Lactobacillus plantarum (KACC 92189) as a Potential Probiotic Starter Culture for Quality Improvement of Fermented Sausages

  • This study was conducted to evaluate the effects of fermenting temperature on the applicability of Lactobacillus plantarum for production of fermented sausages as starter cultures, and its applicable efficiency was also compared with those inoculated with commercial starter culture or non-inoculated control. The L. plantarum isolated from a naturally-fermented meat, identified by 16S rDNA sequencing and again identified by de novo Assembly Analysis method was used as a starter culture. Six treatments: 3 with L. plantarum at different fermenting temperatures (20, 25 and $30^{\circ}C$), and other 3 treatments (1 with commercial starter culture, 1 with its mixture with L. plantarum and 1 non-inoculated control) fermented under the same conditions ($25^{\circ}C$) were prepared. Results revealed that the fermenting temperature considerably affected the pH change in samples added with L. plantarum; the highest pH drop rate (1.57 unit) was obtained on the samples fermented at $30^{\circ}C$, followed by those at $25^{\circ}C$ (1.3 unit) and $20^{\circ}C$ (0.99 unit) after 4 days fermentation. Increasing the temperature up to $30^{\circ}C$ resulted in significantly lower spoilage bacteria count (5.15 log CFU/g) and lipid oxidation level in the products inoculated with L. plantarum. The sensory analysis also showed that the samples added with L. plantarum at $30^{\circ}C$ had significantly higher odor, taste and acceptability scores than those fermented at lower temperatures. Under the same processing condition, although the L. plantarum showed slightly lower acidification than the commercial starter culture, however, it significantly improved the eating quality of the product.

The Effects of Nitrogen Rates on The growth and Yield of Waxy Corn after Cultivating Hairy Vetch in Agricultural Fields with The Stream (하천변 농경지에서 헤어리베치를 재배한 후 질소시비량에 따른 찰옥수수의 생육과 수량에 미치는 영향)

  • This experiment was carried out to figure out the growth and yield of waxy corn after cultivating the hairy vetch which was mostly used as a green manure crop. The waxy corn showed the growth and yield efficiency relative to conventional culture after the hairy vetch was returned to as the green manure and the amount of nitrogen fertilizer increased, and the LAI was similar to the conventional culture when less than 18 kg. Length of ear slightly decreased compared to 188 mm of conventional culture when the hairy vetch was used as the green manure and the amount of nitrogen fertilizer was less than 9 kg, but the length of ear increased relative to the conventional culture when the amount of applied nitrogen fertilizer was more than 18 kg. The highest 100-kernel weight was 35.4 g with the hairy vetch and 27 kg of nitrogen fertilizer treatment. The lowest 100-kernel weight was 27.4 g with the hairy vetch and 0 kg of nitrogen fertilizer treatment. Quantity with the hairy vetch and 9 kg of the nitrogen fertilizer treatment was similar the conventional culture. Suger content with the hairy vetch and more than 18 kg of the nitrogen fertilizer treatment was higher than the conventional culture. Thickness of pericarp showed no difference among treatments.

Efficient Use of Lactose for Production of the Soluble Recombinant Human Epidermal Growth Factor in Escherichia coli. (대장균에서 lactose를 이용한 수용성 재조합 인간 상피 세포 성장 인자의 생산)

  • Recombinant human epidermal growth factor (rhEGF) was produced by E. coli BL2l (DE3) harboring a plasmid pYHB101. The production of rhEGF was 44.5 mg/L when the E. coli BL2l (pYHB101) was cultured at 27$^{\circ}C$ for 48 hr in the modified MBL medium containing 10 $\mu\textrm{g}$/L glucose with 10 $\mu\textrm{m}$ IPTG/lactose induction at 2 hr after inoculation. It was shown that lactose is able to induce the rhEGF expression of E. coli BL2l (pYHB101) with the same efficiency as IPTG. In the batch culture system, when induced with 10 $\mu\textrm{m}$ lactose, E. coli BL2l (pYHB101) produced maximum 45 mg/L of the rhEGF at 28 hr culture in the modified MBL medium containing 10 g/L glucose. In the semi-fed batch culture system, the volumetric yield was 160 mg/L when the culture was added with 0.5% (w/v) lactose and 0.25% (w/v) yeast extract in the late logarithmic phase and 94.3% of rhEGF was secreted as soluble form. However, when the culture was added with them in the early logarithmic phase, the volumetric yield was 120 mg/L and 20.9% of rhEGF was found in cytoplasmic insoluble aggregates. It was found that the addition time of lactose was important for production of soluble rhEGF from E. coli BL21 (pYHB101).

