새송이버섯 재배사의 환경인자 계측(III) -영구재배사를 중심으로- (Analysis of Environmental Factors in Pleurotus eryngii Cultivation House(III) -Based on Panel Type Structure-)

  • Pleurotus eryngii(King oyster) is one of the most promising mushrooms being produced on the domestic farms. The quality as well as quantity of Eryngii is sensitively affected by micro climate factors. To safely produce high-quality Eryngii all the year round, it is required that the environmental factors be carefully controlled by well designed structures equipped with various facilities and control systems. This study was carried out at the commercial mushroom cultivation houses to find out reasonable range of each environmental factor and yield together with economic and safe structures influencing on the optimal productivity of Eryngii. This experiment was conducted from Nov. 10, 2004 to Aug. 27, 2005 in Eryngii. cultivation houses. The environmental factors measured for this study were inside/outside temperature, relative humidity and $CO_2$ concentration in Pleurotus eryngii medium. In addition, the yield and quality of mushroom were made investigation. But the optimal productivity will be evaluated by considering the quality and quantity of mushroom production, energy requirements, facility construction and management cost, etc.

  • Recently, the demand for smart farms is increasing due to the increase in the cultivation area such as horticulture, fruit trees and special crops. However, due to the irregular weather changes and the cultivation method of the crops due to the different cultivation environment, there are frequent occurrence of diseases and insect pests and infectious diseases due to system error or carelessness, and the cycle is also very short. In addition, the Smart Farm business has been built by combining various sensors (temperature, humidity, CO2, illumination) and LED lighting, but it is costly in terms of frequent errors, lack of power supply, And thus the management can not be efficiently managed. Therefore, this paper combines real time sensing technology based on IoT Platform and high performance control technology to control pests and equipment errors and monitor the growth status of crops in real time based on big data analysis and Artificial Intelligence System.

옻나무 노지재배 투자 수익성 분석 (ROI(Return on Investment) Analysis of Open-field Cultivation of Rhus verniciflura)

  • 본 연구의 목적은 옻나무를 재배하고자 하는 사람들이나 재배면적을 늘이고자 하는 사람들에게 옻나무의 경영실태 및 소득과 투자 수익성을 분석하여 옻나무 재배 투자에 대한 정보를 제공하고자 하는 것이다. 수익성 분석 기법은 IRR, B/C Ratio, NPV를 이용하였다. 옻나무 주산단지인 강원 원주, 충북 옥천, 경남 함양지역의 51가구의 옻나무 재배농가를 대상으로 2011년 5월부터 11월까지 작업공정별 투입노동 및 자재비 등에 관한 면접방문조사를 실시하였다. 그 결과 현지판매가격과 투자수익성의 판단기준이 되는 가격을 기준으로 IRR, B/C Ratio, NPV, 조수익, 소득, 순수익, 소득율 등을 구하였다. 할인율 3%를 적용하였다. 옻나무와 옻칠의 최근 5년간 옻나무와 옻칠의 각각의 농가평균판매가격인 20,000원/주, 2,200,000원/관을 적용한 결과 IRR 27.76%, B/C Ratio가 2.5가 되어 투자수익성이 높은 것으로 나타났다.

남조류의 대량배양 및 남조류 독소의 스트립분석법 연구 (A study on the massive cultivation of cyanobacteria and strip analysis of cyanobacterial toxin)

  • 남조류 독소인 마이크로시스틴은 여름철 우리나라 여러 호수들에 존재하여 물고기와 가축 그리고 인간에게 강한 독성을 나타내는 독소이다. 지금까지는 남조류독소인 마이크로시스틴을 분석하기 위하여 HPLC를 사용하거나 ELISA 분석법을 이용하였으나 본 연구에서는 대량배양을 통해서 얻어진 남조류들 속에 미량존재하는 마이크로시스틴을 분석하기 위해 스트립을 이용하는 새로운 분석방법을 시도하였다. 이 분석법은 항원-항체기술을 이용하여 높은 특이도(specifisity)와 테스트 스트립(test strip)의 신속성, 그리고, 형광리더(reader)의 고감도(sensitivity) 분석능력 등의 장점을 이용하였다. 따라서 새로 개발된 분석법을 이용하면 다양한 물시료속에 존재하는 마이크로시스틴을 독소 표준품 없이 단시간에 고감도로 분석할 수 있다.

