The Extracting Methods of the Seeds of Yew(Taxus cuspidata Sieb) by Solvent Extraction and Its Efficacy (주목(朱木)씨앗 추출물의 제조방법 및 효능효과)

  • Yew (Taxus cuspidata Sibe) selected cultivation as drug, food and decorative plant in Gyong-gi province in Korea. To extract the water soluble active ingredients, as a extracting method, there was extracted with 20g of dried seeds with each 20g of butylene glycol(BG) and propylene glycol(PG), and 40 mL of water mixing 72 hours at $40{\pm}5^{\circ}C$, and then they were filtrated by 400 mesh. Appearance of extract of seeds was pale brown, $pH=4.5{\pm}0.5$, $gravity=1.013{\pm}0.05$, a reflective $index=1.373{\pm}0.05$, and yield=75%. Also, to extract the high purity oil from seeds, it minutely pulverized the dried seeds and added the hexane, mixing 2 hours at $20{\pm}57^{\circ}C$. And then, this filtrated it with 400-mesh. It got the purified oil through evaporating them at $55^{\circ}C$ during under vacuum. As the results, appearance was slightly brown, gravity=0.922 acid value=0.12, saponification value=192, and it should be obtained the $40{\pm}5%$ of yield. As the efficacy evalution of cosmetic field, the antioxidative activities by NBT method were stronger 86.0% from extract of talus seeds than 52.0% from green tea extract and 35.0% from skullcap extract as well as the antioxidative activities by DPPH method were stronger 93.7% from extract of seed than 60.3% from extract of green tea and 27.1% from extract of skullcap. These are more effective than other plant extracts. The collagen synthesis rate on the activating fibroblast for Taxus cuspidata Sibe extract showed 35.43%. As the activity of the skin elasticity, PPE(porcine pancreatic elastase)-inhibitory activities of talus extract was 50.8%. Anti-inflammatory activity was more effective to be taken 41.1% of taxus seed oil than 24.2% of steady glycyrrhetinate (SG) as a control.

Basic study on high gradient magnetic separation of nano beads using superconducting magnet for antibody purification

  • The manufacturing process of antibody drugs comprises two main stages: the upstream process for antibody cultivation and the downstream process for antibody extraction. The domestic bio industry has excellent technology for the upstream process. However, it relies on the technology of foreign countries to execute downstream process such as affinity chromatography. Furthermore, there are no domestic companies capable of producing the equipment for affinity chromatography. High gradient magnetic separation technology using a high temperature superconducting magnet as a novel antibody separation and purification technology is introduced to substitute for the traditional technology of affinity chromatography. A specially designed magnetic filter was equipped in the bore of the superconducting magnet enabling the continuous magnetic separation of nano-sized paramagnetic beads that can be used as affinity magnetic nano beads for antibodies. To optimize the magnetic filter that captures superparamagnetic nanoparticles effectively, various shapes and materials were examined for the magnetic filter. The result of magnetic separation experiments show that the maximum separation and recovery ratio of superparamagnetic nanoparticles are 99.2 %, and 99.07 %, respectively under magnetic field (3 T) and flow rate (600 litter/hr).

메밀경작에 의한 논토양 내 아연존재형태 및 탈수소효소-활성도 변화

  • Changes of zinc speciation and dehydrogenase activity in soil were studied before and after a field scale phytoextraction by Fagopyrum esculentum at a paddy soil near a closed zinc mine. The concentrations of zinc in paddy soil, in which Fagopyrum esculentum was planted, ranged from approximately 600 mg/Kg(high Zn soil) to 300 mg/Kg(moderate Zn soil). Despite of severe growth inhibition by Zn at the high Zn region, Fagopyrum esculentum accumulated phytoavailable fraction of Zn absorbed from the soil, and enhanced soil dehydrogenase activity (DHA) that had been inhibited by Zn toxicity. After the plant cultivation of 2 months, the concentrations of phytoavailable Zn in the rhizosphere soil at high and moderate Zn region has decreased 25% and 75%, respectively. The amount of Zn reduction in the rhizosphere soil corresponds to that accumulated in plants (recovery 92$\sim$107%), which implies Fagopyrum esculentum removed Zn from the soil. DHA was inversely correlated to the total Zn concentrations in soil. Before plant cultivation, the DHA in the high Zn soil was twice lower than that in the moderate Zn soil. More than 35% of DHA increase was observed in both soils after the application of phytoextraction with Fagopyrum esculentum.

Changes in Heavy Metal Phytoavailability by Application of Immobilizing Agents and Soil Cover in the Upland Soil Nearby Abandoned Mining Area and Subsequent Metal Uptake by Red Pepper (광산 인근 밭토양에서 중금속 안정화제 처리 및 복토층 처리에 의한 토양 중 중금속 식물유효도 변화 및 고추의 중금속 흡수)

  • At the current situation of continuous utilization of heavy metal contaminated upland for agricultural purpose in Korea, minimizing transport of heavy metals from soil to crops is important for securing safety of human health. The present study (in field scale) examined the efficiency of several soil amendments (dolomite, steel slag, lime, zeolite, and compost) on reduction of phytoavailable heavy metals (Cd, Cu, Pb, and Zn) in soil through comparison with no amendment treatment and clean soil cover treatment. For determination of the phytoavailability, 1 M $NH_4NO_3$ extraction and red pepper cultivation were introduced. Among the amendments, in general, dolomite and steel slag were the most effective in reduction of metal (Cd, Pb, and Zn) phytoavailability resulting in less accumulation of these metals in shoot and fruit of red pepper. However, dolomite and steel slag treatment was not as effective as clean soil cover treatment which showed the least metal accumulation in red pepper fruit. Nevertheless, with taking into account the cost, treatment of dolomite or steel slag can be competitive method because the current study showed that dolomite or steel slag treatment reduced accumulated heavy metal concentration effectively in both shoot and fruit of red pepper compared to those from control soil and the concentration in fruit was within the standard value (<0.2 mg $kg^{-1}$ for both Cd and Pb).

