This study aimed to provide implications required to establish a content strategy by examining the influencing factors affecting the acceptance of curation services for 320 OTT users, and the main results are as follows. First, innovativeness was found to have a positive effect on performance expectations. Second, innovativeness was found to have a positive impact on the effort expectation. Third, performance expectation had a positive effect on the intention to use continuously. Fourth, it was shown that the effort expectation had a positive effect on the intention to use continuously. Fifth, social influence was found to have no significant effect on the intention to use continuously. Sixth, it was found that the facilitating conditions did not significantly affect the intention to use continuously. The above results can be assessed as the higher the OTT users perceive the performance and effort expectations of the curation service, the higher their intention to continue using them. This study is meaningful in that it verified the factors affecting the intention to use the OTT curation service and expanded the UTAUT model.
This study was carried out to select the optimal condition for prothallus propagation and sporophyte formation of Leptogramma pozoi (Lag.) Ching subsp. mollissima (Fisch. ex Kunze) Nakaike and to provide basic data for mass production system. In the propagation, 300 mg of prothallus were inoculated in different kinds of medium [1/4, 1/2, Murashige and Skoog (MS), Knop], sucrose (0, 1, 2, 3, 4%), and cultured for 8 weeks. Sporophyte formation studies were carried out by inoculating blended prothallus into artificial soils. The soils were mixed with horticultural substrate, peatmoss, perlite, and decomposed granite at different ratios or only with the horticultural substrate. Then the prothallus was cultivated for 12 weeks to figure out the formation and growth of the sporophytes. The growth and development of prothalli were excellent on MS medium. According to medium components, prothallus growth was favorable in all treatments except 0% sucrose treatment and the highest in 2% sucrose. In the experiment of soil mixtures, sporophyte formation was the highest in the horticultural substrate:perlite 2:1 (v:v) and horticultural substrate:decomposed granite 2:1 (v:v) treatment, and the overall growth was good in the horticultural substrate:decomposed granite 2:1 (v:v) mixed soil.

Decay fungi can decompose plant debris to recycle carbon in the ecosystem. Still, they can also be fungal pathogens, which can damage living trees and/or wood material and cause a large amount of timber loss. We isolated and identified basidiomycetous fungi from the decayed bark of Mongolian oak wrapped with sticky roll traps. The degrading enzyme activities were then tested for all fungal isolates. The decay ability of selected isolates was assessed based on the weight loss of wood discs after inoculating with culture suspension of decay fungi under the different humidity levels. A total of 46 basidiomycetous fungal isolates belonged to 12 species, and 10 genera were obtained from Jong Myo (16 isolates), Chang Kyung palace (7 isolates), Cheong Gye (10 isolates), and Gun Po (13 isolates). Gymnopus luxurians was the most dominant fungus in the present study, and this species distributed in all survey sites with 9 isolates in Jong Myo, followed by 3 isolates in Chang Kyung palace, while Cheong Gye and Gun Po had only 1 isolate each. Among 46 isolates, 44 isolates secreted at least one enzyme, while 25 isolates produced both cellulase and phenol oxidase enzymes, and 2 isolates produced neither. The assessment of decay ability by artificial inoculation indicated that the weight loss of wood discs was significantly influenced by humidity conditions when inoculated with bark decay fungi. The percent weight losses by G. luxurians inoculation in RH of 90-100% and RH of 65-75% were 4.61% and 2.45%, respectively. The weight loss caused by Abortiporus biennis were 6.67% and 0.46% in RH of 90-100% and RH of 45-55%, respectively. The humidity reduction approach should be applied for further studies to control the growth and spread of bark decay fungi on the trunks wrapped with sticky roll traps.
This paper is an essay on Chaekmun(Answer papers of official examination in Pre-modern period). Especially I tried to point out the possibility of practical use in the field of politics. For this purpose, I analyzed an examination paper written by Lee Ga-hwan, and the title is "a debate about construction of the Miang palace". Exquisite writing skill is also important in Chaekmun, but it is not everything. The subject of Chaekmun concerns various fields such like politics, economy, society, culture, and history. So that writers should have sufficient knowledge and opinion. The King, as an examiner, wants to test retainers' ability both in writing and politics. In this paper, I focused on using in field of politics between the King and the retainers. And as an good example, I analyzed a paper which dealt the event of building huge palace named Miang palace. That is because the King reflects his concerns by setting exam questions. And his concerns also could not be free of contemporary political conditions. Therefore we should be careful of reading those articles. Regarding this, Lee Ga-hwan's article had a distinctive characteristic. Unlike any other articles dealing with same event, he tried to access through creative point of view. And his thought were much close to the King's heart.
