Generation and Retention of Defective RNA3 from Cucumber Mosaic Virus and Relevance of the 2b Protein to Generation of the Subviral RNA

  • A defective RNA3 (D3Yα) of strain Y of cucumber mosaic virus (CMV-Y) was examined on host-specific maintenance, experimental conditions, and a viral factor required for its generation in plants. D3Yα was stably maintained in cucumber but not in tomato plants for 28 days post inoculation (dpi). D3Yα was generated in Nicotiana tabacum or N. benthamiana after prolonged infection in the second and the third passages, but not in plants of N. benthamiana grown at low temperature at 28 dpi or infected with CMV-Y mutant that had the 2b gene deleted. Collectively, we suggest that generation and retention of D3Yα depends on potential host plants and experimental conditions, and that the 2b protein has a role for facilitation of generation of D3Yα.

  • Tomato aspermy virus(TAV-B)는 두 개의 다른 CMV균주 D 및 K에서 분리한 satellite RNA의 증식 및 encapsidation에 대하여 helper virus의 역할을 하였다. 이 두 satellite RNA는 역시 ??? system에서 보이는 바와 같이 TAV가 감염된 담배에서 TAV 병징을 크게 감소시켰으며, 이는 감염된 식물내의 Virus 함량의 감소와 관계가 있었다. 또한 CMV가 감염된 토마토에서 lethal necrosis를 유발시킴을 볼 수 있었다.

아마리리스에 모자익병을 일으키는 CMV에 관한 연구 (Isolation and Properties of Cucumber Mosaic Virus Inducing Mosaic Symptoms in Hippeastrum hybridum Hort)

  • 아마리리스에 모자익병을 일으키는 병원바이러스의 기주범위, 물리적성질, 혈청반응을 조사하고 바이러스 입자를 전자현미경으로 관찰을 하였다. 바이러스의 지표식물 검정 결과 C. amaranticolor, V. unguiculata, C. sativus G. globosa둥 21종의 CMV감수성식물에 병징이 나타났다. 바이러스의 내열성은 55C(10분간)이며 내희석성은 $10^{-3}$이고 내보존성은 실온에서 2일이었다. 한천내 확산법을 이용한 항혈청반응결과 뚜렷한 반응대가 형성되었으며, 바이러스흡광도는 260nm에서 최고였으며, 245nm에서 최저였다. 전자현미경에 의한 바이러스입자의 검경결과 직경이 90nm내외인 소구형 입자가 관찰되었다. 이상의 결과를 종합하면 아마리리스에 모자익병을 일으키는 것은 CMV에 의한 것으로 보인다.

Transgenic cucumber expressing the 54-kDa gene of Cucumber fruit mottle mosaic virus is highly resistance and protect non-transgenic scions from soil infection

  • Cucumber fruit mottle mosaic tobamovirus (CFMMV) causes severe mosaic symptoms with yellow mottling on leaves and fruits, and occasionally severe wilting of cucumber plants. No genetic source of resistance against this virus has been identified. The genes coding for the coat protein or the putative 54-kDa replicase were cloned into binary vectors under control of the SVBV promoter. Agrobacterium-mediated transformation was peformed on cotyledon explants of a parthenocarpic cucumber cultivar with superior competence for transformation. R1 seedlings were evaluated for resistance to CFMMV infection by lack of symptom expression, back inoculation on an alternative host and ELISA. From a total of 14 replicase-containing R1 lines, 8 exhibited immunity, while only 3 resistant lines were found among a total of 9 CP-containing lines. Line 144 homozygous for the 54-kDa replicase was selected for further resistance analysis. Line 144 was immune to CFMMV infection by mechanical and graft inoculation, or by root infection following planting in CFMMV-contaminated soil. Additionally, line 144 showed delay of symptom appearance following infection by other cucurbit-infecting tobamoviruses. Infection of line 144 plants with various potyviruses and cucumber mosaic cucumovirus did not break the resistance to CFMMV. The mechanism of resistance of line 144 appears to be RNA-mediated, however the means is apparently different from the gene silencing phenomenon. Homozygote line 144 cucumber as rootstock demonstrated for the first time protection of a non-transformed scion from soil inoculation with a soil borne pathogen, CFMMV.

Variation in the Pathogenicity of Lily Isolates of Cucumber mosaic virus

  • Two isolates of Cucumber mosaic virus (CMV) originated from lily plants, named Ly2-CMV and Ly8-CMV, were compared with their pathological features in several host plants. Ly2-CMV and Ly8-CMV could induce systemic mosaic symptom in Nicotiana benthamiana, but Ly2-CMV could not systemically infect tomato and cucumber plants that have been used for CMV-propagative hosts. While Fny-CMV used as a control infected systemically the same host plants, producing typical CMV symptoms. Ly8-CMV could infect systemically two species of tobacco (N. tabacum cv. Xanthi-nc and N. glutinosa) and zucchini squash (Curcubita pepo), but Ly2 failed systemic infection on these plants. As resulted from tissue-print immunoblot assay, different kinetics of systemic movement between Ly2-CMV and Ly8-CMV were crucial for systemic infection in tobacco (cv. Xanthi-nc). Sequence analysis of full-length genome of two lily isolates showed Ly2 and Ly8 belonged to subgroup IA of CMV. The lily isolates shared overall 98 % sequence identity in their genomes. Coat protein, 3a protein, and 2b protein involved in virus movement was highly conserved in genomes of the isolates Ly2 and Ly8. Although there is the low frequency of recombinants and reassortants in natural CMV population, phylogenetic analysis of each viral protein among a number of CMV isolates suggested that genetic variation in a defined population of CMV lily isolates was stochastically produced.

