Development of Cosmetic Ingredient by Fermented Paprika Juice (파프리카 발효즙의 화장품 소재개발 연구)

  • In this study, cosmetic materials were developed using a new method of making juice through the fermentation of raw natural materials with microorganisms in order to supplement the advantages and disadvantages of an organic solvent extraction method and a microbial fermentation method. The natural products were selected from two colors (red, green) of paprika known to be rich in various colors and vitamins. The microorganisms used for fermentation were fermented by inoculating paprika with lactic acid bacteria (Lactobacillus plantarum) having sugar-hydrolyzed ability. First, we investigated the changes of physiologically active substances of two kinds of paprika juice and two kinds of fermented paprika juice. Total phenols content and total flavonoids content were higher in the fermented paprika juice than in the paprika juice, and especially in the fermented red paprika juice. Free radical scavenging effect and lipid peroxidation inhibitory effect were also showed an excellent antioxidative effect on paprika fermented juice, among which the effect of red paprika fermentation juice was the highest. The expression of MMP-1 in fermented red paprika juice with high antioxidant activity was inhibited by concentration-dependent expression of MMP-1 mRNA and MMP-1 protein. In the glycation experiments with aging, the anti-glycation effect of fermented paprika juice was highly inhibited by the production of advanced glycation end-products (AGEs), which was closely related to the antioxidant effect. In addition, the activity of senescence-associated ${\beta}$-galactosidase (SA-${\beta}$-gal), an indicator of cell senescence, was measured using human dermal fibroblast (HDF). The results showed that the cell senescence was inhibited when the cells were treated with fermented paprika juice. In conclusion, fermented paprika juice using lactic acid bacteria showed better antioxidative and anti-aging effects than paprika juice. Among them, fermented red paprika juice has the best antioxidant and anti-aging effect and can be applied as natural new material of antioxidant and anti-aging.

The comparative study on physiological activity of White ginseng, Red ginseng and Black ginseng extract (백삼, 홍삼, 흑삼 추출물의 생리활성 비교 연구)

  • This study presents a comparative study for differences in efficacy and ingredient of white ginseng(WG), red ginseng(RG), and black ginseng(BG) using anti-oxidative activity test and anti-inflammatory activity test. In the results of cytotoxicity test for WG, RG, and BG, the survival rate of all cells was more than 95%. In the total polyphenol analysis, DPPH, ABTS radical scavenging test, and ROS production test, BG showed higher anti-oxidant activity than WG and RG. RG and WG showed higher inhibition activity of NO production and $PGE_2$ production, respectively. As results of the test for the effect on reduction of inflammatory cytokine production, WG and RG were effective on reduction of IL-$1{\beta}$ production, and BG was effective on reduction of IL-6. In the case of TNF-${\alpha}$ production, there was no difference among samples. This study could be useful basic data for the development of functional food and the fabrication of safe cosmetic.

Antioxidant and Anti-inflammatory Effects of Bletilla striata Reichenbach fil. Fractions as Cosmetic (백급(Bletilla striata Reichenbach fil.) 분획물의 항산화, 항염증 효과와 화장품소재로서의 연구)

  • The antioxidant and anti-inflammatory effects of active ingredients in cosmetics are very important. The effects are closely related to the prevention of skin aging. Among medicinal plants, Bletilla striata Reichenbach fil. has well-known pharmacological activity. Extracted samples were prepared using sequential fractionation of ethyl acetate, butanol, and water. The antioxidant effect of the fractions was confirmed by DPPH and ABTS+ radical scavenging. Among the various fractions of B. striata Reichenbach fil., ethyl acetate was associated with a reduction in nitric oxide production, which was induced by LPS (lipopolysaccharide) treatment in a dose-dependent manner. In addition, the production of iNOS (inducible nitric oxide synthase) and COX-2 (cyclooxygenase-2), which are upstream regulators of nitric oxide production, was also inhibited. Thus, the ethyl acetate fraction of B. striata Reichenbach fil. appears to be a potential active ingredient for use in cosmetics.

