Although flotation techniques are often used for the removal of algal particles, the practicality of algae-harvesting technologies is limited owing to the complex and expensive facilities and equipment required for chemical coagulation. Here, we examined the feasibility of an approach to separating algal particles from water bodies without the need for chemical coagulants, depending on the condition of the algae, and to determine the optimal conditions. Using Anabaena sp., a cyanobacterium causes algal blooms in lakes, we stimulated auto-flocculation in algal particles without coagulants and conducted solid-liquid separation experiments of algal particles under various conditions. The six cultivation columns included in our analysis comprised four factors: Water temperature, light intensity, nutrients, and carbon source; auto-flocculation was induced under all treatments, with the exception of the treatment involving no limits to all factors, and algal particles were well-settled under all conditions for which auto-flocculation occurred. Meanwhile, flotation removal of auto-flocculated algal particles was attained only when nutrients were blocked after algae were grown in an optimal medium. However, no significant differences were detected between the functional groups of the extracellular polymeric substances (EPSs) of floated and settled algal particles in the FT-IR peak, which can cause attachment by collision with micro-bubbles.
Optimal cultivation conditions for the production of phenylalanine ammonia-lyase(PAL) from Rhodotorula aurantiaca K-505 were selected, and the kinetic parameters of the produced PAL were determined. The most suitable carbon and nitrogen sources were glucose and tryptone, respectively. The strain expressed PAL constituttively when using the optimized semi-complex media. High cell density culture could be critical for maximal production of PAl since the PAL ynthesis was growth associated. maximum PAL activity was observed at initial pH 6.0. although the ll growth was not markedly affected by temperature between 22 and 28$^{\circ}C$, the cells yielded the maximum PAL activity when cultivated at 22$^{\circ}C$. The maximum activity for deamination of L-phenylalnine to trans-cinnamic acid was observed around pH 8.8. The PAL activity gave the maximum at 45$^{\circ}C$, and greatly decreased at higher than 5$0^{\circ}C$. Activation energy({TEX}$E_{a}${/TEX}) calculated from Arrhenius equation was 6.28 kcal/mol in the range of 22$^{\circ}C$ to 4$0^{\circ}C$. A oolf plot showed that the enzyme reaction follows Michaelis-Menten equation, whose {TEX}$K_{M}${/TEX} and {TEX}$V_{max}${/TEX} values were 4.65$\times${TEX}$10^{-3}${/TEX} M and 0.89$\mu$ mol/mg-min respectively.
In order to explain the connection between ginseng rust spot and soil ecological conditions, the bed soils and ginseng roots were sampled at different microrelief units and the reducing substances of the bed soils and iron forms of the ginseng root epi dermises were determined. The results showed that the occurrence of the rust spot was connected with the ecological conditions of the soils and the metabolism of the plant which was caused by the excessive $Fe^{2+}$ in the soil solution. Ginseng rust spot was the enrichment of iron which was mainly composed of organic complex irons. Including active ferrous active ferric and non active ferric forms and they were transformed into each other following the change of soil moisture and temperature regimes. According to the regularity of growth and decline of reducing substances in soil and rust index of ginseng roots as well as the difference of adaptability to excessive $Fe^{2+}$ in soil among different year-old seeding, a new comprehensive measure based on the connection of ameliorating soil and improving cultivation system was recommended to prevent the occurrence of ginseng rust spot.
The gastrointestinal (GI) tract, including the rumen and the other intestinal segments of cattle, harbors a diverse, complex, and dynamic microbiome that drives feed digestion and fermentation in cattle, determining feed efficiency and output of pollutants. This microbiome also plays an important role in affecting host health. Research has been conducted for more than a century to understand the microbiome and its relationship to feed efficiency and host health. The traditional cultivation-based research elucidated some of the major metabolism, but studies using molecular biology techniques conducted from late 1980's to the late early 2000's greatly expanded our view of the diversity of the rumen and intestinal microbiome of cattle. Recently, metagenomics has been the primary technology to characterize the GI microbiome and its relationship with host nutrition and health. This review addresses the main methods/techniques in current use, the knowledge gained, and some of the challenges that remain. Most of the primers used in quantitative real-time polymerase chain reaction quantification and diversity analysis using metagenomics of ruminal bacteria, archaea, fungi, and protozoa were also compiled.
In order to know the residual pattern of pesticides and predict to the degradation period until below MRL we experimented procymidone and chlorothalonil for grape which were the most detected pesticide in grape by NAQS(National Agricultural product Quality management Service) survey. In this experiment we sprayed those pesticides 10 days before harvest and analyzed 0, 1, 2, 3, 5, 7, 10 day sample to establish logical equation and to calculate $DT_{50}$. Also the same day samples stored at $4^{\circ}C$ and $20^{\circ}C$, which were compared their degradation patterns. During the cultivating period, the residue amount of procymidone was changed from 1.85 mg/kg (0 day) to 0.33 mg/kg (10 day), $DT_{50}$ was 3.5 days, and chlorothalonil was changed from 5.5 mg/kg (0 day) to 3.49 mg/kg (10 day), $DT_{50}$ was 4.4 days. During the storage period, $DT_{50}$ of procymidone and chlorothalonil at $4^{\circ}C$ were 10.5 and 7.6 days, and 6.3 and 6.1 days at $20^{\circ}C$, respectively.

