Study on the Purification of 5'-Ribonucleotide (5'-Ribonucleotide의 분리(分離) 정제(精製)에 관(關)한 연구(硏究))

  • The purification of 5'-riboncleotide using ion exchange resins has been studied and the optimum conditions were determined. The amount used of Amberlite IR120 ion exchange resin in 2nd resin tower could be reduced up to 20% by pretreating the hydrolyzed RNA solution in 1st resin tower. The amount used of regenerant could be also reduced up to 20% by desalting the hydrolyzed RNA solution in the 1st tower, because the desalted solution eluted easily by the water in the 2nd tower. The crystal obtained in this experiment was the mixed-crystals of $5'-IMP.Na_2\;and\;5'-GMP.Na_2$. The crystallization of the complexes formed from $5'-IMP.Na_2\;and\;5'-GMP.Na_2$ gave the best result at pH 7.6. The yield of crystal complexes formed from $5'-IMP.Na_2\;and\;5'-GMP.Na_2$ was obtained higher in high MeOH concentration. However, in higher than 60% MeOH concentration the products was amorphous. The higher content of MeOH for the crystallization of the product gave the smaller value of $5'-IMP.Na_{2}/5'-GMP.Na_2$.

Study on the Storage Stability of Horse Fat in Jeju (제주산 말지방(Horse Fat)의 저장 안정성 향상에 관한 연구)

  • Horse fat is known to be an effective ingredient in Asia, and the horse fat itself, which is mixed with other ingredients at the additive level, is often sold as a finished product. In this case, physical properties of the horse fat raw material are important. Many horse fats produced in Korea (Jeju) have low temperature stability, so if not stored at low temperatures, segregation may occur. In the case of Japanese horse fat, it is partially hydrogenated or is used the solid phase as the horse fat by separating the liquid phase and the solid phase that is harder and more stable than the horse fat of Jeju. In this study, the physical properties were tested to improve the temperature stability even without the partial hydrogenation process of Jeju horse fat. Various oil gelling agents were used in the study. Results confirmed that the physical properties of the hydroxystearic acid added Jeju horse fat were improved. In addition, stability evaluations at temperatures of 25 ℃, 40 ℃, 45 ℃ and flow behavior evaluations at temperatures of 25 ℃, 30 ℃, 40 ℃ were performed for Jeju horse fat with hydroxystearic acid, 100% Jeju horse fat, and 100% Japanese horse fat. Results showed that the Jeju horse fat improved in flow behavior by adding hydroxystearic acid similar to that of Japanese horse fat. In addition, when the crystal state was observed under a microscope, the thermal stability was improved by decreasing the size of the needle-type crystals with the addition of hydroxystearic acid. Jeju horse fat containing hydroxystearic acid was found to have no physical problems even when stored at room temperature for a long time.

The Relative Effectiveness of Various Radiation Sources on the Resistivity Change in n-Type Silicon

  • Resistivity changes of n-type float-zone silicon crystals with 6.4$\times$10$^{14}$ to 1.25$\times$10$^{17}$ phosphorus atoms/㎤ due to irradiation by (1) 1 MeV electrons, (2) two types of research reactors, and (3) $Co^{60}$ ${\gamma}$-ray sources were investigated. The results were analyzed on the basis of a simple exponential formula derived by Buehler. While the formula gave a fair fit in the low fluence range in most cases, the deviation was quite appreciable in the case of 1 MeV electron irradiation, and a linear change gave better fit in some cases. The large change in the carrier removal rate in electron-irradiated samples in the high fluence range was analyzed in detail in terms of the Fermi level cross-over of the defect levels. Based on the damage constants evaluated from the initial portion of data where the formula was applicable, the relative effectiveness of various radiation sources in causing the resistivity change in n-type silicon was compared. The TRIGA Mark II reactor neutrons, for example, were found to be about 40 times more effective than 1 MeV electrons. The dependence of the damage constant on the initial carrier concentration was also examined. The physical basis of the exponential law and the effect of the Fermi level cross-over of the defect levels on the resistivity change in the high fluence ranges are discussed.

Redescription of Two Terrestrial Hypotrichous Ciliates from Jindo Island, Korea (진도의 육상 하모섬모충류 2미기록 종의 재기재)

  • Two hypotrichous ciliates inhabiting the representative terrestrial habitats of Jindo Island, Holostichides chardezi Foissner, 1987 and Rigidocortex octonucleatus (Foissner, 1988) are found from Jindo Island. These two species are newly recorded from Korea. The description was based on the observation of living specimens, protargol impregnated specimens and biometric analysis. The morphological variations among the populations of these species were investigated with morphometry. The diagnostic characteristics of these species are as follows: Holostichides chardezi : elongate body flexible and twisted, yellowish subpellicular granules scattered entire cortex, adoral zone of membranelles covering about $28\%$ of body length, left and right rows of mid-ventral cirri paired, left row ended at buccal vertex and right row extending to center of body, no transverse cirri, both marginal rows of cirri not confluent at posterior end, 28-42 macronuclei and 1-5 micronuclei scattered entire body. 3 caudal cirri and 4 dorsal kineties. Rigidocortex octonucleatues: elongate-oval body rigid and not contractile, yellowish cytoplasmic crystals and fat globules densely packed, adoral zone of membranelles covering about $40\%$ of body length, 5 transverse cirri, both marginal rows of cirri not confluent at posterior end, 8 macronuclei longitudinally arranged in median of cell, 3 caudal cirri and 7-10 fragmented dorsal kineties.