The Tobacco Ubiquitin-activating Enzymes NtE1A and NtE1B Are Induced by Tobacco Mosaic Virus, Wounding and Stress Hormones

  • Recent characterization of several genes involved in plant defense responses suggested that ubiquitin-mediated protein degradation has a role in these responses. We isolated two cDNAs (NtUBA1 and NtUBA2) encoding ubiquitin-activating enzyme (E1) from Nicotiana tabacum cv. BY-2. The open reading frames of both encoded 1080 amino acids, corresponding to molecular masses of 120 kDa. The E1s and corresponding transcripts were upregulated by infection with tobacco mosaic virus (TMV) and tomato mosaic virus (ToMV), and to a lesser extent by cucumber mosaic virus (CMV). Furthermore, they were also upregulated by wounding stress, and the plant hormones salicylic acid, jasmonic acid and the ethylene precursor, aminocyclopropane-1-carboxylic acid (ACC). Our findings support the idea that the ubiquitin-proteasome system plays a role in plant disease defenses.

  • A virus causing stunt, yellowing, severe mosaic, malformation symptoms on leaves and uneven development and malformation on fruits of zucchini was prevalent around Goseong, Gyeongsangnam-do, Korea. A survey conducted (2004) in the Goseong area revealed about 20% virus infection rate. The disease causative identified as Cucumber mosaic virus (CMV-Z1) was further characterized. The isolate induces mosaic symptoms on Cucumis sativus, while severe mosaic, stunt and malformation on C. pepo. Thin section analyses have shown that virus inclusions are formed in the cuticle layers as well as epidermal, parenchyma and collenchymas cells in virus-infected Nicotiana tabacum. CMV-Z1 isolate induced specific cytoplasmic inclusion bodies such as irregular clumps (IC), crystal (Cr) and irregular chloroplasts (ICh). IC was made up of virus particles interspersed with a darkly stained amorphous material and found both in the cytoplasm and vacuoles, whereas ICh and Cr were rarely found in the vacuoles. The genome of CMV-Z1 RNA-1 consists of 3359 nucleotide (nt) encoding 1a protein of 993 amino acids (aa). The CMV-Z1 RNA-2 was 3050 nt in length containing 2a (857 aa) and 2b (110 aa), while RNA-3 encoding 3a movement protein (279 aa) and coat protein (218 aa) was 2215 nt in length. Phylogenetic analyses of nucleotide sequences of CMV-Z1 isolate appeared it is more closely related to subgroup IA than to subgroup IB or II.

Phylogenetic Analysis of New Isolates of Cucumber mosaic virus from Iran on the Basis of Different Genomic Regions

  • Molecular characterization of Cucumber mosaic virus (CMV) was done by using samples from tomato and cucurbitaceous plants collected from different locations in the northwest region of Iran. After screening by enzyme-linked immunosorbent assay, 91 CMV-infected samples were identified. Biological properties of eight representative isolates were compared with each other revealing two distinct phenotypes on squash and tomato plants. Phylogenetic analyses based on nucleotide sequences of the coat protein (CP), movement protein (MP) and 2b of the new isolates, together with that of previously reported isolates, led to the placement of the Iranian isolates in subgroups IA and IB according to CP and MP genes, but in subgroup IA according to the 2b gene. These data suggest that reassortment may have been a major event in the evolution of CMV in Iran, and that the Iranian isolates are derived from a common recent ancestor that had passed through a bottleneck event.

오이모자익 바이러스의 순화와 항혈청 제조 (Purification and Serology of Cucumber Mosaic Virus)

  • 순수분리된 오이모자익 바이러스(CMV)를 함배 (KY-57)에서 증식하여 개량된 순화방법으로 순화하였다. 순화된 바이러스를 Complete adjuvant와 1: 1로 섞어 토끼에 10일 간격으로 2회 근육주사하여 높은 역가의 CMV 항혈청을 제조하였다. 한천내확산법에 의한 항혈청의 역가는 1/1280이었다. 제조된 CMV 항혈청은 앞으로 채소작물이나 그밖에 식량작물에 이병된 CMV를 분리동정하는데 이용될 것이다.

오이모자이크바이러스 2b 유전자 발현 담배의 형태 및 전사체 분석 (Phenotypic and Transcriptomic Analysis of Nicotiana benthamiana Expressing Cucumber mosaic virus 2b gene)

  • 오이모자이크바이러스 2b 유전자는 전사후유전자침묵(PTGS)을 억제하는 기능을 가진 억제인자이다. 식물체내 2b 유전자 기능을 분석하기 위해 Nicotiana benthamiana에 형질전환하였고 형태변화와 유전자 발현변화를 분석하였다. 8계통의 2b 유전자 형질전환체 중에서 1개의 유전자가 T0 개체에 삽입된 계통은 3개였다. 2b 유전자 형질전환체는 일반적으로 종자 확보가 어려웠지만 다행히 일부 배수화되지 않은 계통(hemizygote)에서는 소량의 종자가 확보되어 계통유지가 가능하였다. 고정계통의 전사체를 해독하여 대조와 비교분석한 바, 2b 유전 자는 특정유전자의 발현을 선택적으로 증대시키는 것이 아닌 다수의 유전자를 비선택적으로 발현을 증대시키는 것으로 판단되었다. 이러한 결과는 2b 유전자가 세포질에 존재하는 다양한 RNA의 대사중 분해를 억제하여 세포질내 RNA가 축적되고 이로 인해 단백질 합성도 증대되어 정상적 생장발달이 저해되고 기형적인 형태의 식물체가 되는 요인으로 판단된다.
