  • In this study, viral disease samples were obtained between 2006 and 2010 from pepper fields in 11 major pepper-growing districts in Gangwon-do, and in 83 areas from other provinces, with the exception of Gyeongsangnam-do and Jeju island in Korea. In order to assess the type of infection, field surveys were conducted with regard to viral disease severity and virus type, based on typical symptoms on leaves. The means of single and mixed-virus infections were 46.6% and 48.0%, respectively, during those periods, suggesting that viruses are the agents that most severely decrease pepper production in field cultivation in Korea. In terms of single infection, Cucumber mosaic virus (CMV) was the most prevalent virus based on its disease severity ratings (34.8%). Next, Pepper mild mottle virus (PMMoV) and Pepper mottle virus (PepMoV) were shown to cause severe viral diseases in pepper, with disease severities of around 5-10%. On the other hand, Tomato spotted wilt virus (TSWV) occurs in a limited area in Chungcheongnam-do and Jeollanam-do. Thus, the viral disease caused by CMV, PMMoV, and PepMoV in pepper can be severe, and these virus types should remain considered critical reasons for decreased pepper production in field cultivation in Korea. In addition to single infection, mixed infections are frequently observed in collected pepper samples from all areas. The ratios of mixed infection were therefore studied to evaluate the disease severity of mixed infections and to define individual virus types. These data showed that different types of viruses were present, and CMV was the most abundant virus for mixed infection, as in the case of single infection. Among mixed infections, the highest disease severity was seen with CMV+Broad beam wilt virus 2 (BBWV2), followed by other types of mixed infection such as CMV+PepMoV and CMV+PMMoV. However, further work is needed to reduce the severe damage caused by viruses and to assess mixed infection types involving three or more viruses.

  • In the greenhouse fields for fruits and vegetables during the winter in Korea, there are serious damages by the sclerotium diseases due to the low temperature and humidity. This study was carried out to select an antagonic agent for the biological control of the sclerotium diseases. The 55 antagonic agents were selected from the rhizosphere in soil where the fruits and vegetables were cultivated in the green house fields, and strain AB02 among the tested isolates was estimated to be the strongest antagonist against the sclerotium disease. Using strain AB02, the antifungal spectrum was tested against 5 different plant pathogens. According to the results of the test, strain AB02 . showed the high antagonistic effect against Botrytis cinerea and Sclerotinia sclerotiorum. For the experiment of biological control against the sclerotium disease, it was estimated the suppression effect and the control effect by the strain in the pot experiment using the green perilla. According to the result of the pot experiments, the suppression effect was 40% and the control effect was 62%, respectively. For the stimulation effect of the tested plant growth by strain AB02 compared to the control, it was improved as 120% for the total length, 141% for the liveweight, 121% for the total number of leaves, 185% for the leaf area, and 327% for the liveweight of the root, respectively. Strain AB02 showing the antagonistic effect against the sclerotium disease and the stimulation effect for the plant growth was identified as Bacillus sp.

  • In order to investigate the variations on the seed hardness of Pisum sativum distributed in southern area of Korean peninsula, 10 local strains (Choonseong. Ganghwa, Pocheon, Hadong, Haman, Geumneung, Yeongyang, Boseong, Gochang, Namweon), which located from $34^{\circ}26"N$ to $38^{\circ}11"N$, were selected according to their latitudes and geographical distances. The seeds of these strains were collected and their contents of mannose and galactose were analyzed. Mannose contents in the seeds were variable in the range between the highest 10.351 mg/g (Ganghwa) and the lowest 5.962 mg/g (Yeongyang). The contents of galactose were also represented remarkable differences from 7.050 mg/g (Yeongyang) to 19.314 mg/g (Hadong). The local strains were classified into 3 variation types such as the south central type (Namweon, Yeongyang, Geumneung, Gochang), the central type (Choonseong. Ganghwa, Pocheon) and the southern type (Haman, Hadong) and 1 strange strains (Boseong) according to the geographical climatic type, isopleth of warmth index and the ratio of mannose to galactose, which indicate the hardness of seeds in Leguminosae, ranged from 0.46 to 0.94. The variation types are very significant genecologically as an evidence for microevolution related to natual and artificial selection in cultivated plants.

