Electrohydraulic Pump-Driven Closed-Loop Blood Pressure Regulatory System

  • An electrohydraulic (EH) pump-driven closed-loop blood pressure regulatory system was developed based on flow-mediated vascular occlusion using the vascular occlusive cuff technique. It is very useful for investigating blood pressure-dependant physiological variability, in particular, that could identify the principal mediators of renal autoregulation, such as tubuloglomerular feedback (TGF) and myogenic (MYO), during blood pressure regulation. To address this issue, renal perfusion pressure (RPP) should be well regulated under various experimental conditions. In this paper, we designed a new EH pump-driven RPP regulatory system capable of implementing precise and rapid RPP regulation. A closed-loop servo-controlwas developed with an optimal proportional plus integral (PI) compensation using the dynamic feedback RPP signal from animals. An in vivo performance was evaluated in terms of flow-mediated RPP occlusion, maintenance, and release responses. Step change to 80 mmHg reference from normal RPP revealed steady state error of ${\pm}3%$ during the RPP regulatory period after PI action. We obtained rapid RPP release time of approximately 300 ms. It is concluded that the proposed EH RPP regulatory system could be utilized in in vivo performance to study various pressure-flow relationships in diverse fields of physiology, and in particular, in renal autoregulation mechanisms.

Antihypertensive Properties of Dried Radish Leaves Powder in Spontaneously Hypertensive Rats (자연발증적 고혈압 흰쥐에서 무청 건조분말의 항고혈압 효과)

  • The study aim was to investigate the antihypertensive effect after oral supplementation of dried radish leaves powder (DRLP). Angiotensin-converting enzyme (ACE) activity was measured by spectrophotometric assay. The systolic blood pressure (SBP) was measured in spontaneously hypertensive (SHR) and normotensive rats (Wistar) by the tail cuff method after a 4-week diet with DRLP at the level of 2.5% or 5%. The supplementation of DRLP decreased SBP of SHR although the 5% supplementation level did not show any more pronounced effect than the 2.5% level did. The decrease in the SBP observed for both 2.5% and 5% DRLP was accompanied by significant increases of the urinary Na and K excretion. The DRLP supplementation showed a potent ACE-inhibitory activity in pulmonary tissue from both hypertensive and normotensive rats. However, the DRLP supplementation did not affect the SBP in normotensive rats. These results indicated that DRLP exerted an antihypertensive effect in SHR due to the decreased ACE activity and increased urinary Na excretion.

  • We report a case of an 85-year-old woman with tuberculous subdeltoid bursitis. Tubeculous bursitis without adjacent joint tuberculosis may occur any site but it is rare. It is common in bursae subjected to trauma like trochanteric, prepatellar, olecranon and so on. Because the clinical findings or physical examination of tuberculous subdeltoid bursitis are similar to rotator cuff disorder and the MRI or arthroscopic findings are similar to synovial diseases, careful differential diagnosis is necessary.

Continuous Blood Pressure Prediction Using PTT During Exercise (PTT를 이용한 자전거 운동 중 지속적인 혈압의 예측)

  • The purpose of this work is to predict the systolic blood pressure (BP) during exercise from pulse transit time (PTT) for warning of possible danger. PTT was calculated as the time between R-peak of ECG and the peak of differential photoplethysmograph (PPG). For the PTT-BP model, we used regress equations from previous studies and 3 kinds of new models combining linear and nonlinear regress equation. The model parameters were estimated with the data measured under low to middle intensity exercise, and then was tested with the data measured under high intensity exercise. Predicted BP values after high intensity exercise were compared with those measured by cuff-type sphygmomanometer. The results showed that the error between measured and predicted values were acceptable for the monitoring BP. We tested PTT-BP models 1 month after the identification without further calibration. Models could predict the BP and the errors between measured and predicted BP were about 5mmHg. The suggested system is expected to be helpful in recognizing any danger during exercise.

Effect of Soybean Hydrolysate on Hypertension in Spontaneously Hypertensive Rats (대두 가수분해물에서 분리한 UF-peptide가 in vivo에서 자발성 고혈압 흰쥐의 혈압 강하에 미치는 영향)

  • Effect of soybean hydrolysate prepared by ultrafiltration(UF-peptide) was evaluated regarding its action on blood pressure, selected biochemical changes in spontaneously hypertensive rats(SHR). Male SHR were divided into three groups and fed diet with 0, 3, 10% UF-peptide for 6 weeks. Development of hypertension was determined by tail cuff method during the experimental period. After the experimental period, angiotension I converting enzyme(ACE) activity in serum and vascular tissue, and lipids level in serum were measured. Spontaneous elevation of blood pressure was suppressed by the UF-peptide supplementation after two weeks of feeding. After five weeks of feeding the development of hypertension was significantly inhibited in the UF-peptide administered groups compared to the control. In addition, the UF-peptide was effective for lowering serum lipids level such as triglyceride and total cholesterol, as well as ameliorating some pathological changes in SHR.

