한국 약용 및 식용식물들의 항산화성 식물탐색 (Screening of Korean Medicinal and Food Plants with Antioxidant Activity)

  • 60종(種)의 자생 약용식물과 식용작물을 대상으로 항산화활성의 생리활성을 조사함으로써 약용식물과 식용작물의 유용성 측면의 확인뿐만 아니라 새로운 생리활성물질 탐색의 가능성을 검토하기 위해서 실시한 실험 결과는 다음과 같다. 자생 약용식물 및 식용작물에 대한 50% EtOH 추출물을 이용하여 TBA법. DPPH법에 의하여 1차 활성검정 결과 처리방법에 따라 활성값에 차이가 있으나 TBA법에서는 검정콩의 추출액이 87.3%, DPPH법에서는 까마중 추출액이 80.6%로서 가장 높은 활성을 보였고 두가지 방법에서 동시에 활성이 높은 것은 검정콩을 비롯하여 10종 이었다. SOD활성정도도 검정식물에 따라 큰 차이가 있으나 검정콩이 53.5%로서 가장 높은 SOD활성을 나타내었으며 SOD종류는 Cu/ZnSOD, FeSOD을 포함하는 것으로 나타났다. Cu/ZnSOD만을 함유하고 있는 잔대는 SOD활성 정도가 10.37%로서 실험 대상식물 중 가장 낮은 활성을 보였다.

Antiproliferative Effect of Artemisia argyi Extract against J774A.1 Cells and Subcellular Superoxide Dismutase (SOD) Activity Changes

  • The water and methanol extracts of Artemisia argyi showed significant cytotoxicities against J774A.1 cells but not so much against normal leukocytes. The cytotoxicities were found to be dependent on the extract concentration and the incubation time. The concentration of water and methanol extracts inhibiting 50% of cell proliferation ($IC_{50}$) were estimated to be 44.2 mg/ml and 71.6 mg/ml, respectively. In the presence of Artemisia argyi water extract, total superoxide dismutase (CuZnSOD and MnSOD) activities of media, cytoplasmic and mitochondrial fractions of J774A.1 cells increased in accordance with cytotoxicity. MnSOD was found to be the main component of enhanced total SOD activities, particulary in the mitochondrial fraction. In contrast to SOD, catalase and glutathione peroxidase (GPx) were not found in any instance of the current investigation. In addition, substantial amount of $O_2^-$ appeared to be generated in the mitochondrial fraction under the influence of Artemisia argyi. All data put together, it is postulated that Artemisia argyi extracts seem to stimulate $O_2^-$ generation in mitochondria of J774A.1 cells with concomitant increases of SODs. Since $H_2O_2$, the reaction product of SOD on $O_2^-$, is known to be readily converted to very toxic $OH{\cdot}$ in the absence of catalase and/or GPx cooperation, toxicity derived from ROS such as $O_2^-$, $H_2O_2$, and $OH{\cdot}$ may be the main cause of necrosis and/or apoptosis of J774A.1 cells.

Characterization of Copper/Zinc-Superoxide Dismutase (Cu/Zn-SOD) Gene from an Endangered Freshwater Fish Species Hemibarbus mylodon (Teleostei; Cypriniformes)

  • Gene structure of copper/zinc-superoxide dismutase (Cu/Zn-SOD; sod1) was characterized in Hemibarbus mylodon (Teleostei; Cypriniformes), an endangered freshwater fish species in Korean peninsula. Full-length cDNA of H. mylodon SOD1 consisted of a 796-bp open reading frame sequence encoding 154 amino acids, and the deduced polypeptide sequence shared high sequence homology with other orthologs, particularly with regard to metal-coordinating ligands. Genomic structure of the H. mylodon sod1 gene (hmsod1; 1,911 bp from the ATG start codon to the stop codon) was typical quinquepartite (i.e., five exons interrupted by four introns); the lengths of the exons were similar among species belonging to various taxonomic positions. The molecular phylogeny inferred from sod1 genes in the teleost lineage was in accordance with the conventional taxonomic assumptions. 5'-flanking upstream region of hmsod1, obtained using the genome walking method, contained typical TATA and CAAT boxes. It also showed various transcription factor binding motifs that may be potentially involved in stress/immune response (e.g., sites for activating proteins or nuclear factor kappa B) or metabolism of xenobiotic compounds (e.g., xenobiotic response element; XRE). The hmsod1 transcripts were ubiquitously detected among tissues, with the liver and spleen showing the highest and lowest expression, respectively. An experimental challenge with Edwardsiella tarda revealed significant upregulation of the hmsod1 in kidney (4.3-fold) and spleen (3.1-fold), based on a real-time RT-PCR assay. Information on the molecular characteristics of this key antioxidant enzyme gene could be a useful basis for a biomarker-based assay to understand cellular stresses in this endangered fish species.

