Healing of mandibular through-and-through osseous defects by Guided Tissue Regeneration in ferrets

  • 본 연구의 목적은 관통된 뼈의 결손부위를 각기 다른 membrane(Gore-tex, Vicryl, Guidor)을 사용하여 보다 나은 골 재생을 얻을 수 있는 가를 평가하는 데 있다. 여섯 마리 흰 족제비의 12개 하악 소구치를 근관치료한 후, 치근단 절제술을 실시하여 $3mm\times5mm$ 크기의 관통된 뼈의 결손을 소구치의 근첨부에 형성하였다. 전부 12개의 결손부위가 형성되었고, 이를 3개의 군으로 나누었다. 대조군으로 결손부위를 membrane barrier없이 점막골막피판으로 덮었다. 다른 각 군은 결손부위를 각 Gore-Tex, Vicryl, Guidor membrane을 사용하여 설측과 협측 모두 덮었다. 각 군을 치유기간 6주와 12주로 두 아군으로 분리하였다. 방사선학적 소견으로 6주군에서 대조군은 $65\%$, Gore-Tex군은 $90\%$, Vicryl군은 $95\%$, Guidor군은 $65\%$의 결손부위의 치유를 보였다. 12주 군에서 대조군은 $80\%$, Gore-Tex군은 $95\%$, Vicryl군은 $95\%$, Guidor군은 $90\%$의 치유를 보였다. 조직학적 소견으로 대조군에서는 완전한 골 재생이 일어나지 않았으며, 결손부위로 결합조직이 자라들어온 것이 관찰되었다. Gore-Tex 6 주군에서는 대부분 fibrillar bone이 관찰되었고, 12 주군에서는 부분적으로 lamellar bone이 형성되었다. Vicryl군에서는 거의 완전한 골의 재생이 관찰되었다. 6주군에서는 재생된 뼈는 fibrillar bone이고 부분적으로 lamellar bone을 관찰되었고, 12주 군에서는 주로 lamellar bone으로 구성되었다. Guidor군에서는 제한적인 골 재생과 함께 심한 염증이 관찰되었다. 본 연구의 결과에서 조직재생유도술은 일반적으로 골 재생을 증진시킴을 볼 수 있었고, Vicryl membrane이 가장 뛰어난 골 재생유도 가능성을 보였다.

  • In the past, radio astronomers have sought isolation from man-made signals by placing their telescopes in remote locations. These measures may no longer safeguard scientific observations, since NGSO satellite systems, particularly low-Earth orbit (LEO) systems, are usually designed to provide global or wide regional coverage. Further, radio astronomers have historically made their observations in the frequency bands allocated for their use by the member countries of the International Telecommunication Union (ITU). The science of radio astronomy could be adversely impacted by the deployment of large constellations of new non-geostationary orbiting (NGSO) satellites for telecommunications, navigation and Earth observation, and the proliferation of new, high-power broadcasting and telecommunication satellites in geostationary (GSO) orbits. Radio telescopes are extremely sensitive, and, in certain situations, signals from satellites can overwhelm the signals from astronomical sources. This paper describes the problem in detail and identifies ways to mitigate it without adversely affecting the continued vigorous growth of commercial space-based telecommunications.

Redundant Operation of a Parallel AC to DC Converter via a Serial Communication Bus

  • The redundant operation of a parallel AC to DC converter via a serial communication bus is presented. The proposed system consists of three isolated CUK power factor correction modules. The controller for each converter is a dsPIC30F6010 microcontroller while a RS485 communication bus and the clock signal are used for synchronizing the data communication. The control strategy of the redundant operation relies on the communication of information among each of the modules, which communicate via a RS485 serial bus. This information is received from the communication checks of the converter module connected to the system to share the load current. Performance evaluations were conducted through experimentation on a three-module parallel-connected prototype, with a 578W load and a -48V dc output voltage. The proposed system has achieved the following: the current sharing is quite good, both the transient response and the steady state. The converter modules can perform the current sharing immediately, when a fault is found in another converter module. In addition, the transient response occurs in the system, and the output voltages are at their minimum overshoot and undershoot. Finally, the proposed system has a relatively simple implementation for the redundant operation.