Panax ginseng (Ginseng or Korean ginseng) is one of the most important medicinal herbs in the world. We made a high-quality whole genome sequence of P. ginseng using 'Chunpoong' cultivar, which is the first cultivar registered in Korea Seed and Variety Service (KSVS) with relatively similar genotypes and superior phenotypes, representing approximately 3 Gbp and 60,000 genes. Genome sequence analyses of P. ginseng and related speciesrevealed the origin of Korean ginseng and the ecological adaptation of 18 Panax species around the world. Korean ginseng and American ginseng (P. quinquefolius) are tetraploid species having 24 chromosome pairs, while the other 16 species are diploid species with 12 chromosome pairs. Panax and Aralia are the closest genera belonging to the Araliaceae family that diverged approximately 8 million years ago (MYA). All Panax species evolved as shade plants adapting to cool climates and low light conditions under the canopy of deep forests from Southeast Asia such as Vietnam to Northeast Asia such as Russia approximately 6 MYA. However, through recurrent ice ages and global warming, most diploid Panax species disappeared due to the freezing winter, while tetraploid P. ginseng may have appeared by allotetraploidization, which contributed to the adaptation to cold temperaturesin Northeast Asian countries including the Korea peninsula approximately 2 MYA. American ginseng evolved by the adaptation of P. ginseng in Northeast America after the intercontinental migration 1 MYA. Meanwhile, most of diploid Panax species survived in high-altitude mountains over 1,600 meters in Southeast Asia because they could not endure the hot temperature and freezing cold. The genome sequence provides good basisto unveil the origin and evolution of ginseng and also supports practical gene chips which is useful for breeding and the ginseng industry.
This study intended to identify causes of poor tree vigor in the Hanbando coastal forest by investigating its geographical environment, climate condition, soil physicochemical characteristics, and growth condition of Pinus thunbergii. It divided the forest into an area with poor tree vigor or dead standing trees and a control area with good tree vigor and examined them separately. The survey showed that stand density was significantly higher in the area with poor tree vigor. In contrast, the crown width in the area with good tree vigor was wider. The number of dead standing trees and the stand density showed a negative correlation. The stand density and diameter at breast height (DBH), tree height, crown height, and crown width also showed a negative correlation. The result indicated that, as the tree's stand density increases, the crowns of individual trees overlapped and the lower branches died. Then crown height and crown width decreased, and the number of leaves and photosynthesis was reduced, leading to lower tree height and weaker growth of breast diameter. As a result, tree vigor weakened, and combined with environmental pressures from the lack of moisture and nutrients in coastal soil and salty wind, P. thunbergii in coastal areas is expected to die massively. Although the causes of dead standing trees and poor tree vigor of P. thunbergii in the Hanbando coastal forest are complicated, poor management of adequate tree density in response to the growth of P. thunbergii is the primary cause. The secondary cause is external environmental pressures, including unfavorable soil conditions and salty and strong wind that obstruct the growth of P. thunbergii.
An electromagnetic generator with variable stimulation parameters is required to conduct basic research on magnetic flux density and frequency for pulsed electromagnetic fields (PEMFs). In this study, we design an electromagnetic generator that can conduct basic research by providing parameters optimized for cell and animal experimental conditions through adjustable stimulation parameters. The magnetic core was selected as a solenoid capable of uniform and stable electromagnetic stimulation. The solenoid was designed in consideration of the experimental mouse and cell culture dish insertion. A voltage and current adjustable power supply for variable magnetic flux density was designed. The system was designed to be adjustable in frequency and pulse width and to enable 3-channel output. The reliability of the system and solenoid was evaluated through magnetic flux density, frequency, and pulse width measurements. The measured magnetic flux density was expressed as an image and qualitatively observed. Based on the acquired image, the stimulation area according to the magnetic flux density decrease rate was extracted. The PEMF frequency and pulse width error rates were presented as mean ± SD, and were confirmed to be 0.0928 ± 0.0934% and 0.529 ± 0.527%, respectively. The magnetic flux density decreased as the distance from the center of the solenoid increased, and decreased sharply from 60 mm or more. The length of the magnetic stimulation area according to the degree of magnetic flux density decrease was obtained through the magnetic flux density image. A PEMF generator and stimulation parameter control system suitable for cell and animal models were designed, and system reliability was evaluated.
