대전기영동법(帶電氣泳動法)에 의한 유청단백질(乳淸蛋白質)에 관(關)한 연구(硏究) (Studies on Milk Protein by Zone Electrophoresis)

  • 본연구(本硏究)에서는 한우유(韓牛乳)와 Holstein cow milk의 단백질(蛋白質)을 Zone electrophoresis에 의하여 분석(分析)하여 그 결과(結果)를 다음과 같이 얻었다. (1) 한우유(韓牛乳)와 Holstein cow milk의 이화학적(理化學的) 성질(性質)이 Table 1, Table 2에 나타나 있다. (2) 한우유(韓牛乳)의 유청단백질(乳淸蛋白質)의 단백질(蛋白質) 분포(分布)는 Lactoalbumin 1.74mg %, Alpha-1 globulin 1.19mg%, Alpha-2 globulin 0.68mg %, Beta globulin 2.57mg %, Gamma globulin 0.93mg %이었다. (3) Holstein cow milk protein의 단백질(蛋白質) 분포(分布)는 Lactoalbumin 1.23mg %, Alpha-1 globulin 1.93mg %, Alpha-2 globulin 0.86mg %, Beta globulin 2.56mg %, Gamma globulin 1.74mg %를 얻었다. (4) 한우(韓牛)(female)와 Holstein cow(male)와의 교잡종인 F-1인 유청단백질(乳淸蛋白質) 분포(分布)는 Lactoalbumin 1.30mg %, Alpha-1 globulin 1.58mg %, Alpha-2 globulin 2.46mg %, Beta globulin 1.39mg %이었고 F-2의 그것은 Lactoalbumin 1.57mg %, Alpha-1 globulin 1.74mg %, Alpha-2 globulin 2.19mg %, Beta globulin 1.17mg %, F-4에서는 Lactoalbumin 2.46mg %, Alpha-1 globulin 1.24mg %, Alpha-2 globulin 1.57mg %, Beta globulin 1.74mg %를 각각(各各) 얻었다.

새로운 분류법에 따른 소아 위장관 우유 알레르기 질환에 관한 임상적 고찰 (Clinical Observations of Gastrointestinal Cow Milk Allergy in Children According to a New Classification)

  • 목 적: 효율적인 진단과 치료 및 적절한 의학적 교류를 목적으로 소아 위장관 알레르기 질환의 새로운 분류법이 발표되었다(J Pediatr Gastroenterol Nutr 2000;30:S87-94). 그러나 명칭과 분류의 변경으로 이미 알려진 질환들이 새로운 질환으로 보고되기도 하는데, 저자들은 새로운 분류법에 따른 소아 위장관 알레르기 질환의 질병군에 관하여 알아보고, 국내에서 이미 보고된 관련 문헌을 함께 고찰하여 의학적 교류에 혼선을 최소화 하고자 한다. 방 법: 2003년 3월부터 7월까지 계명대학교 의과대학 소아과를 방문하여, 우유 유발 및 제거시험, 내시경을 이용한 조직생검 등을 이용하여 소아 GI-CMA로 진단된 37례(남 19, 여 18)를 대상으로 후향성 조사하였다. 새로운 분류에는 포함되지 않았으나 위식도역류 관련 알레르기를 추가하였다. 국내 문헌은 소아과학회지, 소아소화기영양학회지, 소아알레르기호흡기학회지를 근거로 고찰하였다. 결 과: 1) 대상 환아의 연령은 2주~15개월, 평균 $5.4{\pm}4.8$개월이었다. 2) 출생체중은 전례에서 10~90백분위수를 차지하였으며, 25~75백분위수가 25례(68%)를 차지하였다. 내원 당시 체중은 3백분위수 이하가 18례(49%)이었다. 3) 증상발현에서 진단까지 걸린 시간은 2주~12개월, 평균 $2.4{\pm}3.3$개월이었다. 4) IgE 군인 IgE 매개형 우유알레르기(IGE) 6례(16%), IgE와 Non-IgE 혼합군인 호산구성 위장관염(EOS) 2례(5%), Non-IgE군인 전형적 우유 단백질 유발 장관염(CMPIE-T) 7례(19%), 비전형적 우유 단백질 유발 장관염(CMPIE-AT) 5례(14%), 알레르기성 대장염(AC) 12례(32%)이었다. 위식도역류증 관련 우유 알레르기(GERA)는 5례(14%)이었다. CMPIE-T 전례에서 소장 조직생검상 장병증이 관찰되었다. 5) 진단 당시 연령은 IGE $4.3{\pm}0.8$개월, EOS 생후 2주와 14개월, CMPIE-T $3.8{\pm}4.6$개월, CMPIE-AT $10.4{\pm}3.8$개월, AC $3.4{\pm}3.9$개월, GERA $7.8{\pm}5.7$개월로 질환군간 유의한 차이를 보였다(p<0.05). 6) 내원 당시 3백분위수 이하를 보인 경우가 IGE 17%, EOS 0%, CMPIE-T 86%, CMPIE-AT 60%, AC 25%, GERA 100%로 질환군간 유의한 차이를 보였다(p<0.05). 7) 국내 문헌을 고찰하여 소아 위장관 우유 알레르기 질환은 만성설사, 난치성설사, 우유 불내성, 우유 알레르기, 위장관 알레르기, 우유 과민성 장병증, 호산구성 위장관염, 알레르기성 대장염 등의 용어로 보고되었다. 결 론: 새로운 분류법에 따른 소아 위장관 우유 알레르기 질환은 연령, 임상 증상, 위장관 침범 부위에 따라 특징적인 유형으로 구분할 수 있으며, 임상적으로 드물지 않다. 국내의 문헌을 통하여 다양한 용어로 각 유형의 알레르기 질환들은 연구 보고되어 왔다.

