Characterization of Physiological Functionalities of Codonopsis lanceolata, Cornus officinalis S. et Z, and Their Mixtures

  • These studies were carried out to characterize the physiological functionalities of Codonopsis lanceolata, Cornus officinalis, and their mixtures. We investigated the antioxidative, fibrinolytic, and ${\alpha}$-glucosidase inhibitory activities of them. The antioxidative activities of Codonopsis lanceoiata and Cornus officinalis were 87% and 90%, respectively. Addition of salt to Codonopsis lanceolata and Cornus officinalis did not affect its antioxidative activities. In spite of fourfold addition of Codonopsis lanceolata to Cornus officinalis, the antioxidative activity was conserved at 90%. The fibrinolytic activities of Codonopsis lanceolata and Cornus officinalis were 0.78 plasmin unit/ml and 1.74 plasmin unit/ml, respectively. Addition of salt decreased the fibrinolytic activities of both Codonopsis lanceolata and Cornus officinalis. A mixture (3:1) of Codonopsis lanceolata and Cornus officinalis exhibited a 21% increase in activity. The ${\alpha}$-glucosidase inhibitory activities of Codonopsis ianceoiata and 100-fold diluted Cornus officinalis were 25% and 73%, respectively. The addition of salt to Codonopsis lanceolata and Cornus officinalis slightly decreased their ${\alpha}$-glucosidase inhibitory activities. According to the addition of Cornus officinalis to Codonopsis lanceoiata, the ${\alpha}$-glucosidase inhibitory activities of the resulting mixture were highly increased. We anticipate that these results will be used as basic data for the development of new bifunctional foods.

The Characteristic of Vascular Plants in Cornus officinalis Farmland in Gurye which Designated as Korea's Important Agricultural Heritage System (국가중요농업유산 구례 산수유 농업지의 관속식물상 특성)

  • This study has been conducted to understand the relationships such as interspecific interactions between species and landscapes within and outside of agricultural land by identifying the flora features of Cornus officinalis farmlands in Gurye which designated as Korea's Important Agricultural Heritage System. For this, the flora of vascular plants was surveyed in every land uses of two representative districts. These two districts represent the own features of Cornus officinalis farming area which including land uses like forests(especially pine tree), valley, farmland(rice paddy, farms, Cornus officinalis farmland), and residence. Also differences in flora by stone wall's existence and Cornus officinalis's DBH were surveyed. As a result, Cornus officinalis farmlands showed the various vascular plants than the other land uses. According to the existence of stone wall in Cornus officinalis farmlands, there were no differences in appearance flora. But the flora in Cornus officinalis farmlands which have different Cornus officinalis' DBH(Divided into average over 15cm and under 8cm), various plants were showed in average over 15cm than average under 8cm. In conclusion, Cornus officinalis farmlands have been actively engaged in various types of land use and species exchanges, including forests, valleys, villages, and fields, and it has been confirmed that it acts as an ecological axis connecting forests to villages.

Quality Characteristics of Sugar Snap-Cookie with Added Cornus fructus (산수유 분말을 첨가한 쿠키의 품질 특성에 관한 연구)

  • This study examined the effects of Cornus fructus addition to suger snap cookies on WRC (water rentention capacity) and AWRC (alkaline water retention capacity). WRC and AWRC increased as more Cornus fructus powder was added, whereas the sedimentation value and Pelshenke value decreased. Regarding cookie color, L value decreased, whereas the a and b values increased. As the cookie diameter became smaller, thickness increased and spread factor decreased. Hardness of the cookies decreased, and preference was highest for cookies with 5% added Cornus fructus. The preferences for cookies were in the following order: 7% < 3% < 1% < 0% < 5% addition. In conclusion, addition of less than 5% Cornus fructus powder was the most desirable. To complement cookie appearance, it seems necessary to adjust the water content of Cornus fructus powder or add an emulsifier.

Anti-Inflammatory Effect of Cornus Walteri (말채나무의 항염증 효과)