Angle-resolved Photoemission Study of Epitaxial Graphene on Cu(111)

  • Copper is considered to be the most promising substrate, especially Cu(111), for the growth of high quality monolayer graphene. Since interactions between graphene and Cu substrates will influence on the orientation, quality, and electrical properties of synthesized graphene, we experimentally determine a weak interfacial interaction between Cu(111) substrate and graphene using angle-resolved photoemission spectroscopy (ARPES). The measurement was conducted from the initial stage to the formation of a graphene monolayer. Graphene growth was initiated along the Cu(111) lattice, and two rotated graphene domains were grown, where no significant differences were observed in the band structure depending on different orientations. The interaction, including electron transfer from the Cu(111) to graphene, was limited between the Shockley state of the Cu(111) surface and the ${\pi}$ bands of graphene. These results provide direct information on the growth behavior and interactions between the Cu(111) and graphene.

ESL-𝚪-Z- Source Inverter

  • On the basis of the traditional ZSI (Z-source inverter), this paper presents a ESL-${\Gamma}$-ZSI, which uses a unique ${\Gamma}$-shaped impedance network and an extended SL network for boosting its output voltage in addition to their usual voltage-buck behavior. The inverter can increase the boost factor through adjusting shoot-through duty ratio and increasing the number of inductors. Capacitor voltage stress of ESL-${\Gamma}$-ZSI is a constant when 1>D>0, and ESL-${\Gamma}$-ZSI has small inductor current stress. The working principle of ESL-${\Gamma}$-ZSI and comparison with the classical ZSI and SL- ZSI are analyzed in detail. The power loss comparison between ESL-${\Gamma}$-ZSI and Cuk converter is analyzed detailedly. Simulation and experimental results are given to demonstrate the operation features of the inverter.

Analysis of Blood Flow-dependent Blood Nitric Oxide Level and Half-life of Nitric Oxide in Vivo

  • Endothelial release of nitric oxide (NO) contributes to the regulation of vascular tone by inducing vascular relaxation. To estimate the blood flow-dependent nitric oxide level and half-life (T1/2) of nitric oxide in vivo state, we investigated the change of aortic NO currents during the change of aortic blood flow rate using NO-selective electrode system and electromagnetic flowmeter in the aorta of anesthetized rats. Resting mean aortic blood flow rate was $49.6{\pm}5.6ml/min$ in the anesthetized rats. NO currents in the aorta were increased by the elevation of blood pressure and/or blood flow rate. When the aortic blood flow was occluded by the clamping, aortic NO currents were decreased. The difference of NO concentration between resting state and occluded state was $1.34{\pm}0.26{\mu}M$ (n=7). This NO concentration was estimated as blood flow-dependent nitric oxide concentration in the rats. Also, while the aortic blood flow was occluded, NO currents were decreased with exponential pattern with $12.84{\pm}2.15$ seconds of time constant and $7.70{\pm}1.07$ seconds of half-life. To summarize, this study suggested that blood flow-dependent NO concentration and half-life of nitric oxide were about $1.3{\mu}M$ and 7.7 seconds, respectively, in the aorta of anesthetized rats. The nitric oxide-selective electrode system is useful for the direct and continuous measurement of NO in vivo state.

Nafamostat Mesilate: Can It Be Used as a Conduit Preserving Agent in Coronary Artery Bypass Surgery?

  • Background: Graft vessel preservation solution in coronary artery bypass surgery is used to maintain the graft conduit in optimal condition during the perioperative period. Nafamostat mesilate (NM) has anticoagulation and anti-inflammatory properties. Therefore, we investigated NM as a conduit preservative agent and compared it to papaverine. Methods: Sprague-Dawley (SD) rat thoracic aortas were examined for their contraction-relaxation ability using phenylephrine (PE) and acetylcholine (ACh) following preincubation with papaverine and NM in standard classical organ baths. Human umbilical vein endothelial cells (HUVECs) were cultured to check for the endothelial cell viability. Histopathological examination and terminal deoxynucleotidyl transferase dUTP nick end labeling assay were performed on the thoracic aortas of SD rats. Results: The anti-contraction effects of papaverine were superior to those of NM at PE (p<0.05). The relaxation effect of NM on ACh-induced vasodilatation was not statistically different from that of papaverine. Viability assays using HUVECs showed endothelial cell survival rates of >90% in various concentrations of both NM and papaverine. A histopathological study showed a protective effect against necrosis and apoptosis (p<0.05) in the NM group. Conclusion: NM exhibited good vascular relaxation and a reasonable anti-vasocontraction effect with a better cell protecting effect than papaverine; therefore, we concluded that NM is a good potential conduit preserving agent.