뽕잎 첨가수준에 따른 두부의 관능평가 및 일반성분분석에 관한 연구 (A Study on the Changes of Physicochemical Characteristics of Soybeans Curd with Cow's Milk According to the Adding Levels of Mulberry Leaf Powder)

  • In this experiment, the curd was made from soybean and cow's milk in the ratio of 7 to 3, and coagulated with Glucono-delta-lacton(GDL) and $CaSO_4&.$2H_{2}O& in the 1:1 ratio. During the procedure, the mulberry leaf powder was added. The amount of the added powder varied from 0.1 % to 0.5 %. This experiment aimed to show the change of yield, general components, minerals, amino acid, changes of pH, sensory properties of these products. As mulberry leaf powder addition increased, crude protein, crude fat, crude ash content, minerals and amino acids increased. Whereas moisture content decreased, and in particular,$Ca^{++}$, $K^+$ and $Na^+$ increased. On the other hand, in the sensory properties, the levels of satisfaction about the color, appearance, flavor, texture and savory were lower in mulberry leaf powder added than under control.

ON FARM EVALUATION OF UREA MOLASSESED STRAW (UMS) FEEDING TO LACTATING COWS

  • Two on farm feeding trials were conducted at the Pabna milkshed area during July to October 1993, to evaluate the response of feeding urea molassesed straw (UMS) on milk production of Pabna milking cows and its economics. A feeding trial of 60 days with 50 Pabna milking cows showed that the cows fed UMS as a replacer of dry straw increased daily milk yield by 1.37 litre. Moreover, it reduced the daily requirement of concentrate by 1.0 kg/head. In the second trial, a herd of 52 animals including 14 milking cows were fed UMS in place of traditionally used dry straw. Introduction of UMS increased the milk production by 1.0 litre/cow/day and saved concentrate by 1.5 kg/cow daily. Urea molassesed straw can safely and economically be fed to lactating cows.

Organoleptic Properties of Cow Milk, Yoghurt, Kefir, and Soy Milk When Combined with Broccoli Oil: A Preliminary Study

  • Broccoli sprouts are an excellent source of health-promoting phytochemicals, such as glucosinolates, phenols, and vitamins. In this investigation, oil extracted from broccoli was adjusted to various concentrations (control, 1%, 2%, 3%, 4%, and 5%, respectively) and added directly to dairy products (cow milk, yoghurt, and kefir) and non-dairy products (soy milk), and their organoleptic properties assessed. The results showed that when the amount of broccoli oil was increased, the organoleptic properties (texture, color, and flavor) and overall acceptability tended to decrease. Cow milk, yoghurt, kefir, and soymilk supple-mented with 1% broccoli oil showed the best organoleptic properties when compared to the control group. The fermented products such as yoghurt and kefir with added broccoli oil showed good organoleptic properties. Overall, the results of this study provide evidence for the use of broccoli oil in dairy and non-dairy products. Further research will be required to assess the various physiological active functions of broccoli oil.

전환기 젖소의 사료첨가제 급여가 사료섭취량, 산유량 및 대사성장애 발생에 미치는 영향 (The Effect of Feed Additives Supplement on Prerpartum and Postpartum Feed Intake, Milk Production and Metabolic Disorders of Dairy Cows)

  • 본 연구는 분만전후 40두의 젖소에 무첨가, Aspergillus oryzae(T2), Saccharomyces cerevisiae 혼합제(T3) 및 효소분비촉진제(T4) 급여시 사료섭취량, 산유량 혈중대사물질 및 대사성 질병에 발생에 미치는 영향을 구명하기 위하여 완전임의배치법 10반복으로 수행하였다. 주요 연구결과는 다음과 같다. 분만전 건물섭취량은 처리간에 별 차이가 없었지만 분만일과 분만후 3주 동안에는 Aspergillus oryzae, Saccharomyces cerevisiae 혼합제 및 효소분비촉진제를 급여했을 때가 대조구보다 다소 많았다. 산유량은 사료첨가제를 급여한 구가 무첨가구보다 더 높았으며 특히 효소분비촉진제에서 산유량이 가장 높았다. 혈중 글루코스 및 NEFA 함량은 분만전에는 처리간에 큰 차이가 없었지만 분만일과 분만후 1, 2 및 3일에는 사료첨가제 급여구에서 대조구보다 혈중 글루코스 함량은 더 높고 반면에 NEFA 함량은 더 낮았다. 혈중 Ca 수준은 시험기간동안 처리간에 차이가 없었다. 대사성질병 발생은 대조구에서만 케토시스 1두 발생하였으며, 후산증체는 효소분비 촉진제구는 전혀 발생하지 않았으나 다른 구에서는 공히 2두가 발생하였다. 자궁 내막염은 T2구에서 1두 발생하였다. 결론적으로 전환기 젖소에 사료첨가제를 급여하면 사료섭취량 및 산유량 증가와 더불어 대사성질병 발생이 저하될 것으로 사료된다.