  • This research seeks a basis for developing new anti-inflammatory medicine by investigating Cornus Walteri extract for its anti-inflammatory effects. After the injection of LPS in to rats with Cornus Walteri extract, its anti-inflammatory effects were compared among the treatment groups. The plasma concentration of IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ peaked at 5h after LPS injection, and the values of the Cornus Walteri extract groups were lower than those of the control group. In the increment of concentration of these cytokines at 2h and 5h after LPS injection, the Cornus Walteri groups were lower than that of control group. The plasma concentration of IL-10 peaked at 5h after LPS injection, and the values of the Cornus Walteri extract groups were higher than those of the control group. In the increment of cytokines concentration at 2h and 5h after LPS injection, the Cornus Walteri groups were higher than that of control group. Liver cytokines measurement was done at 5h after LPS injection. The concentration of liver IL-$1{\beta}$ and IL-6 in the Cornus Walteri groups was lower than that of the control group. The concentrations of liver TNF-${\alpha}$, and IL-10 showed no significant differences among all the treatment groups. In the studies of lipopolysaccharide-exposed Raw 264.7 cells, the concentration of IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ in the lipopolysaccharide-exposed cells groups was higher than that of control group (normal group). However, in lipopolysaccharide-exposed cells groups, they showed lower values than those of control group and these values showed a tendency to decrease in the Cornus Walteri groups. The concentration of IL-10 in the lipopolysaccharide-exposed cells groups was higher than that of control group (normal group), and among the lipopolysaccharide-exposed cells groups, all Cornus Walteri extract groups showed higher values than single lipopolysaccharide-exposed cells groups. This studies have shown that in vitro and in vivo Cornus Walteri extracts are significantly more sensitive to inflammatory cytokines and LPS induced lethality. We conclude that the Cornus Walteri extracts have an functional material for inflammatory activities.

Effects of Stem Bark Extracts of Cornus walteri Wanger on the Lipid Lowering, Anti-oxidative Activity and Concentration of Proinflammatory Cytokines in Rat Fed High Fat Diet (모래지엽(毛棶枝葉)(Cornus walteri Wanger) 추출물이 비만 쥐의 지질강하, 항산화효과 및 전염증성 cytokines 농도에 미치는 영향)

  • Objective : This study was designed to examine the effects of stem bark extracts of Cornus walteri Wanger on the lipid lowering, anti-oxidative activity and concentration of proinflammatory cytokines in hyperlipidemic rat. Methods : Male rats weighing $195.21{\pm}5.85g$ fed high fat diet for 8 weeks and 40 rats(above 400 g) were divided into 4 groups. Each groups were divided into a control group and 3 experimental groups. We fed a control group of rats a basal diet and administered normal saline(100 mg/kg, 1 time/1 day) for 4 weeks. And we fed each experimental group of rats Basal diet and administered an extract of Cornus walteri Wanger(100 mg/kg, 200 mg/kg, 300 mg/kg, 1 time/1 day) for 4 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. We measured lipid of plasma and liver, concentration of proinflammatory cytokines, anti-oxidative activity and gene expression. Results : 1. Concentration of plasma free fatty acid, LDL-cholesterol showed a tendency to decrease in Cornus walteri Wanger ext. groups. Concentration of plasma triglyceride, total cholesterol showed a significantly decrement in all Cornus walteri Wanger ext. group than that of control group. HDL-cholesterol showed a significantly increment in the 300 mg/kg Cornus walteri Wanger ext. group. 2. Concentration of liver total cholesterol showed a tendence to decrease in Cornus walteri Wanger ext. groups. Concentration of triglyceride liver showed a significantly decrement in all Cornus walteri Wanger ext. group than that of control group. 3. Concentration of plasma and liver TBARS showed a tendence to decrease in Cornus walteri Wanger ext. groups. The values of GSH-Px, SOD and CAT activity showed a significantly increment in the 300 mg/kg Cornus walteri Wanger ext. group than that of control group. 4. The values of plasma AST and ALT activity showed no significantly different in all treatment groups. 5. Concentration of plasma $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ showed a tendency to decrease in the Cornus walteri Wanger ext. groups. However the concentration of IL-10 in the 200 and 300 mg/kg Cornus walteri Wanger ext. groups showed a significantly increment than that of control group. Concentration of liver $IL-1{\beta}$, $TNF-{\alpha}$ and IL-10 showed no significantly difference in all treatment groups. However concentration of IL-6 in the 200 and 300 mg/kg Cornus walteri Wanger ext. groups showed a significantly decrement than that of control group. 6. In the analysis of RT-PCR, gene expression of $TNF-{\alpha}$, Apo-B, Apo-E and leptin in the Cornus walteri Wanger ext. groups showed a lower expression than that of control group. 7. The ratio of $TNF-{\alpha}$, Apo-E and leptin expression per $\beta$-actin expression in the 200 and 300 mg/kg Cornus walteri Wanger ext. showed a significantly decrement than that of control group. The ratio of Apo-B expression per $\beta$-actin expression in the 300 mg/kg Cornus walteri Wanger ext. showed a significantly decrement than that of control group. Conclusions : According to above results, in lowering lipid effect, antioxidative activity and antiinflammatory effect, the Cornus walteri Wanger ext. gives positive effect.