Study on Scientific Analysis about Red Pigment And Binder - The Korean Ancient Red Pottery - (한국 고대 붉은 간토기의 적색 안료 및 교착제에 대한 과학적 분석)

  • From the collection of the National Kimhae Museum, qualitative analyses using microscopic observation, SEM-EDS, Raman spectroscopy, FT-IR-ATR spectroscopy, and GC-MS were conducted on three burnished red potteries-Jeoksaekmaoyeonwa burnished red pottery (Neolithic age red pottery), Dandomaoyeonwan burnished red pottery(Bronze age red pottery) and Jeoksaekmaoyeongajimun burnished red pottery(Bronze age red pottery)-to investigate the components of the red pigments and the binder. After the layers of the primer were separated from the red surface, crystals of red pigment particles and minerals were found on the red surface. Through SEM-EDS, Raman estimates that the red pigment is Among soil pigments with iron oxide(Fe2O3) as the main color development source, Red Ocher(Fe2O3). A band characteristic of the Urushiol polymer was detected in the FTIR-ATRspectra(4000~600cm-1), GC-MS analysis confirmed the presence of the benzenemethanol-2-prophenyl, 4-heptylphenol, 1-tetracecanol, heptafluorobutyric texidecane, all of which are the ingredients of the directional structure of the lacquer present in the red layer. Therefore, it seemed that the three burnished red pottery: Jeoksaekmaoyeonwan pottery(Neolithic age burnished red pottery), Dandomaoyeonwan pottery(bronze age burnished red pottery) and the Jeoksaekmaoyeongajimun pottery(bronze age burnished red pottery) made by mixing minerals and Red Ocher(Fe2O3), with lacquer.

Analysis on Material Characteristics of Restored Areas with Mortar and Basis of Surface Deterioration on the Stupa of State Preceptor Jigwang from Beopchensaji Temple Site in Wonju, Korea (원주 법천사지 지광국사탑 복원부 모르타르 재료학적 특징 및 표면손상 기초 해석)

  • The Stupa of State Preceptor Jigwang from Beopcheonsa Temple Site in Wonju (National Treasure) is a representative stupa of the Goryeo Dynasty, with outstanding Buddhist carvings and splendid patterns, clearly indicating its honoree and year of construction. However, it was destroyed by bombing during the Korean War (1950-1953) and repaired and restored with cement and reinforcing bars in 1957. The surface condition of the original stone shows long-term deterioration due to the m ortar used in past restorations. In order to identify the exact causes of deterioration, the m ortar and surface contaminants on the original stone were analyzed. Portlandite, calcite, ettringite, and gypsum from the mortar were identified, and its ongoing deterioration was observed through pH measurements and the neutralization reaction test. Analysis of surface contaminants identified calcite and gypsum, both poorly water-soluble substances, and their growth in volume among rock-forming minerals was observed by microscopy. Based on those results, semi-quantitative analysis of Ca and S contents significantly influencing the formation of salt crystals was conducted using P-XRF to analyze the basis of surface deterioration, and cross-validation was performed by comparing the body stone affected by the mortar and the upper stylobate stone unaffected by the mortar. Results indicate that the elements are directly involved in the surface deterioration of the body stone.

An Analysis of Material Property on Eartherwares Excavated at Auraji site in Jeongseon (정선 아우라지 출토 토기의 재료과학적 특성 분석)

  • In this study, we aimed to elucidate the materialistic characteristics of 11 pieces of earthenware belonging to the Neolithic and Bronze Age excavated from Jeongseon Auraji, South Korea. As a result, the chemical composition of earthenwares belonging to the early Bronze Age was distributed in the intermediate area between the Neolithic and Bronze Age earthenwares, but no significant difference was confirmed based on their manufacturing period. Upon comparison, the earthenwares excavated from Jeongseon Auraji site were found to comprise less acidic components than those excavated from Yeongdong, and are characterized by the alkaline components depending on the excavated site. In the rare earth elements distribution pattern, all the analyzed earthenwares exhibited similar pattern, confirming that the raw materials present in the clay were the same. As a result of microstructure analysis, the clay particles and voids were found to be irregularly distributed in the analyzed earthenwares. Neolithic earthenwares exhibited many irregular voids, and an arrangement of aluminosilicate, including feldspar, was observed along with the clay substrate. Furthermore, we confirmed that the empty space in early Bronze Age earthenwares was filled with fine particles and cube crystals. Moreover, the main mineral phase of earthenwares excavated from Jeongseon Auraji exhibited similar composition, and therefore, there was no significant difference in the firing temperature of these earthenwares. The firing temperature of the earthenwares ranged from 750 to 850℃.

