Objectives : This in vitro research was conducted to investigate the effect of the Ginseng Radix plus Crataegi Fructus. on the cytokine protein release and Nitric oxide release in releted to Alzheimer's disease. Methods : Specifically, the effects of the Ginseng Radix plus Crataegi Fructus extract on $IL-1{\beta}$, IL-6, $TNF-{\alpha}$ of BV2 microglia cell line treated with lipopolysacchride. Results: The Ginseng Radix plus Crataegi Fructus extract suppressed the production of inflammatory cytokine protein $IL-1{\beta}$, IL-6, $TNF-{\alpha}$ and in BV2 microglia cell line treated with lipopolysacchride. Conclusion: These results suggest that the Ginseng Radix plus Crataegi Fructus extract may be effective for the prevention and treatment of Alzheimer's disease. Investigation into the clinical use of the Gin-CHF extract for Alzheimer's disease is suggested for future research.
Objectives The purpose of this study is to investigate the effects of Crataegi Fructus Pharmacopuncture(CFP) on the adipogenesis in 3T3-L1 cells, lipolysis in rat epididymal adipocytes and histological changes in porcine adipose tissue. Methods Inhibiton of preadipocyte differentiation and/or stimulation of lipolysis play important roles in reducing obesity. 3T3-L1 preadipocytes were differentiated with adipogenic reagents by incubating for 3days in the absence or presence of CFP ranging from 0.01 to 1mg/mL. The effect of CFP on adipogenesis was examined by measuring GPDH activity and by Oil Red O staining. Mature adipocytes from rat epididymal fat pad was incubated with CFP ranging from 0.01 to 1mg/mL for 3 hrs. The effect of CFP on lipolysis was examined by measuring free glycerol released. Fat tissue from pig skin was injected with CFP ranging from 0.1 to 10mg/mL to examine the effect of CFP on histological changes under light microscopy. Results The following results were obtained from present study on adipogenesis of preadipocytes, lipolysis of adipocytes and histological changes in fat tissue. 1. Crataegi Fructus Pharmacopuncture inhibited adipogenic differentiation at the concentration of 1.0mg/mL. 2. Crataegi Fructus Pharmacopuncture decreased the activity of glycerol-3-phosphate dehydrogenase(GPDH) at the concentration of 0.1mg/mL. 3. Crataegi Fructus Pharmacopuncture ok. lipolysis at the concentration of 0.1mg/ml. 4. Crataegi Fructus Pharmacopuncture ranging 0.1 to 10mg/mL failed to exert lysis of cell membrane in porcine fat tissue. Conclusions These results suggest that Crataegi Fructus Pharmacopuncture at relatively high concentration inhibited adipogenesis and increased lipolysis of adipocytes. However, Crataegi Fructus Pharmacopuncture didn't exert any effect on lysis of cell membrane in fat tissue.
Crataegi Fructus has often been cited in medical literature for its medicinal effects. The purpose of this study was to investigate the possibility of Crataegi Fructus application as an edible medicinal (nutritional supplement) food resource. In this study, Crataegi Fructus, which has been used in oriental medicine and folks remedy, was investigated to characteristics of nutritional composition(protein, lipid, ash, fiber, free sugar and minerals). The approximate composition of low Crataegi Fructus was crude protein, 0.26%, crude lipid, 0.30% and crude ash, 0.66%. And total dietary fiber 5.60%(insoluble 4.66%, soluble 0.94%), contents of glucose and fructose were 5.02mg% and 6.21 mg%, Nutritional composition of fermented liquid was crude protein 0.24%, crude lipid, 0.03%, crude ash, 0.53% and total dietary fiber, 0.24%. And glucose, fructose contents were 14.77mg% and 7.30mg%. The other hand, nutrition contents in water extract of Crataegi Fructus were significantly lower than low Crataegi fructus and fermented liquid. The above results showed that Crataegi Fructus and fermented liquid have sufficient values to use as a food stuff for medicinal food and nutritional supplement.
본 연구에서는 산사 발효액의 품질특성 및 항산화 및 소화활성 효과를 평가하였다. 산사 발효액의 수분함량은 $39.3{\pm}0.06%$이었으며, 조회분 $0.20{\pm}0.01%$, 조지방 $1.40{\pm}0.59%$, 조단백질 $1.77{\pm}0.04%$이었다. 그리고 산사 발효액의 총페놀 함량 분석 결과 $3,015{\pm}250$(GAE ${\mu}g/g$)로 나타났다. 색도 분석에서 L값은 79.24으로 나타났으며, a값과 b값은 각각 1.58과 31.28으로 측정되었다. 다양한 항산화 평가 모델(DPPH, FRAP, 환원력, ABTS)을 통하여 산사 발효액의 항산화 활성을 측정한 결과, 산사 발효액의 농도가 증가함에 따라 항산화능이 유의적으로 증가하는 것으로 나타났다. 한편, 산사 발효액의 혈당 조절능을 평가하기 위해, Porcine pancreas, Human saliva 및 Bacillus licheniformis 기원의 ${\alpha}$-amylase와 효모 기원의 ${\alpha}$-glucosidase에 대한 추출물의 저해 활성을 조사한 결과, ${\alpha}$-amylase에서는 저해활성을 보이지 않은 반면 ${\alpha}$-glucosidase에서는 10.4%의 저해활성을 보였다. 산사 발효액의 단백분해능을 측정한 결과, 대조품으로 사용한 pancreatin보다 우수한 활성을 나타내었다. 산사 발효액의 우수한 생리활성을 나타내는 성분에 대해서는 더욱 연구가 필요할 것으로 사료되며, 이러한 결과로 보아 산사 발효액을 소화제 등의 천연소재로 이용할 수 있을 것으로 사료된다. 따라서 산사의 발효액을 제조 시 기능성 향상을 위한 발효공정의 개선 및 최적화 등의 추후 연구가 필요한 것으로 사료된다.