Antibacterial Activity of the Ethanol Extract from Cornus officinalis against Some Bacteria Related to Foodborne Illness and Food Spoilage (산수유 에탄올 추출물의 식중독 및 식품 부패 세균에 대한 항균 활성 분석)

  • The antibacterial activity of the ethanol extract of Cornus officinalis against some bacteria related to foodborne illness and food spoilage was investigated. In the case of the disc diffusion assay, clear zones due to the inhibition of proliferation by Cornus officinalis extract were 8.5~18.3 mm at $4,000{\mu}g/disc$. The inhibitory effect of Cornus officinalis was as follows: Escherichia. coli ${\geq}$ Listeria monocytogenes ${\geq}$ E. coli O157:H7 > Bacillus cereus > Staphylococcus aureus > Pseudomonas aeruginosa > Salmonella typhimurium. In addition, the MIC of Cornus officinalis extract toward B. cereus was the lowest at 1,000 ppm. The extract inhibited the growth of E. coli, L. monocytogenes, E. coli O157:H7, and Bacillus cereus throughout the growth stage. However, the growth of P. aeruginosa, S. aureus, and S. typhimurium was only inhibited by the extract during the lag phase. The constituents of all cells tested were released, especially E. coli and E. coli O157:H7. Observation of the cells using SEM demonstrated a morphological change and disruption of cells in response to treatment with Cornus officinalis extract. Based on these findings, the ethanol extracts of Cornus officinalis showed strong antimicrobial activity against all tested bacteria, indicating that Cornus officinalis can be a useful natural antimicrobial reagent.

Effect or Cornus officinalis Sieb. et Zuccha Extracts on Physiological and Antioxidative Activities in Streptozotocin Induced Diabetic Rats

  • This investigation was performed to study the antioxidant activities of Cornus officinalis Sieb extracts and the effect of Cornus officinalis Sieb extracts on glucose, lipid metabolism in diabetic rats. DPPH free radical scavanging activity and superoxide anion radical scavenging of Cornus officinalis Sieb extracts were 94.7% and 92.1%, respectively. Streptozotocin (45 mg/kg body weight, i.p.) induced diabetic rats showed a significant increases of plasma glucose, triglyceride and total cholesterol, concomitantly significant decrease of plasma high density lipoprotein. Glutathione level were decrease in cytosol of liver, lung and brain tissue of rats. Lipid peroxide were increase in microsome of liver cells. Group 1 and 2 were treated with Cornus officinalis Sieb extracts 200 mg/kg body weight and 100 mg/kg body weight for 24 days, individually. Group 1 and 2 rats showed decreased plasma glucose, triglyceride, total cholesterol and lipid peroxide in microsome of liver, and increased plasma high density lipoprotein and glutathione in cytosol of liver, lung and brain. The result suggest that Cornus officinalis Sieb extracts may normalize the Impaired antioxiants status in streptozotocin induced diabetic rats. Cornus officinalis Sieb extracts were used to improve the imbalance between free radicals and antioxidant system due to the diabetes.

Tricyclic Pyridone Alkaloids from Cultures of Cornus officinalis Fruits-associated Fungus, Fusarium lateritium SSF2 (산수유(Cornus officinalis) 과실에서 분리한 Fusarium lateritium SSF가 생산하는 Tricyclic Pyridone계 알칼로이드)

  • Three tricyclic pyridone alkaloids, (-)-4,6'-anhydrooxysporidinone (1), (-)-6-deoxyoxysporidinoCne (2), and (-)-sambutoxin (3) were isolated from cultures of Fusarium lateritium SSF2 derived from the fruits of Cornus officinalis. Chemical structures of the isolated compounds were determined by spectroscopic method and comparisons with reported data values. The compounds 1-3 were isolated for the first time from this fungal strain.

A Study on the Cosmetic Preservative Effects of Cornus officinalis seed Extracts (산수유 종자 추출물의 화장품 방부효능에 관한 연구)

  • To evaluate the possibility as a natural preservative of Cornus officinalis seed extracts, we investigated the anti-microbial activity and preservation effect of cosmetics. Cornus officinalis seed extracted with 1,3-Butylene Glycol(1,3-BG) and Ethanol(EtOH). As a result, Cornus officinalis seed extracts showed anti-microbial activity at Staphylococcus aureus, Staphylococcus epidermidis, Propionibacterium acnes and Cornus officinalis seed 1,3-BG extracts(COS-A) showed $26.7{\pm}4.0mm$ clear zone at the $1000.0{\mu}l/ml$. Minimum inhibitory concentration of S. epidermidis was observed at Cornus officinalis seed EtOH extracts(COS-B) $25.0{\mu}l/ml$. The result of challenge test for preservation effect on cosmetics, S. aureus and S. epidermidis was 100% eliminated from emulsion containing COS-A and COS-B after 7 days. These results suggest that Cornus officinalis seed extracts may have possibility as the natural preservative instead of synthetic perservative on cosmetics.

  • Dimethyltetrahydrofuran cis-2,5-dicarboxylate a furan derivative has been isolated from the fruits of Cornus officinalis, and it was isolated for the first time in the nature. The structure was elucidated by NMR spectroscopic techniques.