The soil ecosystem is a complex system performing particularly complicated and varied functions, such as providing a habitat for organisms, acting as a medium for plant cultivation and growth, and functioning as a buffer against external materials in the environment. To assess whether these important functions of the soil ecosystem are executed appropriately, the concept of soil ecosystem health has been introduced, which is defined as the ability to perform the specific functions of the soil ecosystem. Understanding soil properties and soil indicators related to soil functions is essential to assess the soil health. In this study, systems, the indicators, and evaluation factors for assessing soil ecosystem health employed in a number of countries were investigated and discussed. In particular, it is necessary to introduce a technique for the evaluation of soil ecosystem health in Korea and to develop techniques and indicators appropriate to the soil ecosystem and status in Korea.
Herbal medicines have been used worldwide, particularly in the Far East countries, for thousands of years. Herbal medicines comprise complex mixtures of different phytochemical substances, and their nature may vary substantially depending on many factors related to the cultivation condition, area and harvest time. The processing of the herbal medicines significantly affects their pharmacological and toxicological properties. In this paper, we discuss current research that deals with the techniques for processing Chinese herbal medicines. These results will provide a scientific basis for developing new techniques for the processing of herbal medicines.
To investigate the occurrence and species diversity of mycobacteria in waters, surface water samples were collected monthly from the Han River and tap water samples at the terminal sites of the distribution system. Mycobacteria in each water sample were isolated by decontamination using cetylpyridinium chloride (CPC) and cultivation on Middlebrook 7H10 agar, and then identified by polymerase chain reaction-restriction fragment length polymorphism analysis (PRA) and sequencing of the 65-kDa heat-shock protein gene (hsp65 gene). Mycobacteria were detected in 59% of the surface water samples and 26% of the tap water samples. Over half of the 158 isolates could not be identified by hsp65 PRA and gene sequencing, and several identification discrepancies were observed between the two methods. The most frequently isolated species was Mycobacterium gordonae in surface water and M. lentiflavum in tap water. M. avium complex (MAC), the most important pathogen among environmental mycobacteria, was detected in the surface water samples but not found in the tap water samples. The result demonstrated that water is an important environmental source of mycobacteria and the combined application of hsp65 PRA and sequencing was more reliable than hsp65 PRA alone to accurately identify mycobacteria present in water.
Strawberries are a popular economic crop, and one of the major plantations and exporting countries is Korea in the world. The Fusarium oxysporum species complex (FOSC) is a soil-borne pathogen with genetic diversity, resulting in wilt disease in various crops. In Korea, strawberries wilt disease was first reported in the 1980s due to the infection of FOSC, causing significant economic damage every year. The causal agent, F. oxysporum f. sp. fragariae, is a soil-borne pathogen with a characteristic of FOSC that is difficult to control chemically and mutates easily. This study obtained genetic polymorphism information that was based on AFLP, of F. oxysporum f. sp. fragariae 91 strains, which were isolated from strawberry cultivation sites in Gyeongsangnam-do and Chungcheongnam-do, and compared strains information, which was the isolated location, host variety, response to chemical fungicide, and antagonistic bacteria, and mycelium phenotype. As a result, AFLP phylogeny found that two groups were mainly present, and group B was present at a high frequency in Gyeongsangnam-do. Group B proved less sensitive to tebuconazole than group A through Student's t-test. In addition, the fractions pattern of AFLP was calculated by comparing the strain information using PCA and PERMANOVA, and the main criteria were separated localization and strawberry varieties (PERMANOVA; p< 0.05). And tebuconazole was different with weak confidence (PERMANOVA; p< 0.10). This study suggests that the F. oxysporum f. sp. fragariae should be continuously monitored and managed, including group B, which is less chemically effective.

In particular, in order to solve problems such as increasing productivity and production of agricultural products, improving quality, and saving energy, it is difficult to solve them with traditional farming methods. Recently, with the wind of the 4th industrial revolution, ICT convergence technology and artificial intelligence have been rapidly developing in many fields, but it is also true that the application of new technologies is somewhat delayed due to the unique characteristics of agriculture. However, in recent years, as ICT and artificial intelligence utilization technologies have been developed and applied by many researchers, a revolution is also taking place in agriculture. This paper summarizes the current state of research so far in four categories of agriculture, namely crop cultivation environment management, soil management, pest management, and irrigation management, and smart farm research data that has recently been actively developed around the world.