As these combinations of natural products will activate some parts of body, theymay be applied to pharmaceutical applications, functional foods, antiaging tea, alsoexpected to control bacterial growth for fermentative beverage bearing multifunction.

Objectives : Crataegi Fructus has been used as traditional medicines for more digestion action, amenorrhea due to blood stasis, and hyperlipemia. The aim of this study was to compare of Crataegi Fructus in South Korea collected during three years according to the standards in monographs of the Korean Pharmacopoeia Eleventh edition (KP 11). Methods : Crataegi Fructus was carried out identification test (Qualitative reaction, Thin layer chromatography), heavy metal test, and total ash registered at KP. Add to we tested loss on dry, contents of ethanol-soluble extracts, and HPLC profiling. Results : Identification test (TLC) was on comparing with ursolic acid standard solution in $R_f$ value, all samples showed red purple spot ($R_f$ value 0.9). Ursolic acid spot in $R_f$ value 0.35 showed by changing mobile phase condition. Heavy metals showed contents for Pb, Cd, As, and Hg range of 0.0 ~ 0.5 ppm, 0.0 ~ 0.2 ppm, 0.0 ~ 0.3 ppm, and 0.0 ~ 0.1 ppm. Loss on drying was ranged from 5.5 to 11.9 %, total ash was between the range 2.7 ~ 4.0 %. Contents of ethanol-soluble extracts was ranged from 17.8 to 44.9 %. The content of chlorogenic acid was ranged from 0.0 to 0.1 % based on the chlorogenic acid standard curve. Conclusion : We have verified the current specification standard of Crataegi Fructus and standard that is not set. We hope that it will help the standardization of Crataegi Fructus.
The study was performed by examining the effects of fermented liquid of Crataegi Fructus on the lipid profile improvement in rats fed high fat diets. Sprague-Dawley rats of weighting $180.0{\pm}30g$ were randomly divided into five groups : basal diet (Normal control group, NCG), only high fat diet (High fat control group, HFC), high fat diet and supplemented with 1.69 mg/100 g body weight, 3.38 mg/100 g body weight, 6.76 mg/100 g body weight by fermented liquid of Crataegi Fructus - HFL, HFM, HFH group). These experimental diets were fed for 6 weeks. The fermented liquid of Crataegi Fructus fed groups had more significantly decreased in the levels of serum total cholesterol, triglyceride, LDL-cholesterol and atherogenic index than the high fat control group, while the HDL-cholesterol was higher when compared to the normal control group. Total lipid, total cholesterol, triglyceride, LDL-cholesterol contents in liver were decreased in high fat experimental groups. But the degree of increment was reduced by administration of fermented liquid of Crataegi Fructus. while the fermented liquid of Crataegi Fructus fed group had ore significantly increased in the level of HDL-cholesterol than the high fat control group. The singularity of the unsaturated fatty acid contents attracted our attention. Especially, the polyunsaturated fatty acid compositions were 36.36%, 34.70%, 20.31%in serum, liver and fecal of fermented liquid of Crataegi Fructus fed groups, respectively. These results imply that the fermented liquid of Crataegi Fructus can be used as possible food resources and medicinal food materials.
In this study we investigated the biological activities of Crataegi Fructus, including antioxidative, fibrinolytic, $\alpha$-glucosidase inhibitory, and thrombin inhibitory activities. Crataegi Fructus, hot water extract was fractionated into hexane, $CHCLl_3$, ethyl acetate, butanol, and water fractions, and each of these was assayed individually. The water fraction showed the highest extraction yield at 4.08% (w/w). The antioxidative activities of the water, ethyl acetate, and butanol fractions were 31.07%, 45.87%, 50.28%, and 91.74%, respectively. Assays for fibrinolytic activity indicated that only the butanol fraction has significant efficacy at 1.93 plasmin units/ml. Thrombin inhibitory assays indicated that the 10-fold dilutions of the $CHCLl_3$, ethyl acetate, and butanol fractions had inhibitory activities of 34.97%, 41.43%, and 58.10%, respectively. The 10-fold dilutions of the only ethyl acetate fraction demonstrated $\alpha$-glucosidase inhibitory activities of 75.07%. From the above results, we propose that extracts of Crataegi Fructus can be used as a material for the development of biofunctional foods.
Objectives : This research was investigated the effect of the Ginseng Radix plus Crataegi Fructus on the injury of brain tissue in the mice with Alzheimer's disease induced by ${\beta}A$. Methods : Observed a change of the injury of brain tissue and reduced the infarction area of hippocampus in the mice with Alzheimer's disease induced by ${\beta}A$. Results : 1. The Gin-CF extract reduced the infarction area of hippocampus, and controlled the injury of brain tissue in the mice with Alzheimer's disease induced by ${\beta}A$. 2. The Gin-CF extract reduced the Tau protein, GFAP protein, and presenilin1/presenilin2 protein (immunohistochemistry) of hippocampus in the mice with Alzheimer's disease induced by ${\beta}A$. Conclusion : These results suggest that the Ginseng Radix plus Crataegi Fructus extract may be effective for the prevention and treatment of Alzheimer's disease. Investigation into the clinical use of the Ginseng Radix plus Crataegi Fructus extract for Alzheimer's disease is suggested for future research.
