Comparative Analysis of Callus Induction and Plant Regeneration Rates Using One-step and Two-step Cultures for Rice Anther Cultivation (벼 약배양 1단계 및 2단계 배양을 이용한 캘러스 유도 및 식물 재부화율 비교 분석)

  • Anther cultivation for crop breeding is a method of rapid production of homozygosities by greatly reducing the time required for at least six generations to develop new varieties using conventional breeding methods. This technique of producing anther culture provides an opportunity to obtain more green plants from a methodological point of view, and the techniques that save time and effort in anther culture are also important because they increase the efficiency of culture. This study compared the callus induction rate and green plant regeneration rate of a one-step and a two-step culture that differ in their culture media and culture methods. One-step culture allows callus induction and plant regeneration in one medium, whereas two-step culture requires induction and plant regeneration in two different media. In this study, we compared the callus induction and plant regeneration rates of rice anthers as one-step and two-step cultures. The callus formation rate was 13.0% for one-step cultures and 8.6% for two-step cultures, so the rate was 4.4% higher for one-step cultures than for two-step cultures. The plant regeneration rate was 1.0% in one-step cultures and 3.0% in two-step cultures, so the regeneration rate was three times higher for the two-step cultures than for one-step cultures. This suggests that the two-step cultures are more efficient than the one-step cultures for haploid production.

Effect of Growth Rate and Cultivation Temperature on the Yeast RNA Accumulation and Autolysis Efficiency (생육속도 및 배양온도가 효모 RNA 축적과 autolysis 효율에 미치는 영향)

  • Continuous fermentations were performed in order to investigate the effect of culture condition on the yeast RNA accumulation and autolysis efficiency. The content of intracellular RNA increased with increasing dilution rate, showing its maximum value of 14.8% at D=0.35 $h^{-1}$. Also, both RNA productivity and specific RNA productivity tended to increase with the increase of dilution rate. The maximum biomass was obtained at $30^{\circ}C$ in the fixed dilution rate of 0.2 $h^{-1}$, whereas the maximum RNA content appeared at the lowest temperature experimented. Growth rate affected significantly on the yeast autolysis efficiency such that the extraction ratio(TN/TN) increased with increasing growth rate, whereas the hydrolysis ratio(AN/TN) was reversed. On the other hand, its efficiency was little affected by cultivation temperature.

Establishment of an Efficient Agrobacterium Transformation System for Eggplant and Study of a Potential Biotechnologically Useful Promoter

  • An efficient and reliable Agrobacterium transformation procedure based on TDZ (thidiazuron)-induced organogenesis was established and applied to six Brazilian eggp1ant varieties. Optimum transgenic plants recovery was achieved upon the study of the following parameters affecting transformation efficiency, using F-100 variety as a model: i) explant source; ii) pre-culture period; iii) physical state of the pre-culture medium and iv) coculture conditions. The highest frequency of kanamycin-resistant calli derived from leaf explants (5%) was obtained without a pre-culture period and co-cultivation for 24 h in liquid medium followed by five days on solid RM (regeneration medium). For cotyledon explants, best results were achieved upon a pre-culture of 24 h in liquid RM and a co-cultivation period of 24 h in liquid RM followed by three days in solid RM, resulting in a transformation Sequency of 22.7%. Kanamycin-resistant organogenic calli were also obtained from cultivars Emb, Preta Comprida, Round nose Shaded, Campineira and Florida Market. The expression pattern of an epidermis-specific promoter was studied using transformants expressing a chimaeric construct comprised by the promoter Atgrp-5 transcriptionally fused to the coding region of the gus gene. The expression pattern was similar to that previously observed in tobacco and Arabidopsis thaliana, with preferential expression at the epidermis and the stem phloem. These results support the idea that the Atgrp-5 promoter can be used to drive defense genes in these tissues, which are sites of pathogen interaction and spread, in programs for the genetic improvement of eggplant.

