Transgenic rice cells using RAmy3D promoter can provide high productivity, and the production of recombinant protein is induced by sugar starvation. In this system, productivity was reduced during the scale-up processes. To ensure the influences of shear stress and oxygen transfer rate, working volume and mixing performances were investigated under various agitation speeds and working volumes. In addition, inoculation methods including suspended cells and filtered cells were compared. Working volumes and shaking speeds were 300, 450 mL and 80, 120 rpm, respectively. Hydrodynamic environment of each condition was measured numerically like mixing time and $k_La$. Good mixing performance and high shear stress were measured at high agitation speed and low volume. The highest level of hCTLA4Ig was 30.7 mg/L at 120 rpm, 300 mL. When conditioned medium was used for inoculation, increased cell growth was noticed during the day 0~4 and decreased slower than filtered cells. Compared with filtered cells, the maximum hCTLA4Ig level reached 37.8 mg/L at 120 rpm, 300 mL and lower protease activity level was observed. In conclusion mixing performance is critical factor for productivity and conditioned medium can have a positive effect on damaged cells caused by hydrodynamic shear stress.
Partial quenching structure of diffusion flames in a turbulent mixing layer has been investigated by the method of flame hole dynamics in oder to develope a prediction model for the phenomenon of turbulent flame lift off. The present study is specifically aimed to remedy the shortcoming of the stiff transition of the conditioned partial burning probability across the crossover condition by employing the level-set method which enables us to include the effect of finite flame edge propagation speed. In light of the level-set simulation results with two models for the edge propagation speed, the stabilizing conditions for turbulent lifted flame are suggested. The flame hole dynamics combined with the level-set method yields a temporally evolving turbulent extinction process and its partial quenching characteristics is compared with the results of the previous model employing the flame-hole random walk mapping based on three critical scalar dissipation rates. The probability to encounter reacting state, conditioned with scalar dissipation rate, demonstrated that the conditional probability has a rather gradual transition across the crossover scalar dissipation rate. Such a smooth transition is attributed to the finite response of the flame edge propagation.

In this study, we conducted a study on characteristics of exhaust gas emissions from boiler when water-bunker oil mixed by homogenizer was burned in boiler. The results showed that NOx concentration and CO concentration of the homogenized bunker oil was decreased by 19% and 54% compared to pure bunker oil pretreatment was not being performed. And, in the case of water-bunker A oil, the NOx concentration was decreased with increasing water mixing ratio in bunker A oil. In particular, the NOx concentration in exhaust gas of 20 %water-80 %bunker A oil decrease by 45 % compared with pure bunker-A. However, the CO concentration in exhaust gas of 20 %water-80 %bunker A oil shows irregular changes. This means that the mixing of water more than a certain amount can cause a decrease in combustion performance. From this result, it can be found that critical mixing ratio of water in bunker A oil for normal combustion is 15% in this study. Deposition amount of soot that is collected in the vicinity of the chimney was decreased with increasing water mixing ratio.
Soil washing techniques coupled with high pressure air jet were applied for diesel-contaminated soils sampled by an underground oil reservoir of which the initial total petroleum hydrocarbon (TPH) ($2,828{\pm}206\;mg/kg$) exceeded 5 times of current standard level (500 mg/kg) regulated by the Soil-Environment Conservation Law. Through several tests, we found that the position of impeller has a critical impact for washing efficiencies. The highest washing efficiency was obtained at an oblique angle (30 degree) for the impeller and optimized mixing speed (300 rpm) that could have high shearing forces. Considered economical and feasible aspects, the optimum mixing time was 10 min. Rate constants of TPH removal derived from the first-order equation were not linearly increased as mixing speed increased, indicating that mechanical mixing has some limits to enhance the washing efficiencies. Application of high-pressure air jet in washing process increased the washing efficiency. This increase might be caused by the fact that the surface of micro-air bubbles strongly attached hydrophobic matters of soil particles. As the pressure of air jet increased, the separation efficiencies of TPH-contaminated soil particles increased. In the combined process of high-pressure air jet and mixing by impeller, the optimum mixing speed and air flow-rate were determined to be 60 rpm and $2\;kg/cm^2$, respectively. Consequently, the washing technique coupled with high-pressure air jet could be considered as a feasible application for remediating petroleum-contaminated soils.

The metal-ferroelectric-semiconductor (MFS) structure is widely studied for nondestructive readout (NDRO) memory devices, but conventional MFS structure has a critical problem. It is difficult to obtain ferroelectric films like PZT on Si substrate without interdiffusion of impurities such as Pb, Ti and other elements. In order to solve these problems, the metal-ferroelectric-insulator-semiconductor (MFIS) structure has been proposed with a buffer layer of high dielectric constant such as MgO, $Y_2O_3$, and $CeO_2$. In this study, the etching characteristics (etch rate, selectivity) of MgO thin films were etched using $Cl_2/Ar$ plasma. The maximum etch rate of 85 nm/min for MgO thin films was obtained at $Cl_2$(30%)/Ar(70%) gas mixing ratio. Also, the etch rate was measured by varying the etching parameters such as ICP rf power, dc-bias voltage, and chamber pressure. Plasma diagnostics was performed by Langmuir probe (LP) and optical emission spectroscopy (OES).