논 재배 시 한국 콩 품종의 수량성 관련요인분석에 의한 품종군 분류 (Cluster Analysis of Korean Domestic Soybeans based on Yields Characters in Paddly Field Cultivation)

  • In order to select the suitable soybean cultivars for paddly field cultivation, growth response and yields characters were investigated with 37 Korean domestic soybean cultivars, in Yeoncheon, Gyeonggi province. The results can be summarized as follows ; When the soybeans were cultivated in paddy field, significant differences were observed in growth responses at each growth stage and yields among the cultivars. Yields of six cultivars, such as Danbaekkong, Paldokong, Pungsannanulkong, Myeongjunamulkong, Saeolkong, and Namhaekong, were less than 200kg/10a, however, seven cultivars, such as Shinpaldalkong 2, Sohokong, Dormikong, keumkangkong, Bukangkong, Dajangkong, and Geomjeongkong 2, showed highest yields ranged from 301~385kg/10a. At V5 stage, highly positive correlation(r=091$^{{\ast}{\ast}}$) was observed between the top dry weight and leaf area while negative correlation was existed between T/R ratio and root dry weight. However, at R2 age, leaf area was correlated with both top and root dry weight while T/R ratio was correlated with leaf area and top dry weight. Thirty-seven soybean cultivars used in this experiments were classified into 3 different clusters based on 19 important agronomic characters, and genotypic properties of the cultivars were similar in each cluster. In results, high yields could be achieved by selected cultivation of suitable soybean cultivars such as Shinpaldalkong 2 which showed early maturation date and highly ripening ratio.

Effect of Herbicide Combinations on Bt-Maize Rhizobacterial Diversity

  • Reports of herbicide resistance events are proliferating worldwide, leading to new cultivation strategies using combinations of pre-emergence and post-emergence herbicides. We analyzed the impact during a one-year cultivation cycle of several herbicide combinations on the rhizobacterial community of glyphosate-tolerant Bt-maize and compared them to those of the untreated or glyphosate-treated soils. Samples were analyzed using pyrosequencing of the V6 hypervariable region of the 16S rRNA gene. The sequences obtained were subjected to taxonomic, taxonomy-independent, and phylogeny-based diversity studies, followed by a statistical analysis using principal components analysis and hierarchical clustering with jackknife statistical validation. The resilience of the microbial communities was analyzed by comparing their relative composition at the end of the cultivation cycle. The bacterial communites from soil subjected to a combined treatment with mesotrione plus s-metolachlor followed by glyphosate were not statistically different from those treated with glyphosate or the untreated ones. The use of acetochlor plus terbuthylazine followed by glyphosate, and the use of aclonifen plus isoxaflutole followed by mesotrione clearly affected the resilience of their corresponding bacterial communities. The treatment with pethoxamid followed by glyphosate resulted in an intermediate effect. The use of glyphosate alone seems to be the less aggressive one for bacterial communities. Should a combined treatment be needed, the combination of mesotrione and s-metolachlor shows the next best final resilience. Our results show the relevance of comparative rhizobacterial community studies when novel combined herbicide treatments are deemed necessary to control weed growth.

Quantitative Analysis of Dammarane-type Ginsenosides in Different Ginseng Products

  • Ginseng products available in different forms and preparations are reported to have varied bioactivities and chemical compositions. In our previous study, four new dammarane-type ginsenosides were isolated from Panax ginseng, which are ginsenoside Rg18 (1), 6-acetyl ginsenoside Rg3 (2), ginsenoside Rs11 (3), and ginsenoside Re7 (4). Accordingly, the goal of this study was to determine the distribution and content of these newly characterized ginsenosides in different ginseng products. The content of compounds 1 - 4 in different ginseng products was determined via HPLC-UV. The samples included ginseng roots from different ginseng species, roots harvested from different localities in Korea, and samples harvested at different cultivation ages and processed under different manufacturing methods. The four ginsenosides were present at varying concentrations in the different ginseng samples examined. The variations in their content could be attributed to species variation, and differences in cultivation conditions and manufacturing methods. The total concentration of compounds 1 - 4 were highest in ginseng obtained from Geumsan ($185{\mu}g/g$), white-6 yr ginseng ($150{\mu}g/g$), and P. quinquefolius ($186{\mu}g/g$). The results of this study provide a basis for the optimization of cultivation conditions and manufacturing methods to maximize the yield of the four new ginsenosides in ginseng.