Cytotoxicity, Antibacterial and Antioxidant Activities of the Prescription Cheongyeolsodokum and Its Constituent Herbs (청열소독음(淸熱消毒飮)과 구성약재의 암세포 독성, 항균 및 항산화 효과)

  • Objectives : The purpose of this research was physiological activities, cytotoxicity, and antibacterial activities of the Cheongyeolsodokum and its constituent herbs. Methods : Physiological activities, cytotoxicity, and antibacterial activities were examined through the Cheongyeolsodokum and its constituent herbs. Results : In the physiological activities, the electron donating ability(EDA) of the water and ethanol extracts from the Cheongyeolsodokum were over 60% and 80% at 100 ppm, respectively. The EDA of the water and ethanol extract from the Cheongyeolsodokum ingredients were gradually increased as well. Water and ethanol extracts from the Cheongyeolsodokum and its constituent herbs inhibited xanthine oxidase activity, they showed superoxide dismutase(SOD)-like activity. The Cheongyeolsodokum and its constituent herbs inhibited cancer cell growth in a dose-dependant manner. Also, the clear zones against Staphylococcus aureus and S. epidermidis were clear shown at 2.5 and 5 mg/disc. Its constituent herbs showed the clear zone against various bacteria such as Candida albicans, S. aureus, S. epidermidis, and S. mutans were shown at 0.5 and 1mg/disc. Conclusions : We observed physiological activities, cytotoxicity, and antibacterial activities of the Cheongyeolsodokum and its constituent herbs. The results also indicated that water extract and ethanol extract of Cheongyeolsodokum and its constituent herbs can be used as a natural ingredient in food or cosmetic industry.

Characteristics of Galactooligosaccharide Production Using Cellulases (셀룰라제를 이용한 갈락코올리고당의 생산 특성)

  • Galactooligosaccharlde (GOS) is a kind of functional oligosaccharides that can be used as a food ingredient and a cosmetic additive. In this paper, characteristics of GOS synthesis by cellulase, using lactose as a substrate, were investigated. Penicillium funiculosum cellulose was found to be the most efficient for GOS production among six cellulose tested. The optimum pH and temperature for GOS production were 5.0 and $50^{\circ}C$, respectively. There was an optimum ratio of lactose concentration to enzyme loading; the value was 10 (w/w). The reaction pattern of P. funiculosum cellulase is consistent with that of microbial ${\beta}$-galactosidase which shows transgalactosylation activity. Amounts of GOS produced from 20% (w/v) lactose after 6 h incubation at $50^{\circ}C$, were 23% (w/w) based on total saccharide in the reaction medium. The GOS % increased with initial lactose concentration in the range of 5 to 20%. The products mainly consisted of a trisaccharide and tetrasaccharide from HPLC and TLC analysis. Among enzymes involved in transgalactosylation reaction, high molecular weight fractions over 50,000 Da, presumably ${\beta}$-glucosldase, were considered to be responsible for GOS production. Using this cellulose, a direct synthesis of galactosyl g1ucoside including GOS could be readily achieved with lactose as a galactosyl donor.

Quantitative Analysis of Ellagic Acid in Nymphaea tetragona Georgi Root by HPLC-UVD (HPLC-UVD를 이용한 수련(Nymphaea tetragona Georgi)의 Ellagic Acid 정량 분석)

  • A high performance liquid chromatography (HPLC) method for the quantitation of ellagic acid in Nymphaea tetragona was developed for the quality control of functional cosmetic ingredient, the extract of N. tetragona. Separation and quantitation were successfully achieved with a Kromasil C18 column ($5{\mu}m$, $250mm{\times}4.6mm$, i.d.) by isocratic elution of a mixture of acetonitrile containing 0.1% trifluoroacetic acid and water containing 0.03% phosphoric acid at a flow rate of 1.0 ml/min. The UV detector was used for the detection and the wavelength for quantitation was set at 254 nm. The presence of ellagic acid in the extract was determined by comparison of retention time and spiking with authentic standard. Analytical results showed good linearity ($R^2=0.99996$) in relatively wide concentration ranges. The R.S.D. for precision test was less than 3.0%. Recovery of the compound was 98.55~101.72% with R.S.D values less than 4.0%. In conclusion, this method has been successfully applied to the determination of ellagic acid in N. tetragona.

Anti-wrinkle Activities Verification of Buplerum falcatum Extracts on CCD-986sk (CCD-986sk세포내 시호 추출물의 항주름 활성 검증)

  • The electron donating ability, elastase inhibitory, procollagen synthesis and Matrix metalloprotease-1 (MMP-1) activities were measured in order to verify the anti-wrinkle properties of extracts from Buplerum falcatum as a functional ingredient for cosmetic products. Electron donating ability and elastase inhibition activities were 80 and 52% at a dose of $1,000{\mu}g/mL$ of B. falcatum 70% ethanol extract. Pro-collagen synthesis was increased with the increase concentration of B. falcatum extract on CCD-986sk in addition to decrease the amount of protein of MMP-1. The results suggested that B. falcatum extract can be used to reduced electron donating ability, elastase, pro-collagen synthesis and MMP-1 activity and is a potential candidate for cosmedical materials.