Effects of Extracellular Matrix Protein-derived Signaling on the Maintenance of the Undifferentiated State of Spermatogonial Stem Cells from Porcine Neonatal Testis

  • In general, the seminiferous tubule basement membrane (STBM), comprising laminin, collagen IV, perlecan, and entactin, plays an important role in self-renewal and spermatogenesis of spermatogonial stem cells (SSCs) in the testis. However, among the diverse extracellular matrix (ECM) proteins constituting the STBM, the mechanism by which each regulates SSC fate has yet to be revealed. Accordingly, we investigated the effects of various ECM proteins on the maintenance of the undifferentiated state of SSCs in pigs. First, an extracellular signaling-free culture system was optimized, and alkaline phosphatase (AP) activity and transcriptional regulation of SSC-specific genes were analyzed in porcine SSCs (pSSCs) cultured for 1, 3, and 5 days on non-, laminin- and collagen IV-coated Petri dishes in the optimized culture system. The microenvironment consisting of glial cell-derived neurotrophic factor (GDNF)-supplemented mouse embryonic stem cell culture medium (mESCCM) (GDNF-mESCCM) demonstrated the highest efficiency in the maintenance of AP activity. Moreover, under the established extracellular signaling-free microenvironment, effective maintenance of AP activity and SSC-specific gene expression was detected in pSSCs experiencing laminin-derived signaling. From these results, we believe that laminin can serve as an extracellular niche factor required for the in vitro maintenance of undifferentiated pSSCs in the establishment of the pSSC culture system.

Immobilized Condition of Suchwowces cerevisiae for Ethanol Production from Persimmon Juice. (감 즙으로부터 에탄을 생산을 위한 Saccharomyces cerevisiae의 고정화 조건)

  • The immobilized culture system of Saccharomyces cerevisiae was examined to improve the efficiency of vinegar production from persimmon juice. Optimum concentration of Na-alginate for the immobilization was 2%. When the 1eakage of yeast from get beads was checked by turbidity of culture medium with varying concentration of Na-alginate from 1 to 4%, turbidity of culture medium increased from 8 hrs of cultivation with 1% Na-alginate concentration showing optical density of 0.82 at 20 hrs. However, the increase in turbidity of culture medium was slow with 2-4% Na-alginate showing optical density of 0.55-0.58 at 20 hrs. Microscopical analysis of gel matrix showed that the immobilized yeast was grown well regardless of Na-alginate concentration. Optimum size of gel bead and amount of inoculation were 2-3 m and 33mg, respectively. For ethanol production aerobic cultivation for 121hrs using cohen plug followed by anaerobic cultivation using silicon plug equipped with a check valve was the most effective.

Pathogenicity of Lactobacillus pentosus PL11 isolated from eel (Anguilla japonica) intestine and single oral toxicity of its culture broth in rats (랫트에서 뱀장어로부터 분리된 Lactobacillus pentosus PL11의 병원성 및 배양액에 대한 단회 경구독성 시험)

  • The pathogenicity and acute toxicity of Lactobacillus (L.) pentosus PL11 from eel (Anguilla japonica) were investigated using male and female Sprague-Dawley rats. The pathogenicity of L. pentosus PL11 was examined after treating the rats with $10^{11}$ CFU/mL, $10^9$ CFU/mL or $10^7$ CFU/mL doses of L. pentosus PL11 culture or 0.85% NaCl (Control) intragastrically. For acute toxicity studies, rats were treated with dried culture broth of L. pentosus PL11 at doses of 5,000 mg/mL, 2,500 mg/mL, 1,250 mg/mL or 625 mg/mL or Lactobacilli MRS broth (Control), and clinical signs or mortalities were monitored for two weeks. The results of the present investigation revealed no mortalities or obvious clinical signs in rats administered with the live bacterial cultures or dried culture broth at any investigated dose level. Also, no significant differences were observed in net body weight gain, gross pathological findings, feed and water consumption and body temperature among the different treatment groups and between the treated and control rats. It can be concluded from the above findings that L. pentosus PL11 is a safe probiotic strain with potential as feed additive to increase the feed efficiency or health of fish.

