현호색의 이온삼투요법 활용에 관한 연구 (Study on Corydalis Tuber for iontophoresis)

  • 이온삼투요법이란 지속적인 직류의 사용으로 피부를 통해 이온화된 약물을 무통성으로 환부에 도입하여 질병을 치료하는 요법이다. 한의학에서 이온삼투요법에 적용하기 위한 한약물의 제조 방법 및 기준이 아직 구체적으로 설정되어 있지 않다. 이에 저자는 이온삼투요법에 자주 활용되고 있는 현호색 증류 추출물의 정량분석을 실시하였다. HPLC에서 corydaline 표준품의 크로마토그램 peak는 5.95분에서 나타났으나, 현호색 증류 추출물에서는 corydaline 고유의 peak인 5.95분에서 나타나지 않는 것으로 보아 corydaline이 함유되지 않은 것을 알 수 있다. CG/MS에서 corydaline 표준품의 스백트럼의 peak는 7.89분에서 나타나고 분자량은 m/z 369가 나타났으나, 현호색 증류 추출물에서는 산성 조건, 중성 조건, 알카리 조건 모두에서 corydaline 표준품 고유 스펙트럼의 peak가 나타나지 않았고 분자량도 m/z 369가 나타나지 않은 것으로 보아 corydaline이 함유되지 않은 것을 알 수 있다. 이상의 결과를 볼 때 현호색 증류 추출물의 모든 샘플에서 corydaline이 검출되지 않은 것은 증류하는 과정에서 corydaline이 가진 지용성(脂溶性) 특징 때문에 함유되지 않거나, 극미량(極微量)만이 함유되어 있기 때문일 것으로 보인다. 그러므로 이온삼투요법에 응용하는 약물을 추출할 때에는 해당 약물의 화학적 특성을 고려하여 추출방법을 선택해야 목적하는 유효성분을 얻을 수 있다. 그리고 현호색의 경우 corydaline 이외의 다른 성분이 약효에 작용하는지에 대한 여부가 지속적으로 연구되어야 한다.

Neuroprotective mechanism of corydaline in glutamate-induced neurotoxicity in HT22 cells

  • Glutamate-mediated oxidative stress causes neuronal cell death by increasing intracellular Ca2+ uptake, reactive oxidative species (ROS) generation, mitogen-activated protein kinase (MAPK) activation, and translocation of apoptosis-inducing factor (AIF) to the nucleus. In the current study, we demonstrated that corydaline exerts potent neuroprotective effects against glutamate-induced neurotoxicity. Treatment with 5 mmol/L glutamate increased cellular Ca2+ influx, ROS generation, MAPK activation, and AIF translocation. In contrast, corydaline treatment decreased cellular Ca2+ influx and ROS generation. Western blot analysis revealed that glutamate-mediated MAPK activation was attenuated by corydaline treatment. We further demonstrated that corydaline treatment inhibited the glutamate-mediated translocation of AIF to the nucleus. We propose that corydaline is a promising lead structure for the development of safe and effective neuroprotectants.

면역시스템을 통한 세포수명연장 천연물질 스크린 (Screening of Natural Product Libraries for the Extension of Cell Life-span through Immune System)

  • 한약진흥재단(NIKOM)의 640-천연물 라이브러리를 PC12세포에 10 μg/ml/day로 처리하였다. 면역강화에 관여하는 ERAP1 (Endoplasmic Reticulum AminoPeptidase 1)과 수명연장에 관여하는 FOXO1 (FOXO-family transcription factor) 유전자발현을 RT-PCR로 확인하였다. ERAP1 유전자를 1.5-2배까지 상승시킨 것은 22개, 2배 이상 발현시킨 것은 14개였다. FOXO1 유전자를 1.5-2배까지 상승시킨 것은 총 24개, 2배 이상 발현시킨 것은 4개였다. 동시에 ERAP1과 FOXO1 유전자를 2배 이상 상승시키는 것은 족도리풀(Asiasari radix)에서 분리된 (±)-Car-3-ene-2,5-dione, 두꺼비(Bufonis venenum)에서 분리된 Cinobufagin, 대국(Euphorbiae pekinensis)에서 분리된 Corilagin, 현호색(Corydalis tuber)에서 분리된 Corydaline 4종이다. 이들 4종의 천연물은 초파리를 이용한 개체의 수명연장실험에 사용될 것이다.