Detection of Microbial Growth in an Automated Culture System (자동배양기를 이용한 미생물 검출)

  • Modern automated culture systems have increased the isolation rate of microorganisms and shortened the time to detection, reducing experimental errors in diagnosis of infecting agents. BacT/ALERT 3D system is based on the colorimetric detection of $CO_2$ produced by the growing microorganisms. In order to evaluate the efficiency of the detection system, sterility test were performed using 6 bacteria. With standard aerobic and anaerobic bottles containing the liquid media, both three aerobic bacteria (P. aeruginosa, M. luteus, B. subtilis) and a facultative bacterium S. aureus were detected up to 1 CFU in 31.44 hr. In addition, growth of anaerobic C. sporogenes was recognized up to 1 CFU in 15.96 hr. The slowly growing bacteria P. acnes was detected up to 10,000 CFU in 129.36 hr. In comparison with conventional culture method, BacT/ALERT 3D automated culture system was more sensitive and saved detection time up to$2\sim10$ hr. Therefore, this automated culture system enables to efficiently detect bacteria in clinical samples and biological medicines.

  • PURPOSE : Club dance is one of the most popular cultures worldwide and is getting popular in Korea over time. However, club still has negative awareness in the country due to the media spreading only harmful aspects of club culture. At any rate, nowadays many young aged people can easily have chances to experience clubs so we tried to find out the bad effects caused by clubs. METHOD : We conducted a survey and the number of participants who agreed to participate in this survey was 500, including both men and women. There are a total of 50 questions in the questionnaire form used by this study and the questions were divided into three categories: general(18), psychologic-related(7), social-cultural(9), and physical-related(16) questions. Data collected for this study was analyzed via frequency analysis using SPSS. RESULT : In our survey, most students who had already experienced the club culture tend to go to club once or twice a month, and they answered that they would feel bored if they were nor able to do so. In terms of social life, most respondents said that they had lower efficiency in working and studying the next day after going to clubs because of fatigue. In physical aspect, club users use then pelvis and lower back a lot while dancing. The respondents answered that they experienced muscle soreness, a loss in weight, and changes in their body as a consequence of intense dancing. They complaint pain mostly in lower limbs, so we could find out the fact that legs were primarily used for the club dance. CONCLUSION : To sum up, the club culture in undergraduate students affects on their daily living and body, and also has good effects as well as bad ones. Therefore, we need to look at the club culture positively and to improve bad effects of club culture.

A Method of Using Digital Map to Improve Multicultural Education Efficiency: A Design of 'e-Atlas of Society and Culture' for Marriage Immigrant Women to Overcome Contextual Limitations (다문화교육 효율성 제고를 위한 전자지도의 활용방안 -결혼이주여성의 맥락적 한계 극복을 위한 'e-사회문화지도'설계-)

  • In this paper, we study on an 'e-Atlas of Society & Culture(e-ASC)', to develop and implement educational courseware for marriage immigrant women, to adapt Korean society. The 'e-ASC' can help cultural adaptation, such as the understanding of Korean people's common values and behaviors and can likewise provide opportunities to contribute to Korean society, for marriage immigrant women living in Korea. E-cultural map is a useful tool for providing educational experiences and sharing a sociocultural homogeneity, using three dimensional space, for both immigrants and residents. The 'e-Atlas of Society & Culture', suggested in this paper, using the notion of an E-cultural map, has a strong point. That is to say, the immigrants have contextual limitations about Korea and Korean culture, caused by some particular events in their motherland. This atlas provides an opportunity to overcome this contextual limitations and to help understand and adapt to Korean society for these immigrants. This 'e-Atlas of Society & Culture' can be an efficient educational tool for immigrants and can help marriage immigrant women understand and adapt Korean social and cultural phenomena. They might have contextual limitations about these phenomena in their motherland.

