INFLUENCE IN CHOOSING BOX-COX TRANSFORMATION

  • A procedure for investigating the influence of observations in choosing Box-Cox transformation for multivariate data is suggested. It is effective in spotting influential observations. A numerical example is provided for illustration.

Paclitaxel Stimulates Cyclooxygenase-2 Expression via MAP Kinase Pathway in Rabbit Articular Chondrocytes

  • Paclitaxel, an antimicrotubule agent, binds to beta-tubulin in the microtubule and stabilizes the polymer, thereby repressing dynamic instability. Here, we have demonstrated that microtubule cytoskeletal architecture involved in regulation of the COX-2 expression in chondrocyte treated with paclitaxel. Paclitaxel enhanced COX-2 expression and prostaglandin E2 production, as indicated by the Western blot analysis, reverse transcriptase PCR(RT-PCR) and immunofluorescence staining, and $PGE_2$ assay, respectively. In our previous data have shown that paclitaxel treatment stimulated activation of ERK-1/2 and p38 kinase(Im et al., 2009). SB203580, an inhibitor of p38 kinase, blocked the induction of COX-2 expression by paclitaxel. Also PD98059, an inhibitor of ERK-1/2 kinase was blocked the induced COX-2 expression. These results indicate that activation of ERK-1/2 and p38 kinase is required for COX-2 expression induced by paclitaxel in rabbit articular chondrocytes.

  • Inappropriate up-regulation of cyclooxygenase-2 (COX-2) has been implicated for pathogenesis of various types of human cancer. COX-2 expression is known to be regulated by the eukaryotic transcription factor NF-$textsc{k}$B. In an attempt to link the NF-$textsc{k}$B activation and COX-2 induction, we have examined the kinetics of phorbol ester-induced activation of NF-$textsc{k}$B and COX-2 expression in the immortalized human breast epithelial cell line (MCF10A).(omitted)

  • 병원의 임상연구실험에서 종종 환자들의 치료에 따른 병세의 호전상태를 여러단계로 분류하여 상이한 치료방법에 따른 치료효과간의 차이를 알고자 하는 경우가 있다. 이와 같이 다중상태의 생존자료분석을 위한 한가지 방법으로 본 논문에서는 비동형의 Markov 모형에 Cox 회귀모형을 적용하여 회귀계수와 기저생존함수, 그리고 이를 바탕으로 반응확률함수를 추정하고 아울러 이들 추정량들의 대표본 성질들을 셈과정(Counting process) 기법을 이용하여 알아 보았다. 그리고 본 논문의 결과에 대해 실제 예를 들어 보였다.

  • Engle과 Russell (1998)의 ACD 모형은 재무학에서 가격과 거래 시간의 밀접한 관계에 대한 관심을 불러 일으켰다. ACD 모형은 GARCH 모형과의 유사성을 바탕으로 Box-Cox 변환과 충격 함수 곡선(shocks impact curve)을 적용시켜 Log ACD, Power ACD, Box-Cox ACD 등과 같은 보다 유연한 모형으로 일반화될 수 있다. 본 연구에서는 이와 같이 일반화된 ACD 모형들을 국내 주식시장에서 거래되고 있는 주식의 price duration에 적용시켜 그 성능을 비교해보고자 한다.

  • The present study was designed in order to determine whether Lonicerae flos (LF) could mitigate rheumatoid arthritis through inhibition of cyclooxygenase (COX)-2 and matrix metalloproteinase (MMP)-9 by regulation of macrophage migration inhibitory factor (MIF). We found that MIF mRNA expression in synoviocytes stimulated with phorbol-12-myristate-13-acetate dose-dependantly decreased by LF extract treatment (0.4 - 1.0 mg/$m{\ell}$). The distribution of MIF, COX-2 and MMP-9 positive reacted cells in LPS induced arthritis of mice were decreased by LF (45 mg/kg/day) treatment for 28 days. These data likely indicate that LF may act as MIF inhibitor and may be possible to develop useful agent for rheumatoid arthritis.

  • In order to understand the mechanism of action of TCDD, we have examined the effect of COX-inhibitors on cyplal activity. We observed the effect of COX-inhibitor on EROD activity in C57BL/6 mouse in vivo. And we also evaluated the effect of COX-inhibitors on cyplal mRNA, mouse cyp1a1 promoter activity and EROD activity in Hepa cell.(omitted)

Intraspecific variation of gene structure in the mitochondrial large subunit ribosomal RNA and cytochrome c oxidase subunit 1 of Pyropia yezoensis (Bangiales, Rhodophyta)

  • Red algal mitochondrial genomes (mtDNAs) can provide useful information on species identification. mtDNAs of Pyropia / Porphyra (Bangiales, Rhodophyta) have shown diverse variation in their size and gene structure. In particular, the introns and intronic open reading frames found in the ribosomal RNA large subunit gene (rnl) and cytochrome c oxidase subunit 1 gene (cox1) significantly vary the mitochondrial genome size in Pyropia / Porphyra species. In this study, we examined the exon / intron structure of rnl and cox1 genes of Pyropia yezoensis at the intraspecific level. The combined data of rnl and cox1 genes exhibited 12 genotypes for 40 P. yezoensis strains, based on the existence of introns. These genotypes were more effective to identify P. yezoensis strains in comparison to the traditional DNA barcode cox1 marker (5 haplotypes). Therefore, the variation in gene structure of rnl and cox1 can be a novel molecular marker to discriminate the strains of Pyropia species.

종대황 추출물의 COX-2 활성 억제 효과 (Extracts of Rheum undulatum L. Inhibits COX-2 Activities in Lipopolysaccharide-stimulated Raw 264.7 Cells)

  • Rheum undulatum L. has been used as Rhei Radix in Korean Pharmacopea although their pharmacological effects were not studied much. In this studym, we tested anti-inflammatory effect as a representative activity of Rheum undulatum L. extracts using cyclooxygenase (COX)-2 inhibition. Murine macrophage, Raw 264.7 cells were incubated with lipopolysaccharide (1 $\mu\textrm{g}$/ml) to induce COX-2. The prostagladin $E_2$ ($PGE_2$) levels as an indicator of COX-2 activity were determined in the culture medium using ELISA. Inhibition of acetylsalicylic acid (ASA) as a standard, aloe-emodin, chrysophanol, rhein, 80% ethanol extract of Rheum undulatum L. (Ex) and ether fraction (Fr) after acid hydrolysis of Rheum undulatum L. were tested in induced COX-2 described above. $IC_{50}$ values were 0.082 $\mu\textrm{g}$/ml for ASA. 181 $\mu\textrm{g}$/ml for aloe-emodin, 3.65 $\mu\textrm{g}$/ml for emodin, 144 $\mu\textrm{g}$/ml for chrysophanol, 39.8 $\mu\textrm{g}$/ml for rhein, 141 $\mu\textrm{g}$ of herb/ml for Ex, and 95.7 $\mu\textrm{g}$ of herb/ml for Fr. We found that Ex and Fr of Rheum undulatum L. were more effective than other anthraquinones, since their $IC_{50}$ are lower than others.
