과 의복의 쾌적성에 미치는 영향 -한랭혈관반응, 온랭감각, 한랭통증을 중심으로- (Effect of Cooling Hands in the Cold Water for the Physiological Responses and Clothing Comfort -Focused on Vascular Hunting Reaction, Thermal Sensation and Pain Sensation-)

  • This study was conducted to compare the hunting reaction of finger in the cold water. Finger skin temperature is measured the left middle finger tip immersion in cold water of 5℃ for 30 minutes and measurements were made on finger skin temperature(Ts), thermal comfort, and cold pain sensations during the experiment at the spring (March) and Winter(December). Results were follows. Is before immersion was at the highest in spring and at the lowest in winter and was closely related to the indoor temperature Ts during immersion and recovery. Mean of finger skin temperature(MST), the skin temperature at the first rise(TTR) and amplitude of finger skin temperature reaction during immersion(AT) were significant higher in spring than that in winter(P<.01). The lowest skin temperature(LST) during the cold water immersion were significantly higher in spring than that in winter (P<.05). The frequency of the appearance of cold-Induced vase dilation(CIVD) was higher in spring than that in winter. However, time for the first temperature(TTR) and recovery time(RT) had no seasonal variation. In addition, cold pains during immersion were felt more strongly in spring than in winter. Local thermal sensation, finger thermal sensation in dynamic state during hand immersion was different from that in the Winter. Spring was slowly cold in cold water immersion.

The Neural-Fuzzy Control of a Transformer Cooling System

  • In transformer cooling systems, oil temperature is controlled through the use of a blower and oil pump. For this paper, set-point algorithms, a reset algorithm and control algorithms of the cooling system were developed by neural networks and fuzzy logics. The oil inlet temperature was set by a $2{\times}2{\times}1$ neural network, and the oil temperature difference was set by a $2{\times}3{\times}1$ neural network. Inputs used for these neural networks were the transformer operating ratio and the air inlet temperature. The inlet set temperature was reset by a fuzzy logic based on the transformer operating ratio and the oil outlet temperature. A blower was used to control the inlet oil temperature while the oil pump was used to control the oil temperature difference by fuzzy logics. In order to analysis the performance of these algorithms, the initial start-up test and the step change test were performed by using the dynamic model of a transformer cooling system. Test results showed that algorithms developed for this study were effective in controlling the oil temperature of a transformer cooling system.

슈의 팽화에 대한 저온 조건의 영향 (Effect of Low-Temperature Conditions on Expansion of Choux)

  • This study examined the effects of low temperature conditions on the expansion of choux when mixing the dough with egg fluid. The egg fluid was tested at 5 and $17^{\circ}C$, and the dough temperature was 20, 30, 40, 50, 60 or $70^{\circ}C$. The expansion decreased with decreasing temperature of the egg fluid and dough, with the concomitant formation of membranes in the cavities of choux because of the poor emulsion stability of the choux paste. In addition, the structure of the choux pastry was not dense and the cracks in the surface were partial and narrow. The shape, expansion and cracks of choux were the best at a dough temperature of 60 and $70^{\circ}C$ added with egg fluid at $17^{\circ}C$. The temperature of the choux paste in these two samples was higher than the melting point of the butter used to produce the paste. Therefore, the practical limit temperature when mixing the egg fluid and dough is 20 and $60^{\circ}C$, respectively, and care should ve taken to maintain a low temperature when making choux in confectionary.

보리 생육온도조건과 등숙기간에 따른 미숙종자의 특성 및 세대촉진을 위한 배배양시 유묘 생육의 변이 (Variation of Immature Kernel as Affected by Growth Temperature and Grain -filling Period, and of Seedlings Obtained from Barley Embryo Culture for Shortening Generation)

  • 본 시험은 미숙종자의 배배양을 이용한 보리의 세대촉진을 위하여, 재배조건(15/10, 25/15$^{\circ}C$)과 등숙기간에 따른 미숙종자의 형질을 조사하고 이들 형질들간의 상호 관계를 구명함과 동시에, 미숙종자의 배배양시 유묘의 생육 반응을 검토하였던 바, 그 결과를 요약하면 다음과 같다. 1. 출수 후 21일이 경과한 미숙종자의 배장은 이삭의 하부보다는 중상부가, 저온(15/10$^{\circ}C$)보다는 고온(25/15$^{\circ}C$) 조건에서 길었는데, 특히 저온 조건의 하부에서 짧았다. 그러나 부위별 차이보다는 생육 온도 조건에 따른 차이가 컸다. 2. 배배양 후 유아장과 근수 및 근장은 고온의 경우 이삭 부위별 차이가 적었으나, 저온조건에서는 중, 상부보다 하부의 생육이 현저히 불량하였다. 3. 등숙 기간에 따른 배장의 변리는 고온 조건에서 21일경에 최장에 이르러 더이상 신장하지 않았지만 저온 조건에서는 29일까지도 계속적으로 신장하여 저온 조건의 29일배가 고온 조건의 17일배와 유사한 경향치를 보였다. 4. 고온 조건에서 17일이 경과한 배의 배장은 입폭, 유아장, 근수 및 근장과 정의 상관이 인정되었으나, 21일이 경과한 것은 어느 형질과도 상관이 인정되지 않았다. 5. 미숙종자의 배배양을 이용한 세대촉진시에는 25/15$^{\circ}C$의 고온 조건에서 출수후 17일이 경과한 배를 이용하는 것이 적당할 것으로 보였다.