  • To investigate the response on feeder cell cultures, protoplasts isolated from cell suspensions initiated from mature seed scutellum-derived callus of the Japonica rice variety Taipei 309, were cultured on filter membranes under various conditions. The effects of various factors, such as gelling agents, feeder cell and protoplast densities, species of feeder cells and heat shock treatment, have been investigated to improve protoplast plating efficiencies on filter membranes. Maximum protoplast plating efficiencies were obtained when protoplasts were cultured on KPR medium semi-solidified with Sea Plaque agarose at a density of $5\;\times\;10^{5}\;ml^{-1}$ protoplasts in the presence of Lolium multiflorum as feeder cells (0.5 ml pcv per 10 ml of protoplast culture medium). Pre-culture heat shock treatments for 1 min. and 5 min. to the protoplasts did not give any appreciable increase on the plating efficiency of protoplasts in the presence of feeder cells. Maltose-supplemented medium was superior for plant regeneration from protoplast-derived colonies compared with medium containing only sucrose. The plants were transferred to the glasshouse, flowered and were fertile.

  • The effect of several factors on efficiency of anther culture of broccoli was studied. The medium supplemented with 14% sucrose, 125mg/L of silver nitrate and 0.1mg/L of each of NAA and 2,4-D was best in embryogenesis in all varieties tested. The effect of auxin was affected by silver nitrate, that is, it lowered embryogenesis when the concentration of NAA and 2,4-D was low, but it enhanced when their concentrations were high. Benzyl aminopurine increased embryogenesis in some varieties, but decreased in others. Although microspore embryogenesis was observed in anthers of which heigest was 1 to 0.6 - 1.0 of petal heigest in the bud, which corresponds to the early uninucleate to early binucleate stage of the microspores, it was much better when the ratio of heigest was 1 : 0.8. More embryos ware inducted from culture in March than in February in general. Because there was a big yearly variation, it was assumed that microspore embryogenesis was influenced greatly by the culture condition and the physiological condition of donor plants.

  • This study was based on the research contents of previous studies. and After operatively defining public culture and arts institutions in terms of combining the characteristics of public institutions and culture and arts, the necessity of analyzing the priority of performance evaluation indicators was raised in accordance with this. The results of priority analysis based on the AHP axiom are as follows. The priorities for the performance evaluation index of public culture and arts institutions were in the order of artistry, expertise, marketing, publicity, organizational management, exchange and cooperation, management efficiency, operating system, and convenience. Based on the analysis results, the following implications were presented. First, it is necessary to first pay attention to the items of artistry that appear as the highest priority and seek strategies. Second, it is necessary to put a variety of experts in the original plan so that the audience and the public can have a high interest in this. However, in order not to exceed the 10 items recommended by the AHP axiom, we had to intentionally limit the evaluation items. Therefore, it is necessary to analyze priorities by deriving various evaluation items in subsequent studies.

Development of a Simple Cell Lysis Method for Recombinant DNA Using Bacteriophage Lambda Lysis Genes

  • In this study, we describe the development of a simple and efficient method for cell lysis via the insertion of a bacteriophage lambda lysis gene cluster into the pET22b expression vector in the following order; the T7 promoter, a gene for a target protein intended for production, Sam7 and R. This insertion of R and Sam7 into pET22b exerted no detrimental effects on cellular growth or the production of a target protein. The induction of the T7 promoter did not in itself result in the autolysis of cells in culture but the harvested cells were readily broken by freezing and thawing. We compared the efficiency of the cell lysis technique by freezing and thawing to that observed with sonication, and determined that both methods completely disintegrated the cells and released proteins into the solution. With our modification of pET22b, the lysis of cells became quite simple, efficient, and reliable. This strategy may prove useful for a broad variety of applications, particularly in experiments requiring extensive cell breakage, including library screening and culture condition exploration, in addition to protein purification.