Analysis and Monitoring of Environmental Parameters in a Single-span Greenhouse during Strawberry Cultivation

  • In this study, strawberry cultivation environment in a greenhouse located in Jeonju was monitored and internal environmental parameters were analyzed. Temperature, humidity, RAD, and PPF sensors were installed to monitor environmental conditions in the test greenhouse. Data were collected every 10 minutes during four winter months from sensors placed across the greenhouse to assess its permeability and environmental uniformity. Temperature and humidity inside the greenhouse were relatively uniform with negligible deviations among the center, south, and north; however, it was judged that further analysis of gradients of these parameters from the east to the west of the greenhouse would be needed. Both RAD (Total solar radiation) and PPF (Photosynthetic photon flux) had high values on the south and were low on the north and the reduction rate of these parameters was 54% and 61%, respectively, indicating that a significant amount of light could not be transmitted. This implied a significant decrease in the amount of light entering the greenhouse during winter. Therefore, it is concluded that environmental control devices and auxiliary lighting are needed to achieve uniform greenhouse environment for efficient strawberry cultivation.

Studies on the Analysis of Dammarane Aglycones of Korean Ginseng

  • To establish a convenient quantitative method for dammarane glycosides in Korea ginseng, the ginseng roots harvested at the cultivation areas of Ga Pyeong, Geum San and Jeung Pyeong were dried, powered, extracted with methanol and hydrolyzed. The ginseng root obtained at Gang Hwa was divided into three parts: main root, laterial root and cortex, and then these were treated in the same manner as the above. The various hydrolysates thus obtained were subjected to the analysis by an instrument coupled with flame ionization detector. The results showed that panaxadiol contents in the ginsengs of the three different cultivation sites were similar. However, the lateral root of Gang Hwa ginseng was found to contain the largest amount of panaxadiol among the three parts of ginseng. This method of the analysis for panaxadiol in ginseng was found to be one with relative rapidity and ease.

An Overview of Different Techniques on the Microbial Community Structure, and Functional Diversity of Plant Growth Promoting Bacteria

  • Soil is a dynamic biological system, in which it is difficult to determine the composition of microbial communities. Knowledge of microbial diversity and function in soils are limited because of the taxonomic and methodological limitations associated with studying the organisms. In this review, approaches to measure microbial diversity in soil were discussed. Research on soil microbes can be categorized as structural diversity, functional diversity and genetic diversity studies, and these include cultivation based and cultivation independent methods. Cultivation independent technique to evaluate soil structural diversity include different techniques such as Phospholipid Fatty Acids (PLFA) and Fatty Acid Methyl Ester (FAME) analysis. Carbon source utilization pattern of soil microorganisms by Community Level Physiological Profiling (CLPP), catabolic responses by Substrate Induced Respiration technique (SIR) and soil microbial enzyme activities are discussed. Genetic diversity of soil microorganisms using molecular techniques such as 16S rDNA analysis Denaturing Gradient Gel Electrophoresis (DGGE) / Temperature Gradient Gel Electrophoresis (TGGE), Terminal Restriction Fragment Length Polymorphism (T-RFLP), Single Strand Conformation Polymorphism (SSCP), Restriction Fragment Length Polymorphism (RFLP) / Amplified Ribosomal DNA Restriction Analysis (ARDRA) and Ribosomal Intergenic Spacer Analysis (RISA) are also discussed. The chapter ends with a final conclusion on the advantages and disadvantages of different techniques and advances in molecular techniques to study the soil microbial diversity.