Validation of Nafamostat Mesilate as an Anticoagulant in Extracorporeal Membrane Oxygenation: A Large-Animal Experiment

  • Background: Unfractionated heparin is commonly used for anticoagulation in extracorporeal membrane oxygenation (ECMO). Several studies have shown that nafamostat mesilate (NM) has comparable clinical outcomes to unfractionated heparin. This study compared anticoagulation with NM and heparin in a large-animal model. Methods: Beagle dogs (n=8; weight, 6.5-9 kg) were placed on venovenous ECMO. Blood samples were taken every hour and the following parameters were compared: hemoglobin level, activated partial thromboplastin time (aPTT), thromboelastography (TEG) data, platelet function, and inflammatory cytokine levels. Results: In both groups, the aPTT was longer than the baseline value. Although the aPTT in the NM group was shorter than in the heparin group, the TEG parameters were similar between the 2 groups. Hemoglobin levels decreased in both groups, but the decrease was less with NM than with heparin (p=0.049). Interleukin $(IL)-1{\beta}$ levels significantly decreased in the NM group (p=0.01), but there was no difference in the levels of tumor necrosis factor alpha or IL-10 between the 2 groups. Conclusion: NM showed a similar anticoagulant effect to that of unfractionated heparin, with fewer bleeding complications. NM also had anti-inflammatory properties during ECMO. Based on this preclinical study, NM may be a good alternative candidate for anticoagulation in ECMO.

Vascular Cell Responses against Oxidative Stress and its Application

  • The history of studies in biology regarding reactive oxygen species (ROS) is approximately 40 years. During the initial 30 years, it appeared that these studies were mainly focused on the toxicity of ROS. However, recent studies have identified another action regarding oxidative signaling, other than toxicity of ROS. Basically, it is suggested that ROS are reactive, and degenerate to biomolecules such as DNA and proteins, leading to deterioration of cellular functions as an oxidative stress. On the other hand, recent studies have shown that ROS act as oxidative signaling in cells, resulting in various gene expressions. Recently ROS emerged as critical signaling molecules in cardiovascular research. Several studies over the past decade have shown that physiological effects of vasoactive factors are mediated by these reactive species and, conversely, that altered redox mechanisms are implicated in the occurrence of metabolic and cardiovascular diseases ROS is a collective term often used by scientist to include not only the oxygen radicals($O2^{-{\cdot}},\;{^{\cdot}}OH$), but also some non-radical derivatives of oxygen. These include hydrogen peroxide, hypochlorous acid (HOCl) and ozone (O3). The superoxide anion ($O2^{-{\cdot}}$) is formed by the univalent reduction of triplet-state molecular oxygen ($^3O_2$). Superoxide dismutase (SOD)s convert superoxide enzymically into hydrogen peroxide. In biological tissues superoxide can also be converted nonenzymically into the nonradical species hydrogen peroxide and singlet oxygen ($^1O_2$). In the presence of reduced transition metals (e.g., ferrous or cuprous ions), hydrogen peroxide can be converted into the highly reactive hydroxyl radical (${^{\cdot}}OH$). Alternatively, hydrogen peroxide may be converted into water by the enzymes catalase or glutathione peroxidase. In the glutathione peroxidase reaction glutathione is oxidized to glutathione disulfide, which can be converted back to glutathione by glutathione reductase in an NADPH-consuming process.

p66shc Adaptor Protein Suppresses the Activation of Endothelial Nitric Oxide Synthase in Mouse Embryonic Fibroblasts

  • Among the Shc proteins, p66shc is known to be related to oxidative stress responses and regulation of the production of reactive oxygen species (ROS). The present study was undertaken to investigate the role of p66shc on endothelial nitric oxide synthase (eNOS) activity in the mouse embryonic fibroblasts (MEFs). When wild type (WT) or p66shc (-/-) MEFs were transfected with full length of eNOS cDNA, the expression and activity of eNOS protein were higher in the p66shc (-/-) MEFs. These phenomena were reversed by reconstitution of p66shc cDNA transfection in the p66shc (-/-) MEFs. The basal superoxide production in the p66shc (-/-) MEFs was not significantly different from that of WT of MEFs. However, superoxide production induced by NADPH in the p66shc (-/-) MEF was lesser than that in WT MEFs. When compared with WT MEFs, cell lysate of p66shc (-/-) MEFs showed significantly increased H-ras activity without change of endogenous H-ras expression. Our findings suggest the pivotal role of p66shc adaptor protein played in inhibition of endothelial nitric oxide production via modulation of the expression and/or activity of eNOS protein.