Anti-oxidant Activity of Akebia quinata fruit extract and the Effects of Skin (으름열매 추출물의 항산화 활성 및 피부미용 효과)

  • Akebia quinata fruit(AQF), as identified in the preceding paper, polyphenols and other phenolic components of saponin also has similar or higher levels. The purpose of this study is to analyze the effect of akebia quinata fruit extract, well-known for soothing, anti-oxidizing effects, on the improvement of the moisture, sebum, melanin, erythema content of facial skin. As a result of measuring DPPH radical scavenging activity to examine independent anti-oxidation of AQF, there was a slight scavenging activity. Compared to before the usage of cream, a group who used cream with akebia quinata fruit extract showed a very slight increase in the moisture content and slight decrease in the sebum, melanin, erythema on their faces after 4 weeks of tests, indicating that there was some statistically significant changes found. This study proves that the akebia quinata fruit(AQF) extract has a positive effect on the overall improvement of facial skin and it also implies that the akebia quinata fruit extract has high potential as an ingredient of cosmetic products.

Effect of Expression of Genes in the Sphingolipid Synthesis Pathway on the Biosynthesis of Ceramide in Saccharomyces cerevisiae

  • Ceramide is important not only for the maintenance of the barrier function of the skin but also for the water-binding capacity of the stratum corneum. Although the exact role of ceramide in the human skin is not fully understood, ceramide has become a widely used ingredient in cosmetic and pharmaceutical industries. Compared with other microorganisms, yeast is more suitable for the production of ceramide because yeast grows fast and is non-toxic. However, production of ceramide from yeast has not been widely studied and most work in this area has been carried out using Saccharomyces cerevisiae. Regulating the genes that are involved in sphingolipid synthesis is necessary to increase ceramide production. In this study, we investigated the effect of the genes involved in the synthesis of ceramide, lcb1, lcb2, tsc10, lac1, lag1, and sur2, on ceramide production levels. The genes were cloned into pYES2 high copy number vectors. S. cerevisiae was cultivated on YPDG medium at $30^{\circ}C$. Ceramide was purified from the cell extracts by solvent extraction and the ceramide content was analyzed by HPLC using ELSD. The maximum production of ceramide (9.8 mg ceramide/g cell) was obtained when the tsc10 gene was amplified by the pYES2 vector. Real-time RT-PCR analysis showed that the increase in ceramide content was proportional to the increase in the tsc10 gene expression level, which was 4.56 times higher than that of the control strain.

ROS Scavenging and Anti-Wrinkle Effects of Clitocybin A Isolated from the Mycelium of the Mushroom Clitocybe aurantiaca

  • Clitocybin A, an isoindolinone from Clitocybe aurantiaca, was investigated to assess its anti-wrinkle properties, through reactive oxygen species (ROS)-scavenging and elastase inhibitory activities, procollagen synthesis, and matrix metalloproteinase-1 (MMP-1) expression, in human primary dermal fibroblast-neonatal (HDF-N) cells. Clitocybin A exhibited no significant cytotoxicity up to 10 ppm in HDF-N cells, with cell viability and cell proliferation activity greater than 94.6% and 91.9%, respectively. Strong and concentration-dependent ROS radical scavenging activities of clitocybin A were observed following irradiation with UVB at $30mJ/cm^2$. Furthermore, clitocybin A treatment of cells at 0.1, 1, and 10 ppm exhibited decreased elastase activity, in a concentration-dependent manner, by 1.97%, 6.6%, and 8.31%, respectively, versus the control group. The effects of clitocybin A on procollagen synthesis and MMP-1 expression were investigated. Clitocybin A treatment of cells at 1, 5, and 10 ppm increased procollagen synthesis, by 67.9%, 74.4%, and 112.9%, respectively, versus the control group. At these concentrations, MMP-1 expression decreased significantly following UV irradiation. Together, these findings suggest that clitocybin A may be an effective ingredient for use in anti-wrinkle cosmetic products.