부추의 수확시기에 따른 클로로필, 무기질 및 superoxide dismutase 유사활성의 변화 (Chlorophyll, Mineral Contents and SOD-like Activities of Leeks Harvested at Different Times)

  • 경남 김해에서 수확되는 부추의 가공에 기초자료로 활용하기 위하여 초벌 2벌, 3벌, 4벌, 5벌에 재배되는 부추의 성분들을 분석하였다. 부추의 엽록소 함량은 2벌 762.54, 3벌 775.09, 4벌 763.71, 5벌 798.47mg%로 5벌에서 가장 많이 함유되어 있었다. 무기질 함량은 초벌 부추에서 철, 칼륨, 구리의 함량은 각 10.61, 627.13, 0.14mg%로써 가장 높았으며 수확횟수가 증가함에 따라 감소하였고 칼슘, 망간, 인, 아연, 나트륨의 함량은 각 117.34, 1.27, 92.61, 0.73, 21.52 mg%로 5벌에서 가장 높은 간을 나타내었다. 중금속오염을 나타내는 납은 검출되지 않았다. SOD 유사활성은 2벌 1600, 3벌 1357.32, 4벌 1317.65, 5벌 1291.71unit/g으로 2벌에서 가장 높은 값을 나타내었으며 수확이 거듭될수록 활성이 낮아지는 경향을 보였다.

B16F10 Murine Melanoma Cell에서 Myricetin이 항산화효소의 m-RNA 발현에 미치는 영향 (Effect of Myricetin on mRNA Expression of Different Antioxidant Enzymes in B16F10 Murine Melanoma Cells)

  • Flavonoids are class of polyphenolic compounds widely distributed in the plant kingdom, which display a variety of biological activities, including antiviral, antithrombotic, antiinflammatory, antihistaminic, antioxidant and free-radica 1 scavenging abilities. The antioxidant enzyme (AOE) system plays an important role in the defense against oxidative stress insults. To determine whether flavonoid, myricetin can exert antioxidative effects not only directly by modulating the AOE system but also scavenging free radical, we investigated the influence of the flavonoid myricetin on cell viability, different antioxidant enzyme activities, ROS level and the expression of different antioxidant emzyme in B16F10 murine melanoma cells. Myricetin in a concentration range from 6.25 to $50\;{\mu}M$ decreased superoxide dismutase (SOD) and glutathione peroxidase (GPx) enzyme activities, but catalase (CAT) activity was increased. In the myricetin-treated group, ROS levels were decreased dose-dependently. Antioxidant enzyme expression was measured by RT-PCR. Myricetin treatment of B16F10 cells increased catalase expression. Expression levels of copper zinc superoxide dismutase (CuZn SOD) were not affected by exposure of myricetin. Manganese superoxide dismutase (Mn SOD) and GPx expression levels decreased slightly after myricetin treatment. In conclusion, the antioxidant capacity of myricetin was due to CAT and free-radical scavenging.