In Vivo Disease Control Efficacy of Isoquinoline Alkaloids Isolated from Corydalis ternata against Wheat Leaf Rust and Pepper Anthracnose

  • Phytochemicals have been considered as alternatives for synthetic fungicides because of their biodegradability and low toxicity. In this study, we found that the methanolic extract of Corydalis ternata suppressed the development of plant diseases caused by Puccinia triticina and Colletotrichum coccodes. As the antifungal substance, three isoquinoline alkaloids (dehydrocorydaline, stylopine, and corydaline) were isolated from C. ternata. These active compounds also exhibited in vivo antifungal activity against P. triticina and C. coccodes. Taken together, our results suggest that C. ternata and its active compounds can be used to control plant diseases.

천연물에 의한 초파리수명연장 효과 (The Effect of Natural Compounds on the Longevity Extending in the Insect, Drosophila melanogaster)

  • 우리는 이전실험에서 배양세포를 사용하여 4종류의 천연물(Corydaline, (${\pm}$)-Car-3-ene-2,5-dione, Cinobufagin, Corilagin)이 ERAP1와 FOXO1 (DFA16) 유전자발현을 2배 이상 상승시키는 것을 증명하였다. 본 실험은 1% agar, 5% sucrose, natural compound $20{\mu}l$를 넣은 먹이를 만들어 4시간 starvation후에 4시간 동안 먹였다. Cinobufagin와 Corilagin를 먹이면 대조군에 비하여 6-8일 정도 더 생존하였다. RT-PCR 실험결과 ERAP1와 FOXO1 유전자 발현을 조절하는 것이 증명되었다. 산업곤충질병 진단과 치료에 사용될 것이며, 초파리를 대신한 생쥐에서도 동일한 결과를 실험하여 수명연장을 위한 신약으로 발전시킬 것이다.

Aldose Reductase Inhibitory Alkaloids from Corydalis ternata

  • A methanolic extract of Corydalis ternata having aldose reductase inhibitory activity was examined as a possible aldose reductase (ALR2) inhibitor, a key enzyme involved in diabetic complications. Seven alkaloids, tetrahydrocoptisine (1), corydaline (2), tetrahydropalmatine (3), isocorybulbine (4), corybulbine (5), dehydrocorydaline (6), and N-methyltetrahydroberbinium (7) were isolated from $CHCl_3$ fraction of C. ternata methanol extract. Among them, compounds 1, 5, and 7 exhibited $5.04{\pm}1.97%$, $5.00{\pm}1.26%$, and $1.80{\pm}2.33%$ inhibitions, respectively at $40{\mu}M$. The activities of the single compounds were not comparable to that of the whole extract, suggesting that the whole combination of each single compound was responsible for the activity of the extract as shown in many cases of natural medicines. Even though this is the second report on aldose reductase inhibition activity of C. ternata, recombinant human aldose reductase was employed in this study unlike in the previous report. Furthermore, the aldose reductase inhibitory activities of isocorybulbine, corybulbine, and N-methyltetrahydroberbinium, to the best of our knowledge, were evaluated for the first time in this study. These results suggest a use of the extract of C. ternata for ameliorating diabetic complications.

HPLC Separation of Isoquinoline Alkaloids for Quality Control of Corydalis species

  • A simple and rapid analytical method was developed for the determination of eight isoquinoline alkaloids in Corydalis species. Eight isoquinoline alkaloids, including 2 aporphine alkaloids (isocorydine and glaucine) and 6 protoberberine alkaloids (coptisine, palmatine, berberine, canadine, corydaline, and tetrahydrocoptisine) were used as chemical markers which have a various biological activity and determined for quality control of Corydalis (C.) species (C. ternata, C. yanhusuo, and C. decumbens). To evaluate the quality of these herbal medicines, LC chromatographic separation of alkaloids were preferentially investigated on reversed-phase C18 column with pH variation and composition of mobile phase. In addition, the separation of these alkaloids in herbal extracts was found to be significantly affected on mobile phase composition using gradient elution. Especially for C. yanhusuo extract, berberine was seriously interfered with other alkaloid extracted from sample matrix when mobile phase composition was not optimized. As results, these compounds were successfully separated within 28 min using 10 mM ammonium acetate containing 0.2% triethylamine (adjusted at pH 5.0) as a mobile phase with gradient elution. On the basis of optimized HPLC conditions, 23 different Corydalis species samples were analyzed for the determination of alkaloid levels. In addition, principal component analysis (PCA) combined with the chromatographic data could be successfully classified the different geographic origin samples.