  • Instead of fertilizing the lands they moved to find other fertile lands and lived a feudal life agriculture and various machincs played a main role in the land reclamation. The best method of land clearing, the time and capacity in the operation and the effect of growing crops should systematically analized prior to the time of 3rd Five-year Economic Development(1972-1976) in order to cultivate 210,000ha of wasteland or the modernization of our country. The present study was investigated to find out a new working-system of mechanical land clearing and development of fertile soil. The results are as follows: 1) The first stage of productive power increases when weeds were mixed soil in the land clearing work in order to supply organic materials and to make soil swollen instead of burning or just removing. 2) The equipments such as bulldozers, harrows, power tillers and so on should be prepared in order to do a systematic work in the land clearing. 3) The work of pulling-up roots is dependent upon the forms of roots spreading under the ground. The work of the pulling-up of the straight roots was most difficult. But before the roots are pulling-up we do not know the forms of roots. 4) The cutting of soil from upside to under side land clearing method (II) Spent 6 hr 7 min 43 sec per 1 Danbo in doing land clearing and control method spent 4 hr 52 min 30 sec per 1 Danbo in doing land cearing. No significant difference could be found between II treatment type and I treatment type. 5) The volume of soil carried by bulldozer $270.38m^3$ per 1 Danbo in I type and in II type, $368.58m^3$ per 1 Danbo or about 36% increased in II type then in I type. But that is not a significant difference when considering the whole productive power of soil. 6) The land clearing of terrace by bulldozers cannot escape making the embankment slope of about $45^{\circ}$, and 25.3% should be decreased in the area. It is recommended to make use of the embankment slope by planting grass for cows. 7) The time of retary tilling increased 2.3 times or 1 hr 42 min 22 sec per 1 Danbo compared to the timesrequired in the harrowing. Because it mixed the Organic matter on ground and the harrowing of subsoil. 8) The havest of pasture growing on the land reclamation in natural slope is as follows. (a) The none fertilizers with reclaimed block of productivity decresed 1/10 than that of the farmland. Therefore the none fer tilizing in the cultivated pasture is unprofitable. (b) The havest of the manured IV treatment block was the best not only in the kinds of fertilizers but also in the kinds of pasture grass.

  • Mungbean should be harvested several times according to its physiological characteristics and weather conditions of cultivation region. In Korea, mungbean is usually sown in June and harvested three or four times, and the cultivated area is being rapidly reduced. Therefore, the author developed cultivation techniques of mechanical harvesting suitable for the weather conditions of the southern part of the Korean peninsula. The optimum sowing time of mungbean for mechanical harvesting in southern part of Korea is around July 20. When sown around July 15, mungbean should be harvested twice and then the mechanical harvesting of mungbean was not possible. Meanwhile, when sown after July 25, the mechanical harvesting was possible but the maturing period was longer and the seed yield was decreased. Therefore, it is safe to say that in Korea the mechanical harvesting of mungbean is possible for the middle part of Korea when the plant is sown before July 20 and for the southern coastal region of Korea when sown after July 20 (if July 20 is set up as the baseline for the southern part of Korea). Out of Keumseong and Owool, which are popularized cultivars in Korea most, Owool is determined to be most appropriate for mechanical harvesting. Owool is favorable for mechanical harvesting because, when compared to Keumseong, it is higher both in plant height and in pod height, and also the seed yield is better.

  • The great economic losses caused by Cucumber mosaic virus (CMV) infection of peppers has led to the development of genetically modified (GM) CMV-resistant peppers. We developed virus-resistant pepper plants using Agrobacterium tumefaciens -mediated transformation. The expressed recombinant protein was purified using nickel-nitrilotriacetic acid resin and immunoaffinity chromatography, and purity was assessed by sodium dodecyl sulfate polyacrylamide gel electrophoresis. Immunoblot analysis revealed the purified CMV coat protein (CMV-CP) had a molecular mass of 25 kDa. After in-gel digestion and desalting, the internal peptide fragments of CMV-CP were sequenced by matrix-assisted laser desorption/ionization-time of flight. Most GM pepper and Escherichia coli BL21 internal peptides had identical peptide sequences and contained 137 of 183 whole peptides in CMV-CP. A quantitative enzyme-linked immunosorbent assay was performed to detect CMV-resistant GM peppers. We also provide basic information about the expressed protein in GM peppers for further safety assessment. The contents of soluble protein and CMV-CP were measured in GM and control peppers cultivated in three different areas of Korea. Statistical significance in terms of cultivation areas, harvest times, generations, and plant tissue origin were determined based on a P value of 0.05. The highest amount of CMV-CP was detected at the seedling stage from plant grown in each region. T3 and T5 showed significantly different levels of CMV-CP from T4 in leaves in the whorl stage. No statistical differences were observed among GM peppers at different stages of maturity in any cultivation area. The results from this study contribute to the safety evaluation of newly designed CMV-resistant GM peppers and provide a standard against which to compare other virus-resistant GM peppers.