Food waste (FW) has been recognized as a critical problem in Korea and many research was conducted to efficiently utilize or treat FW. Main purpose of this research was to evaluate a feasibility for producing fermented organic fertilizer with vegetable waste (VW). Three different organic materials (saw dust, coco peat, and waste mushroom media) were mixed with VW at the rate of 30, 40, 50% respectively. Total days of composting experiment were 35 days and each sub samples were collected at every 5 days from starting of composting. Result showed that inner temperature of composting was increased to $60{\pm}4^{\circ}C$ within 5~10 days depending on varied organic materials and mixing ratio. Among different treatment, the highest increase of inner temperature was observed when 30% of saw dust was mixed with VW. After finishing composting experiment, maturity of each compost was evaluated with solvita and germination test. Maturity index (MI) of each treatment was ranged between 5~7 indicating that manufactured fertilizer was curing or finished stage. Calculated germination index (GI) was at the range of 57.83~101.16 depending on organic materials and mixing ratio. Both MI and GI showed that manufactured fertilizer was met for fertilizer criteria while control (VW only) was not adequate for composting. Overall, VW can be utilized for making organic fertilizer mixing with saw dust, coco peat and more research should be conducted to make high quality of organic fertilizer with vegetable waste.

Among different treatment, the highest increase of inner temperature was observed in treatment mixed with food waste and EC 30%. After finishing composting experiment, maturity was evaluated with solvita and germination test. Maturity index (MI) of each treatment was ranged between 5~7 indicating that manufactured fertilizer was curing or finished stage. Calculated germination index (GI) was at the range of 104~116 depending on mixing ratio of EC. Both MI and GI showed that manufactured fertilizer was suitable for fertilizer criteria while control (FW only) was not adequate for composting. Overall, earthworm cast can be utilized for improving compost maturity by mixing with food waste and more research should be conducted to make high quality of food waste compost with earthworm cast in agricultural fields.
Thermal mixing by steam jets in a pool is dominantly influenced by a turbulent water jet generated by the condensing steam jets, and the proper prediction of this turbulent jet behavior is critical for the pool mixing analysis. A turbulent jet flow induced by a steam jet discharged through a vertical upward single hole into a subcooled water pool was subjected to computational fluid dynamics (CFD) analysis. Based on the small-scale test data derived under a horizontal steam discharging condition, this analysis was performed to validate a CFD method of analysis previously developed for condensing jet-induced pool mixing phenomena. In previous validation work, the CFD results and the test data for a limited range of radial and axial directions were compared in terms of profiles of the turbulent jet velocity and temperature. Furthermore, the behavior of the turbulent jet induced by the steam jet through a horizontal single hole in a subcooled water pool failed to show the exact axisymmetric flow pattern with regards to an overall pool mixing, whereas the CFD analysis was done with an axisymmetric grid model. Therefore, another new small-scale test was conducted under a vertical upward steam discharging condition. The purpose of this test was to generate the velocity and temperature profiles of the turbulent jet by expanding the measurement ranges from the jet center to a location at about 5% of $U_m$ and 10 cm to 30 cm from the exit of the discharge nozzle. The results of the new CFD analysis show that the recommended CFD model of the high turbulent intensity of 40% for the turbulent jet and the fine mesh grid model can accurately predict the test results within an error rate of about 10%. In this work, the turbulent jet model, which is used to simply predict the temperature and velocity profiles along the axial and radial directions by means of the empirical correlations and Tollmien's theory was improved on the basis of the new test data. The results validate the CFD model of analysis. Furthermore, the turbulent jet model developed in this study can be used to analyze pool thermal mixing when an ellipsoidal steam jet is discharged under a high steam mass flux in a subcooled water pool.
Recombinant Pseudomonas lipase was used to study protein aggregation and adsorption upon in vitro refolding. Protein adsorption as well as aggregation was responsible for major side reactions upon in vitro refolding as a function of protein concentration. The optimal range of protein concentration was determined by the relative contribution of protein aggregation and adsorption. Above the optimal range, the yield of active lipase inversely correlated with protein aggregation, showing a competition between folding and aggregation. However, adsorption of protein rather than protein aggregation is thought to contribute as a major side reaction of the refolding process at sub-optimal concentrations at which the formation of aggregates should be more reduced. Protein aggregation was influenced by the amount of guanidine hydrochloride in the refolding solvent. The refolding temperature was a critical factor determining the extent of protein aggregation. The refolding yield was also affected by the dilution fold and dilution mode, which suggests that the refolding process might kinetically compete with the rate of mixing.

The erodibility of soil is an important factor to scour, especially in fine-grained soils. In this study, the erosion characteristics of kaolinite are quantified through the scour rate tests using the Erosion Function Apparatus called EFA. The basic soil property tests are also performed. The kaolinite samples are prepared by mixing with distilled water and formed to the designed maximum consolidation pressure of 60, 110, 160, 240, 360kPa, respectively. The results of the scour rate tests are presented in a format of a plot showing the relationship between erosion rates and shear stresses. Erosion properties of kaolinite showed a striking contrast according to the maximum consolidation pressure, and a correlation was established between the erosion properties of kaolinite and the soil properties; water content, undrained shear strength, dry density.