  • The aim of this study was to evaluate the short-term outcomes of the trans anal one-stage pull-through procedure(TOP) in Hirschsprung's disease. Eight patients aged 3 weeks to 8 months with Hirschsprung's disease underwent this procedure. A rectal mucosectomy was performed from just proximal to dentate line to the level of peritoneal reflexion, where muscle layer was incised circumferentially. Rectosigmoid was mobilized out through the anus, and full-thickness frozen biopsy was taken for confirmation of ganglionic cells. After the rectal muscular cuff was divided longitudinally in the posterior aspect, aganglionic bowel was removed and ganglionic colon was anastomozed to the anus. The mean operating time was 161 minutes, and the mean hospital stay after operation was 3.8 days. Five patients had three to four bowel movement per day without other therapy at mean postoperative 39.2 days. Although long-term follow-up will be required, the TOP might be the new alternative surgical procedure for Hirschsprung's disease.

  • Evaluation of the heart work is starting to emerge as a new diagnostic tool for arterial diseases. The aim of this study is to develop a mathematical model for the estimation of heart work utilizing the pulse waves between two points of a vessel. In order to calculate heart work, medical data such as blood pressure waveforms (which are measured using a cuff) are utilized. The heart work is calculated by employing the modified Windkessel model together with the viscosity models of Casson or Herschel-Bulkely (H-B). The results indicate that the compliance values at the proximal and distal locations differ for the Casson and H-B models.

A novel model of THO/TREX loading onto target RNAs in metazoan gene expression

  • The THO/TREX complex consists of several conserved subunits and is required for mRNA export. In metazoans, THO/TREX binds a subset of mRNAs during RNA splicing, and facilitates their nuclear export. How THO/TREX selects RNA targets is, however, incompletely understood. In our recent study, we reported that THO is loaded onto Piwi-interacting RNA (piRNA) precursor transcripts independent of splicing, and facilitates convergent transcription in Drosophila ovary. The precursors are later processed into mature piRNAs, small noncoding RNAs that silence transposable elements (TEs). We observed that piRNAs originating from dual-strand clusters, where precursors are transcribed from both strands, were specifically affected by THO mutation. Analysis of THO-bound RNAs showed enrichment of dual-strand cluster transcripts. Interestingly, THO loading onto piRNA precursors was dependent on Cutoff (Cuff), which comprises the Rhino-Deadlock-Cutoff (RDC) complex that is recruited to dual-strand clusters by recognizing H3K9me3 and licenses convergent transcription from he cluster. We also found that THO mutation affected transcription from dual-strand clusters. Therefore, we concluded that THO/TREX is recruited to dual-strand piRNA clusters, independent of splicing events, via multi-protein interactions with chromatin structure. Then, it facilitates transcription likely by suppressing premature termination to ensure adequate expression of piRNA precursors.

General Pharmacology of Bamboo Salt (죽염의 일반약리작용)

  • This study has been carried out to investigate general pharmacological action of bamboo salt (jukyom) in terms of effects on central nervous system and cardiovascular system in experimental animals. After bamboo salt, crude salt or reagent-grade NaCl were orally administered into male ICR mice with dose of 2.0 g/kg, general behavioural syndromes such as body weight and locomotor activity, spontaneous motor activity, pento-barbital-induced sleeping time, muscle incoordination, electroshock-induced convulsion, body temperature and writhing response caused by 0.6% acetic acid solution were observed. Bamboo salt had no influences in these indices for examinition of effect on central nervous system. Additionally, conscious male Sprague Dawley rats fastened overnight won ere treated with bamboo salt, crude salt or reagent-grade NaCl (2.0 g/kg, p.o.) to examine the effect of these salts cardiovascular system. Systolic, median and diastolic food pressure and heart rate were dertemined using tail cuff indirect method. Treatment with Hydralazine (50 mg/kg, p.o) as a positive control produced the decreases in systolic, median and diastolic blood treasure and an increase in heart rate. whereas no changes were observed in bamboo salt, crude salt and reagent-grade NaCl treated groups. These results strongly suggest that bamboo salt may have no effects on general pharmacology of central nervous systems and cardio-vascular systems.

Automatic blood pressure measurement device using oscillometric method and Korotkoff sounds

  • The oscillometric method and Korotkoff sound method are the most common ways to measure the blood pressure. A new automatic blood pressure measurement device, which uses both oscillometric method and Korotkoff method, was developed. A pressure sensor was used to obtain cuff pressure and oscillation signal, and a microphone was used to detect Korotkoff sounds. Forty-five measurements from fifteen subjects were used for analysis. Correlation coefficients between the traditional auscultatory method and Korotkoff sound method were 0.9820 and 0.9721 for the systolic and diastolic blood pressure values, respectively. Standard deviations of differences for the systolic and diastolic blood pressure values were 1.3019 and 1.4495, respectively. Correspondingly, correlation coefficients between the traditional auscultatory method and oscillometric method using newly developed algorithm were 0.9651 and 0.9136 for the systolic and diastolic blood pressure values, with the standard deviations of 1.42 and 1.73, respectively. The results showed that the newly developed algorithm for oscillometirc method provide accurate blood pressure values, moreover, Korotkoff sound method using microphone provides even higher accuracy. Therefore, a new automatic device which utilizes both oscillometric method and Korotkoff sound method would provide the accurate and reliable blood pressure values.