The objective of this study was to optimize industrial-grade media for improving the biomass production of Weissella cibaria JW15 (JW15) using a statistical approach. Eleven variables comprising three carbon sources (glucose, fructose, and sucrose), three nitrogen sources (protease peptone, yeast extract, and soy peptone), and five mineral sources (K2HPO4, potassium citrate, ⳑ-cysteine phosphate, MgSO4, and MnSO4) were screened by using the Plackett-Burman design. Consequently, glucose, sucrose, and soy peptone were used as significant variables in response surface methodology (RSM). The composition of the optimal medium (OM) was 22.35 g/l glucose, 15.57 g/l sucrose, and 10.05 g/l soy peptone, 2.0 g/l K2HPO4, 5.0 g/l sodium acetate, 0.1 g/l MgSO4·7H2O, 0.05 g/l MnSO4·H2O, and 1.0 g/l Tween 80. The OM significantly improved the biomass production of JW15 over an established commercial medium (MRS). After fermenting OM, the dry cell weight of JW15 was 4.89 g/l, which was comparable to the predicted value (4.77 g/l), and 1.67 times higher than that of the MRS medium (3.02 g/l). Correspondingly, JW15 showed a rapid and increased production of lactic and acetic acid in the OM. To perform a scale-up validation, batch fermentation was executed in a 5-l bioreactor at 37℃ with or without a pH control at 6.0 ± 0.1. The biomass production of JW15 significantly improved (1.98 times higher) under the pH control, and the cost of OM was reduced by two-thirds compared to that in the MRS medium. In conclusion, OM may be utilized for mass producing JW15 for industrial use.

The repeated monthly or weekly subculture of plant callus is labor intensive and increases the risk of somaclonal variation from the parental callus line. The most effective method for preserving plant callus is cryopreservation, which involves storage in liquid nitrogen. However, this method cannot be applied to the callus of different plant species in the same manner, so it is difficult to develop a standardized cryopreservation method. In addition, the survival rate of the frozen callus after thawing and the regeneration rate after survival are uncertain. Therefore, it is necessary to develop a method to extend the subculture interval of plant callus in an active state. In this study, active plant calli of various species without freezing was incubated at 15℃ for 4 to 12 weeks without subculture. After 12 weeks, 8 lines of plant callus grew less than 2-fold when cultured at 25℃, but at least 2 times as much when cultured at 15℃. Moreover, total antioxidant activity did not differ significantly between plant callus recovered at 25℃ after culturing at 15℃ or at 25℃. These results show that the subculture interval can be extended at a temperature of 15℃ without need for modified medium composition or additional processes. In addition, positive results in all calli of several plant species are expected to reduce labor as well as somaclonal variation by increasing the subculture.
Background and objective: Plants in temples are the results of cultural symbolization that embraces the experience and enlightenment of humans about life. As a way to improve the acceptance of the foreign religion, China gave cultural symbolization to plants in temple gardens through integration with traditional cultures and the understanding of the nature of plants themselves. This study aimed to identify cultural symbolism and signs of acculturation associated with Buddhist plants, targeting Bolisu, the most essential and symbolic plant in temple garden forests in China and Korea. Methods: The morphological and ecological characteristics of plants, functions, the texts that contained the history of Buddhism and literary works were examined through literature review, and the relation of Ficus religiosa with its planting conditions and nature, and Buddhist culture was explored. In addition, the cultural value of Buddhist plants themselves in establishing temples and the reason why Bolisu was planted in temples were reviewed through time series analysis. The obtained results were interpreted using an inductive method to identify substitutes for F. religiosa, cultural symbolism and signs of acculturation. Results: F. religiosa as one of the three holy trees and the five trees and six flowers in Buddhism is known as the original Bolisu. Since it grows well and is widely distributed in regions that accepted Indian Buddhism, it became the most representative holy tree in Buddhism. From the perspective of tree shape and nature, F. religiosa is in line with the Buddhist spirit of saving those in need with mercy and redeeming mankind, and figuratively shows that perfection can be attained like the fruit of Bolisu. Chines Buddhism had adopted highly symbolic plants for a long period of time as a means to spread the same belief and doctrines as Indian Buddhism. In China, however, there were only limited areas suitable for the growth of F. religiosa, and for this reason, borrowed Bolisu trees including Tilia. miqueliana, T. mandshurica and T. amurensis and other plants such as F. virens Ait. var. sublanceolata, G. biloba and M. alba were planted as a substitute in most regions, having been given with symbolism and belief as Bolisu. Conclusion: Chinese Buddhism planted the same plants as Indian Buddhism in order to enhance symbolism and also similar substitutes to express the same symbolism. This is a kind of acculturation and its influence and customs were not limited to China, but were introduced to Korea, The difference between China and Korea was that G. biloba was excluded from the substitute for Bolisu in Korea.