  • Yield of potato is largely influenced by drought stress. This study was conducted in Gangneung and Cheongju during the spring cropping of potato. Potatoes in the Gangneung area were affected by drought but there was no damage due to drought in Cheongju. During the early-growth stage, the contents of inorganic components like available phosphate and growth characteristics of the potato leaf in Cheongju were significantly higher than those in Gangneung but there was no difference after the flowering stage. It was considered that the potato plants cultivated in Cheongju could vigorously grow than that of Gangneung under drought stress. In addition, the content of calcium (Ca), which is a secondary messenger related to aging, was found to be higher in potato plants grown in Cheongju than in Gangneung and accumulated more quickly in potato plants of Cheongju. Because magnesium (Mg) was also found to be higher in potato plants from Gangneung by a wide margin, this phenomenon was thought be related with drought stress. The amounts of all inorganic components absorbed from soil were higher in Cheongju than in Gangneung, showing a relatively higher plant biomass in Cheongju. Correlations of development indexes related to leaf showed less or no relation in Gangneung. According to yield characteristics of the harvest stage, although yield was greatly reduced under drought stress condition, the rate of commercial yield was not significantly affected under the drought stress condition. Consequently, it was considered that these responses to drought stress could be utilized to stabilize potato production under the stressful conditions associated with abnormal climate.

  • Luffa cylindrica Roem (sponge gourd) belongs to the Cucurbitaceae family and has been cultivated as an ornamental plant in Korea. Recently, its cultivation area has been increased with an increase in demand for it as an ingredient in cosmetics, herbal medicines, and health supplements. We analyzed inorganic components of sap collected from land race sponge gourds. We also measured antioxidant enzyme activities and antimicrobial activities of the plant, seed, and sap to examine functional properties of sponge gourd. The sap of the sponge gourd contained high levels of K, Ca, P, and Mg, with the most abundant mineral in the sap being K ($470mg{\cdot}L^{-1}$). The amounts of Ca and Mg were 2 and 1.7 times more than those found in cucumber (Cucumis sativus), respectively. Ascorbate peroxidase was more active than catalase and superoxide dismutase in various plant parts of sponge gourd. Antioxidant activities were much lower in stems than in other plant parts such as leaves, roots, flowers, fruits, seeds, and sap. In addition, sap showed a very low level of antimicrobial activity against two food-borne pathogens, Vibrio parahaemdyticus and Propionibacterium acne, and none against the other eight tested food-borne pathogens. Antimicrobial activities against Candida albicans and Malassezia furfur, which causes dermatitis, appeared to be higher in sap than in other parts of sponge gourd plants. Overall, the antimicrobial activity against Malassezia furfur appeared to be higher than against Candida albicans.

  • This study was conducted to provide a basis for raising farm income by increasing the yield and extending the cultivation period by creating an environment where crops can be cultivated normally during high temperatures in summer. The maximum cooling load of the multi-span greenhouse with a floor area of 504 ㎡ was found to be 462,609 W, and keeping the greenhouse under 32℃ without shading the greenhouse at a high temperature, it was necessary to fog spray 471.6 L of water per hour. The automatic fog cooling control device was developed to effectively control the fog device, the flow fan, and the light blocking device constituting the fog cooling system. The fog cooling system showed that the temperature of the greenhouse could be lowered by 6℃ than the outside temperature. The relative humidity of the fog-cooled greenhouse was 40-80% during the day, about 20% higher than that of the control greenhouse, and this increase in relative humidity contributed to the growth of cucumbers. The relative humidity of the fog cooling greenhouse during the day was 40-80%, which was about 20% higher than that of the control greenhouse, and this increase in relative humidity contributed to the growth of cucumbers. The yield of cucumbers in the fog-cooled greenhouse was 1.8 times higher in the single-span greenhouse and two times higher in the multi-span greenhouse compared to the control greenhouse.