  • The purpose of this study is to analyze the change of hospitals that patients safety and quality improvement by accreditation process and to examine the impact or interrelation of leadership, organizational culture, hospital management activities and recognition of hospital management performances. The data were collected through a review of the literature, and selfadministered survey with a structured questionnaires to 714 subjects from several medical staff members, administration staff members, nursing staff members, medical technicians and other staff members working in 23 accredited hospitals in Korea. In this analysis hierarchical multiple regression and structural equation model were used. The conclusion of this study provides a theoretical model for understanding organizational changes brought about by accreditation system. Factor on improvement of efficiency and raise the morale, rather than increase of medical income and reduce of the cost factors, had a stronger influence on the accreditation process. In the future, the hospital's participation to induce the accreditation program voluntarily will come up with an alternative policy concern about financial perspective. Also, the hospitals which preparing accreditation program to achieve the goal efficiently, will make use of transformational leadership through enhancing individual consideration and intellectual development to leading members participation. Additionally, non-accredited hospitals should aim at professional culture by innovative and creative approaches, and inviting members to learning and growth in the organization.

  • The carbon dioxide, nitrogen, and phosphate removals from wastewater using microalgae have extensively been studied. Especially, the green colonial algae Botryococcus braunii is characterized by unusual high hydrocarbon contents, ranging from 15 to 75% of dry weight, as long-chain unsaturated hydrocarbons. These hydrocarbons suggest that the possibility of renewable biofuels to be converted into useful fuels such as gasoline by simple catalytic cracking. The poor recovery (18 - 32%) of hydrocarbon from B. braunii culture in two-phase bubble column seems to be caused by insufficient mixing between two phases, which was operated using only aeration on the narrow interface between hydrophobic solvent and cell suspension. In addition, hydrocarbon was entrapped tightly in cell-matrix (formed by exopolysaccharide) of algal colony, which make difficult to extract using two-phase system. In order to overcome low recovery efficiency, two-stage extraction culture system including culture vessel and two-phase separator is now under development, resulted improving contact between solvent phase and cell suspension. Hydrocarbon recovery using this process was more than two times as that using two-phase extraction culture.

Effects of Dimethylsulfoxide on the Cell Wall Regeneration and Cell Division of Protoplasts Isolated from Panax ginseng Callus (인삼 캘러스 원형질체의 세포분열과 세포벽 재생에 미치는 Dimethylsulfoxide의 효과)

  • Isolated protoplasts from ginseng (Panax ginseng C. A. Meyer) callus tissue were cultured in modified MS media supplemented with various concentrations of dimethylsulfoxide (DMSO). The cell wall regeneration rate and cell division efficiency of the protoplasts were increased significantly by 1% DMSO treatment. However, there was no difference in the viability of protoplasts between the DMSO treatment and non-treatment. Transmission electron microscopy revealed that the microtubules were oriented in parallel manner to the plasmalemma after 3 days of culture in medium with 1% DMSO. Further, interconnected cellulose microfibrils were observed on the outer surface of the 3-day-cultured protoplasts by scanning electron microscopy These structures shown by electron microscopy were not observed in protoplasts cultured on DMSO-free media. This studies indicates that DMSO supplemented in culture media seemed to stimulate the cell wall regeneration and cell divisions of protoplasts by forming microtubule organizing centers (MTOC).

Growth Characteristics of Tissue Cultured Plantlets by Lighting Direction and tight Intensity (광조사 방향 및 광도에 따른 배양식물체의 생장특성)

  • The present study was carried out to determine the effects of lighting directions and light intensities on the growth of in vitro plantlets of strawberry, potato and lily. The growth of plantlets was affected by two different lighting systems, downward and sideward lighting system. There were no difference for the plantlets of strawberry regardless of lighting directions. However, leaf area, fresh weight and dry weight have increased as the light intensity increased. Also, plant height and root length on node culture of potato decreased at high intensity of the sideward lighting, while thickness of stem and root was thicker, fresh weight and dry weight were heavier, and leaf area was increased. Also, bulblet formation on scale culture of lily has been abundant, and fresh weight and dry weight were increased. Thus, the sideward lighting system which loaded three stories for culture vessels with raising light intensity into culture vessels instead of conventional downward lighting system promoted growth and raised the efficiency of production of high quality microplant.