  • This study investigated the mobility of inorganic elements (As, Cd, Pb, and Zn) that existed as acid extractable and reducible forms in flooded soils with a pot experiment involving rice cultivation. In general, it is known that soil inorganic elements that existed as an acid extractable form which includes exchangeable, carbonates, non-specifically sorbed, and specifically sorbed have mobility. However, the result of the experiment revealed that each inorganic elements of rice roots grown from flooded soils had different characteristics. The concentrations of Arsenic existed as both forms and the concentrations of cadmium and lead existed as a reducible form in the soils showed a high causal relationship with the concentrations of those elements in the roots of rice plants. The concentrations of zinc, an essential plant element, didn't show a causal relationship. Therefore it is necessary to consider the soil's environmental characteristics such as drained/flooded condition, oxidation/reduction condition, etc. for the mobility assessment of inorganic elements. The concentrations of the reducible form of arsenic, cadmium, and lead in flooded environment such as a paddy field should be also considered because the mobility of these elements combined with Fe/Mn increases in the reduction condition.

Use of Discriminant Analysis to Identify Soil Quality Variation by Land Use (판별분석을 이용한 토지이용별 토양 특성 변화 연구)

  • The physical and chemical characteristics of soils in a small watershed were investigated and the effect of geology and land use on soil quality were examined by using multivariate statistical methods, principal components analysis and discriminant analysis. The soil developed from andesite had finer texture and higher contents of water extractable inorganic components, clay, and mafic minerals than the soil developed from granite. It is considered that the accumulation of salts in the farmland soils indicated by electrical conductivity, contents of cations and anions and pH was caused by fertilizer input during cultivation. The low contents of organic matter in the farmland soils was due to the enhanced oxidation of organic matter by tillage and by the harvest of crops. The contents of inorganic components are increased as following order: upland > orchard > paddy field > forest. The high contents of water soluble $SO_4\;^{2-}$ of paddy soils is due to the oxidation of sulfides mineral formed during the flooding period during the air-dry and extraction. The results of principal components analysis show the difference of soil quality was controlled by geology and land use. PCI indicate the input of fertilizer, mineral weathering and ion exchange reaction by application of nitrogenous fertilizers. The results of two discriminant analyses using water extractable inorganic components and their ratios by land use were also clearly classified by discriminant function 1 and 2. In discriminant analysis by components, discriminant function 1 indicated the effect of fertilizer application and increased as following order: upland > orchard > paddy field > forest soil. The investigated and predicted data for land use from discriminant analysis showed similar results. The discriminant analysis can be used as a useful method certifying the change of land use.

A Criteria on Nitrate Concentration in Soil Solution and Leaf Petiole Juice for Fertigation of Cucumber (Cucumis sativus L.) under Greenhouse Cultivation (시설 오이의 관비재배를 위한 토양용액과 엽병즙액중 질산태 농도 기준 설정)

  • To develope a technique for efficiently managing fertilizer for cucumber, a quick test method to quantify nitrate content in soil solution and leaf petiole juice using a simple instrument was investigated. Among the nitrate analyzing instruments such as compact ion meter, nitrate ion meter, and test strip with reflectometer, the paper test-strip used in conjunction with a hand-held reflectometer was most closely correlated with ion chromatography method in nitrate content, and then it would be suggested with a tool that a farmer can use rapidly, conveniently and accurately for nitrate analysis in a field. Nitrate content in soil solution collected by porous cup was very variable on the lapsed time after drip irrigation and the sampling positions such as soil depth and the distance from dripper. As a result, a significant correlation between nitrate contents of soil solutions and 2M KCl soil extract was not found. However, nitrate content in soil solution extracted with a volume basis (soil:water=1:2) showed the highly significant correlation with that in 2M KCl extract. Nitrate contents of cucumber leaf petiole juices was greatly different between upper and lower leaves. Eleven to sixteen positioned-leaf would be a proper sampling position to determine nitrate content in leaf petiole for evaluating nutrient state by plant tissue analysis. From the secondary regression equations between nitrate contents of soil and petiole juice and the yield of cucumber, nitrate levels for real time diagnosis were estimated as $400mg\;l^{-1}$ soil solution by porous cup. $300mg\;l^{-1}$ in a soil volume extraction, and $1400mg\;l^{-1}$ in petiole juice from spring to summer season. In addition, the maximum yield of cucumber fruit in pot test was obtained in nitrate $1500mg\;l^{-1}$ level of petiole juice, which was similar to nitrate $1400mg\;l^{-1}$ in greenhouse trial.