Thermoelectric Properties of the Reaction Sintered n-type β-SiC (반응소결법으로 제조한 n형 β-SiC의 열전특성)

  • Silicon carbide is considered to be a potentially useful material for high-temperature electronic devices, as its large energy band gap and the p-type and/or n-type conduction can be controlled by impurity doping. Particularly, electric conductivity of porous n-type SiC semiconductors fabricated from ${\beta}-SiC$ powder at $2000^{\circ}C$ in $N_2$ atmosphere was comparable to or even larger than the reported values of SiC single crystals in the temperature region of $800^{\circ}C$ to $1000^{\circ}C$, while thermal conductivity was kept as low as 1/10 to 1/30 of that for a dense SiC ceramics. In this work, for the purpose of decreasing sintering temperature, it was attempted to fabricate porous reaction-sintered bodies at low temperatures ($1400-1600^{\circ}C$) by thermal decomposition of polycarbosilane (PCS) impregnated in n-type ${\beta}-SiC$ powder. The repetition of the impregnation and sintering process ($N_2$ atmosphere, $1600^{\circ}C$, 3h) resulted in only a slight increase in the relative density but in a great improvement in the Seebeck coefficient and electrical conductivity. However the power factor which reflects the thermoelectric conversion efficiency of the present work is 1 to 2 orders of magnitude lower than that of the porous SiC semiconductors fabricated by conventional sintering at high temperature, it can be stated that thermoelectric properties of SiC semiconductors fabricated by the present reaction-sintering process could be further improved by precise control of microstructure and carrier density.

Analysis of the Dead Layer Thickness effect and HPGe Detector by Penelope Simulation (Penelope Simulation에 의한 불감층 두께 효과 및 HPGe 검출기 분석)

  • Germanium crystals have a dead layer that causes efficiency deterioration because the layer is not useful for detection but strongly weakens the photons. Thus, when the data provided by the manufacturer is used in the detector simulation model, there is a slight difference between the calculated efficiency and the measured efficiency.The shape and dimensions of the high purity germanium (HPGe) detector were determined by CT scans to accurately characterize the shape for the Monte Carlo roll simulation. It is found that the adjustment of the dead layer is a good match with the relative deviation of ${\pm}3%$ between the measurement efficiency and the simulation efficiency at the energy range of 50 - 1500 keV. Simulation data were compared by varying the thickness of the dead layer. The new Monte Carlo simulations were compared with the experimental results to obtain new blank layer thicknesses. The difference in dead layer results for the 1.5 mm thick end cap simulation model in 1.4 and 1.6 mm thick End Cap simulation models was a systematic error due to the accuracy of the end cap dimensions. After considering all errors including statistical errors and systematic errors, the thickness of the detector was calculated as $1.02{\pm}0.14mm$. Therefore, it was confirmed that the increase in the thickness of the dead layer causes the effect to be effected on the efficiency reduction.

Catalytic Hydrogenation of Triglyceride in a Semi-batch Reactor (Semi-batch 반응기에서의 트리글리세라이드 접촉 수소화 반응)

  • The aim of this study is to investigate the feasibility of an Ni-SA catalyst, which was prepared from nickel, kieselguhr, and alumina, for the hydrogenation of triglyceride in a bench-scale reactor. Ni-SA powders were prepared by precipitating nickel precursors on a silica and alumina support. The powder was reduced in a hydrogen flow, mixed with a saturated palm oil, and then cooled to prepare an Ni-SA catalyst tablet. The sizes of NiO crystals of a commercial Pricat catalyst and the Ni-SA catalyst prepared in this study were $35{\AA}$ and $38{\AA}$, respectively. The pore volume and pore size of the Ni-SA catalyst was much larger than the pore volume and pore size of the Pricat catalyst. In addition, the average particle size of the Ni-SA catalyst was much smaller than that of the Pricat catalyst. The triglyceride hydrogenation reaction was carried out in a semi-batch reactor using catalysts impregnated with oil and molded into tablets. It was found that the Ni-SA catalyst was superior to the commercial Pricat catalyst in triglyceride hydrogenation, which could be ascribed to the raw material and the products being less influenced by the diffusion resistance in the pores of the Ni-SA catalyst. The Ni-SA catalyst prepared in this study has the potential to replace the Pricat catalyst as a catalyst for use in the commercial process for hydrogenation of triglyceride.