일부 여대생의 구리와 아연 영양상태 평가 (Evaluation of Zinc and Copper Status in Korean College Women)

  • This study was conducted to assess dietary intake and nutritional status of zinc and copper in Korean college women. Dietary survey was conducted by 24-hour recall method and fasting serum samples were collected from 111 apparently healthy subjects. Intake levels of zinc and copper were calculated using newly developed database for Zn & Cu of Korea food. Serum levels of Zn, Cu and activities of ALP, EC-SOD were measured from fasting serum sample. Mean daily zinc and copper intakes were 6.72mg/day(56.0% RDA) and 1.11mg/day respectively. Mean values of serum ALP activity, zinc and copper concentration were 43.9U/L, 14.8umol/1, 15.5umol/1and these values were mostly within normal range. EC-SOD activitis of the subjects were low and had no correlation with intake or serum levels of Zn, Cu. In conclusion, these results show that zinc and copper intake of Koran college women are lower than those from other counties but higher than those of adults in rural area of Korea. Their serum levels of Zn, Cu, ALP are relatively normal. These results indicate that marginal deficiency of Zn and Cu may be quite prevalent in these subjects but serum indicators measured may not be sensitive enough to detect such marginal deficiency. Further study in needed to develop a biochemical index sensitive enough to evaluate Zn and Cu status.

비만 청소년의 SOD 활성도 및 혈청 항산화무기질 농도에 관한 연구 (A Study on SOD Activity and Serum Antioxidant Mineral Concentrations in Obese Adolescents)

  • The purpose of this study was to investigate the relationship between obesity and, erythrocyte SOD (superoxide dismutase) activity and serum antioxidant mineral (Fe, Zn, Cu, Mn and Se) concentrations of adolescents. Subjects were assigned to one of two groups such as obese ($BMI{\geq}25$, 32 boys, 24 girls) and normal group (18.5 < BMI < 23, 27 boys, 30 girls) Subjects were evaluated based on anthropometric measurements, 24-hr dietary recalls and blood analysis. The mean age of the total subjects was 13.8 years. The mean weight (p < 0.001), BMI (p < 0.001) and body fat (p < 0.001) of obese were higher than those of normal group. There was no significant difference in nutrient intake between obese and normal groups. SOD activity of obese group was not significantly different from normal groups, in both males and females. However, in the males, serum Cu concentration of obese were significantly lower than those of normal group. In the females, Serum Mn concentration of obese were significantly lower then those of normal group. In the correlation analysis, BMI of the subjects had significantly negative correlations with serum Cu, Zn and Mn. To summarize the results, increase of obesity may lead to decrease of serum antioxidant minerals such as Cu, Zn and Mn.

사료 중 크릴 밀을 급여한 육계의 생산성과 SOD 활성에 미치는 급성기 반응의 영향 (Effect of the Acute Phase Response on the Performance and Superoxide Dismutase Activity in Broiler Chicks Fed on Dietary Krill Meal)

  • 급성 기 반응중인 육계에서 사료 중 크릴 밀의 생산성과 SOD 활성에 미치는 영향을 조사하기 위하여 크릴 밀 A와 B를 이용한 실험 1과 크릴 밀 A를 이용한 확인 실험 2가 실시 되었다. 실험사료는 옥수수 대두박 위주의 기초 사료 와 기초 사료 중 대두박과 2.0%의 크릴 밀 A(수입) 또는 크릴 밀 B(수입)를 대치한 사료이다. 실험 사료를 1 일령 육계 병아리에 급여하여 Salmonella typhymurium lipopolysaccharide (LPS)를 복강 내 주입하여 급성 기 반응을 발생시킨 2 주령 육계의 생산성과 Superoxide dismu- tase(SOD) 활성에 미치는 영향을 Saline을 주입한 대조와 비교하였다. 증체량, 사료 섭취량 및 사료효율은 크릴 밀 A와 크릴 밀 B 사료를 급여한 육계사이에 차이가 있었다. 크릴 밀 사료를 급여한 육계에서 급성기 반응은 간장과 비장 무계를 높였다. 그리고 급성기 반응은 크릴 밀 사료를 급여한 육계 병아리의 간장과 적혈구 세포액의 MnSOD와 Cu/ZnSOD 활성을 높였다. PHA-p 반응에 미치는 크릴 밀 사료의 영향은 없었다. 크릴 밀 B는 면역반응 후 회복중인 육계의 생산성을 향상시켰으나 크릴 밀 A에서는 이러한 영향이 없었다. 이상과 같이 크릴 밀의 종류에 따라 급성기반응시 와 회복시의 생산성은 SOD 활성의 변화로부터 산화 스트레스의 영향을 받고 있다는 것을 시사하고 있다.