Complete Genome Sequence of Enterococcus faecalis CAUM157 Isolated from Raw Cow's Milk

  • Enterococcus faecalis CAUM157, isolated from raw cow's milk, is a Gram-positive, facultatively anaerobic, and non-spore-forming bacterium capable of inhabiting a wide range of environmental niches. E. faecalis CAUM157 was observed to produce a two-peptide bacteriocin that had a wide range of activity against several pathogens, including Listeria monocytogenes, Staphylococcus aureus, and periodontitis-causing bacteria. The whole genome of E. faecalis CAUM157 was sequenced using the PacBio RS II platform, revealing a genome size of 2,972,812 bp with a G+C ratio of 37.44%, assembled into two contigs. Annotation analysis revealed 2,830 coding sequences, 12 rRNAs, and 61 tRNAs. Further, in silico analysis of the genome identified a single bacteriocin gene cluster.

Effect of phytol in forage on phytanic acid content in cow's milk

  • Objective: Bioactive compounds in ruminant products are related to functional compounds in their diets. Therefore, this study aimed to explore the effect of forage sources, Italian ryegrass (IR) silage vs corn silage (CS) in the total mixed ration (TMR), on milk production, milk composition, and phytanic acid content in milk, as well as on the extent of conversion of dietary phytol to milk phytanic acid. Methods: Phytanic acid content in milk was investigated for cows fed a TMR containing either IR silage or CS using 17 cows over three periods of 21 days each. In periods 1 and 3, cows were fed CS-based TMR (30% CS), while in period 2, cows were fed IR silage-based TMR (20% IR silage and10% CS). Results: The results showed that there were no differences in fat, protein, lactose, solids-not-fat, somatic cell count, and fatty acid composition of milk among the three experimental periods. There were no differences in the plasma concentration of glucose, triglycerides, total cholesterol, and nonesterified fatty acids among the three experimental periods, while the blood urea nitrogen was higher (p<0.05) in period 2. The milk phytanic acid content was higher (p<0.05) in period 2 (13.9 mg/kg) compared with periods 1 (9.30 mg/kg) and 3 (8.80 mg/kg). Also, the phytanic acid content in the feces was higher (p<0.05) in period 2 (1.65 mg/kg dry matter [DM]) compared with period 1 (1.15 mg/kg DM), and 3 (1.17 mg/kg DM). Although the phytol contents in feces did not differ among the three feeding periods, the conversion ratio from dietary phytol to milk phytanic acid was estimated to be only 2.6%. Conclusion: Phytanic acid content in cow's milk increases with increasing phytol content in diets. However, phytol might not be completely metabolized in the rumen and phytanic acid, in turn, might not be completely recovered into cow's milk. The change of phytanic acid content in milk may be positively correlated with the change of phytol in the diet within a short time.

아토피피부염에서 모유를 통한 식품 알레르겐에의 감작 (Sensitization of Food Allergen in Breastfed Infant with Atopic Dermatitis)

  • Breastfeeding has been known as the best feeding practice to prevent allergies including atopic dermatitis (AD) However, the benefit on the prevention of allergic disease is still controversial. The objectives of this study were to examine the rate of sensitization to the protein of eggs, cow's milk and soy in exclusively breastfed infants and to evaluate antigen-antibody reaction between breast milk and serum of AD infant. Data on feeding and food hypersensitivity were obtained for 62 AD infants (32 male, 30 female) aged < 6 month who had visited Samsung Medical Center from September 2001 to May 2003. Food hypersensitivity was determined by measuring specific IgE to egg, cow's milk and soy. Specific IgE levels > 0.7 kU/L by CAP assay (Pharmacia, Uppsala, Sweden) were considered positive. The rates of sensitization in breastfed infants were $41.9\%$ (26/62) to egg, $30.6\%$ (19/62) to milk and $18.0\%$ (11/62) to soy. Immunoblotting analyses were performed using breast milk with the matched serum of seven AD infants (4 male/3 female). Binding patterns of AD infant's IgE to breast milk extract showed visible specific band for immunoglobulin, especially in case of a lactating mother who did not completely restricted ingestion of egg, milk and soy. These results indicate that sensitization to food allergen develops via breast milk feeding. Breast milk feeding should be recommended in infants at risk of developing allergic disease, but maternal intake of highly allergenic food might be restricted for prevention and treatment of food allergy among the babies with AD.
